Cellulosans tillgänglighet och reaktivitet är nyckelparametrar vid framställning av regenererad cellulosa och cellulosaderivat. Det är välkänt att på grund av cellulosans kristallina struktur är tillgängligheten begränsad för lösningsmedel och olika reagens. Till exempel kan en inhomogen substitution av hydroxylgrupperna i cellulosakedjan resultera i cellulosaderivat av sämre kvalitet. Baserat på detta har en del av arbetet i denna studie fokuserat på att förbättra cellulosans tillgänglighet och reaktivitet genom att studera effekten av olika enzymatiska behandlingar med monokomponent endoglukanaser. Resultaten visar att närvaron av en cellulosabindande domän fyller en viktig funktion för att öka cellulosans reaktivitet, men strukturen för den katalytiska domänen visade sig ha den största inverkan på cellulosans tillgänglighet. I kompletterande studier har även effekten av en mekanisk förbehandling i kombination med enzymatisk behandling utvärderats. Kombinationen av förbehandlingarna resulterade i en positiv effekt, cellulosans reaktivitet kunde ökas i större omfattning. I dag används huvudsakligen dissolvingmassor som råvara vid framställning av cellulosaregenerat och cellulosaderivat. Kraven för dessa s.k. specialmassor är högt cellulosainnehåll samt lågt hemicellulosa- respektive lignininnehåll. På grund av dessa specifika krav är produktionskostnaderna för dessa massor högre än konventionella sulfatmassor. Den andra delen av studien har därför fokuserat på möjligheten att använda dessa sulfatmassor som dissolvingmassa. Det har visats att kombinationen av enzymatiska behandlingar med monokomponent endoglukanas och xylanas följt av ett alkaliskt steg kan resultera i massor där kraven uppfylls med avseende på cellulosans reaktivitet, och cellulosa- respektive hemicellulosainnehåll. / The accessibility and reactivity of cellulose are key parameters on the manufacturing of cellulose derivatives and regenerated cellulose. It is well known that, due to the crystalline structure of cellulose, the accessibility of solvents and reagents is limited. For instance, an inhomogeneous substitution of the hydroxyl groups of the cellulose chain might lead to the production of derivatives of low quality. As a consequence, part of this work has focused on improving the accessibility and reactivity on cellulose by studying the effect of different monocomponent endoglucanases. It has been demonstrated that the presence of the cellulose-binding domain plays an important role on the enhancement of cellulose reactivity; however, the structure of the catalytic domain has been showed to have the highest influence on this parameter. Furthermore, the influence of mechanical treatment prior to enzymatic treatment has been examined. The combination of pretreatments showed a positive effect enhancing to a larger extent the cellulose reactivity. Currently, dissolving-grade pulps are commonly used for the production of cellulose derivatives and regenerated cellulose. The requirements for these so-called “special pulps” are a high cellulose content and a low hemicelluloses and lignin content. As a result of these specific demands, the production costs of these pulps are higher than those of common kraft pulps. The second part of this work, therefore, has been focused on the study on the viability of converting kraft pulps into dissolving pulps. It has been demonstrated that the combination of enzymatic treatments using a monocomponent endoglucanase and a xylanase together with the addition of an alkaline step could fulfil the requirements of a commercial dissolving pulp in terms of cellulose reactivity and cellulose and hemicellulose content. / QC 20101117

Therapist Utilization of Evidence-Based Treatment Monitoring

The early identification of non-responding psychotherapy clients and reducing their treatment failure rates are the principal functions of Clinical Support Tools (CST). Nearly two decades of patient-focused research have produced several "evidence-based treatment monitoring" (EBTM) practices, that measure individual patients' responses to theory-guided treatments and alert therapists of clients who are at risk of eventual treatment failure. Clinical Support Tools are a quality management program that bundle several EBTM practices, and have been shown to improve outcomes in failing clients (Shimokawa, Lambert & Smart, 2010). Appropriately, EBTM has generated significant interest as it expands notions of what constitutes evidence-based practice with non-responding patients (APA, 2006). There is a practical need to disseminate Clinical Support Tools to a wider audience of clinicians and practice environments. The current study was designed to advance understanding of CST mechanisms by providing detailed, qualitative data that demonstrate how CST procedures are utilized in routine practice. Eleven doctoral-level psychologists experienced in using Clinical Support Tools at a university counseling center were interviewed about their use of Clinical Support Tools with a recent non-responding client and about their general experiences with past non-responding clients. Clinicians' responses were categorized as Actions, Decisions, or Attitudes, and were subjected to inductive, thematic content analysis. Results were interpreted to indicate which CST functions were active or inactive in the therapists' routine care. Findings indicated that therapists utilized CST resources to monitor patient status, to identify problems that may have explained therapy non-response, and to initiate corrective interventions. Although it was clear that therapists used the CST signal-alarm system to initiate a problem assessment and corrective intervention, it was less clear whether therapists used CST's to determine significance of client change or to determine the client's prognosis. This observation needs to be confirmed through further investigation. Future research that quantifies CST utilization and investigates implementation-outcome relationships is recommended. Finally, practical avenues for increasing the influence and prevalence of EBTM practices in behavioral healthcare are discussed.

Towards application of activated carbon treatment for pharmaceutical removal in municipal wastewater

Many pharmaceuticals are found in municipal wastewater effluents due to their persistence in the human body as well as in conventional wastewater treatment processes. This discharge to the environment can lead to adverse effects in aquatic species, such as feminization of male fish. During the past decade, these findings have spawned investigations and research into suitable treatment technologies that could severely limit the discharge. Adsorption onto activated carbon has been identified as one of the two main technologies for implementation of (future) full-scale treatment. Recent research has put a closer focus on adsorption with powdered activated carbon (PAC) than on granular activated carbon (GAC). Studies where both methods are compared in parallel operation are thus still scarce and such evaluation in pilot-scale was therefore a primary objective of this thesis. Furthermore, recirculation of PAC can be used to optimize the treatment regarding the carbon consumption. Such a setup was evaluated as a separate treatment stage to comply with Swedish wastewater convention. Additionally, variation of a set of process parameters was evaluated. During successive operation at three different wastewater treatment plants an overall pharmaceutical removal of 95% could consistently be achieved with both methods. Furthermore, treatment with GAC was sensitive to a degraded effluent quality, which severely reduced the hydraulic capacity. Both treatment methods showed efficient removal of previously highlighted substances, such as carbamazepine and diclofenac, however in general a lower adsorption capacity was observed for GAC. By varying the input of process parameters, such as the continuously added dose or the contact time, during PAC treatment, a responsive change of the pharmaceutical removal could be achieved. An exploration of Shona traditional healers' conceptualisation and treatment of mental illness

Thesis (Ph.D. (Psychology)) -- University of Limpopo, 2016 / After the World Health Organization (WHO) declared 2010 a decade of indigenous knowledge systems, there has been an increase in the studies on the role of African traditional healers in describing mental illness and its treatment. Studies have pointed to the relevance of traditional healing in primary health care in many developing countries. The aim of the present study was to explore the conceptualisation and treatment of mental illness by Zezuru Shona traditional healers in Goromonzi District in Zimbabwe. A qualitative research design, and in particular, the phenomenological method was used in the present study. Ten Shona traditional healers were selected through purposive and snowball sampling and requested to participate in the study. Data was collected through in-depth interviews and analysed using Hycner’s phenomenological explicitation process. Five major themes related to the traiditonal healers’ conceptualisation on mental illness were identified. These are: a). Types/characteristics of mental illness; b). Causes of mental illness; c). Diagnosis and treatment of mental illness; d). Challenges faced by traditional healers; and, e). Facilitating factors in the work of traditional healers. The study further revealed that there are a number of illnesses that are are not mental illness but could be closely associated with mental illness. These are epilepsy (pfari), locking (kusungwa) and sexual dysfunctions. Culture was found to play a central role in the traditional healers’ conceptualisation and treatment of mental illness. In this regard, spirituality was found to be a critical factor in the work of traditional healers in diagnosing, interpreting, treating and preventing mental illness and the associated conditions of ill health. These results were discussed in the context of indigenous knowledge systems and mental illness. The study is concluded by recommending the need to have a holistic understanding of mental illness and associated conditions.

Complex-Restricted Repetitive Patterns of Vocal Behavior of Individuals with High Functioning Autism: An Innovative Intervention

Repetitive verbal patterns of speech are a trait associated with high-functioning autism (HFA). For some, this higher-order restricted, repetitive behavior impedes learning, social opportunities, and access to work environments. Despite emerging motivation for establishing social relationships, some individuals with HFA lack the behavioral prerequisites to establish meaningful relationships. The purpose of this study is to demonstrate the effectiveness of teaching interaction to decrease higher-order verbal restricted and repetitive behaviors (RRBs) for four adults with HFA. Through a multiple baseline design across participants, individuals were exposed to a function-based intervention (i.e., teaching interaction) that systematically taught and reinforced alternative communicative behaviors while interrupting repetitive speech with specific feedback. Teaching interaction effectively reduced RRBs and increased alternative conversation for all four participants. Three of the four participants elected to participate in post-intervention maintenance sessions that occurred in individualized naturalistic settings. Their conversation behaviors maintained with one participant receiving one booster session.

Efficacy of F10 against amphibian chytrid fungus / Maria Susanna de Jong

Outbreaks of pathogens that threaten both human and nature have increased in recent years. Infectious and transmittable diseases, such as chytridiomycosis, which is caused by the emerging pathogen Batrachochytrium dendrobatidis, has been identified as one of the most important drivers of the current declines in amphibian numbers. This pathogen has spread globally and is not only responsible for the declines in amphibian population numbers, but also for the extinction of species in several countries. As part of the Amphibian Conservation Action Plan, the IUCN recommended ex situ breeding of amphibian species to try and stem the global loss of amphibian species. Due to chytridiomycosis being one of the most eminent threats for amphibians, it poses an additional threat for the ex situ breeding plan. There is thus a need for safe and effective measures to treat chytridiomycosis, especially in breeding programs for endangered species. F10 (Health and Hygiene) is a veterinary antiseptic that has shown to be 100% effective in killing B. Dendrobatidis in vitro. Before any chemical treatment can be applied the efficacy and toxicity of F10 has to be determined to establish if F10 can be effectively applied across different amphibian species and across different life stages. We propose to develop a treatment protocol for F10 for the effective treatment of amphibian chytridiomycosis by challenging juveniles of Amietophrynus gutturalis with B. dendrobatidis and subsequently treating the infection with a proposed concentration of F10. The survival of B. dendrobatidis zoospores was also determined in the presence of F10. The results obtained showed survival of tadpoles at a 1:10,000 concentration of F10 for 30min, and juveniles at a concentration of 1:2000 for 15 min. Furthermore the in vitro tests showed that the B. dendrobatidis zoospores died after 10 min at a 1:10,000 concentration and 30 min at a 1:15,000 concentration. The successful treatment of tadpoles as well as juveniles will increase any species chance for survival, especially when treating tadpoles as the pathogen will then be eradicated before the tadpole metamorphoses and reaches the disease-susceptible life stage. By establishing a partnership between the industry, academic and zoo/wildlife communities we hope to maximise the likelihood of implementing this program in the future and thus ensuring long term sustainability. / MSc (Environmental Sciences), North-West University, Potchefstroom Campus, 2015
