Highly-exact structural characterization is the crucial step in the development and manufacturing process of pharmaceutical materials. Their structural composition is, however, often very complex and hardly identifiable. The eligible way for obtaining definite structural interpretation of these systems appears the high-resolution solid-state nuclear magnetic resonance (ssNMR) spectroscopy. For this purpose the reliable tool - the ssNMR toolbox for comprehensive characterization of various pharmaceutical solids is described. The rigorous optimization of ssNMR techniques is carried out on enormous number of measured samples containing active pharmaceutical ingredients (APIs) with systems ranging from APIs formulated in solid dispersions to pure forms revealing extensive molecular disorder. In this study the influence of polymeric matrix on the creation of solid dispersion type susceptible for finely tuned controlled drug release is likewise discussed. The distinction between variable structural alignments of API molecules in 3D dimension of complicated pharmaceutical solids is allowed via simple strategy - factor analysis applied to hardly describable ssNMR spectra (13 C CP/MAS NMR and 19 F MAS NMR). The diploma thesis is focused on Storm water management in urban areas. The first task for processing thesis was to arrange meeting with MČ Brno - Bohunice Ing. Antonín Crha, who has provided necessary information about the targeted location the and the reservoir for accumulation of rainwater collected. The practical part of the thesis is to capture the rainwater drainage in the existing rainwater sewerage system using the newly built pumping station and then pumping into the existing, unused reservoir. Accumulated water in the reservoir is planned to be used for the needs of the city, gardeners and other customers. The first part of the diploma thesis describes the management of refined water in the Czech Republic, Denmark and Germany. The first part also provides description of measures and technologies for the management of rainwater. Finally, there is description of the pumping station and borehole objects and the program for calculating the rainfall from the urbanized area. In the practical part of the diploma thesis the selected variants are described, rewarded and evaluated. The diploma thesis deals with two variants, that are guided along the same route, with the difference of the construction of the drilled well in the second variant. Both variants are handled by pumping. The option of the gravity inflow was rejected because of the large countersink of rainwater sewerage.

The diploma thesis titled "A health state assessment in the Czech Republic and in states of the European Union" is a theoretically conceived work that offers comprehensive information on assessment activities focused on the sickness insurance in the Czech Republic and other five selected countries of the European Union. A choice of the selected states has been affected both geographical and historical linkages to the Czech Republic as well as differences in the social security systems. The most attention has been devoted to the assessment activities under the sickness insurance of the Czech Republic. For other countries there has been particularly given a course of the health state assessment due to the illness, pregnancy and maternity or care for a household member. The aim of the thesis is to analyze the process of the health state assessment in the Czech Republic and a subsequent comparison of those results with a situation in the selected countries of the European Union, namely the Slovak Republic, Poland, Austria, Great Britain and France. To identify differences and to focus on strengths and weaknesses of the systems or to suggest a more efficient solution within the Czech Republic. The results of the thesis show that in all researched countries a medical certificate of the incapacity for work is the legal condition for an acquisition of claims to sickness benefits and the procedure for an obtaining of the certificate is similar in all six countries. It is the examination and health state assessment by a physician authorized by a body administering the social security in the given country. Despite differences of the social security systems in individual countries, a connecting element for the claim to the benefit is the participation on the social insurance. This thesis may also serve the general public as a summary of findings on the issue of the health state assessment under the sickness insurance in the Czech Republic and selected countries of the European Union and for a better orientation in the given systems.

Etnické/rasové rozdíly ve výskytu kolorektálního karcinomu v USA / Ethnic/race differences in the incidence of colorectal cancer in the USA

This diploma thesis deals with ethnic differences in the incidence of colorectal cancer. One of the primary aims is to analyze the influence of selected socio-demographic factors, health factors and lifestyle factors on the incidence of colorectal cancer. Another main aim is to find out if there are ethnic differences in the incidence of colorectal cancer in the United States to determine the contribution of the influence of selected socio-demographic and lifestyle factors using the data from the PLCO database. PLCO is a case-control study which includes individual data collected from approximately 155,000 respondents in the United States. The main finding is a significant influence of respondents' age structure, gender, ethnicity, education, family history of colorectal cancer as well as diabetes on the incidence of colorectal cancer. It has also been suggested that alcohol consumption, smoking and obesity increase the risk of colorectal cancer. On the contrary, an increased intake of vitamin D and drugs containing acetylsalicylic acid (in this case aspirin) reduces the risk of colorectal cancer. Časově rozlišená fluorescence ve výzkumu interakcí hyaluronanu a koloidních systémů / Time-Resolved Fluorescence in Research of Hyaluronan-Colloidal Systems Interactions

The aim of the doctoral thesis was to study advanced fluorescence techniques and its use in colloids or hyaluronan-surfactant systems and hydrogels based on hyaluronan, respectively. Steady-state and time-resolved fluorescence were used to study excited state proton transfer fluroescen probes in hyaluronan-surfactant systems to asses the influence of hyaluronan hydration to its interactions with oppositely charged surfactants. Firstly, different excited state proton transfer fluorescence probes were discussed to choose the most suitable candidate for next research. The influence of hyaluronan on inner environment of micells was determined based on the sensitivity of excited state proton transfer of chosen fluorescence probe 1-naphtol and, based on above mentioned experiments, the structure of hyaluronan hydration shell was discussed. The next part of doctoral thesis was focused on fluorescence lifetime correlation spectroscopy and on the development of method of nanorheology. Measured correlation functions were transformed to mean square displacement with developed MATLAB script. Firstly, the fluorescence method was compared with well described methods such as videomicrorheology and dynamic light scattering to asses the reliability of fluorescence correlation spectroscopy in microrheology. Secondly, nanorheology method was developed and its use in passive nanorheology of hyaluronan hydrogels was discussed. Based on mentioned experiments, the fluorescence correlation spectroscopy microrheology and nanorheology methods were optimized to use the methods in hydrogel research.
