Oligopeptides construits autour du γ-aminoacide ATC : synthèses, analyses structurales et évaluation biologique / Oligopeptides built around ATC γ-amino acid : syntheses, structural analyses and biological evaluation

Les travaux décrits dans ce manuscrit concernent la synthèse, l’étude structurale et l’évaluation biologique d’oligopeptides abiotiques incorporant le gamma-aminoacide hétérocyclique nommé ATC (acide-4-Aminométhyl-1,3-Thiazole-5-Carboxylique). Les ATCs sont construits autour d’un noyau thiazole et présentent deux points de diversité structurale. De précédents travaux ont déterminé que la présence du noyau thiazole entre les positions alpha et béta bloquait l'angle zéta autour de 0°, structurant les homo-oligomères de poly-(S)-ATCs en une hélice 9 droite et les faisant ainsi entrer dans le domaine des foldamères. Dans une première partie, nous avons entrepris de développer une voie de synthèse simple, flexible et énantiosélective permettant d’obtenir les ATCs stéréochimiquement purs sur une échelle de plusieurs grammes à partir d'alpha-aminoacides commerciaux. L’introduction de la diversité chimique est réalisée via deux étapes-clés que sont la condensation croisée de Claisen et la réaction de cyclisation de Hantzsch. Puis l’identification des marqueurs de structuration RMN et IR-TF des oligomères d'ATCs a été mise à profit pour caractériser le repliement d’hétéro-oligomères combinant ATCs et alpha-aminoacides. Ainsi, une étude structurale par RMN, IR-TF, cristallographie RX et dichroïsme circulaire a démontré que l’enchaînement 1:1 (L)-alpha:(S)-ATC se structurait en un ruban, stabilisé par un réseau intramoléculaire de liaisons hydrogène bifides formant des pseudocycles à 9 et 12 chaînons. La distribution des chaînes latérales le long du squelette principal présente une forte analogie avec l’hélice alpha, ce qui pourrait constituer un atout majeur pour le développement de composés à finalité thérapeutique. La dernière partie de ce travail a porté sur la conception de pseudo-peptides amphipatiques pour des applications en temps qu'antimicrobiens. / This manuscript describes the synthesis, the structural study and the biological evaluation of abiotic oligopeptides incorporating the heterocyclic gamma-amino acid ATC (4-Aminomethyl-1,3-Thiazole-5 Carboxylic acid). This original block is built around a thiazole ring and displays two lateral chains. Previous work in our laboratory highlighted that the presence of the thiazole ring between the positions alpha and beta implied that zeta angle was blocked around 0°, thus structuring poly-(S)-ATCs homo-oligomers in a right-handed 9-helix foldamer. First, development of a simple, flexible and enantioselective synthesis on a few grams scale has allowed to get access of a highly diverse ATC library from commercial alpha-amino acids. Introduction of the chemical diversity occurs via two key steps implying a cross-Claisen condensation and a Hantzsch cyclization. Then identification of NMR and FT-IR structural markers of ATC-containing oligomers was used to characterize the folding propensity of hybrid α:ATC oligomers. We demonstrated that 1:1 (L)-alpha:(S)-ATC heterochiral oligomers are structured in solution in a new ribbon-like shape stabilized by a bidentate intramolecular hydrogen bond network forming 9- and 12-membered pseudorings. The distribution of lateral chains along the main skeleton shows a high analogy with alpha-helix thus constituting a major advantage for potential medicinal applications. The last part of this work has focused on the design of amphipathic ATC-containing pseudo-peptides as antimicrobial agents.

Electronic and Ionic Transport in Carbon Nanotubes and Other Nanostructures

abstract: This thesis describes several experiments based on carbon nanotube nanofludic devices and field-effect transistors. The first experiment detected ion and molecule translocation through one single-walled carbon nanotube (SWCNT) that spans a barrier between two fluid reservoirs. The electrical ionic current is measured. Translocation of small single stranded DNA oligomers is marked by large transient increases in current through the tube and confirmed by a PCR (polymerase chain reaction) analysis. Carbon nanotubes simplify the construction of nanopores, permit new types of electrical measurement, and open new avenues for control of DNA translocation. The second experiment constructed devices in which the interior of a single-walled carbon nanotube field-effect transistor (CNT-FET) acts as a nanofluidic channel that connects two fluid reservoirs, permitting measurement of the electronic properties of the SWCNT as it is wetted by an analyte. Wetting of the inside of the SWCNT by water turns the transistor on, while wetting of the outside has little effect. This finding may provide a new method to investigate water behavior at nanoscale. This also opens a new avenue for building sensors in which the SWCNT functions as an electronic detector. This thesis also presents some experiments that related to nanofabrication, such as construction of FET with tin sulfide (SnS) quantum ribbon. O gênero discursivo tira em atividades de leitura em sala de aula / The gender discursive ribbon in activities of reading of classroom

O presente trabalho tem como objeto de pesquisa o estudo do gênero discursivo tira, presente em livros didáticos e avaliações da rede pública estadual, como o SARESP e o ENEM. Teve como ponto de partida observar as propostas de leitura de tiras em livros didáticos e analisar se apresentam atividades que permitam ao aluno ser capaz de perceber o humor obtido nas tiras que, muitas vezes, é fortemente dependente de intertextualidade, dos implícitos e subentendidos, de conhecimento prévio sobre o perfil psicológico dos personagens e do contexto sócio-histórico em que ele atua; e também observar se essas atividades são capazes de explorar a linguagem verbal e a não-verbal presentes nesse gênero. Para direcionar a trajetória dessa pesquisa, efetuou-se uma análise fundamentandose nas atuais propostas de ensino que tem nos gêneros discursivos, no sentido bakhtiniano do termo, suas unidades básicas de ensino. O corpus escolhido para essa pesquisa constituiu-se em vinte propostas de leitura do gênero discursivo tira dos personagens Mafalda, Hagar, o horrível, Recruta Zero, Mônica, Magali, Cascão e Cebolinha coletados aleatoriamente em vários livros didáticos de Língua Portuguesa de Ensino Fundamental e Ensino Médio, adotados na rede pública estadual paulista. Os resultados da pesquisa mostram que, para se obter uma efetiva leitura do gênero discursivo tira presente nos livros didáticos, é necessário que o aluno conheça e identifique os vários recursos verbais e não-verbais presentes nesse gênero, e saiba utilizá-los como facilitadores dessa leitura de humor. Essa pesquisa pode fornecer subsídios aos professores, para que possam planejar atividades de trabalho utilizando o gênero tira, em sala de aula, de maneira mais abrangente e enriquecedora. / The present work has as research object the study of the gender discursive ribbon, present in text books and evaluations of the state public net, like SARESP and ENEM. He/she had as starting point to observe the proposals of reading of ribbons in text books and to analyze if they present activities that allow to the student to be capable to notice the humor obtained in the ribbons that, a lot of times, it is strongly dependent of intertextuality, of the implicit and implied, of previous knowledge on the characters psychological profile and of the partner-historical context in that he acts; and also to observe those activities is capable to explore the verbal language and the no-verbal presents in that gender. To address the path of that research, he/she occurred an analysis being based in the current ones proposed of teaching that has in the discursive goods, in the sense bakhtiniano of the term, their basic units of teaching. The chosen corpus for that research was constituted in twenty proposed of reading of the characters gender discursive ribbon "Mafalda", "Hagar, the horrible", "Recruta Zero", "Mônica, Magali, Crust and Green onion" collected in several text books of Portuguese Language of Fundamental Teaching and Medium Teaching, adopted in the net state public inhabitant from São Paulo. The results of the research show that, to obtain an effective reading of the gender discursive ribbon present in the text books, it is necessary that the student knows and identify the several verbal resources and no-verbal presents in that gender, and know how to use them as facilitators of that humor reading. That research can supply subsidies to the teachers, so that they can plan work activities using the gender removes, in classroom, of including way and rich.

The subject of this master’s thesis is to design an arch structure of a footbridge. The bridge deck will be formed by a stress ribbon and could be divided into three parts. The middle part will be suspended (on the outer edges) on composite steel and concrete arches. Outer parts of the bridge deck will be formed by stress ribbons. The model of this construction is created in a computational program ANSYS and the solution is non-linear. The design is according to European standards.

Computing the Rank of Braids

We describe a method for computing rank (and determining quasipositivity) in the free group using dynamic programming. The algorithm is adapted to computing upper bounds on the rank for braids. We test our method on a table of knots by identifying quasipositive knots and calculating the ribbon genus. We consider the possibility that rank is not theoretically computable and prove some partial results that would classify its computational complexity. We then present a method for effectively brute force searching band presentations of small rank and conjugate length.

Synthesis and Characterizations of Fe-based Metallic Glassy Systems

This thesis is a study of tailoring amorphous Fe-B-Si based alloy to produce bulk glassy rods by adding Nb. We have prepared rapid quenched thin ribbons (thickness ~12 µm) by melt spinning, and glassy rods of diameter ~1mm by Cu-mold casting based on compositions (Fe0.78B0.13Si0.9)100-xNbx (x=0, 4, 8, 12), and studied their different physical properties. The melt-spun ribbons are found to be X-ray amorphous, whereas some nano-crystallinity is observed in the case of rods. All the ribbons show high saturation magnetization and low coercivity, which are the desirable characteristics of a soft ferromagnet. These ribbons are thus suitable for designing high frequency transformers, and sensors from an applications point of view. With increasing Nb content their saturation magnetization, ferromagnetic Curie temperature, and resistivity are found to decrease as expected. The temperature dependence of electrical resistivity shows small positive temperature co-efficient that is expected for a metallic disordered material. We have also studied the modification of the properties on thermal annealing the (Fe0.78B0.13Si0.9)96 Nb4 ribbon at different temperatures in a neutral atmosphere.
