Caspase-3 in lens epitheliumTalebizadeh, Nooshin January 2016 (has links)
Purpose: To model the time evolution of active caspase-3 protein expression in a healthy lens, and in a lens exposed to UVR-300 nm (UVR-B). To develop an automated method to classify the fluorescent signal of biomarkers in the lens epithelial cells. Methods: Six-week old Sprague-Dawley rats were used. Firstly, expression of active caspase-3 was studied in the lens epithelium of healthy rats. Secondly, rats were unilaterally exposed in vivo to 1 kJ/m2 UVR-B for 15 minutes. At 0.5, 8, 16, and 24 hours after the UVR-B exposure, the exposed and the contralateral non-exposed lenses were removed. Immunohistochemistry was done on three mid-sagittal sections from each lens. The florescent labelling for active caspase-3 in each lens section was counted three times. The time evolution of active caspase-3 expression in response to UVR-B exposure was modelled as a function of cell position in the lens epithelium. An automated objective method was developed to quantify the lens epithelial cells and to classify the fluorescent signal of active caspase-3. Active caspase-3 was selected as a model signal. Results: Active caspase-3 was abundant in the anterior pole of the normal lenses. Spatial distribution of active caspase-3 labelling in the lens epithelium was fitted to a logistic model. The probability of active caspase-3 expression was higher in the UVR-B exposed lenses (95% CI = 0.12 ± 0.01). There was no difference in the expression of active caspase-3 between the 0.5 and the 24 hours groups or between the 8 and the 16 hours groups. A difference was noted, when comparing the 0.5 and 24 hours groups with the 8 and 16 hours groups (Test statistic 7.01, F1;36;0.95= 4.11). Exposure to UVR-B has an impact on the average probability of labelling for active caspase-3 as a function of cell position. The probability of labelling as a function of cell number also varied as a function of time after UVR-B exposure. The automated method counted the lens epithelial cells and estimated the proportion of active caspase-3 labelling in the lens epithelium. Conclusions: Active caspase-3 is present in the healthy lens epithelial cells. Active caspase-3 exhibits higher expression at the anterior pole of the lens and the expression decreases towards the periphery. After UVR-B exposure, the expression of active caspase-3 in the lens epithelium increases with a peak of expression occurring around 16 hours after exposure. The average probability of labelling in the lens epithelium is dependent on both the UVR-B exposure and the time period elapsed after the exposure. The automated method enables objective and fast quantification of lens epithelial cells and the expression of fluorescent signal in the lens cells.

Thesis (MScEng (Civil Engineering))--University of Stellenbosch, 2008. / This thesis is an investigation of the wave power resource on the South African coast,
focusing on the spatial distribution of wave power of the coastal region exposed to the
highest wave power. The study’s main objective is to provide a detailed description of the
spatial distribution of wave power to assist in the selection of locations for deployment of
Wave Energy Converter (WEC) units in this zone. The study methodology employed to
achieve this main objective entails an analysis of measured wave data recorded at wave
recording stations distributed along the South African coast. The analysis provided a
general description of wave power at locations for which wave data exist. From this analysis
it was found that the South West Coast is exposed to the highest wave power, with an
average wave power of approximately 40 kW per meter wave crest. The rest of the South
African coast is exposed to average wave power between approximately 18 kW/m to 23
kW/m.
The wave power characteristics on the South West Coast region (from Cape Point to Elands
Bay) were therefore the focus of this thesis. The study objective was achieved by
transferring deep sea wave data into the nearshore South West Coast study area with the
Simulating WAves Nearshore (SWAN) wave model. The deep sea wave data was obtained
from a 10 year period of available hindcast data. A simplified simulation procedure was
required in order to make the study practically feasible. A sensitivity analysis was carried
out to determine the validity of the simplified simulation procedure and it was found that the
procedure slightly overestimate wave power in the shallower water regions due to the
underestimation of energy dissipation processes. This overestimation was deemed
acceptable for the dominant wave conditions and the simplified model was therefore applied
in the study. An appropriate programming system was developed and used to transfer the
available 10 year deep sea wave data into the selected South West Coast region. From this
exercise spatial distribution of wave power and related statistical parameters were obtained
for the study area. The accuracy of the modelled output was investigated by directly
comparing it to wave data recorded during the overlapping recording period. It was found
that the model slightly overestimates the monthly wave power resource compared to the
measured data with a maximum overestimation of 9%; which is sufficiently accurate for the
purpose of the study.
The results of this investigation can be used for the identification of areas of high wave
power concentration within the study area for the location of WEC units. Further numerical modelling is required for the detailed design of wave farms, especially if potential

Thesis (MScConEcol)--Stellenbosch University, 2012. / ENGLISH ABSTRACT: Acacia saligna, or Port Jackson, is an Australian Acacia which has spread throughout the Western
and Eastern Cape provinces of South Africa where it has become invasive and a prominent feature
of the vegetation. In areas where A. saligna establishes it replaces natural vegetation, alters
ecosystem processes and interferes with agricultural practices. Consequently large management
efforts have been made to eradicate this invasive alien tree. However, its large and persistent soil
stored seed bank, even in the presence of management and biological control agents, poses a
serious obstacle to its successful removal. Furthermore the reproductive output and the size of the
seed bank of A. saligna over its whole distribution as well as the variation of seed production and
the seed bank with environmental conditions in time and space is poorly known. Understanding the
seed bank dynamics of A. saligna in time and space is essential for reducing invasive success and
achieving management objectives. This study has filled this knowledge gap through studying the
seed production and seed bank of A. saligna over its invaded range in South Africa, including how
environmental factors influence these factors in time and space. The seed rain of A. saligna was
assessed at 10 sites across its distribution in South Africa. The seed rain of A. saligna at the sites
was determined through the use of seed rain traps. Twenty five traps were placed out at every site
during November 2010 (pre-dehiscence) which was collected again during April 2011 (postdehiscence).
The seed bank of A. saligna was estimated through sampling at 25 sites across its
distribution range in South Africa. The seed bank was sampled during April 2010 (postdehiscence),
November 2010 (pre-dehiscence) and April 2011 (post-dehiscence) through taking 50
litter and soil samples at every site which gave a total sampling size of 3 750 for both the seed in
the soil and litter over its distribution in South Africa. In addition the average tree diameter, tree
density, average number of Uromycladium tepperianum induced galls per tree, the summer aridity
index, De Martonne aridity index, winter concentration of precipitation, temperature of coldest
month and the soil texture for every site was determined. The damage done by the seed feeding
weevil, Melanterius compactus, was also estimated for the seed rain study sites. Water availability
during the hot summer months was assessed as the most important factor governing seed
production and seed bank size. Riparian and non-riparian water regimes were shown to be
important in understanding the seed bank dynamics of A. saligna over its distribution range in the
Cape Floristic Region. In non-riparian A. saligna populations the seed production and consequently
the size of the seed bank and its rate of accumulation is limited by both water and temperature and
in riparian A. saligna populations, only by temperature. Therefore, two environmental gradients
influence the seed bank dynamics of Port Jackson in South Africa. In non-riparian A. saligna
populations the number of seed produced and the accumulation of seed in the seed bank generally
increases along the west coast of South Africa from Clanwilliam towards Cape Town and along the
south coast from Cape Town towards Port Elizabeth. Seed banks are larger closer to the coast,
when A. saligna populations of similar age are compared. In riparian A. saligna populations, the number of seed produced generally increases from Port Elizabeth towards Cape Town and from
Cape Town towards Clanwilliam, again, with larger seed banks being accumulated closer to the
coast, when populations of similar age are compared. This study provides managers with a useful
tool for prioritising management efforts. / AFRIKAANSE OPSOMMING: Acacia saligna of Port Jackson is ‘n indringerplant in Suid-Afrika wat afkomstig is uit Suid-Wes
Australië. Acacia saligna is ‘n onmisbare kenmerk van die plantegroei in die Wes en Oos-Kaap.
Waar Port Jackson vestig, verplaas dit natuurlike plantegroei, verander dit ekosisteem prossesse
en verhinder dit landbou praktyke. Gevolglik is groot bestuurspogings aangewend om hierdie
indringer plant uit te roei. In die teenwoordigheid van bestuursprogramme en biologiese beheer bly
A. saligna lewensvatbaar as ‘n gevolg van die plant se groot blywende grond saadreserwes.
Verder is die saad produksie en saadbank grootte van A. saligna oor sy verspreiding bereike
asook hoe dit met omgewingstoestande in tyd en ruimte interreageer en varieer grootendeels
onbekend. ‘n Goeie en omvattende begrip van die saadbankdinamika is belangrik om die
indringings vermoë van die plant te verlaag en om bestuursdoelwitte te bereik. Hierdie studie vul
die bestaande gaping in die kennis aangaande die saadbank dinamika van A. saligna aan deur die
saadproduksie en die saadreserves van die indringerplant oor sy verspreiding in Suid-Afrika te
bestudeer, insluitend hoe omgewingstoestande die saadbankdinamika beïnvloed in tyd en ruimte.
Die saadreën vir 10 A. saligna populasies was bepaal deur die gebruik van saadlokvalle. Vyf-entwintig
lokvalle was uitgeplaas in elke bestudeerde Port Jackson perseel gedurende November
2010 (voor-saadval) wat weer gaan haal is gedurende April 2011 (na-saadval). Die saadbank van
A. saligna was bepaal in 25 populasies van die boom gedurende April 2010 (na-saadval),
November 2010 (voor-saadval) en April 2011 (na-saadval) deur die neem van 50 blaar- en
grondmonsters by elke perseel wat ‘n totaal van 3 750 blaar- en grondmonster gee oor die hele
verspreiding van A. saligna in Suid-Afrika. Verder is die gemiddelde boomdeursnee, boomdigtheid,
gemiddelde Uromycladium tepperianum geinduseerde galle per boom, die somers droogtheids
indeks, De Martonnes droogtheids indeks, die winter konsentrasie van presipitasie, die
temperatuur van die koudste maand en die grond tekstuur van elke Port Jackson stand bepaal. Die
skade wat die saad voedende kewer, Melanterius compactus, aanrig aan die sade van A. saligna
is ook vir die persele waar die saadreën eksperiment uitgevoer is, bepaal. Die beskikbaarheid van
water vir A. saligna in die droë somermaande is bepaal as die belangrikste faktor wat die grootte
van die saadproduksie en saadbank beïnvloed. Oewer en nie-oewer water omgewings is bevind as
noodsaaklik om die saadbank dinamika van A. saligna oor die indringerplant se verspreidingareas
in die Kaap Floristiese Streek te verstaan. In nie-oewer A. saligna populasies word saadproduksie
en gevolglik die grootte en tempo van akkumulasie van die saadbank deur beide water en
temperatuur omstandighede beperk, terwyl in oewer A. saligna populasies word die indringerplant
slegs deur temperatuur omstandighede beperk. Gevolglik blyk dit dat twee water beskikbaarheids
gradiënte bestaan waarop die plant reageer wat dan uitgedruk word in die plant se saad produksie
en gevolglik ook die plant se saadbank. In nie-oewer Port Jackson populasies neem die
saadproduksie en die tempo waarteen die saadbank akkumuleer algemeen toe langs die weskus
van Clanwilliam na Kaapstad en van Kaapstad na Port Elizabeth, met grootter saadproduksie en saadbank akkumulasie nader aan die kusgebiede, wanneer A. saligna populasies van dieselfde
ouderdom vergelyk word. In oewer Port Jackson populasies neem die saadproduksie en die tempo
waarteen die saadbank akkumuleer algemeen toe van Port Elizabeth na Kaapstad en van

The entry of immigrants into their incoming country as well as their subsequent spread over the country's territory occurs through processes that have a strong spatial bias. The specific migration groups (here defined as the set of immigrants with the same country of origin) tend to reveal different levels and patterns of their spatial concentrations. The understanding of these similarities and dissimilarities in spatial behaviour is important task from both academic and policy perspective. This thesis has a quantitative character and it is based on the study of "spatial relatedness" of migration groups, which is defined as a rate, how much do given migration groups concentrate into the same regions. The thesis is based on a simple assumption that the spatial relatedness mirrors mutual proximity or similarity between these groups in other respects (e.g. cultural or economical). Plenty of datasets describing spatial distribution of migrants within 32 territorial systems on various scales (from global to local) are analysed. These systems will be analysed separately at first and then a synthesis of these partial results will be done with the aim to discover prevailing pattern of the spatial relatedness of migration groups. Influence of catchment spatial subdivision on the accuracy of the snow accumulation and snowmelt model Abstract This thesis is focused on influence of catchment spatial subdivision on the accuracy of rainfall-runoff model simulations. A hydrological model HEC-HMS developed by U.S. Army Corps of Engineers was applied in the thesis. Study area includes two experimental catchments; Bystřice River basin and Zlatý Brook basin. Both catchments are located in the Krušné Mountains. The research in catchments has been carried out by the Department of Physical Geography and Geoecology since 2008. The influence of four different horizontal and vertical catchments delineation on runoff simulations was analyzed. The simulations were performed in two two-year periods 11/2008-10/2010 and 11/2010-10/2012. The attention was mainly aimed to snowmelt component of the model, for which temperature index method was chosen. The results of simulations were compared with water stages data measured in catchments outlets and with data from winter field measurements (snow depth and snow water equivalent). The results from both catchments showed that higher accuracy of simulation of the snow water equivalent was reached by dividing the catchments into more elevation zones. A distribuição espacial de insetos e doenças em campos comerciais é importante, por exemplo, para aplicação racional de pesticidas. Entretanto, não tem sido considerada nas recomendações de manejo da cultura, planejamento de experimentos e estudos amostrais, sendo escassa literatura a esse respeito. Os artigos apresentados nessa tese foram motivados por duas situações diferentes, uma envolvendo dados de contagem e a outra, dados binários. Os dois modelos diferem em relação às estratégias da descrição da estrutura de dependência espacial. No primeiro artigo, a variável resposta é contagem. Para caracterizar o padrão espacial da dispersão do tripes do prateamento da cebola foi feito um levantamento anotando-se o número de insetos por fase de desenvolvimento em folhas de plantas de cebola, em diferentes datas e pontos amostrais dentro de quatro propriedades com fazendas vizinhas apresentando diferentes níveis de infestação e métodos de plantio. O teste de aleatorização de Mantel foi utilizado para testar a presença de padrão espacial, que quando detectado foi descrito por um modelo de Poisson misto espacial com componente aleatório geoestatístico. Tal modelo possibilitou a caracterização do padrão espacial bem como a obtenção de mapas de predição dos níveis de susceptibilidade à infestação na área. No segundo artigo a variável resposta é binária e foi feito um estudo de simulação para verificar o comportamento dos estimadores de pseudo-verossimilhança dos parâmetros do modelo autologístico, considerando diferentes estruturas de covariáveis e de vizinhança, três intensidades de infestação de uma praga e cinco valores para o parâmetro de correlação entre os vizinhos. Uma aplicação dos modelos considerados no estudo de simulação é feita a um conjunto de dados provenientes de um experimento com pimentão. Mostra-se que o método de estimação por pseudo-verossimilhança pode ser usado, com certa cautela, quando o interesse está na contribuição das covariáveis, mas não deve ser usado quando o interesse está na estimação da correlação espacial. Um estudo com diferentes porcentagens de dados faltantes foi feito para verificar a influência na estimação do parâmetro. / The spatial distribution of insects and diseases in commercial fields is important for the efficient application of pesticides. However, in the past this has not been considered in crop management recommendations, experiment planning and sampling plans. The papers presented in this thesis were motivated by two different situation, one envolving count data and the other binary data. The two models used differ in relation to the strategies of the description of the spatial dependence structure. In the the first paper the response variable is a count. In order to characterize the spatial distribution pattern of the onion thrips a survey was carried out to record the number of insects in each development phase on onion plant leaves, on different dates and sample locations, in four rural properties with neighboring farms with different infestation levels and planting methods. The Mantel randomization test was used to test for spatial correlation, and when detected this was modelled by a mixed spatial Poisson model with a geostatistic random component. This model has allowed a spatial pattern characterization as well as the production of prediction maps of susceptibility to levels of infestation in the area. In the second paper the response variable is binary. In this paper a simulation study on pseudo-likelihood estimators of autologistic parameters to verify the effect of different covariate and neighbouring structures is described, with three pest infestation levels and five different spatial correlation coefficient values. An application of the methodology is presented using a bell pepper data set. It is shown that the pseudo-likelihood method can be used when a researcher is interested in the effect of covariates, but should not be used for the estimation of the spatial correlation. A study with different percentages of missing data is made to verify the influency on parameter estimation.

São apresentados resultados de coletas de mosquitos culicídeos realizadas de 1978 a 1983 em área de matas residuais, representando ambiente modificado por ação antrópica, na região do Vale do Ribeira, Estado de São Paulo, Brasil. Pretendeu-se esclarecer aspectos do comportamento de populações de mosquitos (Diptera:Culicidae) na fase adulta, com ênfase na distribuição horizontal e vertical. Como técnica de coleta, foi utilizada a armadilha CDC, com e sem isca animal (galinha doméstica Gallus gallus). Para algumas espécies foram construídos gráficos de distribuição sazonal. No geral, as espécies ou grupos que se destacaram entre as mais frequentes foram: Culex(Culex)spp., Aedes(Ochlerotatus) seratus, Culex(Melanoconion)ribeirensis e Cx.(Mel.) sacchettae. É discutida a capacidade vetora dessas e de outras espécies também consideradas entre as mais frequentes. / Some results of mosquitoes collection from 1978 to 1983 in areas comprising residual forests wich represent environments modified by man in Ribeira Valley, State of São Paulo, Brazil were presented. The aim of this project was to enlighten behavioral aspects of adult mosquitoes (Diptera:Culicidae) giving emphasis to the horizontal and vertical distributions. CDC light traps with and without animal bait (domestic hen- Gallus~gallus) were utilized for collecting. The seasonal distribution of some species were presented graphically. The following species or groups: Culex(Culex)spp., Aedes (Ochlerotatus)serratus, Culex(Melanoconion) ribeirensis, and Cx.(Mel.) sacchettae were distinguished among the most frequent. The vectorial capacity of these and other species considered among the most frequent was discussed.
