Neste trabalho desenvolveu-se uma metodologia analítica para determinação do teor de trialometanos THMs em amostras de águas potáveis, utilizando a técnica de purga e armadilha acoplada à cromatografia a gás (GCPT). Os THMs são subprodutos da cloração da água que devem possuir um limite máximo de 100 μg.L-1 segundo a legislação brasileira, estes compostos são suspeitos carcinogênicos humanos com base em estudos em animais de laboratório. A técnica de purga e armadilha extrai eficientemente estes compostos da água e a separação no cromatógrafo a gás ocorre utilizando coluna de leve polaridade e detector por captura de elétrons. Este detector é seletivo e o mais sensível para estes compostos halogenados. A metodologia desenvolvida foi validada nos itens de: linearidade, seletividade, exatidão, precisão, limite de quantificação, limite de detecção e robustez. O limite de detecção para os THMs foi menor do que 0,5 μg.L-1. A exatidão e precisão foram adequadas para ensaios de compostos traços. As amostras de água potável foram coletadas na cidade de Suzano-SP, que pertence à região do Alto do Tietê, nesta região nas margens do rio a vegetação é predominante. Entre todos os THMs, o composto encontrado na água potável em maiores concentrações foi o clorofórmio onde os resultados quantitativos, utilizando esta metodologia apresentaram teores entre 15,9 à 111,0 μg.L-1 em águas potáveis. / This work shows an analytical methodology developed and optimized to determine trihalomethanes level THMs, in drinking water samples, using purge and trap coupled to gas chromatography (GC-PT). THMs are byproducts water chlorination, these compounds must have a maximum of 100 μg.L-1 under Brazilian law, due these compounds be suspected human carcinogens base on studies in laboratory animals. The technique of purge and trap efficiently extracts these compounds from water, and the gas chromatograph separates the THMs. The GC uses a light polarity column and electron capture detector. This detector is selective and more sensitive in the detection of these compounds. The methodology was validated in terms of: linearity, selectivity, accuracy, precision, quantification limit, detection limit and robustness. The detection limit was less than 0,5 μg.L-1. The accuracy and precision were adequate for testing the trace compounds. The drinking water samples were collected in the city of Suzano-SP, which belongs to Alto do Tietê, in this region lay Tietê river with predominant vegetation. The THMs compound found in drinking water at higher concentrations was chloroform where the spread of values found between 15,9 to 111,0 μg.L-1 in drinking water.

The development of a sensitive method to study volatile organic compounds in gaseous emissions of lung cancer cell lines

The ultimate objective of this research was to develop a low cost, reliable system that would lead to early detection of lung cancer. Tests involved the quantitation of gaseous metabolic emissions from immortalized lung cancer cell lines in order to correlate the chemical markers to be of cancerous origin. The specific aims of the project were the study of gas emissions in selected cancer cell lines and identification of volatile organic compounds (VOCs) in them. Disadvantages of earlier studies were that the measurements were not real time or state specific so that molecular identification was often inconclusive. Furthermore the methods of study used in the past were not quantitative, which limited their practicality for medical applications. We felt the need to prove or disprove these earlier results using a new technique. The method we proposed is different and unique when compared to previous methods because cell lines have not been studied extensively for cancer markers. We have studied cancer cell lines which are adherent, immortalized cultures originating from primary tumors obtained from patients with no prior treatment for lung cancer. We have used an alternative method for the spectrometric analysis and quantitation of the selected chemical markers. The pre-concentration method involved a Purge and Trap unit with a thermal desorber where the vapor concentration was enhanced. The concentrated head space gases were analyzed using a Gas Chromatograph ?? Mass Spectrometer setup. This setup eliminated the bulky apparatus used in earlier studies. It is simpler in design and more comprehensive so that external factors such as patient??s diet, habitat and lifestyle do not contribute to our study of recognition of cancer markers. Based on the results obtained in the above experiments, a more comprehensive, inexpensive study of lung cancer related markers could be made. The first section, after giving an introduction to lung cancer, goes on to explain the background work done by other researchers on cancer. The third section gives a detailed explanation of the experimental setup. This is followed by all the tests conducted with corresponding results. The final section deals with the conclusions drawn from all experiments.

Sensory evaluation and quality assessment of an alternative inner coating film in yogurt cartons

The dairy food industry is continuously striving towards products with higher quality and longer shelf-life available to the customer at low prices. Arla Foods in Linköping, Sweden, is currently investigating the possibilities of changing the material in yogurt packaging containers by replacing the currently used carton with a different and cheaper alternative. A successful switch will give the company an economical advantage without affecting the sensory attributes (smell, taste, sight, and consistency), aroma profile or other important trademarks of the yogurt. This study is designed to examine and compare yogurt that has been stored in different packaging cartons, one coated with a single-layered low-density polyethylene (LDPE) and one coated with a currently used multi-layered ethylene-vinyl alcohol (EVOH). The study was based on the analysis and measurement of sensory attributes performed by experts, physical properties in laboratory and chemical composition in GC-FID/MS together with a discriminative test where a group of people would identify any difference between the yogurts. Together, these analyses would provide an explanation about any differences between the packaging materials by connecting physical, chemical and/or sensory characteristics. The collected results would give a better and more comprehensive picture than each analysis would do separately. The results from the study show that there is a difference between yogurts stored in LDPE-based containers and yogurts stored in EVOH-based containers and that the product was chemically affected, mainly by the level of oxygen in contact with the food. The overall assessment is that the largest difference was discovered in the taste.

Development of analytical methods for ultra-trace determination of total mercury and methyl mercury in natural water and peat soil samples for environmental monitoring

Abstract Mercury is a global pollutant that accumulates easily in forest soils, even in remote areas. Mercury accumulated in soils can be subsequently released into surface waters causing an increased eco-toxicological and human health risk. The most toxic form of mercury to humans and wildlife is methyl mercury (MeHg), which can be formed in the environment via methylation processes. In freshwaters, MeHg is readily accumulated in fish, which are the main source of human exposure to MeHg. The determination of both total mercury and MeHg concentrations in environmental samples, such as natural waters and soils, is important in environmental risk assessment. This study involved the development of analytical methods for the determination of ultra-trace total mercury and MeHg concentrations in humic-rich natural water and peat soil samples. Each developed method was carefully optimized and validated by using real natural water and peat soil samples, certified reference materials and/or reference methods. The cold vapor inductively coupled plasma mass spectrometry (CV-ICP-MS) method developed during this study was found to be a reliable method for the determination of total ultra-trace mercury concentrations in natural freshwaters. Purge and trap gas chromatography, coupled to an ICP-MS, was used in mercury speciation analysis. Together with species-specific isotope dilution this technique proved to be a reliable method in MeHg determinations. Prior to instrumental determination, MeHg was successfully isolated from humic-rich water and peat soil samples using N2-assisted distillation. The analytical methods developed in this study were successfully applied to an investigation of the effects of forest harvesting practices on the mobilization of mercury in boreal forest catchments. / Tiivistelmä Elohopeaa pääsee ilmakehään sekä luonnollisista lähteistä (mm. tulivuorenpurkaukset ja kiviaineksen rapautuminen), että ihmisen toiminnan kautta. Elohopean viipymäaika ilmakehässä on hyvin pitkä, minkä vuoksi se voi kulkeutua kauas päästölähteestä ennen päätymistään maaperään ja vesistöihin. Ympäristössä olevasta epäorgaanisesta elohopeasta voi muodostua erittäin myrkyllistä metyylielohopeaa, joka rikastuu helposti ravintoketjussa. Metyylielohopean muodostuminen on merkittävä osa elohopean biogeokemiallista kiertoa, minkä vuoksi metyylielohopean määrittäminen näytteen kokonaiselohopeapitoisuuden ohella antaa tärkeää tietoa elohopean käyttäytymisestä ympäristössä. Tutkimuksessa kehitettiin analyysimenetelmät, joilla määritettiin ultrapieniä kokonaiselohopea- ja metyylielohopeapitoisuuksia humuspitoisista luonnonvesistä ja turvemaanäytteistä. Tutkimuksessa käytetyt näytteet oli kerätty turvemaametsien valuma-alueilta Sotkamosta. Luonnonvesinäytteiden kokonaiselohopeapitoisuuksien määrityksessä käytettiin kylmähöyrymenetelmää (CV) yhdistettynä induktiiviplasma-massaspektrometriaan (ICP-MS). Vesi- ja turvenäytteiden metyylielohopeapitoisuuksien määrityksessä elohopeaspesiekset erotettiin kaasukromatografisesti (GC) ja määritettiin isotooppilaimennus-ICP-MS:lla. Ennen GC-ICP-MS -määritystä näytteet esikäsiteltiin typpiavusteisella tislausmenetelmällä ja esikonsentroitiin ’purge and trap’ -tekniikalla. CV-ICP-MS ja ’purge and trap’ GC-ICP-MS -menetelmät optimoitiin huolellisesti sekä laiteparametrien, että reagenssimäärien suhteen. Menetelmillä saatavien tulosten oikeellisuus varmistettiin vertailumateriaalien ja/tai vertailumenetelmien avulla. Kehitettyjä analyysimenetelmiä hyödynnettiin tutkimuksessa, jossa seurattiin metsähakkuiden mahdollisia vaikutuksia elohopean huuhtoutumiseen ja metyloitumiseen ojitetuilla turvemailla.
