Many piano teachers agree that suitable repertoire motivates their students to learn and practice their instrument. Asking students to learn different categories of repertoire might be a good approach for piano teachers to motivate and maintain their students’ musical interests. However, how do teachers evaluate what kinds of repertoire is presented in the teaching material that is available to them? The purpose of this study is to review the pieces found in North-American piano method books in order to provide an inventory of the different categories of repertoire found in each series and to calculate the proportion of piano repertoire belonging in each category. Consequently, this research will allow piano teachers to see more clearly which kinds of repertoire are in a method book and help select the most appropriate method books for their students.

The exegesis investigates ways in which major piano works of the 19th and 20th centuries reflect elements of baroque tradition and considers the implications for performance. After the baroque period, from the 18th century to the present, many composers have continued to explore baroque forms, textures and expressive devices. In meinem Vortrag möchte ich zeigen, welche Entwicklungsetappen die Etablierung des Theaters in Tallinn durchgemacht hat. Interessant wäre darüber hinaus zu untersuchen, was dabei im Verhältnis zu anderen deutschsprachigen Theatern ähnlich bzw. was unterschiedlich war. Ich werde mich der Frage widmen, welche Beziehungen es zu anderen Truppen oder Theatern im Ostseeraum gab, welchem Repertoire die Vorliebe des Publikums galt und aus welchen Schichten der Tallinner Einwohner das Publikum bestand.

Chapes / Immunoglobulins are receptors expressed on the outside of a B cell that can specifically bind pathogens and toxic substances within a host. These receptors are heterodimers of two chains: heavy and light, which are encoded at separate loci. Enzymatic splicing of gene segments at heavy and light chain loci within the genomic DNA in every B cell results in a highly diversified and specific repertoire of immunoglobulins in a single host. Spaceflight is known to affect reduce splenic B cell populations and B cell progenitors within the bone marrow, potentially restricting the diversity of the immunoglobulin repertoire (Ig-Rep). The objective of this thesis project was to characterize the impact of spaceflight on the kappa light-chain Ig-Rep of the C57BL/6 mouse. High-throughput sequencing (HTS) technologies have enabled the rapid characterization of Ig-Reps, however, standard Ig-Rep workflows often rely the amplification of immunoglobulin sequences to ensure the capture immunoglobulin sequences from rare B cell clones. Additionally, the Ig-Rep is often assessed in sorted B cell populations. Opportunities for spaceflight experiments are limited and costly, and the exclusive amplification of immunoglobulin sequences prior to HTS results in a dataset that cannot be mined for additional information. Furthermore, due to the difficulties of tissue collection in spaceflight, HTS of sorted B cell populations is not feasible. We optimized a protocol in which the Ig-Rep was assessed from unamplified whole tissue immunoglobulin transcripts. The Ig-Rep was characterized by gene segment usage, gene segment combinations and the region in which gene segments are joined. HTS datasets of ground control animals and animals flown aboard the International Space Station were compared to explore the impact of spaceflight on the unimmunized murine Ig-Rep.

The research was guided by a social constructivist worldview and is an explanatory case study. Data were collected through interviewing five experienced and established choir conductors who conduct secondary school choirs. Five themes crystallised through the data analyses, which are Education, Vocal ability, Type of choir, Programme choice and Audience preference and development. Investigating the immunoglobulin repertoire is a means of understanding the adaptive immune response to infectious disease or vaccine challenge. The data examined are typically generated using high-throughput sequencing on samples of immunoglobulin variable-region genes present in blood or tissue collected from human or animal subjects. The analysis of these large, diverse collections provides a means of gaining insight into the specific molecular mechanisms involved in generating and maintaining a protective immune response. It involves the characterization of distinct clonal populations, specifically through the inference of founding alleles for germline gene segment recombination, as well as the lineage of accumulated mutations acquired during the development of each clone. Germline gene segment inference is currently performed by aligning immunoglobulin sequencing reads against an external reference database and assigning each read to the entry that provides the best score according to the metric used. The problem with this approach is that allelic diversity is greater than can be usefully accommodated in a static database. The absence of the alleles used from the database often leads to the misclassification of single-nucleotide polymorphisms as somatic mutations acquired during affinity maturation. This trend is especially evident with the rhesus macaque, but also affects the comparatively well-catalogued human databases, whose collections are biased towards samples from individuals of European descent. Our project presents novel statistical methods for immunoglobulin repertoire analysis which allow for the de novo inference of germline gene segment libraries directly from next-generation sequencing data, without the need for external reference databases. These methods follow a Bayesian paradigm, which uses an information-theoretic modelling approach to iteratively improve upon internal candidate gene segment libraries. Both candidate libraries and trial analyses given those libraries are incorporated as components of the machine learning evaluation procedure, allowing for the simultaneous optimization of model accuracy and simplicity. Finally, the proposed methods are evaluated using synthetic data designed to mimic known mechanisms for repertoire generation, with pre-designated parameters. We also apply these methods to known biological sources with unknown repertoire generation parameters, and conclude with a discussion on how this method can be used to identify potential novel alleles.
