This thesis mainly examines inflectional morphology of verbal paradigms in Laki, which is considered as one of the Southern varieties of Kurdish language. The association of form and content of morphological markings are viewed from a realizational angle, in which exponents (morphological forms) are associated with the morphosyntactic properties via the application of rules of exponence, appealed by paradigm functions (Stump 2001) and ordered into rule blocks (Anderson 1992). In particular, I applied the paradigm linkage theory proposed and fully developed by Stump (2002 and 2016) to account for Laki verbal paradigms. In this study, it is claimed that alignment pattern and the syntagmatic combination of some of the inflectional exponents such as agreement markers are sensitive to preterite property. Hence, I argue in favor of considering two distinct morphotactic patternings in Laki. As the result of this assumption, depending on whether the inflectional markers attach to a preterite or a non-preterite stem, we should define two separate sets of affix position in this language. Moreover, I examine Laki polyfunctional agreement markers through the consideration of the morphotactics of this language. Applying Stump's analysis (to appear) of Swahili verbal concords, I consider two distinct types of content for these agreement markers: intrinsic content, and positional content. Their positional content is what the morphotactics of the language determines.

Role of polyfunctional and proliferative CD8+ T cell responses in HIV-1 infectionRichmond, Meika 02 1900 (has links)
The limited success of HIV vaccine candidates to date highlights our need to better characterize protective cell-mediated immunity. Understanding correlates CD8+ T cell protection against HIV infection and progressive disease is essential for informing effective vaccine development, design and evaluation. CD8+ T cell responses with a robust polyfunctional and proliferative component are strongly linked to better disease outcomes. However, the specificity of polyfunctional and proliferative CD8+ T cell responses has not been thoroughly investigated. Additionally, the specificity of memory subsets and their connection to polyfunctionality and proliferation responses has not been adequately assessed. We address these gaps in knowledge and provide a better understanding of the fine specificity of HIV-specific CD8+ T cell responses. We hypothesize that the epitopes recognized by central memory (TCM) and effector memory (TEM) CD8+ T cells, defined by functional attributes, differ in chronic HIV-1 infection. Additionally, we hypothesize that polyfunctional and proliferative responses will better correlate with protection in HIV disease progression. The qualities of CD8+ T cell responses were evaluated using polyfunctional flow cytometry measuring both functional and phenotypic attributes of both TEM and TCM subsets in HIV infected individuals. We evaluated the quality and evolution of CD8+ T cell responses in HIV infected individuals shortly after seroconversion through to the chronic phase of infection, finding that early polyfunctional responses may result in better HIV disease outcomes. Additionally, we show that epitope-specificity differs between short-term cytokine/chemokine secretion and long-term proliferative assays. Importantly, we show that, at a cohort level, particular epitopes preferentially elicit specific qualities of CD8+ T cell responses in preference to others. This research improves our understanding of HIV pathogenesis and indicates that we can identify specific epitopes that can elicit protective responses and that early polyfunctional responses may slow HIV disease progression. Understanding the polyfunctional and proliferative capacities of HIV-specific effector and memory cells at various stages of HIV infection is of critical importance to the design of vaccines intended to elicit protective cell-mediated responses.

Inaccusativité et diathèses verbales : le cas du vietnamien / Unaccusativity and verbal diatheses : the case of VietnameseDao, Huy-Linh 04 December 2015 (has links)
Cette thèse se veut une contribution empirique et théorique à l’étude formelle du système verbal vietnamien. Elle se centre plus précisément autour de la problématique de l’inaccusativité et des alternances diathétiques dans cette langue. Elle se propose à cet effet d’examiner de plus près le rôle que joue le marqueur polyfonctionnel bị au sein des constructions à prédicat intransitif. Il sera montré que bị est compatible avec les verbes inaccusatifs et ne se combine avec les verbes inergatifs que si ces derniers sont inaccusativisés à l’aide de matériel lexical approprié. L’hypothèse défendue, qui s’inscrit dans le cadre du Programme Minimaliste de Chomsky (1995 et travaux subséquents), consiste à traiter ce morphème comme une catégorie semi-lexicale/semi-fonctionnelle et comme la tête d’une projection Applicative Haute, située immédiatement au-dessus du vP. Du point de vue sémantique, bị dénote une relation entre un individu et un événement. Il sera établi que bị peut être une tête de Phase ou une tête non-phasale. L’examen d’une sous-classe de prédicats statifs, ceux qui dénotent des états permanents, suggère une réévaluation du rôle de l’Aspect Lexical (Aktionsart) dans le calcul de l’inaccusativité. L’analyse avancée permet non seulement de rendre compte des phrases traditionnellement classées comme des constructions passives, mais aussi d’expliquer l’incompatibilité entre bị et les constructions moyennes. Il sera proposé que cette incompatibilité est due à l’absence d’une projection aspectuelle interne au vP, projection qui accueille dans son Spécificateur des Causateurs Involontaires. On soutiendra également que la marge du vP est exploitable à des fins discursives, ce qui renforce davantage le parallélisme entre CP et vP. / This thesis constitutes an empirical and theoretical contribution to the formal study of the Vietnamese verbal system. More precisely, it concentrates on issues of unaccusativity and diathetical alternations in this language. To this end, it puts under close scrutiny the key role played by the polyfunctional marker bị within intransitive predicate constructions. It will be shown that bị is compatible with unaccusative verbs and can combine with unergative ones only if those verbs undergo unaccusativization when provided with suitable lexical material. The hypothesis put forward, which is couched within the framework of Chomsky’s The Minimalist Program (1995 et seq.), treats this morpheme as a semi-lexical/semi-functional category and as the head of a High Applicative projection, situated immediately above vP. From a semantic point of view, bị denotes a relationship between an individual and an eventuality. It will be established that bị can be a Phase-head or a non-phasal one. A closer inspection of a subclass of stative predicates, those which denote permanent states, suggests a reassessment of the role played by Inner Aspect (Aktionsart) in the calculus of unaccusativity. The analysis proposed not only accounts for sentences that are traditionally classified as passive constructions, but it also gives an explanation for the incompatibility between bị and some middle constructions. Furthermore, it will be put forward that this type of incompatibility is due to the absence of a vP-internal aspectual projection which hosts in its Specifier an Inadvertent/Unintentional Causer DP. It will also be claimed that the vP-edge is active for discursive purposes, which reinforces the parallelism between CP Phase and vP Phase.

Aim of the thesis is evaluation of road network as an integral part of common facilities plan. To get the results I chose two cadastral areas situated at different altitudes in which I examined behaviour of road network based on other common facilities. Basically it was a survey on how the road network in each territory adjoins the elements of Territorial System of Ecological Stability that helps reduce erosion vulnerability and whether it can improve the water situation in the territory. Results are given in the sub-tables that describe the road network in interaction with other common facilities conducted before and after Comprehensive Landscaping. These tables are supplemented by a recommendation which might ensure greater efficiency in the future. Sub-tables also indicate the approach of designers when designing road networks in typologically different territories. All results are graphically presented both in the text and in the annexes.

The dissertation focuses on rehabilitaion of urban space close to historic center of Brno city. One third of the area is taken by brownfields. These final goals were set: finding a new function for unused places, making a use of potential of Svitava canal and spaces in front of old buldings and creation of new urban spaces . Intervention includes creation of riverside designed for pedestrians, bike path construction, connecting area to city’s transportation system, bulding a new railway station and housing blocks with active street level and ability to walk througH area with as little obstacles as possible. Extra emphasis was placed on compactness of development, polyfunctionality and hierarchy of transpotration that respects man, his everyday life and needs. Concept reflects urban designing trends in 21st century.

The territory lies in the City of Brno, in the urban district of Zábrdovice. From the east it is demarcated by the Svitava River, from the south by the Cejl Street, and from the west by the Jan Svoboda Street. Its area is approximately 18,790 m2. From the standpoint of urban planning¸ the intensive house is a separate city block, which is, nevertheless, open in the eastern direction towards the Svitava River, thus communicating with the embankment. In the other direction there is both private and semi-private space inside the block as a relaxation green “garden” in several levels. The mass of the object links up in the prolonged line with the existing neighbouring blocks and thus respects the integrity of the territory. Intensive use of the territory consists in the achievement of high density of built-up area, and at the same time preserving the surroundings of quality for life and habitation. Functions are mixed here and a city of short distances is created here with nearly excluded traffic. In order to achieve such intensity, space urbanism is employed. The principle consists in a simple linear form of masses respecting the shape of the lot, which is vivified with an ellipsoid located in the courtyard, which is an accent towards the linear form. The ellipsoid serves as a small cultural and exhibition centre, at the same time connecting all the arms of the defining block and bringing daylight to the first floor. The block is open in the direction to the river and its half-public space is connected with the embankment with a staircase and verdure. Verdure is also made use of on the roofs. The defining mass is perforated, thus creating covered terraces. The principal architectonic intention was to create an integrated structure, where a significant role is played by the introduction of nature and polyfunctionality to the whole territory, thus increasing its attractivity. The assigned territory is situated in the City of Brno, in the city district of Židenice. From the west it is limited by the railway track Brno-Židenice, from the east by Koperníkova Street, from the north by Lazaretní Street and finally from the south by Bubeníčkova Street. Its area is approximately 45, 480 m2 which is 4,55 ha. From the urban perspective it is an intensive development in a separated block of the city with a protection zone. Total area is permeated by a few lines for pedestrians. The pedestrian zone provides better permeability of the area and better accessibility of the trade parterre. The buildings connect to surrounding development and respect the density of the integrity of the area. Intensive housing structure is regarded as "the city of short distance". The achievement of high density of housing development combines lots of various functions and relatively small built-up surface. While high-quality environment is preserved, people can find job oportunities in the intensive housing development without the need for transport. For achievement conception of compact city applies space urban planning. The shape of the estate directly impacted the principle of the proposal together with the city planning context of the current buildings. In the protective zone there is established a huge park with water reservoirs. The walk-through trade parterre is filled with many various functions, on its walkable roof is created a semi-public zone with skylights and freely introduced verdure. On the walkable roof there are independent blocks of apartments and public facilities. In the northern and southern corner are located public-service utilities and in the middle of this area there is a nursery and a kindergarden. In structural terms the building it is about cast-in-place concrete reinforced frame based on a white concrete foundation which has one underground level and five above-ground levels.
