This master thesis is focused on producing orthopaedic implant materials and measuring their corrosion properties. It describes the bone and its structure, types, bone ossification and healing. It defines functions of orthopaedic implants and mentions the types of implants – biodegradable and non-biodegradable. The thesis interprets what corrosion is, what categories of corrosion exist and how does the corrosion influence orthopaedic implants. Preparing the solution of stimulated body fluid and manufacturing samples of different metal combinations (of iron, manganese, phosphorus, magnesium, silver and zinc) is included in this thesis, together with corrosion measurements, microscopic observations, EDAX analysis, metallographic analysis, microhardness testing of samples and pH changes measurements of solutions, and the results are interpreted and explained.

Due to massive expansion of the mass spectrometry and constant price growth of the human labour the optimalisation of the microbial samples preparation comes into question. This master thesis deals with design and implementation of a machine learning algorithm for segmentation of images of microbial colonies cultivated on Petri dishes. This algorithm is going to be a part of a controlling software of a MBT Pathfinder device developed by the company Bruker s. r. o. that automates the process of smearing microbial colonies onto a MALDI target plates. In terms of this thesis a several models of neural networks based on the UNet, UNet++ and ENet architecture were implemented. Based on a number of experiments investigating various configurations of the networks and pre-processing of the training datatset there was chosen an ENet model with quadruplet filter count and additional convolutional block of the encoder trained on a dataset pre-processed with round mask.

The main aim of this master's thesis is to describe the subject of the implementation of decision boundaries with the help of artificial neural networks. The objective is to present theoretical knowledge concerning this field and on practical examples prove these statements. The work contains basic theoretical description of the field of pattern recognition and the field of feature based representation of objects. A classificator working on the basis of Bayes decision is presented in this part, and other types of classificators are named as well. The work then deals with artificial neural networks in more detail; it contains a theoretical description of their function and their abilities in the creation of decision boundaries in the feature plane. Examples are shown from literature for the use of neural networks in corresponding problems. As part of this work, the program ANN-DeBC was created using Matlab, for the generation of practical results about the usage of feed-forward neural networks for the implementation of decision boundaries. The work contains a detailed description of this program, and the achieved results are presented and analyzed. It is shown as well, how artificial neural networks are creating decision boundaries in the form of geometrical shapes. The effects of the chosen topology of the neural network and the number of training samples on the success of the classification are observed, and the minimal values of these parameters are determined for the successful creation of decision boundaries at the individual examples. Furthermore, it's presented how the neural networks behave at the classification of realistically distributed training samples, and what methods can affect the shape of the created decision boundaries.

Diplomová práce se zabývá zobecněným exponenciálním rozdělením jako alternativou k Weibullovu a log-normálnímu rozdělení. Jsou popsány základní charakteristiky tohoto rozdělení a metody odhadu parametrů. Samostatná kapitola je věnována testům dobré shody. Druhá část práce se zabývá cenzorovanými výběry. Jsou uvedeny ukázkové příklady pro exponenciální rozdělení. Dále je studován případ cenzorování typu I zleva, který dosud nebyl publikován. Pro tento speciální případ jsou provedeny simulace s podrobným popisem vlastností a chování. Dále je pro toto rozdělení odvozen EM algoritmus a jeho efektivita je porovnána s metodou maximální věrohodnosti. Vypracovaná teorie je aplikována pro analýzu environmentálních dat.

An user interface is created for more complex control of each model states of the data being considered.

Pour accéder à la caractérisation des matrices complexes pétrolières, la chromatographie en phase liquide bidimensionnelle couplée à une détection spécifique comme la spectrométrie de masse à couplage inductif (LCxLC-ICP-MS/MS) s’avère être une solution pertinente. Un tel couplage permet d’envisager la spéciation des contaminants soufrés ou encore métallés (vanadium et nickel). Ce couplage reste, à notre connaissance, inédit aujourd’hui et sa mise en place a nécessité en premier lieu d’évaluer différents systèmes d’introduction de l’échantillon en amont de la détection. La comparaison de ces systèmes, au regard de la dispersion qu’ils génèrent, a été effectué afin de conserver la qualité de séparation obtenue en sortie du système LCxLC. La seconde partie du développement instrumental a concerné l’optimisation de la partie LC×LC. Le choix des différents mécanismes de rétention dans les deux dimensions étant primordial au vu de la complexité des échantillons (polarité, solubilité, poids moléculaire…). De plus, l’introduction de matrices organiques dans les sources plasma reste un réel défi qu’il a fallu évaluer, celles-ci pouvant être la cause de nombreuses contraintes analytiques. Enfin, une fois la méthodologie off-line SECxRPLC-ICP-MS/MS développée, elle a été appliquée à différents échantillons montrant qu’elle peut être considérée comme une solution intéressante pour expliquer le comportement de certaines matrices au sein des unités de raffinage, par l’intermédiaire de la comparaison de cartographies 2D / Two-dimensional liquid chromatography coupled with a specific detection such as inductively coupled plasma mass spectrometry (LCxLC-ICP-MS/MS) proves to be a relevant technique for the characterization of petroleum complex matrices. Such coupling makes it possible to consider the speciation of sulfur or metal contaminants (vanadium and nickel). Firstly, the evaluation and the comparison of several sample introduction systems was performed, with regard to the dispersion induced in the system, in order to keep a high efficiency from the LCxLC system. The second part of the instrumental development concern the optimization of both dimensions. The choice of the different retention mechanisms is essential given the complexity of the samples (polarity, solubility, molecular weight...). Additionally, the introduction of organic matrices in the plasma remains a real challenge which could be the cause of many instrumental and analytical issue. Finally, once the off-line SECxRPLC-ICP-MS/MS method was developed, it was applied to different samples showing how it can be considered as an interesting tool to explain the behavior of matrices within the refining units, through the comparison of 2D-contour plots

<p>Senzori vlažnosti zemljišta su od ključnog značaja za sisteme za navodnjavanje kojima je moguće uvećati prinose u proseku za 79%. U okviru ove disertacije razvijena su dva senzorska rešenja za merenje vlažnosti zemljišta. Prvi senzor je namenjen određivanju vlažnosti uzoraka zemljišta u neporemećenom stanju. Rešenje je nastalo nadogradnjom standardne opreme za uzorkovanje zemljišta. Za pomenuti senzor konstruisana je kalibraciona kriva kojom je moguće odrediti vlažnost zemljišta sa relativnom greškom ne većom od 2.5%. Drugo senzorsko rešenje je fabrikovano u LTCC tehnologiji, malih je dimenzija i namenjeno je primeni na terenu. Glavna prednost ovog senzora je da na njega ne utiče provodnost zemljišta koja je usko povezana sa tipom zemljišta. Predloženi senzor je testiran na dva uzorka zemljišta različitog hemijskog sastava. Rezultati merenja su pokazali da tip zemljišta nema uticaj na odziv senzora. Ostvarena relativna greška u odnosu na pun merni opseg iznosi 5.36%.</p> / <p>Soil moisture sensors are of great importance for the irrigation systems that<br />are able to increase the yiel on avarage of 79%. Within this thesis two sensor<br />solutions have been developed. The first sensor, which is intended for<br />laboratory use, has been designed by upgrading existing sampling<br />equipment. The constructed calibration curve for this type of sensor that can<br />be used for soil mositure determination with relative error not larger than<br />2.5% with respect to full scale output. The second sensor solution is<br />designed to be used in the field. It is fabricated in LTCC technology and it is<br />charaterized by small overall dimensions. The main advantage of this sensor<br />is that it is immune to the soil electrical conductivity which is closely related to<br />the soil type. Proposed sensor is tested on two soil sample of different<br />chemical composition and the results have shown that the type of soil does<br />not influence the sensor response. The relative error with the respect of full<br />scale output was only 5.36 %.</p>

The aim of this diploma thesis was the development of a screening method for analysis of 17 benzodiazepines in urine samples using ultra high-performance liquid chromatography with tandem mass spectrometric detection. The partial task was to optimize the conditions for the enzymatic hydrolysis of benzodiazepine glucuronides present in urine using design of experiments (DOE). The optimized chromatographic system consisted of a Zorbax Eclipse Plus Phenyl-Hexyl RRHD column (100 × 2.1 mm, 1.8 μm) and mobile phase consisting of water with 0.1 % acetic acid (component A) and acetonitrile with 0.1 % acetic acid (component B) in various ratios according to the gradient program. Flow rate was 0.2 ml/min, column temperature was 40 řC, and total analysis time was 12 min. Calibration curves for all analytes were measured under optimized conditions in methanol and urine. After optimal detection conditions for oxazepam-glucuronide were found, oxazepam glucuronide was hydrolysed using β-glucuronidase from the abalone to confirm the functionality of the enzyme within the pilot experiment. Optimization of enzymatic hydrolysis conditions via 27 experiments proposed by program Minitab 16 using the Box-Behnken design will be realized later.

Missing Data - A Gentle IntroductionÖsterlund, Vilgot January 2020 (has links)
This thesis provides an introduction to methods for handling missing data. A thorough review of earlier methods and the development of the field of missing data is provided. The thesis present the methods suggested in today’s literature, multiple imputation and maximum likelihood estimation. A simulation study is performed to see if there are circumstances in small samples when any of the two methods are to be preferred. To show the importance of handling missing data, multiple imputation and maximum likelihood are compared to listwise deletion. The results from the simulation study does not show any crucial differences between multiple imputation and maximum likelihood when it comes to point estimates. Some differences are seen in the estimation of the confidence intervals, talking in favour of multiple imputation. The difference is decreasing with an increasing sample size and more studies are needed to draw definite conclusions. Further, the results shows that listwise deletion lead to biased estimations under a missing at random mechanism. The methods are also applied to a real dataset, the Swedish enrollment registry, to show how the methods work in a practical application.
