Morfologická variabilita rodu bříza (Betula L.) ve vybraných oblastech ČR se zaměřením na tetraploidní zástupce / Morphological variability of birch genus (Betula L.) in chosen regions of the Czech Republic with a focus on tetraploid taxa

The aim of this work is to propose reliable method for distinguishing between diploid (2n) and tetraploid (4n) taxa of the genus Betula L., based on leaf measurements. These measurements are realized by common tools, such as ruler or protractor. Main advantage of these methods is that they can be applied even in the field. The further aim of work is to describe and evaluate genetic variability within tetraploid taxa, whose taxonomic relationships are still unclear and under discussions. For statistical analysis, 97 individuals from 6 localities within Šumava region were selected and 4 leaves from every individual were analysed. In total, 20 parameters were measured on every leaf (16 quantitative and 4 qualitative). Each individual was also analysed by flow cytometry method to determine real genome size (ploidy). Differences in selected parameters between individuals with different ploidy were statistically tested. Significant results were observed in 12 of 16 quantitative and 2 of 4 qualitative parameters. For real ploidy prediction, the classification function was designed, whose reliability was 96 % for the primary data. The reliability of classification function was verified on samples from three different regions within the Czech Republic and compared with functions suggested by other authors. Function designed in this work was able to correctly determine real ploidy in 89 %, which is the most of all compared functions. Genetic variability, especially within tetraploid taxa is mentioned in relation to other author's previous studies. Distinguishing between B. pubescens and B. carpatica was not possible using selected genetic markers. Genetic conformity of these taxa is also mentioned in publications of foreign authors, as contrasted to Czech literature, which generally distinguishes B. pubescens and B. carpatica as separated species.

This diploma thesis: Evaluation of qualitative and quantitative indicators in the range of genotypes Lonicera caerulea L. was prepared on fields of genofond Mendel University on experimental crops Žabčice School Farm. The work was focused on the evaluation of phenological, growth and harvest characteristics of different varieties of honeysuckle. From the growth characteristics the work was namely focused on characteristics such as the height of shrubs, bushes yardage, annual increment and flowering evaluation. Harvesting indicators were being monitored based on the average fruit weight, height and width of the fruit, the shape of the fruit, the total amount of the vitamin C presented in content of refractometric dry matter and the dry matter itself of the fruits. From phenological phases we paid the most attention to the date of sprouting, flowering time, ripening time and the length of the developed fruit shoots. The work results are supported by charts and tables of published results. The work also includes a description of each studied variety of honeysuckle accompanied with photo documnetation.

In this work, MC1R, ASIP and TBX3 gene were tested on a sample of 118 Hucul horse mares included in Genetic Resources of Animals in the Czech Republic. We want to determine the genetic structure of mares and to analyse phenotypic data compared to the genotype and to identify possible differences between it. Genetic analysis showed a solid state for all alleles (HW for ASIP P = 0.9360, for MC1R P = 0.1661 and for TBX3 P = 0.4444). The frequency of the allele was as follows: E (0.6780), A (0.5254), and (0.4746), d2 (0.4323), d1 (0.3542), e (0.3220) D (0.2135). The most common genotype was AaEed1d2 and AaEEd1d2. There were very few or no genotypes based on recessive homozygotes in the genes of basic coat colours in the population, we didn´t identify any individual with genotype AaEed1d1. We have publicised genotype dependence within the TBX3 gene with primitive markings, confirming the previous work of other. Alele D was always associated with the occurrence of primitive markings, but primitive markings occur even without allele D in coincidence with the d1 allele. The d2d2 genotype is associated with a phenotype without primitive markings, or with phenotype where we can´t say if the horse has primitive markings or not.

Superfamily Chalcidoidea (chalcid wasps) represent the most species-rich group of the Hymenoptera and includes 22 families. Family Torymidae is one of those families and it's monophyly is currently intensively discussed. This particular thesis aims at the tribe Microdontomerini (Torimidae: Toryminae). Species of this tribe are usually minute inconspicuous wasps (body length ranges from 2 to 4 mm). Recent progress in phylogenetics of chalcid wasps allowes us to formulate robust hypothesis of Microdontomerini evolution, as a component of broader study of Chalcidoidea phylogeny and classification. My approach combines molecular and morphological evidence to formulate the most plausible evolutionary scenario. Dataset of 35 mostly morphological characters for 33 taxa including all existing Microdontomerini genera and 10 outgroups has been scored. Almost all characters has been studied and documented using Hitachi S-3700N-VP scanning electron microscope. In 117 taxa, 5 nuclear genes segments (18S, 28SD2, 28SD3-5, EF1alfa, Wingless) and 3 mitochondrial genes segments (COI-LCOHCO, COI-JerryPat, CytB) were sequenced, to get together the final molecular dataset of 387 Kbp. This work is focused on the evaluation of productivity and growth and phenological phases in the planting of Cornelian cherry. The evaluation was conducted on the experimental plot ŠZP Žabčice. Refraction measurement and level of vitamin C were done in the laboratory of the Institute of Plant Breeding and Propagation. Obtained values were statistically processed and evaluated and were sorted into tables and graphs and subsequently discussed in the text of the thesis. Photographs were taken during observation. These values allow selection of the most suitable type of variety for cultivation and subsequent processing. Findings also allow selecting the variety with the largest fruit, variety with highest crop yields, varieties of fruits richest in vitamin C or variety with the largest bush growth. Jolico variety yields the largest single fruit while Vyšegorodský variety offers highest yield of crop. Highest content of vitamin C is found in Fruchtal variety. Vydubecký variety is suitable for a rapid wood growth. Cornelian cherry is a versatile plant that can find its application in landscaping either for its heavy and hard wood or for its fruit which tastes well and is nutritionally interesting. Plant's resistance to diseases and pests is also significant. Cornelian cherry is not yet very common as a fruit in our country, however; this can change not only due to better awareness but also due to new plant varieties.

This thesis on the topic : Evaluation of growth and harvest data of sweetberry honeysuckle (Lonicera caerulea L.) is focused on the evaluation of growth, harvest and phenological characteristics of different varieties of honeysuckle. In the thesis there were evaluated from the growth characteristics the height of shrubs, cubic capacity of shrubs, annual growth and evaluation of flowering. For harvesting parameters there were observed the height and width of fruits, weight of one fruit, fruit shapes, refractometric dry substance content, the content of vitamin C and the dry matter content in fruits. In phenological phases there were recorded the date of budding, flowering, ripening of the fruits, harvesting and loss of leaves. The thesis is supplemented with charts and tables of published results and with the description in the text part of the work. It also includes the description of some most commonly grown varietes. During the observation there was taken continuous photo documentation. On the base of the findings it is possible now to to select the variety with the largest fruits, the heaviest fruits, the largest content of vitamin C, refractive dry matter or total dry mater content in the fruits or with the largest cubic capacity growth of the shrub.

The aim of this thesis is to find out the growth and harvest features of the chosen set of the cornelian cherry. The summary of history, spread and typical features, that this fruity species distinguished, was given. It describes the chemical structure of fruits, the propagation of cornelian cherry and its cultivation. It also contains its use and processing in chosen products. The assortment of six cultivars ('Elegantní', 'Fruchtal', 'Jolico', 'Lukjanovský', 'Vydubecký', 'Vyšegorodský') was evaluated on the experimental and demonstration area of the institute SŽP Žabčice. The terms of phonological phases, the sizes of bush content, the size of fruity picking, the outer and inner features of fruits were found out. The results of individual measures were processed and statistically evaluated. The acquired values were arranged to charts and graphs and then described in detail in the textual part. The most suitable cultivar for cultivation and succeeding processing is possible to choose according to the acquired values.

When analyzing diversity is an emphasis on primary and secondary characters of diversity: age, gender, ethnicity, mental and physical abilities, marital status, religion, work experience. The work consists of two parts, theoretical and practical. The theoretical part defines concepts: diversity, equal opportunities, primary and secondary characters of diversity, acceptance of the importance of diversity as a value. The practical part deals with the implementation of diversity into practice on the basis of the analytical methods (analysis: workforce, documents and research positions) and draft incorporation into relevant strategic processes of the organization.
