Signal in the Noise? The Effect of Non-Invasive Brain Stimulation on Contrast Perception

A longstanding question in studies of cortical stimulation has been how does stimulation affect brain functioning and cognition, and what are its mechanisms of action. Brain stimulation has been traditionally seen either as a disrupting intervention or as a procedure to enhance cortical excitability and promote improvement in various modality from motor to visual performance. In vision, several hypotheses have been proposed and many experimental paradigms have been used to study how transcranial magnetic stimulation (TMS) and direct current stimulation, particularly transcranial random noise stimulation (tRNS) affect visual discrimination. Psychophysical paradigms are particularly useful to measure visual performance, whereby a stimulus is progressively changed from easy to difficult to perceive it, and accuracy threshold can be measured by titrating the stimulus discriminability. Stimuli that vary in contrast are typically used to study low-level visual functions and it is well known that neurons within the early visual areas in the brain, and primarily V1, are tuned to stimuli involved in contrast discrimination. Here we used an orientation discrimination task to study changes in contrast detection by varying stimulus contrast across different levels (Experiment 1, Chapter 2). We used neuro-navigated single-pulse TMS at different intensities to determine whether behavioral response changed linearly as a function of stimulus discriminability independently of TMSintensity, or whether TMS affected behavior depending on TMS intensity and contrast level. Moreover, we tested whether TMS had an effect selective for the field contralateral to stimulation or whether effects could be seen across the entire visual field. Single pulse TMS was delivered to left V1 while participants performed a 2-alternative forced choice orientation discrimination (OD) of one of two Gabor patches presented on either side of fixation at 5 contrast levels and 4 TMS intensities. Participants' performance on OD increased at all contrast levels in the right visual field (contralateral to stimulation) at 80% of phosphene thresholds (PT, individually measured at baseline). Furthermore, when TMS was delivered at 60% of PT, we found improved performance in the right visual field that was selective for the medium contrast, while performance increased at the highest contrast irrespective of TMS intensity, in the field ipsilateral to stimulation, thus both visual fields were affected by TMS, albeit differently. Since the improvement effects might be explained as the result of added noise to the system that paradoxically improves performance for justbelow threshold stimuli (middle contrasts), in Experiments 1 and 2 (Chapter 3) we used transcranial random noise stimulation, a neuromodulation procedure known to enhance cortical excitability when delivered at high frequencies, to further test the hypothesis that brain stimulation might work through a mechanism of stochastic resonance, whereby adding noise to a nonlinear system, the brain in our case, might paradoxically promote better performance by enhancing stimulus discriminability. This might happen only for selective stimulus intensities and stimulation strength. Based on previous successful work, we tested contrast discrimination changes as a function of four different tRNS low intensity levels of stimulation, and we found a decrease in performance selective for the condition with subthreshold stimuli and at .750 mA stimulation intensity. This result might indicate that low intensity stimulation is not enough to promote enhancement of stimuli under the stochastic mechanism effect, thereby suggesting that higher ranges of stimulation are necessary to create the optimal conditions for improvement.

Využití molekulárních markerů pro studium genetické diverzity u vybraných zástupců Dracaena

Variability in the genetic information DNA tracking individuals carry is easy to detect using molecular markers. In the thesis we examined related Dracaena species. For the study of genetic diversity in the genus Dracaena are used mainly noncoding regions of cpDNA, spacer trnH -- psbA, regions trnL -- trnF and trnS -- trnG -- trnG, which are more variable than coding regions. Also used in this work were coding regions of matK and rbcL. In the genus Dracaena belongs xerophytic species that are characterized by typical shaped treetop. 14 representatives of species were used in thesis growing in tropical regions of the African continent and adjacent islands and the southeastern part of the Arabian peninsula. They are monocotyledonous trees with atypical abilities of secondary thickness of trunk, which I find interesting. The massive trunk is very strong and there is potencial to used it for wood. Trees are very significant for their red plant sap which flowing from demaged trunk. The sap is very precious resource which is used in many areas of industry, for example pharmacy, traditional medicine, dye making etc. Determination of genetic affinity was based on an amplification of cpDNA template of individual Dracaena samples with primers for the studied regions. Data was obtained and evaluated by Multiple alignment program ClustalX and BioEdit after their sequencing. Evalueted data was used to create dendograms affinity. According the resulting phylogenetic tree we find out similarities and identified relationship of the monitored species of the genus Dracaena. The main purpose of research was to get answers to understand phylogenetic relationship between group of Dracaena forestry used trees. The thesis was made in cooperation of Department of Forest Botany, Dendrology and Geobiocoenology, Faculty of Forestry and Wood Technology of Mendel University in Brno.

La phylogénie moléculaire du genre nord-américain Eurybia (Asteraceae : Astereae) et ses proches parents (Oreostemma, Herrickia, Triniteurybia)

Eurybia et ses proches parents Oreostemma, Herrickia et Triniteurybia sont appelés le grade des eurybioïdes. Comprenant 31 espèces vivaces, ce grade appartient au clade Nord-américain de la tribu des Astereae. Les analyses moléculaires antérieures ont montré que ce groupe est à la fois paraphylétique aux Machaerantherinae et un groupe frère aux Symphyotrichinae. Les relations infragénériques partiellement résolues et faiblement supportées empêchent d’approfondir l'histoire évolutive des groupes et ce, particulièrement dans le genre principal Eurybia. Le but de cette étude est de reconstruire les relations phylogénétiques au sein des eurybioïdes autant par l'inclusion de toutes les espèces du grade que par l’utilisation de différents types de régions et de méthodes d'inférence phylogénétique. Cette étude présente des phylogénies basées sur l'ADN ribosomal nucléaire (ITS, ETS), de l'ADN chloroplastique (trnL-F, trnS-G, trnC-ycf6) et d’un locus du génome nucléaire à faible nombre de copie (CNGC4). Les données sont analysées séparément et combinées à l’aide des approches de parcimonie, bayesienne et de maximum de vraisemblance. Les données ADNnr n’ont pas permis de résoudre les relations entre les espèces polyploïdes des Eurybia. Les analyses combinées avec des loci d’ADNnr et d’ADNnr+cp ont donc été limitées à des diploïdes. Les analyses combinées ont montré une meilleure résolution et un meilleur support que les analyses séparées. La topologie de l’ADNnr+cp était la mieux résolue et supportée. La relation phylogénétique de genres appartenant au grade des eurybioïdes est comme suit : Oreostemma (Herrickia s.str. (Herrickia kingii (Eurybia (Triniteurybia - Machaerantherinae)))). Basé sur la topologie combinée de l’ADNnr+cp, nous avons effectué des analyses de biogéographie à l’aide des logiciels DIVA et LaGrange. Ces analyses ont révélé une première radiation des eurybioïdes dans l’Ouest de l’Amérique du Nord, suivi de deux migrations indépendantes dans l’Est de l’Amérique du Nord chez les Eurybia. Due au relatif manque de variabilité de l’ADNnr, l’ADNcp et CNGC4, où le triage de lignés incomplet était dominant, l'origine du grade est interprétée comme récente, possiblement du Pliocène. The present study provides phylogenies based on nuclear ribosomal DNA (ITS, ETS), chloroplastic DNA (trnL-F, trnS-G, trnC-ycf6), and a low-copy nuclear locus (CNGC4), in separate and combined datasets analyzed using maximum parsimony, Bayesian and maximum likelihood approaches. In a separate analysis of the nrDNA dataset, the relationships of polyploids in Eurybia proved to be impossible to resolve. The nrDNA and nr+cpDNA combined analyses therefore were restricted to diploids. The combined analyses provided greater resolution and support than separate analyses. The nr+cpDNA phylogeny was the best resolved and supported. The phylogenetic relationship of genera belonging to the eurybioid grade is as follows: Oreostemma (Herrickia s.str. (Herrickia kingii (Eurybia (Triniteurybia – Machaerantherinae)))). Based on the nr+ cpDNA combined topology, we performed biogeographical analyses using DIVA and LaGrange. These analyses revealed an initial radiation of the eurybioids in western North America, with two independent migrations to eastern North America within Eurybia. MODULAZIONE DELL'AROUSAL MEDIANTE LA STIMOLAZIONE ELETTRICA TRANSCRANICA A FREQUENZE RANDOM / AROUSAL MODULATION BY RANDOM NOISE TRANSCRANIAL ELECTRICAL STIMULATION

Il lavoro di tesi si è focalizzato sullo studio dell’arousal come indice psicofisiologico di attivazione e sull’applicazione della metodica di stimolazione elettrica transcranica (tES) non invasiva con lo scopo di modulare tale indice. L’obiettivo è stato quello di indagare se, applicando la tES, fosse possibile migliorare la performance di soggetti giovani sani in compiti di tipo cognitivo. Il progetto di ricerca si è sviluppato in 2 studi principali per un totale di 4 esperimenti. Tali studi hanno previsto l’acquisizione e la successiva analisi sia di dati comportamentali (tempi di reazione, accuratezza), che di indici psicofisiologici (conduttanza cutanea, diametro pupillare). I risultati hanno evidenziato che è possibile modulare l’arousal con dei “bursts” di stimolazione elettrica transcranica, somministrati in concomitanza di stimoli salienti per il soggetto. Tale modulazione si è manifestata con una riduzione dei tempi di reazione ed un contemporaneo aumento della risposta di conduttanza cutanea. Questi dati supportano la possibilità di utilizzare questo protocollo in pazienti con difficoltà di attenzione o altri problemi cognitivi per aumentare l’efficacia di interventi di riabilitazione. / The thesis analyzed the role of the arousal as a psychophysiological index of activation, and the application of non-invasive transcranial electrical stimulation (tES) technique with the aim to modulate this index. In this work we investigated if the application of tES could increase the performance of healthy young subjects during cognitive tasks. The thesis is based on 2 main studies for a total of 4 experiments with the recording of behavioural (reaction times, accuracy) and psychophysiological (skin conductance, pupil diameter) indeces. The results showed that it is possible to modulate arousal with bursts of tES, administered during the presentation of salient stimuli for the subject. This modulation resulted in a reduction of reaction times and an increase of the skin conductance response. These data support the possibility to use this protocol of stimulation with patients with attentional and other cognitive deficits in a rehabilitative context.
