This diploma thesis deals with rebuild of manual drill-mill machine BF20L into CNC mill machine, 3D model of that and eventual control using PLC. All axes are powered by stepper motors, with encoders, connected to control cards and industrial computer from company Beckhoff. For this control is made full switchboard, supplemented by electrical wiring diagram. Program control along with visualization is made in software TwinCAT 3. Drill-mill machine can be controlled manually or automatically by program.

The thesis deals with the development of systems for intelligent building, focused on wiring system, division of them and comparison of their differences. The advantages of these systems like comfort, energy reduction, etc. are also mentioned. Nowadays several standards of systems for intelligent building exist as KNX/EIB or LON, therefore thesis concerns to trade-off these standards too. Main part of the thesis is focused on system Egon made by company ABB where the system concept as well as bus modules are described that allow different functions, mutual connection of modules and their programming. Tutorial panel with ABB Ego-n wiring system, which will be used for study purpose, is designed in the practical part of the work. It includes scheme of the panel, electric linkage of components and photographs of constructed panel as documentation. In the final part of the work two examples of laboratory exercise for installation and setting are listed, including programming description.

The master thesis includes a literature search, which is focused on questions around panel measure instruments for measuring analog voltage signals. There is the block diagram and the concept of measuring voltage values designed from collected information and defined requirements to the device. In the next step a design of the circuit diagram is created and published and consequently transferred into PCB. The whole board has been fabricated, assembled and tested. The firmware development starts with the definition of basic functional blocks for using of internal elements of the device, which is followed by the implementation of the Modbus slave station service routine. The top part presents graphical user interface on the device.

The cue induced axonal nascent proteome and its translational control mechanisms in neural wiring

Axonal protein synthesis is rapidly regulated by extrinsic cues during neural wiring but the full landscape of proteomic changes and their translational control mechanisms remain unknown. The ability to investigate the nascent proteome on subcellular compartments has been hampered by the low sensitivity of existing methodology on quantity-limited samples combined with the difficulty of obtaining sufficient amounts of pure material. By combining pulsed Stable Isotope Labelling by Amino acids in Cell culture (pSILAC) with Single-Pot Solid-Phase-enhanced Sample Preparation (SP3), I have established an approach to characterize the nascent proteome from quantity-limited somaless retinal axons (~2μg) on an unparalleled rapid time-scale (5 min). The results show that a surprisingly large number of proteins (>350) is translated constitutively in axons, many of which are linked to neurological disease. Axons stimulated by different cues (Netrin-1, BDNF, Sema3A) each show a signature set of up/down newly synthesised protein (NSP) changes (>100) within 5 min. Remarkably, conversion of Netrin-1-induced responses from repulsion to attraction triggers opposite translational regulation for 73% of a common subset corresponding to >100 NSPs. Further, I show that pharmacological increase in cAMP, known to induce chemoattractive response, also leads to rapid and wide-scale remodelling of the nascent axonal proteome (~100 NSP changes). I find that the cAMP-elicited NSP changes underlie the attractive turning but are distinct from those induced by the physiological chemoattractant Netrin-1, suggesting that the same type of chemotropic response can be mediated by different protein synthesis-dependent mechanisms. Finally, I show that Sema3A, but not Slit1, triggers a physiological and non-canonical PERK-eIF2α-eIF2B signalling pathway required in neural wiring to elicit the rapid (< 15 min) local translation control of a specific subset of NSPs. Collectively my findings lead to the general conclusion that guidance molecules rapidly induce cue-specific remodelling of the nascent axonal proteome via distinct regulatory mechanisms.

Investigating wireless network deployment configurations for marginalized areas

In recent years, immense effort has been channelled towards the Information and Technological development of rural areas. To support this development, telecommunication networks have been deployed. The availability of these telecommunication networks is expected to improve the way people share ideas and communicate locally and globally, reducing limiting factors like distance through the use of the Internet. The major problem for these networks is that very few of them have managed to stay in operation over long periods of time. One of the major causes of this failure is the lack of proper monitoring and management as, in some cases, administrators are located far away from the network site. Other factors that contribute to the frequent failure of these networks are lack of proper infrastructure, lack of a constant power supply and other environmental issues. A telecommunication network was deployed for the people of Dwesa by the Siyakhula Living Lab project. During this research project, frequent visits were made to the site and network users were informally interviewed in order to gain insight into the network challenges. Based on the challenges, different network monitoring systems and other solutions were deployed on the network. This thesis analyses the problems encountered and presents possible and affordable solutions that were implemented on the network. This was done to improve the network‟s reliability, availability and manageability whilst exploring possible and practical ways in which the connectivity of the deployed telecommunication network can be maintained. This diploma thesis concerns with problematic of modern intelligent wiring systems in buildings, meanwhile presents their advantages against old classical wiring systems. Also deals with utilization and properties of modern components for regulation, switching and light controlling. The first part deals with light sources and possible ways how to control the power of individual luminary sources. The second part discuss about utilization of radio-frequency systems in new buildings and also in reconstructions. In the next part is described a possibility of bus system for controlling building illumination scope with focusing to control system Nikobus. The last part is concerned on technical-economical evaluation of intelligent wiring systems by various manufacturers according to specific requirements.

The thesis is focused on an analysis of mounting and cable laying fiber optic lines. The first chapter includes an analysis is made comparing parameters fiber and copper cabling, mainly in terms of access networks. This part specifies the various transmission media and access networks that are operated on them. Attention is focused on the transmission rate and its possible impact, provided the capacity and other important values. This section also demands site maintenance, service, or any failure on the routes of these networks. With this goal in the second chapter was created several designs networks that provide insight into possible solutions to the situation for emerging networks, and for upgrading existing networks, using both management and elements of optical and copper, or combinations thereof. Provide information from expert sources and verified directs the work of the future and to clarify further the eventual development of individual technologies. Most other space and offers the potential of optical technology, nature, and therefore the third chapter is fully devoted to only the transmission medium. At the beginning of this section is a theoretical analysis of possible ways of cable laying fiber optic cabling with a comparison of advantages and disadvantages. The most stable is an underground installation, which is costly in standard cases. As a result of these factors proved to be a possible solution using the underground installation of water pipeline shutdown. Therefore, the optical path designed, organized and implemented laying of fiber optic cable. By completing the inspection measurement and evaluation of the cost compared with other ways of installing fiber optic cable.
