Development of a dual Fries-Claisen rearrangement strategy

Synthetic approaches towards linear fused carbon rings have been developed over the last decades due to their high interest as compounds with potential antibiotic activity and as organic electronics. The application of a novel iterative route towards linear fused carbon ring systems could provide a versatile new mode of access to complex substituted ring systems. Studies have previously demonstrated the use of an allylation-double Claisen-RCM sequence to build fused carbon rings (C). In this thesis we described our work to expand the use of this methodology by developing milder reaction conditions that could be applied to a wider range of compounds. This novel methodology was applied towards the synthesis of benzodifuran compounds (D). Our work was extended by developing, for the first time, a mixed Fries-Claisen rearrangement strategy which furnishes template (F), a key intermediate in the synthesis of pyranonaphthoquinones (G). Our work culminated in a new approach to the pyranonaphthoquinone natural products eleutherin (H) and isoeleutherin (I). [Figures missing from abstract].

Detection and Quantitation of Tetracycline Antibiotics in Agricultural Swine Wastes

The spread of tetracyclines through agricultural systems is causing the present bacteria to develop antibiotic resistance. The spread of this bacteria, as well as the tetracycline antibiotics in the environment is dangerous because these antibiotics pose health hazards for humans. The overuse of antibiotics, which are added to livestock feed, results in the antibiotics being released into the environment via animal feces. In this research, we have attempted to design an analytical method to isolate antibiotics from agricultural wastes with subsequent detection using liquid chromatography and mass spectrometry (LC-MS). The antibiotics investigated in this study were tetracycline, chlortetracycline, and oxytetracycline. The analytical procedure involves mixing the agricultural samples with an organic solvent, such as methanol, which solubilizes these antibiotics. Next, samples are centrifuged to remove solid particulates. A polymeric weak cation cartridge was used to concentrate and separate the antibiotics from the unwanted organic chemical compounds found in the samples. The antibiotics were released with methanol with small amounts of acid and then detected and quantified using LC-MS and high performance liquid chromatography with a photodiode array detector (HPLC-PDA).

Nanofibras poliméricas carregadas com oxitetraciclina para tratamento de alveolite seca: estudo em ratos / Oxytetracycline-loaded polymeric nanofibers for the treatment of dry socket: a study in rats

A alveolite seca é uma das complicações pós-operatórias mais comuns e sintomáticas na exodontia, porém, até o momento não possui um protocolo de tratamento definido. A oxitetraciclina (OTC) é um antibiótico de amplo espectro utilizado no tratamento de múltiplas infecções. Entretanto, nos últimos anos as tetraciclinas de primeira geração, tais como a OTC, têm sido evitadas devido ao surgimento de microrganismos resistentes. Afortunadamente, nos últimos anos tem se observado uma tendência contrária graças às medidas de controle no uso de antibióticos. Sistemas de liberação controlada podem evitar o desenvolvimento de resistência bacteriana e diminuir o risco de efeitos adversos. Este estudo teve como objetivo preparar uma nanofibra polimérica de policaprolactona e oxitetraciclina (PCL/OTC), e ainda avaliar suas propriedades físicas e biológicas in vitro e in vivo, visando seu futuro uso no tratamento da alveolite seca. As nanofibras poliméricas carregadas com OTC foram preparadas pelo sistema de eletrofiação, caracterizadas através da microscopia eletrônica de varredura (MEV) e avaliadas sobre seu perfil de liberação do fármaco. A atividade antibacteriana da nanofibra sobre um biofilme misto de Porphyromonas gingivalis, Prevotella intermédia, Fusobacterium nucleatum, Eikenella corrodens, Streptococcus sanguis e Aggregatibacter actinomycetemcomitans foi analisada mediante a contagem das unidades formadoras de colônias viáveis após os tempos de contato de 1, 4, 24, 30 e 48 h. O processo de reparo alveolar induzido pela nanofibra PCL/OTC foi avaliado de forma histomorfológica e histomorfométrica após o tratamento da alveolite seca em molares de ratos, e comparado ao tratamento com nanofibras de PCL pura e Alvogyl®, 7, 14 e 21 dias após a exodontia. Finalmente, foi realizada a identificação e contagem de osteoclastos através da marcação da fosfatase ácida resistente ao tartarato (TRAP), nos mesmos tempos de avaliação. As nanofibras PCL/OTC apresentaram orientação randômica em camadas e morfologia multiforme. O perfil de liberação do sistema apresentou um efeito de explosão nas primeiras 8 horas, seguido por um período de liberação lenta e prolongada da OTC. A nanofibra PCL/OTC (132 ?g de OTC/mL) reduziu mais de 50% das colônias do biofilme nas primeiras horas de liberação, seguido por um período de manutenção em que a porcentagem de colônias bacterianas permaneceu baixa. O grupo tratado com PCL/OTC apresentou a maior porcentagem de tecido conjuntivo no 7º dia (p<0,05) e maior porcentagem de osso neoformado no 14º e 21º dias após a exodontia (p<0,05), em comparação aos outros grupos. O tratamento de alveolite seca com a fibra PCL/OTC permitiu a regeneração tecidual do alvéolo quase em sua totalidade até o 21º dia, sendo que a cronologia de regeneração tecidual com este material foi mais rápida do que com a fibra de PCL pura, e esta última mais rápida que do que o controle positivo Alvogyl®. O grupo PCL/OTC apresentou uma quantidade menor (p<0,05) de osteoclastos por área em relação aos outros grupos nos três tempos experimentais. O perfil de liberação prolongando de OTC e as propriedades biológicas relevantes apresentadas pela nanofibra PCL/OTC, sugerem que esta poderia ser considerada uma alternativa para o tratamento da alveolite seca. / Dry socket is one of the most common and symptomatic postoperative complications in tooth extraction, however, to date, it does not have an established treatment protocol. Oxytetracycline (OTC) is a broad-spectrum antibiotic, employed in the treatment of multiple infections. Nevertheless, in the last few years, first-generation tetracyclines such as OTC, have been avoided due to the development of resistant microorganisms. Fortunately, in recent years there has been a contrary trend thanks to control measures in the use of antibiotics. Controlled release systems can prevent the development of bacterial resistance and decrease the risk of adverse effects. This study aimed to prepare a polymeric nanofiber of polycaprolactone and oxytetracycline (PCL/OTC), and to evaluate its physical and biological properties in vitro and in vivo, aiming its use in the treatment of dry socket at the future. The OTC-charged polymeric nanofibers were prepared by electrospinning, and characterized by scanning electron microscopy (SEM) and drug release profile evaluation. The antibacterial activity of the nanofiber over a mixed biofilm containing Porphyromonas gingivalis, Prevotella intermedia, Fusobacterium nucleatum, Eikenella corrodens, Streptococcus sanguis, and Aggregatibacter actinomycetemcomitans was analyzed by counting the viable colony-forming units after 1, 4, 24, 30 and 48 h of contact. The alveolar repair process induced by the PCL/OTC nanofiber was evaluated histomorphologically and histomorphometrically after the treatment of dry socket in rat molars, and compared to the treatment with plain PCL nanofibers and AlvogylTM, 7, 14 and 21 days after extraction. Finally, the identification and counting of osteoclasts were performed through the tartrate resistant acid phosphatase (TRAP) staining, at the same evaluation time points. PCL/OTC nanofibers presented random orientation in layers and multiform morphology. The release profile of the PCL/OTC system showed a burst effect within the first 8 hours, followed by a prolonged slow release period. The PCL/OTC nanofiber at a concentration of 132 ?g OTC/mL reduced more than 50% of the biofilm colonies in the first few hours of release, followed by a maintenance period in which the percentage of bacterial colonies remained low. The PCL/OTC group presented the highest percentage of connective tissue on the 7th day (p<0.05), and of newly formed bone on the 14th and 21st days (p<0.05) after dental extraction, compared to the other groups. The treatment of dry socket with the PCL/OTC fiber allowed almost complete tissue regeneration up to the 21st day, and the chronology of tissue regeneration with this material was faster than with the plain PCL fiber, and the latter was more rapid than the positive control AlvogylTM. The PCL/OTC group presented a lower amount (p<0.05) of osteoclasts per area than the other two groups in the three experimental periods. The prolonged release profile of OTC and the relevant biological properties presented by the PCL/OTC nanofibers suggest that these may be considered as an alternative treatment of dry socket.

IIntrodução A utilização excessiva de antimicrobianos na medicina humana, veterinária e agricultura resultou no aparecimento da resistência bacteriana. Este fenômeno gera problemas de saúde pública que podem resultar na reemergência de doenças infecciosas. O uso de antibióticos, principalmente de forma profilática, tornou-se prática na aquicultura, como ocorre no Brasil, onde a regulamentação para o uso de medicamentos veterinários é ineficiente. Além disso, há evidências da transferência de organismos resistentes para humanos por meio do consumo de produtos de origem animal, quando, durante a fase de criação e produção destes foram administrados antibióticos. Objetivo Pesquisar a ocorrência de genes de resistência a antibióticos em filés de tilápia comercializados em supermercados do município de São Paulo SP. Material e Métodos Foram coletadas 10 amostras de filé de tilápia e realizada a pesquisa de coliformes termotolerantes como indicadores das condições higiênico-sanitárias do alimento. Em seguida, as amostras foram inoculadas em Caldo Lúria 0,5 por cento e o DNA total das bactérias cultivadas nesse meio foi extraído por meio de choque térmico para pesquisa de genes de resistência aos antibióticos -lactâmicos e tetraciclinas pela PCR. Os genes identificados pela PCR foram confirmados pelo sequenciamento. Resultados Em 100 por cento das amostras analisadas o resultado para coliformes termotolerantes foi < 3 NMP.g-1. Na pesquisa de genes de resistência a -lactâmicos, o gene blaOXY-5 foi detectado em 90 por cento das amostras, blaTEM-1b em 20 por cento , blaLEN-16 em 10 por cento , blaSHV-28 em 10 por cento , blaKPC-2 em 20 por cento , blaMOX-6 em 10 por cento e blaCphA em 60 por cento . Os genes de resistência a tetraciclinas identificados foram tetB em 10 por cento das amostras, tetC em 20 por cento , tetD em 80 por cento , tetE em 50 por cento , tetG em 60 por cento , tetO e tetS em 10 por cento cada e tetW em 20 por cento . The use of antibiotics, mainly prophylactically, has become a practice in aquaculture, including Brazil, where the legislation for the use of veterinary drugs is inefficient. Furthermore, there is evidence of transmission of resistant organisms to humans through consumption of animal products, especially when the antibiotics have been administered during the raising and production of these animals. Objective To search for the occurrence of antibiotics resistance genes in tilapia fillets commercialized in supermarkets in São Paulo city - SP. Material and Methods 10 tilapia fillet samples were collected and submitted to fecal coliform search as an indicator of the sanitary conditions of the food. Then, the samples were inoculated into Luria broth 0,5 per cent and the total DNA was extracted from growing bacteria by thermal shock for the search of resistance genes to -lactam and tetracyclines antibiotics by PCR. The genes identified by PCR were confirmed by sequencing. Results In 100 per cent of samples the result was < 3 NMP.g-1 for fecal coliform organisms. In the study of -lactam resistance genes, blaOXY-5 gene was detected in 90 per cent of samples, blaTEM-1b in 20 per cent , blaLEN-16 in 10 per cent , blaSHV-28 in 10 per cent , blaKPC-2 in 20 per cent , blaMOX-6 in 10 per cent and blaCphA in 60 per cent . The tetracyclines resistance genes identified were tetB in 10 per cent of samples, tetC in 20 per cent , tetD in 80 per cent , tetE in 50 per cent , tetG in 60 per cent , tetO and tetS in 10 per cent each and tetW in 20 per cent . Utilização de eletrodos compósitos na análise voltamétrica de tetraciclinas em amostras biológicas e ambientais / The use of composite electrode in voltammetric analysis of tetracyclines in biological and environmental samples

Neste trabalho, um eletrodo compósito à base de grafite e poliuretana (EGPU) 60% (grafite, m/m) foi avaliado quanto ao seu potencial para determinação dos antibióticos da família das tetraciclinas em amostras de água natural, de urina bovina e humana, e em amostras de leite bovino e humano. Visando desenvolver métodos simples, de baixo custo e com baixa geração de resíduos, o eletrodo foi usado sem modificação, para atender a estes propósitos e considerando os níveis de concentração das tetraciclinas nas amostras biológicas. A otimização dos parâmetros em voltametria de pulso diferencial (DPV) foi realizada para a determinação dos antibióticos tetraciclina (TC), oxitetraciclina (OTC) e clortetraciclina (CTC) em tampão fosfato 0,10 mol L-1 pH 2,3, permitindo atingir limites de detecção de 2,6 &micro;mol L-1, 1,7 &micro;mol L-1 e 0,82 &micro;mol L-1, respectivamente, sem a necessidade de renovação da superfície, entre as medidas sucessivas. Durante as determinações de TC e OTC em amostras de água natural e purificada, foram observadas recuperações entre 92,6% e 100%, após etapa de pré-concentração. As amostras de urina foram acidificadas, em seguida centrifugadas para a remoção de partículas sólidas e proteínas. A TC e OTC foram determinadas diretamente, na amostra de urina tratada e diluída em tampão fosfato 0,10 mol L-1 pH 2,3. Durante a determinação de TC e OTC nas amostras de urina, após o preparo da amostra, foram obtidas recuperações entre 96% e 100%. As amostras de leite bovino e humano foram intencionalmente contaminadas com TC. Após preparo das amostras, estas foram submetidas à extração, usando cartucho para extração em fase sólida (SPE). Após eluição e retomada do analito retido no cartucho, a quantidade de TC presente nas amostras foi determinada pelo método de adição de padrão, usando DPV, com EGPU 60% (grafite, m/m). Foram obtidas recuperações entre 76% e 103% para as amostras de leite bovino e recuperações entre 91% e 100 % para as amostras de leite humano, que mostram que o eletrodo proposto pode ser usado neste tipo de determinação com as características postuladas acima. / A bare graphite-polyurethane composite electrode 60% (graphite, m/m) (GPUE) was evaluated in the determination of tetracycline (TC) class antibiotics in natural water as well as, in human and bovine urine and in milk samples. Thus, a DPV procedure was optimized for the determination of tetracycline, oxytetracycline (OTC) and chlortetracycline (CTC) antibiotics in phosphate buffer 0.10 mol L-1 pH 2.3, resulting in LOD of 2.6 &micro;mol L-1, 1.7 &micro;mol L-1 and 0.82 &micro;mol L-1, respectively. During the determination of TC and OTC in the water samples, recoveries between 92.6 and 100% were found, after pre-concentration stage. The urine samples were submitted to acidification followed by centrifugation to remove solids and proteins. During the determination of TC and OTC in urine samples, after pre-treatment of urine, recoveries between 96 and 100% were found. The composite electrode was also evaluated in the determination of TC in the bovine and human milk samples. During the determination of TC in milk, recoveries between 76 and 103% were found for bovine milk and recoveries between 91 and 100% were found for human milk after pre-concentration stage, showing the feasibility of the procedure.

Regulation of the putative ykkCD riboswitch by tetracycline and related antibiotics in Bacillus subtilis

Multi-drug resistance among bacterial pathogens can be mediated by a number of mechanisms, including multidrug efflux pumps. One such pump in Bacillus spp. is ykkCD, a heterodimer of the SMR family consisting of C and D subunits. Previous studies suggest that the expression of ykkCD is controlled by a putative riboswitch and that the antibiotic tetracycline binds to the riboswitch in vitro. Additional studies have shown that two derivatives of tetracycline also bind to the putative riboswitch. These findings now need to be validated by an in vivo study. In this study, the effects that tetracycline and its commercially available derivatives—doxycycline, minocycline, anhydrotetracycline, and oxytetracycline—have on the expression levels of the ykkCD gene in Bacillus subtilis were explored. The level of ykkCD expression was quantified using two different methods: (1) ykkCD protein levels was determined using a ykkCD RNA--galactosidase reporter gene construct and (2) ykkCD mRNA levels was quantified by quantitative RT-PCR. Although the findings from method (1) were inconclusive, upregulation was observed for tetracycline and minocycline, in agreement with the results of the previous binding studies. / Department of Chemistry

Comparison of the neuroprotective potential of theanine and minocycline

Stroke is one of the most common causes of disability and death worldwide. The most commonly experienced stroke in the clinical setting is focal ischaemia in which the middle cerebral artery (MCA) is occluded and leads to a complex series of various pathophysiological pathways that ultimately lead to neuronal cell death. Several studies have been conducted on various therapeutic agents in the search for a neuroprotective drug and various animal models have been used to carry out this research. While theanine, a component of green tea and minocycline, a tetracycline antibiotic, have been shown to possess some neuroprotective properties, the mechanisms by which these two agents carry out these effects still remains unclear. The objectives of this study were to investigate the mechanisms by which these drugs carry out these neuroprotective effects and their neuroprotective ability in a MCA occlusion model of focal ischaemia. Ischaemia leads to oxidative stress due to the imbalance of free radicals and the endogenous antioxidant defence system. An antioxidant assay using the stable 2, 2-diphenyl-1-picrylhydrazyl (DPPH●) radical was used to assess the antiradical properties of each drug. It was found that minocycline showed superior antioxidant activity in vitro when compared to theanine. Further studies on the drugs‟ ability to attenuate the Fenton reaction (in which iron catalyses the formation of reactive species) were elucidated using electrochemical analysis, UV/VIS studies, ferrozine and ferritin assays. It was found that minocycline, in contrast to theanine, was able to bind to iron ions and thus potentially prevent the participation of iron in metal catalysed radical reaction. The antioxidant activity of both drugs was further investigated by assessing their effect on cyanide-induced superoxide generation and quinolinic acid (QA)-induced lipid peroxidation (LP). Experimental evidence shows that both drugs had no significant effect on the generation of superoxide in vitro and that there was a significant decrease in LP for minocycline in vitro and theanine in vivo. The metal binding and antioxidant properties were postulated to be a possible mechanism through which these agents reduced lipid peroxidation. A study was conducted to determine the effects of the drugs on the biosynthesis of the neurotoxin, QA and it was found that minocycline increases the levels of holoenzyme activity of tryptophan-2, 3-dioxygenase (TDO) in vitro and that theanine reduces the levels of the same enzyme in vivo after treatment for 10 days. TDO is the enzyme that converts tryptophan to other products that enable enzymatic activity to change it to QA. Minocycline was thought to bring about this effect as it has been shown from preceding experimental studies that it is an effective reducing agent. Theanine on the other hand is hypothesised to bring about a reduction in holoenzyme activity by changing the binding of tryptophan to the enzyme or affecting the radicals that participate in the enzymatic degradation of tryptophan. A focal ischaemic model of stroke was induced by occluding the MCA. Histological examination of the hippocampus post -ischaemia shows a reduction in the size of the infarct after pre-treatment with minocycline only. A further study into the effects of the drugs on the generation of superoxide and on the levels of the endogenous glutathione after a stroke was carried out. Pre-treatment of the animals with either theanine or minocycline showed no significant effects on the generation of the radical species or of the endogenous antioxidant which ruled out these as a mechanism of neuroprotection of both drugs, post-ischaemia.The findings of this study provide novel information on the possible mechanisms by which both theanine and minocycline act to bring about neuroprotection. In particular in this study, pre-treatment with minocycline has shown promise in the focal ischaemic model of stroke.
