Previous issue date: 2012-07-31 / Introduction: Heat shock proteins like mycobacterial HSP70 has a protective and anti-inflamatory activity in several animals models diseases, like arthritis and skin allograft. This protein can modulates both the innate and adaptative immune system.
Objective: The aim of this study was evaluate the effect of HSP70 from M. tuberculosis on development of pulmonary allergic reponse in a mice model.
Methods: Female BALBc mice with ovalbumin induced pulmonary disease received intraperitoneal inject or intranasal instillation of mycobacterium HSP70. Treg cell in lung tissue, inflammatory cells profiles and cytokines levels in broncalveolar lavage fluid was measured.
Results: HSP70 show an inhibitory effect on pulmonary allergic response on mice. Eosinophils, IL-5 and IFN- levels was reduced in broncoalveolar lavage fluid in animals that received intranasal instillation of mycobacterial HSP70.
Conclusion: Mycobacterial HSP70 can be efficiently used to treat airway allergic disorders. / Introdu??o: Algumas prote?nas de choque t?rmico, como a HSP 70 micobacteriana, apresentam propriedades anti-inflamat?rias e protetoras em modelos experimentais de doen?as como artrite e aloenxertos cut?neos. Essa prote?na pode modular tanto o sistema imune inato quanto o adaptativo.
Objetivo: O objetivo do presente estudo foi avaliar os efeitos da prote?na HSP70 de M. tuberculosis no desenvolvimento da resposta pulmonar al?rgica em um modelo experimental de asma al?rgica em camundongos.
M?todos: camundongos BALB/c f?meas submetidos a um protocolo de indu??o de asma por ovalbumina foram expostos a prote?na HSP70 de M. tuberculosis atrav?s de diferentes vias de administra??o (intraperitoneal e intranasal). Foram analisados par?metros inflamat?rios (leuc?citos e citocinas IFN, IL-4, IL-5 e IL-10) no fluido do lavado broncoalveolar.
Resultados: A administra??o de MtHSP70 intranasal reduziu significativamente o infiltrado inflamat?rio e os n?veis de IL-5 e IFN- no lavado broncoalveolar.
Conclus?o: A prote?na HSP70 micobacteriana pode apresentar potencial aplica??o no tratamento da asma al?rgica.
