Electrospray ionisation fourier transform ion cyclotron resonance and quadrupole ion trap mass spectrometry of metal-flavonoid complexes

Positive-ion electrospray ionisation Fourier transform ion cyclotron resonance and ion trap mass spectrometry have been used to investigate the reactions of the flavonoids 3-hydroxyflavone, 5-hydroxyflavone, 5-methoxyflavoe, quercetin, quercitrin and rutin with monovalent Li+, Na+, K+ and Cs+, divalent Cu2+, Zn2+ and Pb2+ and trivalent La3+ and Eu3+ metal cations. The effect of capillary-skimmer potential difference and the ion residence time in the hexapole ion trap of the Fourier transform ion cyclotron resonance mass spectrometer are systematically investigated for the flavonoid-alkali and divalent metal ion experiment. It is observed that these variables impact significantly on the type of ions observed in the ESI experiments and hence the mass spectra. The binding selectivity of alkali metal ions towards 3-hydroxyflavone, 5-hydroxyflavone and 5-methoxyflavone are determined using the results from FTICR mass spectrometry experiments. The selectivity order follows the order Li+>Na+>K+ for individual flavonoids. Collision-induced dissociation experiments are carried out by Fourier transform ion cyclotron resonance and ion trap mass spectrometry to compare the fragmentation behaviour of metal-flavonoid complexes. Low energy collision-induced dissociation experiments of the [2L+M]+ for 3-hydroxyflavone, 5-hydroxyflavone and 5-methoxyflavone alkali metal complexes show the loss of ligand only. When the energy is increased only the lithiated dimer [2L+Li]+ for 5-methoxyflavone shows the loss of methyl radical along with the ligand. For quercitrin the predominant dissociation pathways are the loss of rhamnose for Li+, Na+ and K+ complexes although aglycone loss is also observed for the K+ complex. The favourable dissociation pathways for rutin are the loss of disaccharide, aglycone and rhamnose for the Na+ complex and the loss of disaccharide for the K+ complex. Collision-induced dissociation data are also used to determine the threshold dissociation energies for displacement of one flavonoid ligand from alkali metal flavonoid complexes. The threshold dissociation energies for loss of one ligand from [2L+M]+ of 5-methoxyflavone and quercitrin follow the order Li+ > Na+ > K+, rutin follows the order Na+ > K+ > Li+ , and 3-hydroxyflavone and 5-hydroxyflavone follow the order Li+ > Na+. For the same metal cation experiment, 5-methoxyflavone system has the highest dissociation energy compared to the 3-hydroxyflavone and 5-hydroxyflavone experiment. Preliminary DFT calculations show that the calculated dissociation energies follow the same trend as the experimental dissociation energies for the simple flavonoid alkali metal cation experiments. For 5-methoxyflavone-divalent metal cation (Zn2+, Cu2+ and Pb2+) complexes loss of methyl radical is the common process. CO loss is also observed for the Zn2+ complex whereas CHO and H2O losses are observed for Cu2+. For 3-hydroxyflavone and 5-hydroxyflavone divalent metal cation experiments loss of ligand is the dominant process. Zn2+ and Cu2+ complexes also show CO loss. La3+ and Er3+ with the same flavonoids show the ligand as the dominant product. For quercetin-divalent metal cation experiment, ligand loss is the dominant process. For quercitrin and rutin various dissociation products are observed where the dissociation occurs via the loss of the rhamnose and/or the disaccharide moieties. Similar dissociation patterns are also observed for La3+ and Er3+ complexes for quercitrin and rutin.

Em muitos pacientes, o diagnóstico é difícil de ser definido, pelo método clássico de dosagem de sódio e cloro no suor, ou pelo sequenciamento genético, justificando a utilização de novas técnicas de auxílio diagnóstico, como a Diferença de Potencial Nasal (DPN). Este teste proporciona uma forma de avaliação direta e sensível, através do epitélio nasal, do transporte de sódio e cloro das membranas celulares, baseado nas propriedades bioelétricas transepiteliais. O objetivo deste trabalho foi verificar se existe diferença dos valores obtidos no exame de DPN em pacientes com FC em comparação com indivíduos controles saudáveis; e verificar se este teste permite diferenciar pacientes com FC das subclasses funcionais mais graves (I, II, III) das subclasses menos graves (IV, V, VI). Foram incluídos no estudo 15 pacientes FC, 10 com mutações mais graves (grupo A) e 5 com mutações menos graves (grupo B), e 21 controles saudáveis (grupo C). Foram considerados os seguintes parâmetros do teste da DPN: "Finger", PDMax, ?Amilorideo, ?Amilorídeo+livrecloreto e index de Wilchanski. Para a variável "Finger", foi encontrada diferença entre pacientes com FC grupo B - mutações menos graves (classe IV, V ou VI) e indivíduos saudáveis - grupo C. O valor do index de Wilchanski mostrou diferença entre pacientes com FC grupo A - mutações mais graves (classes I, II ou III) e indivíduos saudáveis - grupo C. No nosso estudo, a DPN mostrou valores estatisticamente diferentes entre FC com 2 mutações conhecidas e sujeitos saudáveis. Porém, não conseguiu diferenciar fibrocísticos com mutações mais graves (classes I, II e III) daqueles com mutações consideradas menos graves (classes IV, V e VI) / Abstract: Cystic fibrosis (CF) is an autosomal recessive genetic disease, due to the total absence of protein CFTR (Cystic Fibrosis Transmembrane Conductance Regulator), or due to qualitative or quantitative changes in the gene that transcript this protein in cells of various organs of the human body, resulting in numerous genotypes and phenotypes of the disease. In several patients, the diagnosis is difficult to be defined by the classical method of sodium and chloride dosage in sweat, or by genetic sequencing, justifying the use of new techniques for diagnosis, as the Nasal Potential Difference (NPD). This test provides a way of direct and sensitive assessment of the transport of sodium and chloride ions in cell membranes, via the nasal epithelium, based on transepithelial bioelectric properties. The objective of this work was to verify the difference of the values obtained in the examination of NPD in patients with CF compared with healthy control subjects, and, to verify if this test allows differentiating patients with more severe CF functional subclasses (I, II , III) from patients with less severe CF subclasses (IV, V, VI). This study included 15 CF patients, 10 with more severe mutations (group A) and 5 with less severe mutations (group B), and 21 healthy controls (group C). We considered the following test parameters of NPD: "Finger", PDMax, ?Amiloride, ?Amiloríde+Chloridefree and Wilchanski index. For "Finger" values, it was found difference between patients with CF Group B - less severe mutations (class IV , V or VI) and healthy individuals - group C. The value of Wilchanski index showed difference between group A CF patients, with more severe mutations (class I, II or III) and healthy individuals - group C. In our study, NPD showed statistically different values between CF patients with two known mutations and healthy subjects. Fabrication and use of new solid state phosphate ion selective electrodes for monitoring phosphorylation and dephosphorylation reactions

Highly selective and sensitive phosphate sensors have been fabricated by constructing a solid membrane disk consisting of variable mixtures of aluminium powder (Al), aluminium phosphate (AlPO4) and powdered copper (Cu). Both binary and ternary electrode systems are produced depending on their composition. The ternary membranes exhibit greater selectivity over a wide range of concentrations. The ternary electrode with the composition 25% AlPO4, 25% Cu and 50% Al was selected as our preferred electrode. The newly fabricated ternary membrane phosphate selective electrodes exhibited linear potential response in the concentration range of 1.0 × 10−6 to 1.0 × 10−1 mol L−1. The electrodes also exhibit a fast response time of <60 s. Their detection limit is 1.0 × 10−6 mol L−1. The unique feature of the described electrodes is their ability to maintain a steady and reproducible response in the absence of an ionic strength control. The electrodes have a long lifetime and can be stored in air when not in use. The selectivity of the new phosphate selective electrodes with respect to other common ions is excellent. The results obtained provide further insight into the working principles of the newly fabricated phosphate selective electrodes. Dephosphorylation and phosphorylation reactions were monitored using the preferred phosphate selective electrode. The following reactions were studied and inferences drawn; (a) the reactions between *[{CoN4(OH)(OH2)}]2+ and *[OH(PO2O)]2- for 1:1, 2:1 and 3:1 *[{CoN4(OH)(OH2)}]2+ to *[OH(PO2O)]2- ratios.(b) the reactions between *[{CoN4(OH)(OH2)}]2+ and *[O2NC6H4PO2(O)(OH)]- for 1:1, 2:1 and 3:1 *[{CoN4(OH)(OH2)}]2+ to *[O2NC6H4PO2(O)(OH)]- ratios. Among these are the methods of discontinuous pulse, hybrid pulse and polarized pulse methods. The structures formed by these methods can be applied to sensors and photonic materials. In this work the influences of the pulse parameters for the porous anodic alumina were investigated by polarized pulse potentiostatic anodization. Data analysis was presented from the duty cycle seen in the literature, which provides important information about the structure formed for the applied conditions and, in addition, new ways of analyzing the data related to the pulse, such as the analysis of the potential difference and the pulse frequency, this latter complementing the analysis of the duty cycle. As a result it was possible to observe that the porous anodic alumina thickness obtained is proportional to the duty cycle applied, where the higher duty cycle, the greater the thickness obtained. From the potential difference, the differences in the morphological and optical parameters could be analyzed by the influence of the negative minimum voltage applied during the pulsed anodization. The pulse frequency, also discussed in this work, was used to analyze different pulse periods for the same duty cycle, demonstrating that can be obtained structures with different values by varying the value of the pulse frequency. For the porosity calculation, an application was developed that helped to obtain the pore distance, which is one of the variables used to calculate the porosity. With the porosity values, the effective refractive index of each sample was calculated to find the effective optical thickness (EOT) and, finally, the thickness of the obtained anodic alumina film. / A alumina anódica porosa pode ser obtida, além dos métodos convencionais como potenciostático e galvanostático, ou por métodos pulsados. Dentre estes destacam-se os métodos de pulso descontínuo, pulso híbrido e pulso polarizado. As estruturas formadas por estes métodos podem ser aplicadas em sensores e materiais fotônicos. Neste trabalho foram investigadas as influências dos parâmetros de pulso para a obtenção da alumina anódica porosa pelo método de anodização potenciostática com pulso polarizado. Foram apresentadas análises de dados a partir do ciclo de trabalho visto na literatura, onde traz importantes informações sobre a estrutura formada para as condições aplicadas e, além disto, novas formas de analisar os dados referentes aos pulsos, como a análise a partir da diferença de potencial e a frequência de pulso, este último complementando a análise do ciclo de trabalho. Como resultado foi possível observar que a espessura de alumina anódica porosa obtida é proporcional ao ciclo de trabalho aplicado, onde quanto maior o ciclo de trabalho, maior será a espessura obtida. A partir da diferença de potencial pôde-se analisar as diferenças nos parâmetros morfológicos e ópticos pela influência da tensão mínima negativa aplicada durante a anodização pulsada. A frequência de pulso, também abordada neste trabalho, foi utilizada para analisar diferentes períodos de pulsos para o mesmo ciclo de trabalho, demonstrando que variando o valor da frequência de pulso pode-se obter estruturas com valores distintos. Para o cálculo da porosidade foi desenvolvido um aplicativo que auxiliou na obtenção da distância entre poros, que é uma das variáveis utilizadas para o cálculo da porosidade. E com os valores de porosidade foi calculado o índice de refração efetivo de cada amostra para encontrar a sua espessura óptico efetivo (EOT, do inglês Effective Optical Thickness) e, por fim, a espessura do filme de alumina anódica obtido.

Hur skall en ishallsvägg byggas? : Fuktrisker och mögelanalys / How should an ice hall wall be built? : Moisture risks and mold analysis

In Sweden there are many ice halls and ice halls climate differ from the usual buildings' climate, because temperatures are lower indoors than outdoors during the summer months. This results in a higher vapor content outdoors compared with indoors during the course of the year. Moisture transport takes place from the outside towards the ice halls, which is contrary to what happens in a common building. During the critical period, the vapor transport direction is inward. When the hot air hits the cold surface of the wall structure, it leads to a risk of condensation in the wall that leads to moisture damage. Moisture damage can impair the wall structure and lead to unwanted consequences. The purpose of this study was to dimension a number of walls that meet climate conditions, which are done by dynamic humidity calculation in WUFI Pro 5.1 with adapted climate data and real climate data and then mold analysis in WUFI Bio. The aim of the bachelor thesis work was that the results from this study can then be used as a guide to the future building of ice skating walls. The results from WUFI Pro 5.1 and WUFI Bio have shown that a number of walls designed during this master thesis are judged to work as well as revealed how an ice hallway wall should not be constructed. The conclusion shows that climate conditions control the design of the wall structure. / I Sverige finns det många ishallar och ishallsklimatet skiljer sig från vanliga byggnaders klimat, detta på grund av att temperaturen är lägre inomhus än utomhus under sommarhalvåret. Detta resulterar i en högre ånghalt utomhus jämfört med inomhus under en del av året. Fukttransporten sker då utifrån och in mot ishallen, det vill säga tvärtemot vad som sker i en vanlig byggnad. Under den kritiska perioden är ångtransportsriktningen inåt. När den varma luften träffar den kalla ytan i väggkonstruktionen, medför det en risk för kondens i väggen som leder till fuktskador. Fuktskador kan försämra väggkonstruktionen och leda till oönskade konsekvenser som exempelvis mögelpåväxt och rötskador. Syftet med studien har varit att dimensionera ett antal väggar som klarar av klimatförutsättningarna, detta gjordes genom dynamisk fuktberäkning i WUFI Pro 5.1 med anpassade klimatdata samt verkliga klimatdata och sedan mögelanalys i WUFI Bio. Målet med examensarbetet var att resultaten från denna studie sedan kan komma att användas som en vägledning till framtida byggen av ishallsväggar. Resultaten från WUFI Pro 5.1 och WUFI Bio har visat att ett antal väggar som konstruerats under detta examensarbete bedöms att fungera samt kommit fram till hur en ishallsvägg inte skall konstrueras. Slutsatesen visar att klimatförutsättningarna styr dimensioneringen av väggkonstruktionen.
