Avaliamos a função cardíaca frente a Isquemia e reperfusão, em corações isolados de ratas intactas, com hipotireoidismo (Hipo), hipotireoidismo subclínico (HTS), redução dos níveis de hormônios ovarianos (OO) e a ambas condições (HTS+OO). Na estabilização, a Pressão Desenvolvida pelo Ventrículo Esquerdo (LVDP), Pressão Diastólica Final (EDP) e Pressão de Perfusão não diferiram. Porém, a Primeira Derivada Positiva das Pressões (+dP/dt), Primeira Derivada Negativa das Pressões (-dP/dt) e frequência cardíaca foram menores no grupo Hipo. Na reperfusão, os corações Hipo tiveram EDP, +dP/dt e -dP/dt semelhantes a estabilização. Assim, sugere-se que o HTS, a OO e o HTS + OO não alteraram a recuperação cardíaca após a isquemia. E que o Hipo é resistente à lesão por isquemia, apesar da função cardíaca deprimida. / We evaluate the cardiac function in relation to an ischemia and reperfusion event in hearts isolated from intact rats, With hypothyroidism (Hypo), subclinical hypothyroidism (HTS), decreased levels of ovarian hormones (OO) and both conditions (HTS + OO). At etabilization the Left Ventricle Developed Pressure (LVDP), Final Diastolic Pressure (EDP) and Perfusion Pressure did not differ. However, the First Positive Pressure Derivative (+ dP / dt), First Negative Pressure Derivative (-dP / dt) and heart rate were lower in the Hypo group. On reperfusion the hypothyroid hearts presented EDP, + dP / dt and -dP / dt Similar to stabilization. Thus, it is suggested that HTS, a OO e o HTS + OO Did not change cardiac recovery after ischemia.Is that hipo is resistant to ischemia injury, despite depressed cardiac function.

Independentemente dos resultados encontrados, foram coletadas amostras de leite para análises microbiológicas e de contagem de células somáticas (CCS). O isolamento microbiológico foi realizado em ágar sangue de ovino 5% e em ágar MacConkey, com 72 horas de incubação em aerobiose a 37oC. Foram procedidas, a cada 24 horas, leitura e identificação dos isolamentos. A ocorrência de quadros de mastite clínica foi representada por um único caso (0,14%) entre os 735 quartos mamários analisados, ocorrendo na fase inicial da lactação e envolvendo microrganismos de origem ambiental (Morganella morganii, Enterobacter aerogenes). Foram considerados quadros de mastite subclínica aqueles com resultados positivos ao CMT acompanhados de exame microbiológico positivo. A freqüência observada de quadros de mastite subclínica entre os 734 quartos mamários estudados foi de 20,03%, dos quais isolou-se principalmente microrganismos contagiosos. Os resultados negativos ao CMT predominaram durante todo o estudo (69,07%), independente da fase de lactação ou estação do ano consideradas, tendo sido observados resultados positivos do exame microbiológico em 57,58% das amostras CMT negativas. Foi elevada a freqüência de microrganismos classificados como contagiosos entre as amostras de leite analisadas, independente da estação do ano e da fase de lactação, entre os quais prevaleceu o Corynebacterium spp. (47,67%), entre os 579 microrganismos pertencentes a 19 gêneros bacterianos crescidos em cultura pura ou em associação. (Resumo completo, clicar acesso eletrônicio abaixo) / Abstract: A total of 735 mammary quarters from buffalo cows were submitted to inspection, palpation, strip cup test and California Mastitis Test (CMT) in order to diagnose clinical and subclinical mastitis. Independent of the observed results, milk samples were collected from each mammary quarter to microbiological exams and automatic somatic cell counts (SSC). Milk samples were inoculated in 5% ovine blood agar and MacConkey agar and incubated for 72 hours under 37oC and aerobic condition. Growth observations and identifications were performed every 24 hours. One (0,14%) out of 735 mammary quarters studied presented clinical mastitis. It occurred in first stage of lactation (0 to 60th day post calving) and environmental microorganisms were isolated (Morganella morganii, Enterobacter aerogenes). Mammary quarters presenting positive results to CMT and microbiological exams were considered as subclinical mastitis cases and these represented 20,03% among 734 studied samples. CMT negative results were the most frequent observed during the study (69,07%), independent of lactation stage or season of the year considered. Among these CMT negative milk samples, 57,58% were positive in microbiological exams. There was a high occurrence of contagious microorganisms among the 579 identified bacteria from 19 different genus, isolated in pure or in association. Corynebacterium spp. was the most prevalent (47,67%), in every lactation stage or season of the year. Considering the absence of a normal distribution, somatic cell counts were submitted to a logarithmic transformation, log2(CCS/100)+3, originating the transformed somatic cell count (TSCC). O elevado custo dos tratamentos tradicionais da mastite bovina, associado à redução de produção e inviabilidade de tratamento das mastites subclínicas durante a lactação, bem como a exigência cada vez mais rigorosa da ausência de resíduos de antimicrobianos por parte de instituições nacionais e internacionais, impulsiona o desenvolvimento de novas alternativas terapêuticas que visem minimizar o impacto das medidas tradicionais de tratamento. A homeopatia surge como importante alternativa, sendo aceita nacional e internacionalmente. O objetivo do trabalho foi avaliar o tratamento homeopático com Phytolacca decandra 30CH durante a lactação de vacas com mastite subclínica, utilizando parâmetros de qualidade do leite como Califórnia Mastitis Test (CMT), contagem de células somáticas (CCS) totais, porcentagens de polimorfonucleares (PMN) e de mononucleares (MN), teores de proteína, lactose, gordura, sólidos totais (ST) e extrato seco desengordurado (ESD), além da mensuração da produção leiteira. Foram selecionadas 26 vacas, CMT 2+ e 3+ sem sinais de mastite clínica, entre o terceiro e sexto mês de lactação, pluríparas e divididas em dois grupos, um com tratamentos medicamentosos mensais e outro com tratamentos quinzenais. Mensalmente foram colhidas duas amostras de leite de cada glândula mamária (teto) que apresentava mastite subclínica. Uma das amostras foi utilizada para CCS e análise dos componentes do leite, e a outra para identificação microbiológica. Nos grupos tratados, foi administrado o medicamento homeopático Phytolaca decandra 30CH diluído em água e aspergido nas mucosas oro-nasais e vaginais, enquanto que nos grupos denominados controle foi administrado placebo da mesma maneira. A pesagem da produção láctea de ambos os grupos foi realizada quinzenalmente até o final do experimento. Verificou-se então que não houve diferença significativa entre a produção láctea, CCS e a presença de microrganismos na secreção láctea das glândulas quando comparadas antes e depois do tratamento homeopático, dentro de cada grupo, bem como quando comparados os grupos entre si. As medianas de CMT e porcentagens de PMN foram compatíveis com infecções mamárias agudas, embora os animais tenham sido diagnosticados como portadores de mastites crônicas e em nenhum momento desenvolveram sinais clínicos. / The high cost of traditional treatments of bovine mastitis, with the reduction of production and impracticable treatment of subclinical mastitis during lactation, as well as the requirement of an increased demand for absence of antimicrobial agents residues by national and international institutions, drives the development of new therapeutic alternatives in order to minimizing the impact of traditional treatment measures. The homeopathy appears as an important alternative, accepted domestically and internationally. The objective of this work was to evaluate homeopathic treatment with Phytolacca decandra 30CH during the lactation of cows with subclinical mastitis, using some milk quality parameters as California Mastitis Test (CMT), total somatic cell count (SCC), percentages of polymorphonuclear (PMN) and mononuclear (MN) cells, levels of protein, lactose, fat, total solids and dry defatted matter, in addition to the measurement of milk production. Twenty-six cows were selected with CMT 2 + and 3 + without any signs of clinical mastitis, between the third and sixth month of lactation, pluriparous and they were divided into two groups, with monthly or biweekly drug treatments. Monthly were collected two milk samples from each mammary gland (teat) that presented subclinical mastitis. One of the samples was used for analysis of SCC and milk components, and the other one for microbiological identification. In the treated groups, was given the homeopathic medicine Phytolaca decandra 30CH diluted in water and sprayed in the oral-nasal and vaginal mucosa, while to the control group was given placebo following the same method. The weighing of the milk production for both groups was performed fortnightly until the end of the experiment. There was no significant difference between milk production, SCC and the presence of microorganisms in milk gland secretion compared before and after homeopathic treatment, within each group as well as comparing the groups together. The medians of CMT and percentages of PMN were compatible with acute mammary infection, in spite of they have been diagnosed as carriers of chronic mastitis, and at any time no clinical signs have been developed.

No universo da pecuária leiteira, a mastite representa um importante desafio, sendo responsável por perdas econômicas consideráveis relacionadas principalmente com redução na produção de leite. O gênero Staphylococcus assume elevada importância como agente etiológico das mastites devido à sua ampla distribuição e freqüência de ocorrência. Foram coletadas amostras de leite de fêmeas bovinas com mastite subclínica para exames microbiológicos e contagem de células somáticas (CCS). Após o isolamento e identificação dos micro-organismos, as amostras positivas foram submetidas a análises das medianas das CCS, testes de susceptibilidade \"in vitro\" frente a diferentes antimicrobianos, assim como pesquisa de genes codificadores das adesinas - genes que codificam para proteína ligadora de colágeno (cna), proteína ligadora de laminina (eno), proteína ligadora de elastina (ebp), proteína ligadora de fibrinogênio (fib), proteína A ligadora de fibronectina (fnbA), proteína B ligadora de fibronectina (fnbB) e proteína associada à formação de biofilme (bap), por meio da Reação em Cadeia da Polimerase (PCR). De acordo com os achados do presente estudo, dentre as bactérias do gênero Staphylococcus isoladas, os S. aureus foram verificados com maior freqüência, seguido por Staphylococcus coagulase-negativos - S. intermedius, S. chromogens e S. warneri. Com respeito às medianas das CCS, o gênero Streptococcus spp. apresentou o maior valor. O perfil de sensibilidade e resistência aos antimicrobianos testados foi semelhante entre as espécies de Staphylococcus coagulase-positivos e negativos, sendo os antimicrobianos cefalexina, cefalotina e ceftiofur os que apresentaram maior frequência de sensibilidade, e penicilina, amoxicilina e ampicilina os que apresentaram maior resistência. Com exceção do (fnbA), todos os outros fatores de virulência estudados foram detectados, sendo os genes eno, fib e a associação dos genes \"eno/fib/bap\" os mais freqüentemente detectados. Nas amostras coletadas dos tanques de refrigeração não foram detectadas todas as espécies de Staphylococcus spp. isoladas dos quartos mamários. Milk samples were collected from cows with subclinical mastitis to microbiological examinations and somatic cell count (SCC). After isolation and identification of microorganisms, positive samples were analyzed for the medians of SCC, tested for susceptibility \"in vitro\" against different antimicrobials and polymerase chain reaction will be used to search for genes encoding adhesins - genes that code for collagen-binding protein (cna), lamininbinding protein (eno), elastin-binding protein (ebp), fibrinogen-binding protein (fib), fibronectin-binding protein A (fnbA), fibronectin-binding protein B (fnbB) and protein associated with biofilm formation (bap). O uso de antibióticos para tratamento da mastite bovina subclínica durante a lactação apresenta restrições econômicas e a administração indiscriminada e inadequada destes medicamentos os torna potencialmente tóxicos aos animais e aos consumidores finais dos produtos lácteos. A utilização de medicamentos homeopáticos oferece menor custo, facilidade de administração, não há risco de resistência microbiana e não é necessário o descarte do leite dos animais em tratamento. Além disso, a homeopatia é reconhecida no Brasil como especialidade médica veterinária e aceita para uso no sistema de produção animal orgânico. Portanto, o presente trabalho teve como objetivo avaliar a eficácia de dois protocolos homeopáticos para tratamento da mastite bovina subclínica durante a lactação. O experimento foi realizado no período de seis meses e dividido em duas etapas. Na Etapa I, um grupo foi medicado com Sulphur 30CH, enquanto que o outro recebeu placebo. Na Etapa II, um grupo foi medicado com Calcarea carbonica 30CH e o outro grupo recebeu placebo. Os medicamentos foram administrados a cada trinta dias na Etapa I e a cada quinze dias na Etapa II. Em cada etapa foram coletadas amostras de leite antes, durante e após o tratamento. Para avaliar a eficácia dos protocolos, durante o experimento foram analisadas 138 amostras de leite na Etapa I e 72 amostras na Etapa II, realizando-se prova de Tamis, California Mastitis Test (CMT), contagem de células somáticas (CCS), quantificação de polimorfonucleares (PMN) e mononucleares (MN) por microscopia óptica, exame microbiológico e mensuração da produção leiteira. As análises estatísticas foram realizadas utilizando o software Graphpad Instat 1990-93. Os protocolos homeopáticos testados não diminuíram a celularidade do leite, pois não houve diferenças significativas nos resultados do CMT e da CCS no decorrer do experimento. Também não foi constatada nenhuma alteração significante na produção láctea. O microrganismo isolado com maior frequência no rebanho estudado foi o Corynebacterium bovis. Porém, não houve diferenças significativas em relação à frequência de isolamento deste microrganismo ao longo dos tratamentos, indicando a sua permanência nas glândulas mamárias. Além disso, observou-se durante todo o experimento o predomínio de células PMN em relação às MN. Sugere-se o estudo de novos protocolos homeopáticos com outros medicamentos, potência e frequência de administração, a fim de buscar alternativas para o tratamento de vacas com mastite subclínica / The use of antibiotics for treatment of bovine subclinical mastitis during lactation presents economic restrictions and the indiscriminate and inappropriate administration of these drugs makes them potentially toxic to animals and consumers of dairy products. The use of homeopathic medicine offers lower cost, ease of administration, no risk of microbial resistance and is not necessary to discard milk from animals under treatment. Furthermore, homeopathy is recognized as a medical specialty in Brazil and support for veterinary use in organic livestock production system. Therefore, this study aimed to evaluate the effectiveness of two protocols for homeopathic treatment of bovine subclinical mastitis during lactation. The experiment was performed between six months and divided into two stages. In Phase I, one group was treated with Sulphur 30CH, while the other received placebo. In Phase II, one group was treated with Calcarea carbonica 30CH and the other group received placebo. The drugs were administered every thirty days in the Phase I and every fifteen days in the Phase II. At each step the milk samples were collected before, during and after treatment. To evaluate the effectiveness of the protocols during the experiment were analyzed 138 samples of milk at Phase I and 72 samples at Phase II, performing proof Tamis, California Mastitis Test (CMT), somatic cell count (SCC), quantification of polymorphonuclear (PMN) and mononuclear (MN) by optical microscopy, microbiological examination and measurement of milk production. Statistical analysis was performed using the software Graphpad Instat 1990-93. The homeopathic protocols tested not diminished cellularity of milk because there were no significant differences in the results of CMT and SCC during the experiment. Nor was it found no significant change in milk production. The organism isolated most frequently in the herd studied was Corynebacterium bovis. However, no significant differences were observed in the frequency of isolation of this microorganism during the treatments, indicating his permanence in the mammary glands. Furthermore, it was observed during the experiment a predominance of PMN cells compared to MN. It is suggested the study of new protocols with other homeopathic medicines, potency and frequency of administration, to find alternatives for the treatment of cows with subclinical mastitis

Regulation of Adipocyte Lipolysis by TSH and its Role in Macrophage Inflammation

Elevated Thyroid-Stimulating Hormone (TSH) is associated with an increased risk of cardiovascular disease (CVD). We hypothesized that TSH-stimulated FA release from adipocytes contributes to macrophage inflammation. 3T3-L1 and human subcutaneous differentiated adipocytes were treated with TSH for 4 hours under various conditions and lipolysis assessed via glycerol secretion. Optimal conditions were determined and protein expression of ATGL, HSL and perilipin remained stable. TSH-stimulated 3T3-L1 or human adipocyte-conditioned medium (T-ACM) was placed on murine J774 or human THP-1 macrophages, respectively, and macrophage cytokine mRNA levels (IL-1β, IL-6, MCP-1, and TNFα) were measured by real-time RT-PCR. T-ACM did not change cytokine mRNA expression in J774 macrophages or THP-1 macrophages when compared to ACM. Absence of BSA in the medium may have hindered release of FA from differentiated adipocytes into the medium, BSA may be required to permit adequate FA accumulation in the medium to then evaluate the effect of T-ACM on macrophages. Further investigation is required to determine the effect of FA on J774 and THP-1 inflammatory response.

Altered afferent input resulting from neck joint dysfunction has become a growing area of study. Cervical spine manipulation, specifically in individuals with subclinical neck pain (SNCP); induces neurological changes, suggesting it has a positive neuromodulatory effect on brain processing. The effects of manipulation on motor learning in individuals with SCNP have not been investigated until now. Studies in this thesis sought to develop and investigate a novel motor training task to be coupled with cervical spine manipulation to investigate its effects on individual’s ability to process new task information. The studies revealed significant changes in neural activity specific to the cerebellum and sensorimotor integration following a complex motor training task as compared to a simple repetitive task, suggesting that those specific regions are involved in processing of more complex motor skill learning tasks. This novel task was then coupled with manipulation which revealed significant activation increases in cortical and decreases in subcortical brain regions following manipulation. Regions specific to sensorimotor integration (SMI) showed increased activation in both the manipulation and passive head movement control groups, corroborating with the results from the first study. The use of a complex motor training task is a useful tool for determining intervention effects on neural processing in individuals with SCNP. / UOIT

Regulation of Adipocyte Lipolysis by TSH and its Role in Macrophage Inflammation

Elevated Thyroid-Stimulating Hormone (TSH) is associated with an increased risk of cardiovascular disease (CVD). We hypothesized that TSH-stimulated FA release from adipocytes contributes to macrophage inflammation. 3T3-L1 and human subcutaneous differentiated adipocytes were treated with TSH for 4 hours under various conditions and lipolysis assessed via glycerol secretion. Optimal conditions were determined and protein expression of ATGL, HSL and perilipin remained stable. TSH-stimulated 3T3-L1 or human adipocyte-conditioned medium (T-ACM) was placed on murine J774 or human THP-1 macrophages, respectively, and macrophage cytokine mRNA levels (IL-1β, IL-6, MCP-1, and TNFα) were measured by real-time RT-PCR. T-ACM did not change cytokine mRNA expression in J774 macrophages or THP-1 macrophages when compared to ACM. Absence of BSA in the medium may have hindered release of FA from differentiated adipocytes into the medium, BSA may be required to permit adequate FA accumulation in the medium to then evaluate the effect of T-ACM on macrophages. Further investigation is required to determine the effect of FA on J774 and THP-1 inflammatory response.

Carotid artery disease : plaque features and vulnerability

Background: Atherosclerosis is an important cause of stroke. Ultrasound offers the convenience of real-time and detailed assessment of carotid plaque features as well as arterial wall thickening and composition. Evaluation of these features is important for determining patients’ risk of suffering vascular events and also contributes to selecting the best treatment strategy. Methods: Using ultrasound data analysis we have determined plaque features in the bifurcation and internal carotid artery (ICA), including: surface plaque irregularities, calcification, echogenicity (grey scale median-GSM) and other textural plaque features (Juxtaluminal black area, entropy, coarseness). In addition, intima media thickens (IMT) and its grey scale median (IM-GSM) was measured in common carotid artery (CCA). Using Cone Beam CT (CBCT) we have quantified calcification volume of the carotid plaques extracted after carotid endarterectomy procedure. For the meta-analysis we have used comprehensive meta-analysis software version 3. Study I: We have included 39 patients and we compared carotid plaque features of the contralateral arteries with those located ipsilateral to symptomatic side and arteries of asymptomatic patients. Study II: The accuracy of US to detect atherosclerosis calcification was assessed against CBCT in 88 patients. Study III: Based on the previous vascular events in coronary, carotid and lower extremity arterial system, 87 patients were divided into three groups: asymptomatic, symptoms in one vascular system and symptoms in more that one vascular system. IMT, IM-GSM and plaque features were compared between groups. Study IV: We have meta-analyzed ten cohort prospective studies evaluating carotid plaque echogenicity for cerebrovascular symptoms prediction. Results: Study I. Plaques of the contralateral to symptomatic arteries had similar features to those in symptomatic and more vulnerable than asymptomatic arteries. Study II. Carotid ultrasound was accurate in detecting calcification volumes of ≥8mm3 with very high sensitivity but it was less accurate in detecting lower calcification volumes (<8mm3). Carotid calcification was not different between symptomatic and asymptomatic patients. Study III. Echogenicity of the intima-media complex (IM-GSM), but not its thickness (IMT), was significantly decreased with increasing number of arterial systems affected by atherosclerosis. IM-GSM was lower in patients with prior myocardial infarction and stroke. Study IV. Carotid plaque echogenicity evaluated by US could predict future cerebrovascular events in patients with asymptomatic, relative risk RR 2.72 (95% CI, 1.86 to 3.96), and recurrent symptoms in symptomatic patients, RR 2.97 (95% CI, 1.85-4.78). Conclusion: Plaques located in the contralateral to symptomatic arteries have similar features as symptomatic side and more vulnerable than asymptomatic arteries. Carotid ultrasound could accurately detect larger but not smaller carotid plaque calcification volumes (<8 mm3). Low IM- GSM could identify patients with multi-system atherosclerosis disease, suggesting a better marker for determining systemic atherosclerosis disease burden compared to conventional IMT. Finally, carotid plaque echogenicity predicts future cerebrovascular events in patients with symptomatic and asymptomatic carotid stenosis.
