Adaptive pursuit of harmony in times of crisis: Wang Yangming's (1472-1529) contribution to the syncretization of Chinese thought in the Ming Dynasty (1368-1644)

The pursuit of harmony has always been a great concern of Chinese thinkers. In this process, especially prior to the Ming dynasty, a significant “borrowing” of their basic philosophical elements and their mutually syncretic metamorphosis was a common practice among three religious communities, particularly disseminated during times of crisis. The work of Wang Yangming proved to be an epitome of this philosophical “collaboration”, capable of producing new synthetic teachings that directly or indirectly linked two or more polarized teachings. He succeeded in modifying the existing Buddhist idea of inherited Buddha Nature to be now understood as an innate insight, while also promoting the practice of meditation, as a clear example of Chan and Daoist influence. Wang Yangming is probably best known for his emphasis on the simultaneity of the two functions – knowledge and action, viewed as a reinterpretation of non-Confucian ideas in a new Neo-Confucian framework.

Aging and the built environment: observations from three town centres in Surrey British Columbia

Canada’s population is aging rapidly, such that by 2031 it is estimated that one quarter of Canada’s population will be aged 65 or older (Statistics Canada December 15, 2006). It is argued that urban environments are often not adequately designed for older people and can impair their ability to live independently into their elder years (Harris 2004). Proper planning can help people age successfully in their community. This research practicum assesses three town centres within the City of Surrey to determine whether they are agefriendly based on the perceptions of residents over the age of 65. Results from the research found that areas in need of improvement included public transit, the walking environment, clustering various land uses, parks amenities, and home assistance, among other things. The practicum concludes with recommendations of areas for further study.

Mcl-1 in breast cancer: regulation by the EGF receptor family and role in cell survival and drug resistance

Myeloid Cell Leukemia-1 (Mcl-1) is a widely expressed anti-apoptotic member of the Bcl-2 family that is elevated in a variety of tumour types including breast cancer. Mcl-1 promotes tumour cell survival and drug resistance and was a mechanism of resistance to first generation Bcl-2 family inhibitors. To determine the significance of Mcl-1 in breast cancer, we evaluated the regulation of Mcl-1 by signalling via the epidermal growth factor receptors (EGFRs). EGFR signalling is frequently deregulated in breast cancer and leads to increased proliferation and survival of tumour cells. We aimed to determine whether Mcl-1 is a critical downstream effector of this pathway and therefore an important therapeutic target. We found that Mcl-1 protein and messenger RNA levels were rapidly induced upon stimulation of breast cancer cells with epidermal growth factor. This induction was blocked by inhibitors of the Ras/Raf/Mek/Erk signalling cascade and was dependent upon activation of the transcription factor Elk-1. We found Mcl-1 to be an essential survival protein, as targeted knock-down with small interfering RNA alone was sufficient to induce apoptosis. Mcl-1 may be critical for the survival advantage conferred by EGFR activation, as prevention of its up-regulation by Mek/Erk inhibitors significantly reduced the drug resistance conferred by EGF. Furthermore, we found a correlation between phosphorylated Elk-1 and Mcl-1 protein levels in breast tumour samples. Therefore, we conclude that Mcl-1 is an important downstream effector of survival and drug resistance mediated by elevated EGF signalling, making it an important therapeutic target in breast cancer.

Mechanisms of lipid droplet formation by conjugated linoleic acid (CLA) isomers and its effects on cell viability

The putative peroxisome proliferator-activated receptor (PPAR) α ligand, conjugated linoleic acid (CLA) induced cytoplasmic lipid droplet (LD) formation in H4IIE rat hepatoma cells. Currently, the mechanism(s) by which CLA isomers affects hepatic LD formation is unclear. We have investigated the role of PPARα and fatty acid (FA) activation in the regulation of hepatic LD formation induced by CLA isomers [cis-9,trans-11 (c9,t11), trans-10,cis-12 (t10,c12)] and linoleic acid (LA) in an in vitro model of lipid accumulation. Dose response of c9,t11 and t10,c12 CLA isomers as well as LA in quiescent H4IIE cells was assessed by Oil Red O staining and subsequent quantification after 24 hours. LD formation was induced by the CLA isomers similar to LA in a dose-dependent manner. However, treatment with the acyl CoA synthetase (ACS) inhibitor, triacsin C, resulted in significantly reduced LD formation. A similar reduction in lipid accumulation was observed with the PPARα activator, Wy14643. Furthermore, CLA isomers promoted H4IIE viability at 60 µM but decreased viability at a higher dose of 180 µM. To further understand the role of PPARα in hepatic steatosis, we studied the level and phosphorylation of PPARα in livers of male lean and fa/fa Zucker rats fed either a control diet or fa/fa Zucker rats fed a CLA isomer (0.4% wt/wt c9,t11 or 0.4% wt/wt t10,c12) diet for 8 weeks. Immunoblotting results showed that only the t10,c12 CLA isomer significantly reduced phospho-PPARα S21 compared to the lean control (ln Ctl) and it was associated with a significant increase in the phosphorylation of p38 mitogen activated protein kinase (MAPK).These changes were not observed with the c9,t11 CLA isomer. Taken together, we have shown that CLA isomers directly induce LD formation in quiescent H4IIEs by activation of the lipid storage pathway which was significantly reduced by triacsin C or Wy14643. Also, we demonstrate for the first time that only the t10,c12 CLA isomer significantly reduced PPARα phosphorylation while it increased p38 MAPK phosphorylation. These results indicate that the anti-steatotic effects of the t10,c12 CLA isomer is associated with changes in PPARα phosphorylation and thereby its activity in a MAPK-independent manner.

Les technologies de l’information et des communications (TIC) sont de plus en plus utilisées dans les écoles canadiennes. Lorsque présentes en nombre important, comme dans le cas d’un déploiement massif d’ordinateurs portables en salle de classe, les enseignants nécessitent un appui. Le Conseil scolaire francophone (CSF) de la Colombie-Britannique a effectué un tel déploiement et s’est allié d’une équipe de cyberpédagogues afin d’aider les enseignants dans leur tâche d’intégration des technologies. Ce travail de recherche présente une approche méthodologique mixte à dominante qualitative et est structuré selon quatre questions de recherche qui nous permettent (1) de comprendre le rôle des cyberpédagogues, (2) les changements apportés par l’implantation du postes des cyberpédagogues, (3) les formations offertes aux enseignants et (4) le soutien technique lié aux TIC. Les questionnaires distribués aux deux groupes de répondants (26 enseignants et 12 cyberpédagogues) nous ont permis de constater des opinions très partagées entre les deux groupes. Par contre, la majorité des répondants s’entendent sur la nécessité des cyberpédagogues ainsi que le besoin de modifier leur rôle. La formation continue semble essentielle, mais serait un point à améliorer selon les enseignants (et plusieurs cyberpédagogues).

An analysis of secondary control beliefs and physical and psychological well-being in older individuals

Researchers have struggled to define how to age well since the time of Roman philosopher Cicero in 44 BC, yet today it remains a mystery (Tate, Lah, & Cuddy, 2003). In the context of dealing with age-related declines that often accompany later life, it has recently been suggested that positive reinterpretation, a concept implicit to positive psychology, may be important (Ouwehand, de Ridder, & Bensing, 2007; Seligman & Csikszentmihalyi, 2000). Positive reinterpretation has at times been referred to as “secondary control” in the psychological literature. In their seminal article of 1982, Rothbaum, Weisz, and Snyder theorized that there were multiple ways in which people could positively reinterpret outcomes and gain feelings of secondary control, including believing in the power of others and nature, as well as downgrading importance. These belief patterns become especially important in later life, when faced with age-related challenges. In the present study, secondary control beliefs were examined cross-sectionally in 2003 (n = 223) and prospectively in 2006 (n = 117) in a sample of older adults (M age = 85 years, 62% women) using a variety of outcome measures including severity of chronic conditions score, recent health, self-rated health, positive emotion, life satisfaction, and perceived stress. The unique contributions of this study are twofold. First, the findings suggest that some older adults may emphasize certain secondary control beliefs in combination. Second, the combined beliefs were found to relate cross-sectionally to measures of physical and psychological well-being. These exploratory findings have important implications in applied and theoretical contexts. In applied contexts, they may help to enhance physical and psychological well-being in the very old. In theoretical contexts, they extend contemporary thinking on secondary control.

Biogeography and conservation of the pinnipeds (Carnivora: Mammalia)

This thesis examines the biogeography of world pinnipeds, a unique group of marine mammals that have adapted to marine foraging while maintaining terrestrial (land or ice) habitat links. Comparative analyses of species range sizes controlled for phylogenetic relationships using a multi-gene supertree with divergence dates estimated using fossil calibrations. Adaptations to aquatic mating and especially sea ice parturition have influenced range size distribution, and ranges are larger than those of terrestrially mating and/or pupping species. Small range size is endangering for many taxa, and most at risk pinnipeds are terrestrial species with small ranges. Ancestral state reconstructions suggest that pinnipeds had a long association with sea ice, an adaptation that would have allowed early seals to expand into novel habitats and increase their distribution. Range sizes exhibit a strong Rapoport effect (positive relationship between range size and latitude) at the global scale, even after controlling for phylogeny and body size allometry. A latitudinal gradient in species diversity cannot explain the Rapoport effect for global pinniped ranges, as diversity is highest at mid-latitudes in both hemispheres. These regions are characterized by marginal ice zones and variable climates, supporting a mix of pagophilic and temperate species. The climatic variability hypothesis also did not explain the Rapoport effect. Variability is bimodal, and annual sea surface temperature (SST) variability does explain diversity patterns. Range size has a significant negative relationship with annual mean SST, and the largest ranges are found in areas with low mean SST. Temperature responses are possibly related to thermoregulation, sea ice availability, and ecological relationships with other large marine predators. These results agree with other studies and suggest that ocean temperature, and not productivity, drives marine species richness patterns. Future research needs include studies of physiological tolerances, interactions with sharks as predators and competitors, and the role of climate and sea ice in speciation and evolution. A better understanding of distribution and diversity patterns, and the role of the environment in shaping these patterns, will improve conservation efforts, and studies on the role of SST and sea ice are particularly important given current warming trends and declines in ice extent.

The impact of rural to urban migration on forest commons in Oaxaca, Mexico

This thesis investigates the impact of rural to urban migration on long-standing commons regimes in the Sierra Norte (northern highlands) of Oaxaca – the most biologically and culturally diverse state in Mexico. Since the second half of the twentieth century, local communities have been engaged with regional, national and international markets for wage labour, with many losing a significant percentage of their resident populations. The study shows how demographic and cultural change is impacting the two social institutions – cargos and tequios – that underpin the highly autonomous form of governance the region is famed for. The loss of able-bodied men and women has meant that these customary systems are struggling to remain operational. In response, a number of far-reaching changes have been introduced, including institutional adaptations and the forging of strong translocal ties that show potential for reducing the vulnerability of affected communities. However, while migration was temporary or circular for much of the 1970s, 1980s and 1990s, thus helping to maintain a balance between subsistence production and market engagement, a form of semi-permanent or permanent migration has come to dominate over the past decade and a half. This critical yet poorly recognised shift in migration dynamics has seen new and increased pressures emerge, and served to reduce the effectiveness of adaptive strategies at the community level. Within this context, the implications for commons theory are discussed, with two alternate frameworks (rational choice vs. moral economy) utilised to explain why institutions may persist, transform or fail in the face of change. In addition, a layer of complexity is added to the body of work examining the consequences of rural depopulation on Mexican forest landscapes and associated biological diversity. The study questions the assumption that rural to urban migration necessarily stimulates ecosystem recovery and enhances biodiversity conservation at a landscape scale. In fact, because of abandonment of a mosaic of use, the net effect may be an overall loss of biodiversity. From a policy perspective, the principal contributions of the study are especially pertinent at a time when funding agencies and government programs show belated interest in the consequences of out-migration for environmental management, resource use and rural livelihoods in tropical country settings.

Human milk storage conditions in regard to safety and optimal preservation of nutritional properties

This study assessed losses of the linoleic, alpha-linolenic, arachidonic and docosahexaenoic fatty acids, vitamin B2 and total vitamin C during storage of human milk, pasteurized and unpasteurized. The volatile compounds’ pattern change was monitored. The storage conditions were 8 days at 4oC and 6 months at -20oC and at -80oC, with and without limitation of oxygen. Fatty acids were analyzed by GC-FID; vitamins - by HPLC; volatile compounds’ concentration patterns were obtained using the e-nose machine; ANOVA tests were applied, with the statistical significance assigned to P<0.05. The official recommendations for human milk storage of 5-8 days at 4oC and of 6 or more months at -20oC are appropriate in regard to the analyzed nutrients and can be extended for pasteurized human milk storage. Oxygen limitation and -80oC temperature have no clear benefit for nutrient preservation. The evaluation of the odour cannot be based exclusively on the e-nose analysis.
