Eliminace negativního vlivu tepelného stresu u brojlerových kuřat pomocí rostlinných aditiv

The aim of the diploma thesis was to evaluated the effect of supplementation plant additive based on extracts, from turmeric (Curcuma) and baical skullcap (Scutellaria baicalensis) on broiler chickens exposed to heat stress at the end of fattening. The effect of plant additive on performance parameters, meat quality, lymphatic organ weight, biochemical blood profile, pro and anti-inflammatory cytokine levels, heat shock proteins, leukocyte counts, apparent ileal amino acid digestibility, metabolizable energy and chicken behavior were evaluated. Plant additive fed from the 21st day of chickens age significantly improved (P<0.05) feed conversion ratio at optimal temperatures and significantly increased (P<0.05) body weight gain at elevated temperatures (27–29 ° C). The addition of plant additive has significantly increased (P<0.05) metabolizable energy as well as the apparent ileal digestibility of the amino acids lysine, threonine, arginine, histidine, phenylalanine, valine, leucine, isoleucine, tyrosine, serine, glycine, aspartic and glutamic acid and alanine. Significantly lower (P<0.05) manifestations of thermoregulatory behavior (wings lifting and open beaks) in the experimental group with plant extracts were found in the ethological observation. Results of diploma thesis showed that the addition of a mixture of turmeric and baical scullcap extracts can be used to reduce the negative effects of heat stress on the performance of broiler chickens.

This aim of task deals with study of heat dissipation mechanisms and the description of physical phenomena, which is accompanied by non-stationary measurement of thermal characteristics by the method "hot-wire method". In particular, we observe the coefficient of thermal conductivity and its dependence on various variables such as the temperature of the measured sample, its moisture state, the volume of the sample and its porosity. The above mentioned findings are used for the invention of the measuring device of a nonstationary gauge, which is based on regular heating and is dedicated to measuring the thermal conductivity coefficient and the heat capacity by the "hot-wire method" method. In the last part of the thesis is verified functionality of the proposed measuring device, the suitability of the created algorithm for the processing of the measured data and the evaluation of the results was verified. The reproducibility of the measurements was verified and the measured results were compared with the measurement methods, which are commonly used. the influence of humidity on the coefficient of thermal conductivity.

This diploma thesis deals with the problems of heat transfer on printed circuit boards. The first part consists of a theoretical analysis of the principles of conduction of heat in different environments, comparing the properties of common type’s base materials for PCBs in terms of thermal properties and focuses on heat transfer from the chip. In the following part they are general information about the program ANSYS ® Workbench ™. Next part consists of the basic designs of PCBs and their improving. PCBs thermal properties for different configurations are verified with calculations, simulations and practical measurements. In the last part there are created design recommendations for PCBs design based on the comparison of the results of initial proposals and proposals to improvements.

For thermal and hydraulic calculations a program named HTRI Xchanger Suite® has been used. The thesis includes a brief description of heat exchangers, description of the used program, calculation and selection of input data, solving and conclusions. It contains also a layout of the exchanger.

This diploma thesis discusses fouling processes and mechanisms of industrial heat exchangers, with focus on shell side fouling of heat transfer area by flue gases, and eventually taking fouling into account, or more precisely its modeling and predicting by calculation. Thermal-hydraulic design of heat exchanger is focused mainly on shell side, as flue gases are dominant process medium in terms of fouling and heat transfer. Based on available operational data of evaluated industrial heat exchanger, specific process has been developed to determine by calculation, or more precisely predict change of fouling factor throughout operational period of heat exchanger. Usage and various options of this specific calculation technique are being discussed in conclusion, whether there is a potential to use this technique regarding more complex operational inputs or prediction of fouling process for different operational regime of heat exchanger.

Abstract Regulation of transcription by extracytoplasmic-function (ECF) sigma factors of RNA polymerase is an efficient way of cell adaptation to diverse environmental stresses. Amino acid-producing gram-positive bacterium Corynebacterium glutamicum codes for seven sigma factors: the primary sigma factor SigA, the primary-like sigma factor SigB and five ECF stress- responsive sigma factors (SigC, SigD, SigE, SigH and SigM). The sigH gene encoding SigH sigma factor is located in a gene cluster together with the rshA gene, encoding the anti-sigma factor of SigH. Anti-sigma factors bind to their cognate sigma factors and inhibit their transcriptional activity. Under the stress conditions the binding is released allowing the sigma factors to bind to the RNAP core enzyme. In this thesis, regulation of expression of genes encoding the most important ECF sigma factor SigH and its anti-sigma factor RshA as well as genes belonging to the SigH-regulon were mainly studied. The transcriptional analysis of the sigH-rshA operon revealed four housekeeping promoters of the sigH gene and one SigH-dependent promoter of the rshA gene.
