A mechanical model of an axial piston machine

<p>A mechanical model of an axial piston-type machine with a so-called wobble plate and Z-shaft mechanism is presented. The overall aim is to design and construct an oil-free piston expander demonstrator as a first step to realizing an advanced and compact small-scale steam engine system. The benefits of a small steam engine are negligible NOx emissions (due to continuous, low-temperature combustion), no gearbox needed, fuel flexibility (e.g., can run on biofuel and solar), high part-load efficiency, and low noise. Piston expanders, compared with turbines or clearance-sealed rotary displacement machines, have higher mechanical losses but lower leakage losses, much better part-load efficiency, and for many applications a more favourable (i.e., lower) speed. A piston expander is thus feasible for directly propelling small systems in the vehicular power range. An axial piston machine with minimized contact pressures and sliding velocities, and with properly selected construction materials for steam/water lubrication, should enable completely oil-free operation. An oil-free piston machine also has potential for other applications, for example, as a refrigerant (e.g., CO<sub>2</sub>) expander in a low-temperature Rankine cycle or as a refrigerant compressor.</p><p> </p><p>An analytical rigid-body kinematics and inverse dynamics model of the machine is presented. The kinematical analysis generates the resulting motion of the integral parts of the machine, fully parameterized. Inverse dynamics is applied when the system motion is completely known, and the method yields required external and internal forces and torques. The analytical model made use of the “Sophia” plug-in developed by Lesser for the simple derivation of rotational matrices relating different coordinate systems and for vector differentiation. Numerical solutions were computed in MATLAB. The results indicate a large load bearing in the conical contact surface between the mechanism’s wobble plate and engine block. The lateral force between piston and cylinder is small compared with that of a comparable machine with a conventional crank mechanism.</p><p> </p><p>This study aims to predict contact loads and sliding velocities in the component interfaces. Such data are needed for bearing and component dimensioning and for selecting materials and coatings. Predicted contact loads together with contact geometries can also be used as input for tribological rig testing. Results from the model have been used to dimension the integral parts, bearings and materials of a physical demonstrator of the super-critical steam expander application as well as in component design and concept studies.</p>

Formalisations and applications of business process modelling notation

Business Process Modelling Notation (BPMN) is a standardised diagram notation for modelling interactive workflow processes graphically at the design stage. The primary objective of this thesis is to provide a framework for precise specifications and formal verifications of workflow processes modelled as BPMN diagrams. We provide two behavioural semantics for BPMN in the process algebra Communicating Sequential Processes (CSP). We apply existing CSP refinement orderings to both the refinement of business process diagrams and the verification of behavioural compatibility of business process collaborations. The first semantic model is an untimed model, focusing on the control flow and communication of business processes. The second semantic model extends the first one to capture the timing aspect of behaviour. We also consider the applications of the semantic models. The secondary objective of this thesis is to apply BPMN and the semantic models to reason about long running empirical studies (e.g. laboratory experiments, clinical trials). We introduce a declarative workflow model Empiricol for recording trials and experiments precisely, and define bidirectional transformation functions between BPMN and Empiricol. Using the transformation functions, we make graphical specification, simulation, automation and verification of trials and experiments possible. We provide two case studies on the applications of BPMN’s formalisations.

The performance of insolvency prediction and credit risk models in the UK : a comparative study, development and wider application

Contingent claims models have recently been applied to the field of corporate insolvency prediction in an attempt to provide the art with a theoretical methodology that has been lacking in the past. Limited studies have been carried out in order to empirically compare the performance of these “market” models with that of their accounting number-based counterparts. This thesis contributes to the literature in several ways: The thesis traces the evolution of the art of corporate insolvency prediction from its inception through to the present day, combining key developments and methodologies into a single document of reference. I use receiver operating characteristic curves and tests of economic value to assess the efficacy of sixteen models, carefully selected to represent key moments in the evolution of the art, and tested upon, for the first time, post-IFRS UK data. The variability of model efficacy is also measured for the first time, using Monte Carlo simulation upon 10,000 randomly generated training and validation samples from a dataset consisting of over 12,000 firmyear observations. The results provide insights into the distribution of model accuracy as a result of sample selection, which is something which has not appeared in the literature prior to this study. I find overall that the efficacy of the models is generally less than that reported in the prior literature; but that the theoretically driven, market-based models outperform models which use accounting numbers; the latter showing a relatively larger efficacy distribution. Furthermore, I obtain the counter-intuitive finding that predictions based on a single ratio can be as efficient as those which are based on models which are far more complicated – in terms of variable variety and mathematical construction. Finally, I develop and test a naïve version of the down-and-out-call barrier option model for insolvency prediction and find that, despite its simple formulation, it performs favourably compared alongside other market-based models.

Système d’assistance lors d’errance nocturne chez les personnes âgées atteintes d’Alzheimer

Grâce à l’informatique ubiquitaire, il devient possible d’avoir des environnements de plus en plus intelligents offrant de l’assistance ambiante aux activités de la vie quotidienne. Dans ce mémoire, un système d’assistance est présenté offrant une assistance ambiante personnalisée aux personnes âgées avec démence d'Alzheimer souffrant d’errance nocturne. L’hypothèse de départ est que la nuit, les personnes Alzheimer font de l’errance pour satisfaire des besoins précis même si elles sont incapables de les exprimer. Notre sujet s’intéresse à trois types de besoins dans le contexte d’errance nocturne : avoir faim, avoir envie d’aller aux toilettes et avoir besoin d’être rassuré. Un système d’assistance ambiante a été conçu et implémenté pour chacun. Ce système utilise un réseau de capteurs et d’effecteurs hétérogènes. Dans un premier temps, des données sur les routines de la personne atteinte d’Alzheimer sont collectées par des questionnaires et des capteurs. Ces données sont ensuite validées pour établir les scénarios d’assistance et détecter les épisodes d’errance. Dans un second temps, grâce au profil recueilli avec la participation du proche aidant, des règles sont conçues pour décider de l’assistance à fournir pour accompagner la personne à travers la satisfaction de son besoin, puis l’inciter à retourner se coucher sans toutefois la contraindre. De plus, une application tablette est mise en place pour offrir du contrôle sur l’activation de l’assistance. Cette application est confiée au proche-aidant afin qu’il puisse décider s’il préfère offrir sa propre assistance ou celle automatisée. Le système d’assistance ambiante développé recueille donc des données dans le domicile et offre de l’accompagnement à la personne tout en offrant plus de confort à son proche-aidant. Une expérimentation dans un domicile auprès d’un binôme composé d’une mère atteinte de démence et de sa fille a été réalisée sur une période de cinq semaines. Même si aucun épisode d’errance n’a été détecté pendant cette expérimentation, les données recueillies par les différentes sources ont été comparées à celles du système pour en établir la validité. Une reconstruction des événements a été possible avec les données collectées. Celle-ci permet d’observer les activités de la personne atteinte d’Alzheimer dans son quotidien. Cette approche pour obtenir le suivi des activités est applicable aux activités nocturnes permettant ainsi le suivi pendant l’errance s’il y’a lieu.

Investigating the Structure of FtsZ to Understand its Functional Role in Bacterial Cell Division

<p>FtsZ, a bacterial tubulin homologue, is a cytoskeleton protein that plays key roles in cytokinesis of almost all prokaryotes. FtsZ assembles into protofilaments (pfs), one subunit thick, and these pfs assemble further to form a “Z ring” at the center of prokaryotic cells. The Z ring generates a constriction force on the inner membrane, and also serves as a scaffold to recruit cell-wall remodeling proteins for complete cell division in vivo. FtsZ can be subdivided into 3 main functional regions: globular domain, C terminal (Ct) linker, and Ct peptide. The globular domain binds GTP to assembles the pfs. The extreme Ct peptide binds membrane proteins to allow cytoplasmic FtsZ to function at the inner membrane. The Ct linker connects the globular domain and Ct peptide. In the present studies, we used genetic and structural approaches to investigate the function of Escherichia coli (E. coli) FtsZ. We sought to examine three questions: (1) Are lateral bonds between pfs essential for the Z ring? (2) Can we improve direct visualization of FtsZ in vivo by engineering an FtsZ-FP fusion that can function as the sole source of FtsZ for cell division? (3) Is the divergent Ct linker of FtsZ an intrinsically disordered peptide (IDP)?</p><p> One model of the Z ring proposes that pfs associate via lateral bonds to form ribbons; however, lateral bonds are still only hypothetical. To explore potential lateral bonding sites, we probed the surface of E. coli FtsZ by inserting either small peptides or whole FPs. Of the four lateral surfaces on FtsZ pfs, we obtained inserts on the front and back surfaces that were functional for cell division. We concluded that these faces are not sites of essential interactions. Inserts at two sites, G124 and R174 located on the left and right surfaces, completely blocked function, and were identified as possible sites for essential lateral interactions. Another goal was to find a location within FtsZ that supported fusion of FP reporter proteins, while allowing the FtsZ-FP to function as the sole source of FtsZ. We discovered one internal site, G55-Q56, where several different FPs could be inserted without impairing function. These FtsZ-FPs may provide advances for imaging Z-ring structure by super-resolution techniques.</p><p> The Ct linker is the most divergent region of FtsZ in both sequence and length. In E. coli FtsZ the Ct linker is 50 amino acids (aa), but for other FtsZ it can be as short as 37 aa or as long as 250 aa. The Ct linker has been hypothesized to be an IDP. In the present study, circular dichroism confirmed that isolated Ct linkers of E. coli (50 aa) and C. crescentus (175 aa) are IDPs. Limited trypsin proteolysis followed by mass spectrometry (LC-MS/MS) confirmed Ct linkers of E. coli (50 aa) and B. subtilis (47 aa) as IDPs even when still attached to the globular domain. In addition, we made chimeras, swapping the E. coli Ct linker for other peptides and proteins. Most chimeras allowed for normal cell division in E. coli, suggesting that IDPs with a length of 43 to 95 aa are tolerated, sequence has little importance, and electrostatic charge is unimportant. Several chimeras were purified to confirm the effect they had on pf assembly. We concluded that the Ct linker functions as a flexible tether allowing for force to be transferred from the FtsZ pf to the membrane to constrict the septum for division.</p> / Dissertation

Studies of the two redox active tyrosines in Photosystem II

Photosystem II is a unique enzyme which catalyzes light induced water oxidation. This process is driven by highly oxidizing ensemble of four Chl molecules, PD1, PD2, ChlD1 and ChlD2 called, P680. Excitation of one of the Chls in P680 leads to the primary charge separation, P680+Pheo-. Pheo- transfers electrons sequentially to the primary quinone acceptor QA and the secondary quinone acceptor QB. P680+ in turn extracts electrons from Mn4CaO5 cluster, a site for the water oxidation. There are two redox active tyrosines, TyrZ and TyrD, found in PSII. They are symmetrically located on the D1 and D2 central proteins. Only TyrZ acts as intermediate electron carrier between P680 and Mn4CaO5 cluster, while TyrD does not participate in the linear electron flow and stays oxidized under light conditions. Both tyrosines are involved in PCET. The reduced TyrD undergoes biphasic oxidation with the fast (msec-sec time range) and the slow (tens of seconds time range) kinetic phases. We assign these phases to two populations of PSII centers with proximal or distal water positions. We also suggest that the TyrD oxidation and stability is regulated by the new small lumenal protein subunit, PsbTn. The possible involvement of PsbTn protein in the proton translocation mechanism from TyrD is suggested. To assess the possible localization of primary cation in P680 the formation of the triplet state of P680 and the oxidation of TyrZ and TyrD were followed under visible and far-red light. We proposed that far-red light induces the cation formation on ChlD1. Transmembrane interaction between QB and TyrZ has been studied. The different oxidation yield of TyrZ, measured as a S1 split EPR signal was correlated to the conformational change of protein induced by the QB presence at the QB-site. The change is transferred via H-bonds to the corresponding His-residues via helix D of the D1 protein.

Key Virus-Host Interactions Required For Arenavirus Particle Assembly And Release

Viruses are infectious agents that must infect the cells of living organisms in order to reproduce. They have relatively simple genomes which encode few proteins but can compensate for their simplicity by hijacking components of their cellular hosts. Arenaviruses, a family of zoonotic viruses carried by rodents, encode only 4 proteins. One of these proteins, Z, is responsible for several functions during the virus life cycle including driving the formation and release of new virus particles at the plasma membrane of infected cells. Relatively little is known about how this viral protein is regulated or the complement of host proteins it engages in order to produce new virus particles or augment Z's other functions. To address this gap in knowledge, mass spectrometry was used to identify phosphorylation sites in the Old World arenavirus, lymphocytic choriomeningitis virus (LCMV) Z protein. Phosphorylation sites were identified at serine 41 (S41) and tyrosine 88 (Y88). Functional studies using recombinant (r)LCMV containing mutations at these phosphorylation sites revealed that both were important for the production of defective interfering (DI) particles. DI particles are replication-incompetent virus particles that interfere with the production of infectious virus and mitigate its cytopathic effect. While a mutation that mimics phosphorylation at S41 reduced LCMV's ability to produce both infectious and DI particles, this mutation had a much stronger impact on DI particles. Production of DI particles in Y88-mutant rLCMV was drastically reduced while the impact on infectious virus was minimal. Y88 lies within a type of viral late domain (PPXY) also found in matrix proteins of several disparate virus families where it has been shown to drive infectious virus release by recruiting the membrane scission machinery of the cellular endosomal sorting complex required for transport (ESCRT). Inhibition of the ESCRT pathway drastically reduced LCMV DI particle but not infectious virus release indicating that Z's PPXY late domain and the cellular ESCRT complex are required specifically for the production of DI particles. Mass spectrometry was also used to identify host protein partners of Z as well as the host proteins recruited into virus particles for the New World arenavirus, Junin (JUNV). ESCRT complex proteins were enriched in JUNV virus-like particles (VLPs) and bona fide virions. In contrast to LCMV, inhibition of the ESCRT complex resulted in significantly less infectious JUNV release. This indicates that the ultimate role of ESCRT engagement by the Old World arenavirus, LCMV, differs from that of New World, JUNV. This work represents the first demonstration that a viral protein motif and the host machinery it engages selectively drive DI particle production independently of infectious virus. It also suggests that host cell kinases can dynamically regulate the production of DI particles through phosphorylation of Z. Finally, the late domain-mutant rLCMV generated in these studies represents the first LCMV strain known to produce undetectable levels of DI particles which provides the opportunity to assess the impact that a loss of DI particles has on the ability of LCMV to establish or maintain a persistent infection.

What if ethical buying behavior leads to boycotts? : The buying behavior of Generation Z

The care for the environment has been a hot topic during the last fifteen years. Ecological and Fairtrade products made from sustainable materials and methods can be found in almost every store. It seems like awareness regarding our environmental impact has increased and therefore changed our purchase behavior. The awareness has also created pressure on the companies to behave in an ethical manner, if the consumers feel that their ethical needs are not acknowledged, they will tend to stop purchasing products from that company. In this new aware society there is a new player that in a few years will make up the new buying force. Generation Z is individuals born after 1995 and is expected to bring new demands to the market. The purpose of this thesis is to study if ethical products and ethical consumption have an effect on the buying behavior of Generation Z. To better understand if the ethical awareness affect the purchase behavior of Generation Z and if they are willing to boycott companies that misbehaves, theory within the fields of CSR, consumer behavior and ethical consumption has been collected to form questions for a survey. The findings show that the ethical awareness do not affect the purchase behavior of Generation Z, however there is discovered attributes such as ethical profile that affect the frequency of purchases. The findings also show that Generation Z will actively boycott a company if that company misbehaves.

Estimation of Aerodynamic Parameters in Real-Time : Implementation and Comparison of a Sequential Frequency Domain Method and a Batch Method

The flight testing and evaluation of collected data must be efficient during intensive flight-test programs such as the ones conducted during development of new aircraft. The aim of this thesis has thus been to produce a first version of an aerodynamic derivative estimation program that is to be used during real-time flight tests. The program is to give a first estimate of the aerodynamic derivatives as well as check the quality of the data collected and thus serve as a decision support during tests. The work that has been performed includes processing of data in order to use it in computations, comparing a batch and a sequential estimation method using real-time data and programming a user interface. All computations and programming has been done in Matlab. The estimation methods that have been compared are both built on transforming data to the frequency domain using a Chirp z-transform and then estimating the aerodynamic derivatives using complex least squares with instrumental variables.The sequential frequency domain method performs estimates at a given interval while the batch method performs one estimation at the end of the maneuver. Both methods compared in this thesis produce equal results. The continuous updates of the sequential method was however found to be better suited for a real-time application than the single estimation of the batch method. The telemetric data received from the aircraft must be synchronized to a common frequency of 60 Hz. Missing samples of the data stream must be linearly interpolated and different units of measured parameters must be corrected in order to be able to perform these estimations in the real-time test environment.
