Aminocyclopyrachlor: Weed Control, Soil Dissipation, and Efficacy to Seedling Grasses

Aminocyclopyrachlor was developed for invasive weed control in non-cropland. Weed control, soil dissipation, and seedling grass tolerance with aminocyclopyrachlor were evaluated in field and greenhouse trials. Weed control was evaluated with aminocyclopyrachlor applied at 70 to 210 g ha-1. Absinth wormwood was controlled when treated during vegetative growth, but yellow toadflax was only controlled at flowering. Aminocyclopyrachlor alone did not control houndstongue. Aminocyclopyrachlor dissipation generally increased as either soil moisture or temperature increased. The DT50 values ranged from 3 to > 112 d. Aminocyclopyrachlor applied to cool season grasses at 91 to 112 g ha-1 provided adequate weed control and was safe for use on intermediate wheatgrass, but injured western wheatgrass. Efficacy to green needlegrass could not be determined. Big bluestem, sideoats grama, and switchgrass were difficult to evaluate due to poor grass establishment, but minimal injury was observed when aminocyclopyrachlor was applied at 91 to 168 g ha-1.

Vliv nanočástic na produkci karotenoidních a polyfenolických látek u Chlorella vulgaris

Microalgae are increasingly involved in the diet due to their beneficial effects on human health and therefore they are more often studied. Nowadays, there is a trend of using nanotechnology. The undesirable effects of copper oxide nanoparticles (CuO) attract more and more attention today. Since, there is an increasing demand for fossil fuels and simultaneously the amount of CO2 is increasing in the air, microalgae could serve in future as renewable fuels thanks to its rapid grow properties. In the diploma thesis, I dealt with the amount of carotenoid and polyphenolic substances in the algae Chlorella vulgaris after the influence of CuO nanoparticles. Experimental part of this work summarizes the results, revealing that CuO nanoparticles caused decrease of chlorophyle a by up to 60 % with CuO nanoparticles concetration of 25 mg/l, together with significant decrease of chlorophyle b and carotenoids. The amount of polyphenols and flavonoids in biomass has been changed only minimaly. In contrast, with usage of increasing concentration of CuO nanoparticles I found out by the SPOT test the increase of biomass in algae. Significant changes in selected secondary metabolites has been observed. The greatest statistically significant (p<0,01) growth with concetration 5 mg/l (1,08 ± 0,05 µg/g dry weight) has been observed for pOHbenzaldehyde, with increase by 42 % compared to control. In contrast rapid decrease has been noted with concentration 25 mg/l (0,06 ± 0,04 µg/g dry weight)

Isolation and Characterization of Proteus vulgaris Methylglyoxal Synthetase

Methylglyoxal synthetase, which catalyzes the formation of methylglyoxal and inorganic phosphate from dihydroxyacetone phosphate, was found in extracts of Proteus vulgaris. An efficient purification procedure utilizing ion exchange column chromatography and isoelectric focusing has been developed. Homogeneity of the enzyme preparation was confirmed by polyacrylamide gel electrophoresis and rechromatography.Two components of methylglyoxal synthetase were obtained upon isoelectric focusing. A comparison of the chemical and physical properties of the two components was carried out. The enzyme is a dimer. In the presence of inorganic phosphate, the hyperbolic saturation kinetics with dihydroxyacetone phosphate are shifted to sigmoidal.

Several studies were conducted with Artemisia vulgaris (mugwort). The first study involved the observation of shoot initiation from various length rhizome sections arxi positions. A second study was conducted to observe growth habits of mugwort and to determine the amount of infestation and production in a growing season. The third study was conducted to determine fenac rates that would prevent shoot growth from mugwort rhizomes. Also, various herbicides were evaluated in field experiments for the control of mugwort. The last study involved the effects of fenac on respiration and some respiratory enzymes. The most unifonn plants were observed from two-inch rhizome sections. There was no difference in bud viability of older or younger areas of rhizomes. New rhizome production became rapid about seven weeks after small muqwort plants were transplanted to the field. There was a continuous increase in shoots and rhizomes throughout the growing period. New plants were initiated in the spring of the next year from rhizomes that had overwintered in the soil. Fenac, when mixed with soil at two ppn or above, prevented shoot initiation from rhizome buds. When fenac was present in nutrient solution at rates above two ppm, no shoot growth was observed. In the field control experiments, fenac, dicamba, and Tordon tended to give the best control of mugwort. There tended to be an increase in the enzymatic activity of mugwort after the plants were sprayed with fenac. / Master of Science

O feijoeiro-comum (Phaseolus vulgaris L.) é uma planta leguminosa que se destaca por ser uma das principais culturas agrícolas do mundo, com forte expressão cultural e econômica na agricultura brasileira. Com base na relevância do feijão no contexto mundial, é importante considerar as projeções futuras de produção dessa leguminosa dentro do debate internacional de mudanças climáticas e previsão do aumento populacional global, de 2 bilhões de habitantes, até o ano de 2050. Levando em consideração que o setor agrícola poderá ser um dos mais afetados com os efeitos das mudanças climáticas previstas para o decorrer do século XXI, o presente trabalho teve como objetivo simular a produção brasileira de feijão-comum para locais que representem, ao mínimo, 80% da quantidade produzida deste grão. Para isso, o modelo DSSAT/CROPGRO-Drybean foi calibrado para dois grupos comerciais representativos (\"Carioca\" e \"Preto\"). Esta tarefa foi caracterizada pelo ajuste do modelo através da análise comparativa com dados observados, coletados a partir de experimentos conduzidos em Piracicaba-SP e Santo Antônio de Goiás-GO. As projeções climáticas foram obtidas por meio da metodologia recomendada pelo projeto internacional AgMiP (Agricultural Model Intercomparison and Improvement Project), do qual foram considerados os dados climatológicos estimados por 20 modelos de circulação global (GCM), para os cenários futuros de concentração atmosférica de CO2 RCP 4.5 e RCP 8.5, em 24 zonas homogêneas que representam as características edafoclimáticas dos principais locais de produção no Brasil. Por fim, combinando-se as informações ambientais com o modelo de culturas, foi possível simular para o período de 2040 a 2070, a produtividade futura para cultura do feijão-comum. Observou-se que as mudanças climáticas acarretarão em aumento da produção nacional para a maioria das regiões produtoras, entretanto haverá aumento do risco de se produzir abaixo da média esperada em regiões de grande importância para a produção nacional, como os cultivos de 1ª safra de feijão \"Preto\" no Sul do Brasil, e para cultivos de 3ª safra (grupo comercial \"Carioca\") presentes no Centro-Oeste. / Common beans (Phaseolus vulgaris L.) are leguminous plants which represents one of the main world crops, with strong cultural and economical expression in Brazil\'s agriculture. Based on the common beans world relevance, it\'s important to consider the future projections for this crop production inside the international subject of climate change and the global population increase, predicted to 2 billion inhabitants, until 2050. Being aware that the agricultural sector would be severely affected by the effects of the predicted climate changes in 21st century, the main goal of this work was simulate the Brazilian production of common beans for locals which represents at least 80% of the grain produced quantity. For that, the model DSSAT/CROPGRO-Drybean was calibrated for two representative commercial groups (\"Carioca\" and \"Black\"). This task was caracterized by the adjustment of the model through the comparatively analyse of observed data, collected on field experiments performed in Piracicaba/SP and Santo Antônio de Goiás/GO. The climate projections were obtained through a methodology proposed by the international AgMiP project (Agricultural Model Intercomparison and Improvement Project), which were considered the climatological data estimated by 20 global circulation models (GCM), for scenarios of representative atmospheric CO2 concentration pathways RCP 4.5 and RCP 8.5, in 24 homogenic zones representing the edafoclimatic profiles of main producers in Brazil. Lastly, combining all environmental data with the crop model, was possible to simulate for the period of 2040 to 2070, the future productivity for nacional common bean crops. Was observed that climate changes would accomplish a increase in the mean production for most of the regions, by the other hand, it will also increase the risk of productions above the expected average on key regions, as such the 1st crops of \"Black\" beans in the south, and 3rd crops (commercial group \"Carioca) at the Middle-West.

Aditivos na redução de oxidação enzimática em beterraba minimamente processada / Additives in the reduction of enzymatic oxidation in minimally processed red beet

O esbranquiçamento é o principal problema pós-colheita em beterraba minimamente processada. Esse problema consiste no ressecamento das células superficiais do produto, resultando no aspecto esbranquiçado decorrente do bloqueio das células túrgidas e íntegras das camadas inferiores. As etapas de corte e a manipulação durante o preparo dos produtos minimamente processados (PMPs) podem ser consideradas fonte de estresse abiótico, que estimula a formação de espécies reativas de oxigênio (EROs). O acúmulo de EROs provoca alterações bioquímicas e estruturais nas células, ocasionando alterações visuais e nutricionais que afetam diretamente a qualidade comercial. Aditivos alimentares são utilizados para retardar a degradação dos PMPs, mantendo-os frescos e adequados para o consumo por mais tempo. Ácidos orgânicos e aminoácidos são aditivos do tipo GRAS (Generally recognized as safe), ou seja, seguros para alimentos e são amplamente utilizados em produtos hortícolas. O objetivo deste trabalho foi avaliar a eficácia da arginina no controle do esbranquiçamento em beterraba minimamente processada e comparar com os demais aditivos utilizados comercialmente. Desse modo inicialmente foi determinada a combinação mais eficaz de dose e pH de aplicação da arginina, com avaliação de quatro doses (0, 10, 25 e 50 mM) combinadas com três faixas de pH (inalterado, pH 6,0 e pH 7,0). A aplicação de arginina apresenta resultados promissores em relação ao retardo do esbranquiçamento em beterraba minimamente processada, principalmente para o tratamento com 25 mM de arginina em pH 7,0. Além disso, esse tratamento também reduz a atividade respiratória, atividade de enzimas oxidativas (POD e PPO) e promove a manutenção de compostos bioativos importantes para a hortaliça (Betalaínas, compostos fenólicos e flavonois) por 12 dias após o processamento. Na segunda etapa, quatro aditivos (arginina, cisteína, ácido cítrico e ácido ascórbico) e água destilada, como tratamento controle, foram comparados quanto à eficácia na conservação de beterrabas minimamente processadas. O tratamento com arginina promove os melhores resultados quanto a manutenção do aspecto visual e de compostos bioativos em relação aos demais aditivos. O modo de ação diferenciado da arginina favorece a redução do estresse oxidativo, estendendo a vida útil do produto. / White blush is the main postharvest issue in minimally processed red beet. This issue consists in dehydration of superficial cell layers of the product, resulting in a whitening aspect caused by the blockage of turgid and intact cells on the layers below. During the minimal processing preparation, cutting and manipulation of minimally processed products (MPPs) are considered source of abiotic stress, which induce the formation of reactive oxygen species (ROS). The accumulation of ROS causes biochemical and structural changes that directly affects the commercial quality. Food additives are used to slow the degradation of MPPs by keeping them fresh and suitable for consumption. Organic acids and amino acids are GRAS (Generally reconized as safe) additives widely used in vegetables. This study aimed to evaluate the efficacy of arginine in controlling white blush in minimally processed redbeet and compare with additives used commercially. Thus, initially it was determined the best combination of four doses of arginine (0, 10, 25 and 50 mM) with three pH ranges (unchanged, pH 6,0 and pH 7,0). Arginine treatment presents promising results regarding the white blush delay in minimally processed redbeet. In addition, this additive also reduces the respiratory activity, activity of oxidative enzymes (POD and PPO) and maintains the important bioactive compounds (betalains, phenolic compounds and flavonols) for 12 days after processing. In the second step, four additives (arginine, cysteine, citric acid and ascorbic acid) and distilled water, as control treatment, were compared for efficacy in conservation of minimally processed beets. Arginine treatment promotes the best results regarding the maintenance of the visual aspect and bioactive compounds content in relation to the other additives. The different mode of action of arginine favors the reduction of oxidative stress, extending the shelf life of the product.

Bioacessibilidade de compostos fenólicos e minerais em feijão comum (Phaseolus vulgaris L.) / Bioaccessibility of phenolic compounds and minerals in common bean (Phaseolus vulgaris L)

Os grãos de feijão comum (Phaseolus vulgaris L) são considerados uma das principais fontes de nutrientes em muitas regiões do mundo. Estes grãos podem conter proteínas, lipídios, carboidratos, vitaminas e minerais indispensáveis para a nutrição humana. Além disso, as variações nas colorações do tegumento indicam a presença de compostos fenólicos com características antioxidantes. Estes grãos são consumidos geralmente cozidos pelo fato deste processo ajudar a melhorar as características sensoriais, a digestibilidade e a redução de possíveis constituintes antinutricionais. Sendo assim, a determinação da absorção de um nutriente na dieta não é feita somente pela quantificação do teor total, é necessário saber quanto realmente é liberado da matriz alimentar, e o que aproximadamente pode ser aproveitado pelo organismo. Atualmente, estudos da bioacessibilidade de nutrientes em distintas matrizes alimentares vem aumentado nos últimos anos, mas estudos sobre essa abordagem em grãos de feijão comum ainda são escassos. Diante disso, o objetivo desse trabalho foi avaliar a Bioacessibilidade de compostos fenólicos e minerais em feijão comum (Phaseolus vulgaris L). Em primeiro lugar estudou-se a variação do conteúdo de compostos fenólicos e capacidade antioxidante em cultivares de feijão comum, antes e depois do cozimento. Sendo possível alegar que dependendo do cultivar de feijão, o cozimento dos grãos possibilita o aumento do conteúdo de fenólicos totais e flavonoides. Além disso o cozimento permite a liberação de ácidos fenólicos e flavonoides ligados a outros compostos na matriz alimentar. Da mesma forma, foram avaliadas as concentrações de minerais em cultivares de feijão comum, antes e após do cozimento. Os resultados confirmam que os cultivares de feijão apresentam altos conteúdos de Fe, Zn, Cu, K, Mg, Mn e P; como também uma boa fonte de Ca mesmo após do cozimento. Por último, a digestão gastrointestinal de compostos fenólicos e minerais foi realizada em cultivares de feijão comum, cozidos mediante dialise in vitro. Observou-se que o processo gastrointestinal produz reduções significativas de compostos fenólicos e minerais nos cultivares de feijão comum cozidos desde o processo da digestão gástrica até a digestão intestinal. Além disso, somente entre o 3,08 - 6,90% do conteúdo dos compostos fenólicos totais e 5,56 - 6,73% dos flavonoides totais foram considerados bioacessíveis. Foi observado que o ácido gálico (ácido fenólico) e a epicatequina (flavonoide) possuem as maiores porcentagens de bioacessibilidade entre todos os compostos fenólicos reportados neste estudo. Por outro lado, as porcentagens de bioacessibilidade dos minerais foram: Cu 3,45-36,66 %; Zn 21,50-24,88%; Fe 0,18-2,84%; Mn 3,79-8,68%, Ca 3,84-8,27 %; Mg 13,57-14,92%; K 18,75-25,63% e P 9,52-13,75%. Desta forma, é evidente que o feijão proporciona uma importante quantidade de compostos fenólicos e minerais à dieta. / Common bean grains (Phaseolus vulgaris L) are considered one of the main sources of nutrients in many regions of the world. These grains may contain essential proteins, lipids, carbohydrates, vitamins and minerals to the human nutrition. In addition, variations in the color of teguments indicate the presence of phenolic compounds with antioxidant characteristics. These grains are usually consumed cooked so this process helps to improve sensory characteristics, digestibility and reduces possible antinutritional constituents. Therefore, to estimate the absorption of some nutrients in the diet the determination of total contents it is not sufficient, for this it is necessary to know how much in reality is released from the food matrix and what approximately can be used by the organism. Currently, studies of bioaccessibility of nutrients in different food matrices have increased, but studies on this approach in common bean grains are still scarce. The objective of this study was to evaluate the bioaccessibility of phenolic compounds and minerals in common bean (Phaseolus vulgaris L). Firstly, the variation of phenolic compounds content and antioxidant capacity in common bean cultivars, before and after cooking was studied. It is possible to argue that depending on the bean cultivar, the cooking of the grains makes it possible to increase the content of total phenolics and flavonoids. In addition, cooking allows the release of phenolic acids and flavonoids bound to other compounds in the food matrix. Likewise, the concentrations of minerals in common bean cultivars before and after cooking were evaluated. The results confirm that the bean cultivars present high contents of Fe, Zn, Cu, K, Mg, Mn and P; as well it is a good source of Ca even after cooking. Finally, gastrointestinal digestion of phenolic and mineral compounds in common bean cultivars cooked was done by in vitro dialysis. It has been observed that the gastrointestinal process produces significant reductions of phenolics and mineral compounds in common bean cultivars cooked from the gastric digestion process to intestinal digestion. In addition, only between 3.08 - 6.90% of total phenolic compounds content and 5.56 - 6.73% of total flavonoids were considered bioaccessible. It was observed that gallic acid (phenolic acid) and epicatechin (flavonoid) have the highest percentages of bioaccessibility among all the phenolic compounds reported in this study. On the other hand, the percentages of bioaccessibility of minerals were: Cu 3.45-36.66%; Zn 21.50-24.88%; Fe 0.18-2.84%; Mn 3.79-8.68%, Ca 3.84-8.27%; Mg 13.57-14.92%; K 18.75-25.63% and P 9.52-13.75%. In this way, it is evident that the beans provide an important amount of phenolic compounds and minerals to the diet.

Análise da expressão diferencial de genes relacionados à tolerância ao déficit hídrico em feijoeiro comum / Differential expression analysis of genes related to drought stress tolerance in common bean

O Brasil é o maior produtor mundial de feijão com produção média anual de 3,5 milhões de toneladas, entretanto, um dos maiores problemas enfrentados por essa cultura é o déficit hídrico, que leva a uma redução considerável em seu rendimento. Dessa forma, a identificação de genes que controlam os mecanismos de defesa e adaptação do feijoeiro à falta de água durante o seu desenvolvimento é de grande utilidade. E, como a tolerância à seca é um caráter multigênico, genótipos com diferentes graus de tolerância apresentam expressão gênica diferencial, com ativação e/ou repressão de determinados genes. Diante disso, esse trabalho teve como objetivos: (i) identificar genes diferencialmente expressos em dois genótipos de feijoeiro comum, sendo um tolerante (BAT 477) e o outro suscetível (IACCarioca 80SH) à seca; (ii) verificar a expressão gênica espacial (em raízes, caules e folhas) e temporal (cinco níveis crescentes de déficit hídrico - 24 h, 48 h, 72 h, 96 h e 120 h de exposição ao estresse) por meio de RT-qPCR; (iii) predizer a função dos genes identificados, com base nos genes ortólogos de Arabidopsis thaliana, usando dados públicos de microarranjo disponíveis na plataforma Genevestigator. Para atingir esses objetivos, foi conduzido um experimento em casa-de-vegetação, sendo induzido o déficit hídrico, por meio da suspensão da irrigação, quando as plantas atingiram o estádio fenológico R5, mantendo sob suprimento hídrico adequado as plantas-controle. Foi então utilizada a técnica de cDNA-AFLP para isolar os transcritos diferencialmente expressos entre os dois genótipos, sob seca, a qual aliada ao sequenciamento possibilitou a identificação e anotação de 45 transcritos, sendo 21 exclusivamente expressos no genótipo tolerante e 24 no genótipo suscetível. Dentre os transcritos identificados no genótipo tolerante, podem ser listados pelo menos 11, com potencial de serem usados para transformação genética (chlorophyll A-B binding protein, HSP40, HSP70, glycosyl hydrolase, serine/threonine protein kinase, trehalose-6-phosphate synthase, E3 ubiquitin ligase, fructose biphosphate aldolase, mediator complex subunit 13, aquaporin nodulin MTN-3-related e TCP transcription factor), e no genótipo suscetível, podem ser listados nove (coatomer protein complex, monoamine-oxidase A repressor R1, synaptobrevin, haloacid dehalogenase-like hydrolase, ADP-ribosylation factor, mTERF, serine protease S1C HtrA-related, legume lectin e SWI/SNF-related chromatin binding). Na análise de expressão gênica espacial e temporal, os transcritos mais promissores para uso em trabalhos futuros de transformação genética foram: aquaporin nodulin MTN3, E3 ubiquitin ligase, serine/threonine protein kinase, glycosyl hydrolase e HSP 70 protein, uma vez que tiveram uma expressão bastante pronunciada no genótipo tolerante. Através das análises in silico, baseadas nos genes ortólogos de A. thaliana, foram descobertos processos e vias metabólicas que podem estar envolvidos na resposta do feijoeiro comum ao déficit hídrico. Além disso, foram identificados genes associados com a tolerância à seca, corroborando os dados experimentais. Sendo assim, os resultados obtidos nesse trabalho fornecem o entendimento necessário ao desenvolvimento de ferramentas moleculares (marcadores para genes diferencialmente regulados) para serem utilizados em programas de melhoramento, bem como para informação genética básica na anotação funcional do genoma do feijoeiro e também para utilização desses genes candidatos em trabalhos de transformação genética para obtenção de plantas mais tolerantes à seca. / Brazil is the largest producer of common beans, with average annual production of 3.5 million tonnes; however, one of the biggest problems faced by this crop is drought, which leads to a considerable reduction in their yield. Thus, the identification of genes that control the defense mechanisms and adaptation of common bean to drought during its development is very useful. Drought tolerance is a multigenic character, so genotypes with different degrees of water deficit tolerance exhibit differential gene expression, with activation and/or repression of certain genes. Therefore, this study aimed to: (i) identify differentially expressed genes in two common bean genotypes, one tolerant (BAT 477) and other susceptible (IAC-Carioca 80SH) to drought, (ii) verify the spatial (root, stem and leaves) and temporal (five increasing levels of water deficit - 24 h, 48 h, 72 h, 96 h and 120 h of stress exposition) gene expression by RTqPCR (iii) predict the genes function, based on Arabidopsis thaliana orthologous genes, using available data in the public microarray platform Genevestigator. To achieve these objectives, an experiment was conducted in a green house, being induced water deficit, by withholding water when the plants reached growth stage R5, maintaining adequate water supply under the control plants. To isolate transcripts differentially expressed between the two genotypes under drought was used the cDNA-AFLP technique, which coupled with sequencing enabled the identification and annotation of 45 transcripts, 21 exclusively expressed in the tolerant genotype and 24 in the susceptible one. Among the transcripts identified in the tolerant genotype, may be listed at least 11, with potential to be used in genetic transformation (chlorophyll A-B binding protein, HSP40, HSP70, glycosyl hydrolase, serine/threonine protein kinase, trehalose-6-phosphate synthase, E3 ubiquitin ligase, fructose biphosphate aldolase, mediator complex subunit 13, aquaporin nodulin MTN-3-related and TCP transcription factor), and in the susceptible genotype, can be listed nine (coatomer protein complex, monoamine-oxidase A repressor R1, synaptobrevin, haloacid dehalogenase-like hydrolase, ADP-ribosylation factor, mTERF, serine protease S1C HtrA-related, legume lectin and SWI/SNF-related chromatin binding). In the spatial and temporal gene expression analysis, the transcripts that stood out for use in genetic transformation future studies were: aquaporin nodulin MTN3, E3 ubiquitin ligase, serine/threonine protein kinase, glycosyl hydrolase and HSP 70 protein, since it had an expression quite pronounced in the tolerant genotype. Through the in silico analysis, based on orthologous genes of A. thaliana, was discovered processes and metabolic pathways that may be involved in the common bean response to drought. In addition, we identified genes associated with drought tolerance, corroborating the experimental data. Thus, the present results provide the necessary understanding to develop molecular tools (markers for differentially regulated genes) to be used in breeding programs, as well as basic genetic information in the common bean functional genome annotation and also to use these candidate genes for genetic transformation to obtain drought-tolerant plants.

Atualmente, a produtividade do feijoeiro comum (Phaseolus vulgaris L.) pode ser reduzida devido à ocorrência de doenças em todo o território nacional, destacando-se a murcha de fusário, causada por Fusarium oxysporum f. sp. phaseoli (Fop). No campo, o patógeno é disseminado a longas distâncias através das sementes infectadas e/ou contaminadas e a sua sobrevivência ocorre, principalmente, no solo. Os objetivos deste trabalho foram: avaliar a inibição do crescimento micelial de Fop por Trichoderma spp.; classificar a sensibilidade in vitro de Fop e Trichoderma spp., separadamente, a fungicidas e verificar a compatibilidade entre fungicidas químicos e biológicos para controle de Fop, presente nas sementes e no solo. Para avaliar a inibição do crescimento micelial de Fop, foram utilizados três isolados do patógeno, os quais foram confrontados, in vitro, com três isolados de Trichoderma spp. em testes de cultura pareada e produção de metabólitos voláteis a 20-22°C. Os experimentos foram conduzidos em delineamento inteiramente casualizado, com cinco repetições para cada isolado de Trichoderma. Para a classificação da sensibilidade in vitro de Fop e Trichoderma a fungicidas, foram avaliados os mesmos isolados anteriormente utilizados. Foram comparados dez fungicidas, em doses entre 0 a 100 mg L-1 que foram ajustadas de acordo com a CI50 de cada fungicida. Com base na percentagem de inibição do crescimento micelial, foram estimados os valores da concentração inibitória de 50% (CI50) e 100% (CI100) e selecionaram-se os fungicidas compatíveis com Trichoderma spp. A compatibilidade entre tratamentos químico e biológico foi avaliada através da inoculação artificial de sementes de feijão com um isolado de Fop (IAC 11.299-1) e infestação do mesmo no solo. As sementes foram tratadas com os fungicidas fludioxonil, flutriafol e tiofanato metílico, e com os três produtos biológicos, separadamente e em misturas. Avaliou-se o efeito dos tratamentos por meio dos testes de sanidade, germinação, comprimento de plântulas, massa da matéria seca em laboratório e índice de velocidade de emergência e porcentagem de emergência em estufa não climatizada. O efeito protetor dos tratamentos foi verificado através do teste de transmissão do patógeno solo-planta. Todos os isolados de Trichoderma apresentaram antagonismo in vitro contra Fop. No teste de cultura pareada foi observada uma redução de 15 a 20% no crescimento micelial do patógeno. No teste de produção de metabólitos voláteis, o isolado T12-1086G05 foi responsável pela maior inibição do crescimento micelial de Fop (10 a 48%). Os testes de sensibilidade in vitro mostraram que tiofanato metílico, flutriafol e fludioxonil foram compatíveis com Trichoderma (CI50 > 2 mg L-1). Com exceção do flutriafol e do GF 422 isolados e em mistura, todos os tratamentos foram eficientes na erradicação de Fop nas sementes, sem afetar a sua qualidade fisiológica. No teste de transmissão, verificou-se que a incidência de Fop foi de 5 a 40% no hipocótilo e de 5 a 30% nas raízes de feijoeiro provenientes de sementes tratadas com os produtos. / Currently, the common bean (Phaseolus vulgaris L.) productivity can be reduced due to the occurrence of diseases in all over the country, especially Fusarium wilt, caused by Fusarium oxysporum f. sp. phaseoli (Fop). In the field, the pathogen is spread to long distances through infected and/or contaminated seeds and its survival occurs, mainly, in the soil. The objectives of this work were: to evaluate the micelial growth inhibition of Fop promoted by Trichoderma spp.; to classificate in vitro sensitivity of Fop and Trichoderma spp., separately, to fungicides and to verify the compatibility between chemical and biological fungicides to control Fop, applied on the seeds and on the soil. To evaluate the micelial growth inhibition of Fop, it was used three Fop isolates, which were in vitro confronted with three isolates of Trichoderma spp., by paired culture and antibiosis assays at 20-22°C. The experiments were conducted entirely casualized design, with five replications for each Trichoderma isolate. For the in vitro sensitivity classification, it was evaluated the same isolates of Fop and Trichoderma previously used. It was compared ten fungicides and the concentrations of 0 to 100 mg L-1, which were adjusted according to the IC50 of each fungicide. Based on the percentage of micelial growth inhibition, it was estimated the values of inhibitory concentration of 50% (IC50) and 100% ((IC100) and it was selected the compatible fungicides with Trichoderma spp. The compatibility between chemical and biological treatments were evaluated through artificial inoculation of common bean seeds with the Fop isolate IAC 11.299, following the infestation of the pathogen in the soil. Seeds were treated with fludioxonyl, flutriafol and tiofanate methyl, and with three biological products, separately and mixed. It was evaluated the treatment effects by the following assays: sanity, germination, seedling lenght, seedling dry matter in laboratory conditions and emergence speed rate and percentage of emergence in greenhouse conditions. The protective effect of the treatments was verified through the pathogen transmission experiment from soil to plant. All Trichoderma isolates showed in vitro antagonism against Fop. In the paired culture assay it was observed 15 to 20% reduction in the pathogen micelial growth. In the antibiosis assay, the isolate T12-1086G05 was responsible for the maximum micelial inhibition growth of Fop (10 to 48%). in vitro sensitivity experiments showed that thiofanate methyl, flutriafol and fludioxonyl were compatible with Trichoderma (IC50 > 2 mg L-1). Except the treatments with fluatrifol and GF 422 alone and in mixture, all the products were efficient in eradicating Fop in the seeds, without affect its physiological quality. In the transmission experiment, it was verified that the Fop incidence was 5 to 40% in the hypoticol and 5 to 30% in the common bean roots from seed treated with the products.
