Com relação aos búfalos domésticos (Bubalus bubalis), pouco ou nada se conhece sobre estes parasitas. Este estudo teve como objetivo avaliar por meio de métodos parasitológicos (exame direto do parasita no microscópio), sorológicos (RIFI – reação de imunofluorescência indireta) e moleculares (PCR – reação em cadeia pela polimerase e sequenciamento) a presença de tripanosomatídeos em búfalos. Amostras de sangue e suabe conjuntival foram coletadas de 100 búfalos da Fazenda São Francisco, Andradina, SP e de 10 búfalos de Selvíria, MS. Dos 110 búfalos, 60,9% (67/110) apresentaram hemoflagelares tripanossomatídeos nos esfregaços sanguíneos e 90% (99/110) deles apresentaram anticorpos anti-Leishmania pela RIFI. O DNA destes parasitas foi detectado em 29,09% (32/110) dos búfalos pela técnica de PCR, sendo, 26,36% (29/110) na PCR do sangue e 3,6% (4/110) na PCR do suabe conjuntival. Das amostras positivas para PCR, sete foram enviadas para o sequenciamento gênico, e uma delas foi identificada como Leishmania infantum e três como Trypanosoma theileri, ambas as amostras apresentaram 100% de similaridade com as respectivas espécies após comparação pelo GenBank. Este estudo relata o primeiro registro de infecção por L. infantum em B. bubalis na região de Andradina, SP, e a presença de búfalos infectados por espécies de T. theileri, evidenciando a necessidade de mais estudos sobre tripanosomatídeos em búfalos. / Trypanosomatides are flagellate protozoan parasites that in ruminants include those belonging to the family Trypanosomatidae, with the genera Trypanosoma and Leishmania, but with little or nothing information about these parasite infections in domestic water buffalo (Bubalus bubalis). Tthis study was assessed through the parasitological (direct examination of the parasite under microscope), serological (IFAT – immunofluorescence antibody test), and molecular tests (PCR from blood and conjunctival swab and genic sequencing) to detect possible trypanosomatides in buffalo herds from Andradina, SP and Selvíria, MS. Thus, 60.9% (67/110) buffaloes presented trypanosomatid parasites in the blood smears, 90% (99/110) had anti-Leishmania antibodies by IFAT. This study reports for the first time the presence of parasite L. infantum in B. bubalis from Andradina, SP, and highlights the presence of buffaloes infected with T. theileri, and indicates the need for more studies on trypanosomatid parasites in buffaloes.

Qualitative nichtlineare Zeitreihenanalyse mit Anwendung auf das Problem der Polbewegung

In der nichtlinearen Datenreihenanalyse hat sich seit etwa 10 Jahren eine Monte-Carlo-Testmethode etabliert, die Theiler-surrogatmethode, mit Hilfe derer entschieden werden kann, ob eine Datenreihe nichtlinearen Ursprungs sei. Diese Methode wird kritisiert, modifiziert und verallgemeinert. Das, was Theiler untersuchen will braucht andere Surrogatmethoden, die hier konstruiert werden. Und das, was Theiler untersucht braucht gar keine Monte-Carlo-Methoden. Mit Hilfe des in der Arbeit eingeführten Begriffs des Phasensignals werden Testmöglichkeiten dargelegt und Beziehungen zwischen den nichtlinearen Eigenschaften der Zeitreihe und deren Phasenspektrum erforscht. Das Phasensignal wird aus dem Phasenspektrum der Zeitreihe hergeleitet und registriert außerordentliche Geschehnisse im Zeitbereich sowie Phasenkopplungen im Frequenzbereich. <br /> <br /> Die gewonnenen Erkenntnisse werden auf das Problem der Polbewegung angewendet. Die Hypothese einer nichtlinearen Beziehung zwischen der atmosphärischen Erregung und der Polbewegung wird untersucht. Eine nichtlineare Behandlung wird nicht für nötig gehalten. / In the nonlinear data analysis there is a popular Monte Carlo Test method due to Theiler (it was established about 10 years ago), the Theiler surrogate method, which tests whether a time series is of a linear origin. This method is being criticized, modified and generalized in this thesis. What Theiler wants to test, needs other surrogate methods, which are constructed here. And what Theiler really tests, does not need Monte Carlo methods. With the help of the concept of the phase signal, that is introduced here, other test options are possible. The phase signal helps also in investigating the relations between the nonlinear characteristics of the time series and their phase spectrum. The phase signal is derived from the phase spectrum of the time series and registers extraordinary events in the time domain as well as phase couplings in the frequency domain. <br /> <br /> These theoretical approches are applied to the problem of polar motion. The hypothesis of a nonlinear relationship between the atmospheric excitation and the pole movement is examined. A nonlinear treatment is not considered necessary.

Rio de Janeiro, RJ, Brasil. / Para testar a atividade biológica do interferon beta (INF-β) no tratamento da esclerose múltipla (EM), é importante que se tenha um modelo animal. Como a infecção pelo vírus da encefalomielite murina de Theiler (do inglês TMEV – Theiler’s Murine Encephalomyelitis Vírus) é capaz de evoluir para uma lesão desmielinizantesimilar a da EM em humanos, este estudo se propõe a estabelecer os parâmetros para avaliar a infecção do TMEV em culturas de células de rim de hamster neonato (do inglês BHK-21– Baby hamster kidney cells). Para tanto foi necessário adaptar a amostra viral TMEV BeAn à cultura BHK-21, estabelecer um ensaio de RT-PCR e padronizar um PCR em tempo real.Também foi construido um vetor plasmidial contendo o gen L* do TMEV para expressãotransitoria em células HEK-293-T e esta construção plasmidial foi utilizada para obtenção de um soro policlonal anti-L* utilizando a metodologia de imunização genética. Como resultados foram obtidos estoques virais de células BHK-21 infectadas pelo TMEV e parte destes estoques foram avaliados quanto à presença de moléculas genômica TMEV, indicativa de replicação viral, por ensaios de RT-PCR e quantificação por PCR em tempo real. Asregiões do genoma do TMEV 3A3B e L* foram aquelas que forneceram melhores resultadosnesta avaliação genômica quantitativa, que deverá ser aplicada para todos os estoques TMEVBHK-21 que foram obtidos. O vetor plasmidial de expressão células HEK-293-T pcDNA4His/Max contendo o gene L* expressou com sucesso transitoriamente esta proteína heteróloga, porém não foi capaz de induzir a formação de anticorpos policlonais anti-L* em coelhos, através da técnica de imunização genética. / In order to evaluate the biologic activity of the therapeutic drug beta interferon to multiple sclerosis, an adequate animal model is necessary.inthis aspect the infection of murine encephalomyelitis virus can evolve to desmielinization that is very similar to human’s multiple sclerosis, the proposal of this study was the establishment of parameters to evaluate the TMEV infection in baby hamster kidney cells-BHK-21. Toward that objective was adapted the TMEV BeAn prototype sample to BHK-21 and also was established a RT-PCR and a real time PCR. Additionally, it was constructed a plasmid vector containing the L* gene of TMEV, for the expression in HEK-293-T cells. This plasmid vector was evaluated in the capacity to produce anti-L* antibodies by genetic immunization. It was possible to obtain stocks of BHK-21 TMEV infected and some of these contents were evaluated as of the quantity of genomic molecules of TMEV as an indicative of viral replication, by RT-PCR and real time PCR. The TMEV genomic regions 3A3B and L*provided best results in the quantitative evaluation. In thefuture, the real time PCR could be used to evaluate all the stocks that were produced. The plasmidial vector pcDNA4His/Max containing the L* gene was able to transiently express the L* protein, but unfortunately, it was not able to induce anti-L* in rabbits.
