Fundação Carlos Chagas Filho de Amparo a Pesquisa do Estado do Rio de Janeiro / A aplicabilidade de um método selecionado de medição indireta de vitelogenina (Vtg) em plasma sanguíneo de peixe, baseado na quantificação de fosfato álcali-lábil (alkali-labile phosphate-ALP) para acessar estrogenicidade em água, foi investigada na presente tese. O método foi originalmente desenvolvido para a espécie de peixe Carassius carassius (Carpa cruciana) e aplicado pela primeira vez na espécie Oreochromis niloticus (Tilápia do Nilo) no presente estudo. Com o objetivo de acessar a sensibilidade do método, em uma primeira etapa da investigação foram realizados estudos laboratoriais com soluções estoques de 17-ethinylestradiol (EE2), 17-estradiol (E2), e estrona (E1). Os efeitos destes hormônios foram investigados com base tanto na concentração quanto na carga, utilizando-se para tanto, unidades experimentais com volumes distintos (2 L e 130 L). Após a validação do método de ALP, a estrogenicidade foi avaliada nas seguintes águas contaminadas: (i) afluente e efluente de uma grande estação de tratamento de esgotos convencional (ETE) e de uma estação descentralizada de tratamento de esgoto de pequeno porte (Ecossistema Engenheirado-DEE); (ii) água superficial (SW) e água subterrânea (GW) coletadas em uma área de brejo contaminada com gasolina; (iii) água de uma lagoa urbana (LRF) da cidade do Rio de Janeiro, com alta densidade populacional e descarte clandestino de esgoto. Na segunda etapa foram analisados em microalgas os efeitos (outros que não disrupção endócrina) causados pelos hormônios EE2, E2 e E1. Os hormônios foram testados individualmente e em misturas, em culturas individuais e combinada (S+) das espécies de microalgas unicelulares P. subcapitata e D. subspicatus. Com base nos níveis de ALP para a espécie de peixe e no EC50 para as espécies de algas, os resultados mostraram que o EE2 e o E2 causaram disrupção endócrina superior e foram mais tóxicos do que o E1 para peixes e microalgas respectivamente. Quando em misturas (E+) de concentrações equivalentes (EE2:E2:E1), os estrogênios resultaram em efeito aditivo para as espécies O. niloticus e P. subcapitata, e menos que aditivo para D. subspicatus e cultivo misto de algas (S+). Culturas contendo ambas as espécies de algas (S+) por um longo período de exposição (96 h) resultaram na atenuação dos efeitos tóxicos causados pela exposição, tanto individual (EE2, E2 ou E1), quanto na mistura (E+) dos estrogênios, medidos em termos de EC50 (T0h 0,07; 0,09; 0,18; e 0,06 g mL-1; e T96h 1,29; 1,87; 5,58; e 4,61 g mL-1, respectivamente). O DEE apresentou uma maior eficiência na remoção dos disrutores endócrinos do que a ETE convencional. Foi detectada estrogenicidade em amostras da LRF, e de água SW e GW em área brejosa contaminada com gasolina. Os resultados dos ensaios sugerem que as interações (efeitos aditivos ou menos que aditivo) causadas pela mistura dos estrogênios assim como, as interações entre as espécies de algas afetaram o resultado final dos ensaios ecotoxicológicos. Um fator raramente abordado em estudos ecotoxicológicos que foi destacado na presente tese refere-se à importância de considerar não somente a concentração e a dosagem, mas também a carga aplicada e o volume das unidades experimentais. Devido à boa sensibilidade do O. niloticus quando exposto às concentrações relativamente baixas dos estrogênios, a combinação do método de ALP com os biomarcadores auxiliares (particularmente MN) pode ser um protocolo adequado para a detecção de estogenicidade e genotoxicidade respectivamente em diferentes ambiente aquáticos contaminados, como parte de um programa de monitoramento ambiental / The applicability of one selected method for indirect measurement of vitellogenin (Vtg) in fish plasma based on the quantification of alkali-labile phosphates (ALP method) to assess estrogenicity in water was investigated. The ALP method applied in this investigation was originally developed with Carassius carassius (Crucian carps). This thesis describes the first attempt of using this method with Oreochromis niloticus (Nile tilapia). In a first part of the investigation, laboratory studies were conducted with water spiked with 17-ethinylestradiol (EE2), 17-estradiol (E2), and estrone (E1) in order to assess the method sensitivity. The effects of these estrogens were investigated on the basis of both load and concentration, using experimental units with two different volumes (2 L and 130 L). After validation of the method, the estrogenicity of the following contaminated waters was assessed: (i) affluent and effluent of one large conventional municipal wastewater treatment plant (WWTP) and one small decentralized wastewater treatment plant (Decentralized Engineered Ecosystem-DEE); (ii) surface water (SW) and groundwater (GW) obtained from a gasoline-contaminated marshland; (iii) samples from a urban lagoon (LRF) located in Rio de Janeiro city with high density population and clandestine sewage discharge. An additional goal of the thesis was to assess the effect (other than endocrine disruption) caused by EE2, E2 and E1 to microalgae. Assays with single and mixed estrogens and single and combined cultures (S+) of the green microalgae P. subcapitata and D. subspicatus were carried out. The results have shown that EE2 and E2 were more estrogenic and toxic than E1 to the fishes and to the microalgae respectively. Mixed solutions of estrogens (E+) in equal proportions (EE2:E2:E1) resulted in additive effect on O. niloticus and P. subcapitata and less-than-additive effect on D. subspicatus and S+ measured as ALP (for fish) and EC50 (for microalgae). Combined cultivation of both algae species and longer exposure time (96 h) resulted in attenuation of the toxic effects caused by single (EE2, E2 or E1) and mixed (E+) estrogens according to EC50 (T0h 0.07, 0.09, 0.18, and 0.06 g mL-1; and T96h 1.29, 1.87, 5.58, and 4.61 g mL-1, respectively). The decentralized engineered ecosystem was more efficient than the conventional WWTP regarding estrogenicity removal from the final effluent. Estrogenicity was detected in some samples of the LRF and the SW and GW of the gasoline-contaminated marshland. Therefore, the investigations suggested that interactions (additive and less-than additive effect) take place when different estrogens are present in the water environment and interactions also occur between algae species, which affect the final toxicity. Additionally, the study highlighted the importance of taking into account not only concentration and dose regime but also the mass load and therefore, the volume used in the experimental units, which are rarely addressed in ecotoxicity assays. Considering the good sensitivity of O. niloticus exposed to relatively low concentrations of estrogens, the combination of the ALP method with auxiliary biomarkers (particularly MN) can be a suitable protocol for estrogenicity and genotoxicity detection in different contaminated waters as part of water environmental monitoring programs

Diversité, distribution et activité des diazotrophes planctoniques en mer Méditerranée / Diversity, distribution and activity of planktonic diazotrophs in the Mediterranean seaLe Moal, Morgane 10 December 2010 (has links)
La mer Méditerranée est une des zones océaniques les plus oligotrophes de la planète pour laquelle la fixation biologique d’azote a été proposée comme jouant un rôle important dans les flux de carbone et d’azote. Comme les organismes planctoniques potentiellement responsables de cette activité n’y ont jamais été étudiés, les objectifs de cette thèse étaient de caractériser la diversité (abondance et richesse spécifique), la distribution spatio-temporelle, et les facteurs de contrôle des diazotrophes, en s’axant particulièrement sur les cyanobactéries unicellulaires (UCYN). Pour cela, des hybridations in situ (TSAFISH)sur différentes fractions de taille en combinaison avec des comptages microscopiques et des analyses phylogénétiques sur les gènes 16S ADNr et nifH ont été réalisées, à la fois lors d’un cycle saisonnier à la station côtière SOMLIT de Marseille, et lors du transect Méditerranéen BOUM. Alors que les cyanobactéries filamenteuses Richelia intra cellularis et Trichodesmium sp. ont été détectées seulement ponctuellement dans le temps et dans l’espace, en très faible concentration (0.02 filament.ml-1), la communauté des cyanobactéries diazotrophes était dominée à 99,9% par du picoplancton hybridé avec la sonde Nitro821, spécifique aux UCYN. Ces cellules, de petite (0.7-1.5 μm) ou de grande (2.5-3.2 μm)taille, ont été retrouvées en faible concentration (1-6 cellule.ml-1) dans toute la Méditerranée et tout au long de l’année, à l’exception de l’été 2006 lorsque la concentration des petites cellules a atteint 5300 cellule.ml-1 au cours d’un évènement exceptionnel de pollution atmosphérique urbaine. Des efflorescences similaire sont été observées suite à des enrichissements en poussières sahariennes à une station côtière en Corse et au large au centre de la Méditerranée, simultanément avec des augmentations de l’activité de fixation d’azote.L’affiliation des petites cellules aux UCYN-A a été confirmée par phylogénie dans les eaux du large de la Méditerranée occidentale. Les librairies de clones nifH de la fraction de taille picoplanctonique étaient dominées par des séquences d’α-protéobactéries appartenant à un nouveau groupe marin de Bradyrhizobium. D’autres groupes de rhizobia et des γ-protéobactéries ont été détectés ponctuellement.Alors que l’absence de Trichodesmium sp. au large pourrait être liée aux faibles concentrations en phosphate et en fer, les facteurs inhibant le développement des UCYN-B et –C restent inconnus. Lesdiazotrophes picoplanctoniques (Bradyrhizobium, rhizobia, UCYN-A) pourraient avoir développé des stratégies spécifiques, telles que des associations avec des eucaryotes ou des particules inertes, et/ou la capacité de photosynthèse, pour acquérir le carbone nécessaire au soutien du processus de la diazotrophie / The Mediterranean Sea is one of the most oligotrophic marine areas on earth where nitrogenfixation has been formally believed to play an important role in carbon and nitrogen fluxes. Although thisview is under debate, the diazotrophs responsible for this activity have still not been investigated. The aimsof this PhD were to characterise the diversity (abundance and species richness) and the spatio-temporaldistribution of diazotrophs, as well as factors controlling their development, with a particular focus onunicellular cyanobacteria. A combination of microscopic counts with size fractionated in situ hybridization(TSA-FISH), and 16S rDNA and nifH phylogenies were done, either over a year and a half seasonal cycle atthe coastal SOMLIT station off Marseilles, and across the entire Mediterranean Sea during the BOUMtransect. Low concentrations of diazotrophic cyanobacteria were detected and this community wasdominated at 99.9% by picoplankton hybridized with Nitro821 probe, specific for unicellular diazotrophiccyanobacteria (UCYN). Among filamentous cyanobacteria, only 0.02 filament ml-1 of Richeliaintracellularis and Trichodesmium sp. were detected sporadically in time and space. Small (0.7-1.5 μm) andlarge (2.5-3.2 μm) Nitro821-targeted cells were recovered in low concentrations (1-6 cell ml-1) across theentire Mediterranean Sea and all the year long, except over a month period in summer 2006 whenconcentrations of small cells reached 5300 cell ml-1, during an exceptionally high urban pollution event.Similar blooms of small and large cells were reported after Saharan dust inputs off Corsica and at open Sea,simultaneously with increases in N2 fixation rates. The affiliation of the small Nitro821-targeted cells toUCYN-A was confirmed by 16S and nifH phylogenies offshore in the western Mediterranean Sea. Rhizobiasequences, including the ones of a new marine group of Bradyrhizobium, were dominating nifH clonelibraries from picoplanktonic size fractions. A few sequences of γ-proteobacteria were also detected incentral Mediterranean Sea. While low phosphate and iron concentrations could explain the absence ofTrichodesmium sp. offshore, the factors that prevent the development of UCYN-B and C remain unknown.It is proposed that the dominating Mediterranean picoplankters (Bradyrhizobium, rhizobia, UCYN-A)probably developed specific strategies, such as associations with protists or particles, and/or photosyntheticactivity, to acquire carbon for sustaining diazotrophy.

Malformation in different species of benthic diatoms in three herbicide polluted streams in southern SwedenSpångfors, Helena January 2017 (has links)
Målet med den här studien var att undersöka kiselalger och dess eventuella missbildningar i tre vattendrag i södra Sverige - Höje å, Skivarpsån och M42. Kiselalger används som bioindikator för vattenkvalitet i hela Europa, men de befintliga kiselalgsindexen visar inte eventuell förekomst av miljöfarliga ämnen. Det har dock visats att en förekomst > 1 % av missbildade kiselalger indikerar påverkan av miljöfarliga ämnen, såsom pesticider och tungmetaller. Denna studie är en av få som har undersökt kiselalgers missbildningar i vattendrag som är mer eller mindre påverkade av herbicider. Sex prov från varje vattendrag analyserades - kiselalger räknades och identifierades till artnivå och missbildningar dokumenterades och kategoriserades. Vattendragen delades in efter PTI (Pesticide Toxicity Index), där Höje å hade lägst PTI och ansågs minst påverkad av herbicider. Skivarpsån och M42 hade högre PTI, och ansågs därför ha en högre herbicidpåverkan. Det fanns en signifikant skillnad i missbildningsfrekvens vattendragen emellan, den kunde dock inte kopplas till PTI. Både Höje å och M42 hade > 1 % missbildningar. Skivarpsåns missbildningsfrekvens var < 1 % trots vattendragets relativt höga PTI. Det är möjligt att missbildningsfrekvensen bättre hade reflekterat PTI om herbicidprovtagningen skett någon månad tidigare, då herbicidhalter kan variera och kiselalger har visat sig kunna spegla ett vattendrags mående upp till tre månader bakåt i tiden. En annan förklaring till en varierande missbildningsfrekvens kan vara en lika varierande artsammansättning. Vissa arter är mindre “benägna” att missbildas än andra - ett prov som domineras av sådana skulle därför kunna innehålla få missbildningar trots eventuell miljögiftspåverkan. Det krävs dock ytterligare studier för att bättre förstå kiselalgers missbildningar i förhållande till herbicider.

Synthèse et évaluation biologique de nouveaux inhibiteurs de kinases : identification d‘inhibiteurs de kinases parasitaires / Synthesis and biological evaluation of new kinase inhibitors : identification of inhibitors of several parasite protein kinasesBendjeddou, Lyamin 14 October 2014 (has links)
La phosphorylation des protéines par les kinases est l’une plus importantes modification post-traductionnelle dans les processus cellulaires tels que la division, la différenciation, la prolifération et l’apoptose. Due à leur rôle clef, un dérèglement des protéines kinases peut entrainer de nombreuses pathologies proliférative telles que le cancer et non prolifératives telles que les maladies neurodégénératives. Le travail de thèse s’est construit autour de 2 séries d’inhibiteurs de protéine kinases comportant les noyaux imidazo[1,2-b]pyridazine et imidazo[4,5-b]pyridine. L’objectif est d’inhiber sélectivement les protéines kinases choisies, pour leurs implications dans les pathologies visées au laboratoire. Les imidazo[1,2-b]pyridazines ont été préparées pour identifier des inhibiteurs de CLK1 et DYRK1A, cibles potentielles dans la maladie d’Alzheimer. Parmi les imidazo[1,2-b]pyridazines synthétisées, plusieurs molécules se sont révélées particulièrement sélectives de DYRKs et CLKs, avec des IC50 < 100 nM. Une relation structure-activité basée sur la synthèse de 70 molécules, a permis de dégager des éléments structuraux de la sélectivité des molécules. L’évaluation des produits a également été portée sur les kinases de parasites. Il a ainsi été possible d’identifier quelques inhibiteurs actifs sur PfCLK1. La seconde partie de cette thèse avait pour objectif l’optimisation du protocole de synthèse imidazo[4,5-b]pyridines, analogue de la roscovitine. Des dérivés s’étaient révélés capables d’inhiber la formation de kystes, dans un modèle cellulaire de polykystose rénale. Une synthèse en sept étapes a conduit à plusieurs grammes d’imidazo[4,5-b]pyridine 3,5,7 trisubstitués, qui sont ainsi disponibles pour l’évaluation in vivo. / Phosphorylation by protein kinases is one of the most important post-translational modification in cellular processes such as division, differentiation, proliferation and apoptosis. Kinase deregulation is associated with numerous diseases such as cancer or neurodegenerative diseases. Imidazo[1,2-b]pyridazine and imidazo[4,5-b]pyridine were prepared to inhibit protein kinases involved in diseases targeted in the laboratory. The imidazo[1,2-b]pyridazines were synthesized to identify inhibitors of CLK1 and DYRK1A, potential targets in Alzheimer's disease. Among the imidazo[1,2-b]pyridazines synthesized, several molecules were found selective of DYRKs and CLKs, with IC50 < 100 nM. A structure-activity relationship based on the synthesis of 70 molecules, led to the identification of the structural bases of the selectivity. Products were also evaluated against parasite kinases. It was possible to identify some highly potent inhibitors on PfCLK1. The aim of second part of this thesis was to optimize the synthetic process to obtain imidazo[4,5-b]pyridines, which are close analogues of roscovitine. Derivatives had proved capable of inhibiting the formation of cysts in a cellular model of polycystic kidney disease. A seven-step synthesis has led to several grams of 3,5,7-trisubstituted imidazo[4,5-b]pyridine which is now available for evaluation in vivo.
