The modulating effect of fatty acids on the lipid profile in colon epithelial mucosa in Vivo

Magister Scientiae - MSc / Several abnormal conditions, including some cancers, have been associated with changes in the membrane lipid and FA composition. Dietary fat serves as a major source of lipids and FA, particularly the polyunsaturated fatty acids (PUFA), n-6 and n-3. High intakes of n-6 PUFA have been linked to the development of colon cancer in association with low n-3 PUFA intake. Therefore understanding the differences in the lipid and FA profiles between cancer and normal cells in the colon, and the role diet plays in these factors may be invaluable in understanding their role in carcinogenesis. This study compares the lipid profile of azoxymethane (AOM) induced colon polyps to that of the surrounding mucosa tissue in rats fed a diet high in n-6 PUFA. Male Fischer rats were fed the AIN-76A diet containing sunflower oil that has high n-6 PUFA content for a period of nine months. Results indicate that the lipid and FA content of the colon polyps differs significantly from the surrounding mucosa. Colon polyps had an increase in membrane phopholipids phosphatidylcholine (PC) and phosphatidylethanolamine (PE). Changes in membrane fluidity were indicated by the decrease (0.05) in the PC/PE and cholesterol/phospholipids (chol/PL) ratios, and increase (0.05) in the polyunsaturated FA/saturated FA (P/S) ratio. Metabolism of FA was significantly altered in the polyps favouring n-6 FA metabolism and the production of prostaglandin E2. No clear indication of impaired & Delta;6-desauturase enzyme activity was noticed. Increases in the n-6 PUFA content could be a reflection of the dietary FA intake that increases FA incorporation in the polyps. Changes in the FA parameters of the polyps, particularly an increase in C20:4n-6 and the n6/n3 ratio have been shown to contribute to the rapid growth of cancer tissue. These lipid changes associated with the development of colon polyps could provide unique targets for developing strategies in chemoprevention by dietary manipulation. / South Africa

Comparison of the physicochemical characteristics and flavonoid release profiles of Sutherlandia frutescens phytosomes versus liposomes

Magister Pharmaceuticae - MPharm / Sutherlandia frutescens is a traditional plant medicine widely used in South Africa. Traditionally, the leaves of S. frutescens are mainly used as a tea, but these traditional dosage forms have several disadvantages, including that they are not particularly convenient to prepare and store, encourage dosage inaccuracy and are highly susceptible to microbial contamination. To solve these problems, dried aqueous extract forms, e.g. freeze dried aqueous extract (FDAE) of S. frutescens were prepared, but they, in turn, may still suffer from instability and contain mainly hydrophilic phytoconstituents that are poorly absorbed and delivered for in vivo activity. Modified forms of the FDAE, i.e. the active phytopharmaceutical ingredient (API), may be a better solution. Therefore this study sought to prepare liposomes and phytosomes of the freeze dried aqueous extract of Sutherlandia frutescens, as a means of increasing the total the surface area of the API, thus improving its release and dissolution in gastrointestinal fluids. Liposomes and phytosomes of the FDAE of Sutherlandia frutescens obtained were prepared using a thin film hydration method at ratios of lecithin: S. frutescens (3:1) and phosphatidylcholine: S. frutescens (2:1) respectively. The physical characteristics (i.e. particle size, size distribution, zeta potential, and morphology), of flavonoid glycosides (i.e. sutherlandins A to D; API) as well as content and release profiles of each dosage form (i.e. FDAE liposome or phytosomes) at pH 1.2 and pH 6.8 was determined. A validated HPLC assay was used to determine and compare the flavonoid glycoside content and release profiles of the liposomes and phytosomes. Both liposomes and phytosomes were successfully prepared, in moderate yields (± 30 %, and ± 50 %, respectively), using the thin film hydration method. The liposomes had a significantly smaller size, lower size distribution, higher zeta potential and better stability than the phytosomes (p < 0.05). The phytosomes, however, had significantly higher flavonoid glycoside encapsulation efficiency than the liposomes (±50 % vs ±26 %; p < 0.01). In addition, the release at 120 minutes, of flavonoid glycosides from the liposomes (63%, 58%, 76% and 46% % at pH 1.2, and 78%, 76%, 87% and 89 % at pH 6.8 for sutherlandins A, B, C and D, respectively) was significantly higher and faster than that of the phytosomes (52%, 41%, 51% and 39 % at pH 1.2, and 31% 31%, 33%and 45% % at pH 6.8, for sutherlandins A, B, C and D, respectively). The differences in release were likely due to differences in particle size and size distribution of the two modified API forms. Overall, liposomes and phytosomes can be considered promising vehicles for delayed delivery of herbal crude extracts. Based on its characteristics (i.e. narrower size distribution, and better stability), the liposomes were preferred compared to the phytosomes offering a better kinetic release profile. The phytosomes had higher encapsulation than the liposomes that may be due to complex formation between the API and the lipid.

Storm Induced Beach Profile Changes along the Coast of Treasure Island, West-Central Florida, U.S.A.

Storms play a significant role in beach morphodynamics. Storm-induced beach-profile changes and their longshore variations are investigated in this study. The impacts of four summer tropical storms and two series of winter storms over the last 10 years along the coast of Treasure Island were documented. Tropical storms Alberto in 2006, Fay in 2008, Debby in 2012, Hermine in 2016 and winter storms in winter seasons of 2014 and 2015 are discussed in this study. In general, the Treasure Island beach experienced more erosion generated by tropical storms with greater intensity, but shorter duration, as compared to winter storms due to lower waves, weaker wind and smaller storm surge. Winter storms typically do not generate high storm surge and generally do not cause erosion at the dune and back beach unless the pre-storm beach is very narrow. Based on pre- and post-storm beach-profile surveys along the coast of Treasure Island, the northern end of the barrier island, located directly downdrift of the John’s Pass tidal inlet, experienced erosion along the entire profile during the storms. Along the middle part of Treasure Island, dry beach suffered erosion during both the tropical storms and winter seasons while the nearshore zone suffered erosion during the tropical storms and experienced deposition during the winter seasons. Sunset Beach at the southern end experienced severe erosion during tropical storm Debby, but not during other storms. Winter seasons caused relatively small changes to the morphology of Sunset Beach. Deposition happened in the nearshore zone along Sunset Beach during winter storms. Survey line R143 at the very south end of Treasure Island suffered erosion in tropical storm Alberto, Debby and Hermine. Beach profile changes induced by Tropical storm Fay was different as compared to other tropical storms. Considerably less beach erosion occurred due to the large distance of the storm path from the study area. Overall, Sunshine Beach, bounded by John’s Pass inlet at northern end of Treasure Island, was influenced both by wave conditions and the tidal flows. Sediment transport was to the north along the coast of Sunshine Beach when wind direction was from south, e.g. during tropical storm Fay. However the northward sediment transport was blocked by the John’s Pass jetty. Therefore, deposition occurred at Sunshine Beach during tropical storm Fay. When wind direction was from north (e.g. during tropical storms Alberto and during the winter seasons), southward sediment transport was generated. Erosion occurs during the northerly approaching storms. The morphodynamics of the middle section of Treasure Island are influenced by the sand supply at the attachment point of John’s Pass ebb delta. Sunset Beach experienced various levels of erosion during the tropical storms not only because of the high wave, strong wind and high water level generated by storms, but also due to the higher waves associated with an offshore dredged pit.

Online dating in a South African context : a psychological study of the persona profile

Little is known about online dating relationships, or the online dating persona that intentionally forms and develops interpersonal relationships through online matchmaking services. Online dating relationships are becoming more prevalent, especially in South Africa. However, research studies focussing on online dating relationships seems to be lacking. Hence, a scientific psychological enquiry into online dating relationships, and more specifically the online dating persona, is indicated. Focussing on 100 online dating members from DatingBuzz, this comparative research study investigated online dating relationships and the online dating persona. More specifically, this study examined, first, online dating relationships specifically within the South African context compared to online dating relationships internationally; and second, the South African online dating persona. This was done by creating a profile focussing on (a) sociodemographic characteristics, (b) personality characteristics, (c) level of emotional intelligence and, (d) life effectiveness skills. A quasi-comparative research design was used with a non-probability, non-random sample of convenience. The realised sample for this research was 100 participants (N=100), with the total sample (n=100) reporting on online dating relationships and 67 participants (n=67) reporting on the online dating persona. The online matchmaking service, DatingBuzz, consented to post an advertisement for research participation. Using an Internet-based questionnaire with preset response categories in a Likert type format, data collection continued consecutively for six months in 2005. Instruments included (a) a set of self-constructed questions assessing online dating relationships and the sociodemographic characteristics of the online dating persona, (b) Sauciers’ 40 Mini-marker Set to examine the Big Five personality traits, (c) the 33-item EIS scale to investigate emotional intelligence and, (d) the LEQ – H to examine life effectiveness skills. The data were statistically analysed using the SAS programme and included descriptive statistics and analysis of variance (ANOVA) in terms of gender, age, relationship status, geographical location and relationship type. Consistent with previous findings, this research study found comparisons between South African and international online dating relationships. Looking at the online dating persona profile, significant findings pertaining to sociodemographic traits, Big Five personality traits, level of emotional intelligence and life effectiveness skills. Thesis deals with measurement of positioning in marketing, specifically the method of semantic differential. The main objective of this work is to verify how it changes the semantic profile of selected wine samples tasted by young wine consumers in changing contextual conditions of this experiment. The experiment consists of two separate measurements, during which there is a change in the contextual conditions. The theoretical part is description of his positioning and measurement, which continuously follows the method of semantic differential, where he presented Seman-tick differential as a tool by which we can measure and record the positioning of his value by creating semantic profiles. The practical part explains the connection and answer research questions determined through experiment, where it was shown that the change in the contextual conditions tasted wine samples young wine consumers, changes the semantic profile of the wine samples.

Metabolite profiling of defence-related secondary metabolites in tobacco cells, in response to ergosterol, a steroid from fungal membranes

M.Sc. / Plants have the ability to continuously respond to various stimuli which alter their physiology, morphology and development. These stimuli may be abiotic or biotic and range from essential to toxic in their effects. One of these stimuli is a steroid from fungal membranes, ergosterol (C28H44O), which does not occur in plants. Ergosterol acts as a pathogen-associated molecular pattern molecule and triggers defence mechanisms in plants, characterised by highly regulated and interrelated events that include the elicitation of the oxidative burst and expression of a number of defencerelated genes. However, the ergosterol-induced global cellular reprogramming of the host has not been fully investigated in all aspects. No metabolomic study has previously been conducted to elucidate, for instance, the effect of ergosterol on plant metabolism. A clear and broader understanding of the molecular mechanisms involved in plant : ergosterol interactions is of paramount importance, for it would open up possibilities of developing novel, more effective and sustainable strategies to control or eradicate fungal diseases in plants. In plants, the metabolome is a compilation of all primary and secondary metabolites. The latter are the final recipients of genetic information, and their levels can influence gene expression and protein stability. Metabolite patterns reveal the actual cellular dynamic environment. Hence, qualitative and quantitative measurements of extra- and intracellular metabolites yield insights into the cellular processes that control the biochemical phenotype of the cell, tissue or whole organism. Metabolomics, the most recent of the ‘omics’ approaches, is the holistic analysis of metabolites present within a biological system under specific physiological conditions. In the present study a metabolomic approach was used to elucidate and analyse changes in the metabolism of tobacco (Nicotiana tabacum) cells following ergosterol treatment. Special attention is given to sesquiterpenoids since the antimicrobial compounds (phytoalexins) isolated from plants within the Solanaceae are mostly bicyclic sesquiterpenoids. Suspension of tobacco cells were treated with different concentrations (0 - 1000 nM) of ergosterol and incubated for different time periods (0 - 24 h). A viability assay, based on the ability of viable cells to reduce 2,3,5- triphenyltetrazolium chloride (TTC), was used to determine whether cell death occurred due to ergosterol treatment. No loss of cell viability was observed over the concentration range and time periods used in this study, indicating that the observed responses were due to the treatment alone and possible secondary responses due to cell death could be excluded. Intracellular metabolites were extracted with two methods: a selective dispersive liquid-liquid micro extraction and a general methanol extraction. Chromatographic techniques (TLC/HPTLC, GC-FID, GC-MS, GC×GC-TOF-MS, UPLC-MS) and 1H NMR spectroscopy were used for quantitative and qualitative analyses. Multivariate data analyses (PCA and OPLS-DA models) were used to extract interpretable information from the multidimensional data generated from the aforementioned techniques.

Efeitos da mutação mdx no background 129/Sv / Effects of the mdx mutation on the 129/Sv background

O camundongo mdx, modelo murino para a Distrofia Muscular de Duchenne (DMD) possui uma mutação de ponto no gene da distrofina que resulta na ausência da proteína no músculo, porém seu fenótipo é brando o que o torna um bom modelo genético e molecular, mas não um bom modelo funcional. Esperando obter um modelo para DMD que tivesse um fenótipo mais fiel ao apresentado pelos pacientes humanos, optou-se por transferir a mutação mdx para o background 129/Sv. Através de cruzamentos sucessivos foram obtidas 3 gerações de animais mdx com background 129/Sv (mdx129) e cada geração foi avaliada funcionalmente por 6 meses. Desde a primeira geração é possível observar que os animais mdx129 são mais fortes do que os mdx originais em background C57BL (mdxC57BL), sendo o oposto do esperado no início dos experimentos. O estudo então foi redirecionado para tentar entender o motivo dessa melhora. Em relação ao padrão histológico, em geral há diferenças entre o mdxC57BL e mdx129. Observa-se também que os animais mdx129 entram no processo de degeneração mais tardiamente que os animais mdxC57BL e seu processo de regeneração se estende por mais tempo. Através de estudos de microarray foi possível observar que os animais 129/Sv apresentam poucos genes diferencialmente expressos (GDEs) em relação aos animais C57BL, portanto os dois backgrounds são muito semelhantes. O mdxC57BL apresenta muito mais GDEs em relação ao seu selvagem (C57BL) do que o mdx129 em relação ao 129/Sv, entretanto, ambos os modelos apresentam mais genes superexpressos do que subexpressos, indicando que as alterações distróficas e regenerativas estão mais associadas com a ativação do que a repressão de genes. Quando os GDEs de ambos os modelos de mdx são distribuídos em categorias funcionais, há o predomínio de genes ligados ao sistema imune e quando essa categoria é omitida para melhor visualização das restantes, observa se que ambos os modelos apresentam categorias funcionais semelhantes, porém com proporções diferentes. No modelo mdx129 se destaca a diminuição da participação da categoria de rota endo/exocítica (tráfego de vesículas) e homeostase e aumento da participação das categorias de matiz extracelular e atividade enzimática. Cada modelo apresenta genes exclusivos, destacando os genes SPP1 e IL1RN na comparação 129/Sv x mdx129F3. O gene SPP1 codifica a proteína osteopontina (OPN) e o polimorfismo rs28357094 neste gene é utilizado como biomarcador de prognóstico para DMD. O papel da OPN na progressão da distrofia não é bem conhecido. Alguns estudos afirmam que a ausência dessa proteína melhora a força muscular de camundongos mdx, enquanto outros apontam que sua participação é necessária para a regeneração muscular. Assim sendo, mais estudos serão necessários para verificar qual seria a via responsável pela melhora fenotípica do modelo mdx129. Já o gene IL1RN codifica a proteína IL-1Ra, a qual é um antagonista de interleucina 1 (citocina pró-inflamatória e pró fibrótica). Portanto o aumento da expressão do gene de seu antagonista sugere que os animais mdx129F3 podem estar mais protegidos do processo inflamatório causado por essas moléculas. Quando analisadas as listas filtradas para músculo esquelético das comparações C57BL x mdxC57BL e 129/Sv x mdx129F3 para a formação de vias metabólicas, foi gerada apenas uma via em ambas as comparações com número relevante de moléculas. A via gerada pela análise da lista C57BL x mdxC57BL possui mais moléculas do que a via gerada pela analise da lista 129/Sv x mdx129F3, porém, todas as moléculas presentes nesta via, estão presentes na via C57BL x mdxC57BL, indicando que mesmo com número diferente de moléculas envolvidas, os genes participam das mesmas vias. Tanto a comparação de cada geração de mdx129 com o 129/Sv como a comparação das gerações entre si mostram que os efeitos da mudança de background estão presentes desde a primeira geração e não se alteram significativamente com os cruzamentos sucessivos. / The mdx mouse, murine model for Duchenne Muscular Dystrophy (DMD) has a point mutation in the dystrophin gene that results in the absence of the protein in the muscle, however its phenotype is mild, which makes it a good genetic and molecular model, but not a good functional model. Hoping to obtain a model for DMD with a phenotype that is more similar the patients\', it was chosen to transfer the mdx mutation to the 129/Sv background. Through successive breedings, 3 generations of mdx animals with 129/Sv background were obtained and each generation was functionally evaluated for 6 months. Since the first generation it is possible to observe that the mdx129 animals are stronger than the original mdx with C57BL background. The results were the opposite of what was expected in the beginning of the experiments, therefore the study was redirectioned to try to understand the reason of the improved phenotype. About the general histological pattern, there are differences between mdxC57BL and mdx129. It can be observed that the mdx129 animals enter the degenerative process later than the mdxC57BL animals and the regenerative process lasts longer. Through microarray studies it was possible to observe that the 129/Sv animals present few differentially expressed genes (DEGs) in comparison to the C57BL animals; therefore both backgrounds are very similar. The mdxC57BL presents many more DEGs in comparison to C57BL than mdx129 in comparison to 129/Sv, however both models present more super expressed genes than sub expressed, indicating that the dystrophic and regenerative alterations are more associated to the activation rather than the repression of genes. When the DEGs of both mdx models are distributed in functional categories, there is the predominance of genes related to the immune system and when this category is omitted for the better visualization of the remaining, it can be observed that both models present similar functional categories, but with different proportions. In the mdx129 model we can highlight the decrease in participation of the endo/exocytic pathway (vesicle traffic) and homeostasis categories, and increase in participation of the extracellular matrix and enzymatic activity categories. Each model presents exclusive genes, highlighting SPP1 and IL1RN in the comparison 129/Sv x mdx129F3. SPP1 encodes the protein osteopontina (OPN) and the polymorphism rs28357094 in this gene is used as a DMD prognostic biomarker. The role of OPN in the dystrophy progression is not well known. Some studies claim that the absence of OPN increases the muscle strength of the mdx mouse, while others indicate that its participation is necessary to muscle regeneration. More studies are needed to ascertain what pathway is responsible for the phenotypic improvement of the mdx129 model. The IL1RN gene encodes the protein IL-1Ra, and interleukin 1 antagonist, which is a pro-inflammatory and pro-fibrotic cytokine. Therefore, the increase in the expression of its antagonist suggests that the mdx129F3 animals may be more protected from the inflammatory process caused by these molecules. When the filtered lists for skeletal muscle of the comparisons C57BL x mdxC57BL e 129/Sv x mdx129F3 were analyzed for the formation of metabolic pathways, only one pathway was generated in both comparisons. The pathway generated in the analysis C57BL x mdxC57BL has more molecules that the one generated by the 129/Sv x mdx129F3 list, but all molecules present in the latter are also present in the former, indicating that even with different numbers of molecules involved, the genes participate in the same pathways. The comparisons of each generation of mdx129 with the 129/Sv and the comparison of the generations among each other show that the effects of the background change are present since the first generation and are not altered with the successive breedings.

Estudo dos genes de resistência e integrons em cepas de Salmonella multidrogas resistentes (MDR) isoladas em lifonodos de suínos

Orientador: João Pessoa Araujo Junior / Resumo: A segurança dos alimentos é essencial para o desenvolvimento da sociedade e as doenças transmitidas por alimentos (DTAs) acometem milhares de pessoas todos os anos. Dentre os agentes de DTA, Salmonella se destaca como a principal causa de hospitalizações e mortes. A carne suína é uma importante fonte de nutrientes, sendo a mais consumida no mundo. Vários relatos apontaram o isolamento de Salmonella multidrogas resistentes (MDR) associados com a cadeia produtiva dos suínos, e a ocorrência de elementos que possibilitam a transferência de genes de resistência entre populações bacterianas estão diretamente relacionados a essa característica. O presente trabalho objetivou determinar os genes associados com resistência a antimicrobianos de 9 isolados de Salmonella MDR provenientes de linfonodos mesentéricos de suínos, além de aplicar um protocolo in silico para predizer a localização de cada gene (plasmidial ou cromossomal) e detectar e caracterizar a presença de Integrons de resistência. Diversos genes de resistência para a mesma classe de antimicrobianos foram encontrados em cada cepa, e a localização presumida dos genes foi heterogênica, com exceção para os genes de resistência a sulfonamidas, todos classificados como plasmidiais. Cinco cepas apresentaram integrons classe 1, agrupados em três diferentes perfis. A comparação destes perfis com sequências do GenBank revelou que bactérias de diversas espécies, isoladas em diversos países e proveniente de diferentes fontes, inclusive... Among the main foodborne disease agents, Salmonella stands out as the most associated with hospitalizations and deaths. Pork products are an important source of nutrients for population and the most consumed meat in the world. Several reports reveal the presence of Multidrug Resistant (MDR) Salmonella strains in the pork production chain, and the occurrence of elements that allows transference of resistance genes between bacterial populations is related to this characteristic. The present study aimed to determinate the genetic bases associated with Antimicrobial Resistance (AMR) in nine MDR Salmonella strains isolated from swine mesenteric lymph-nodes, beside applying an in-silico protocol to determinate location of the AMR genes (chromosomal or plasmidial) and characterize resistance integrons. Different resistance genes for the same antimicrobial class were present in each strain, and gene localization was heterogenous, except for sulphonamides resistance genes, where all were classified as plasmidial. Five isolates harbored Class 1 ix integrons, which were classified in three distinct Integron Profiles (IPs). Comparison of IPs with sequences of GenBank database revealed that different species of bacteria, from different countries and isolation sources, including human clinical isolates had the same integrons. Stereotypical Science: Exploring High School Occupational Preferences for Science by Sex, Personality, and Cognitive Ability

Circumscription and Compromise theory suggests self-concept and sex stereotype explain occupational preferences, including preferences for science, technology, engineering and mathematics (STEM). Support exists for sex differences between males and females in both science degrees and science careers. The main thrust of observed sex differences in science lies in the development of occupational interest, as it has been suggested females are encouraged away from science due to stereotypes and social pressure. The present study evaluates high school juniors and seniors (n = 295) to explore their preference for science as indicated by science motivation, attitude, academic experience, and interest. Latent Profile Analysis was used to model profiles of preferences for science with a person-centered approach. Then, the impact of self-concept variables was explored and four profiles of science interest were identified. Sex differences were identified based on science interest, but were not always in favor of males. Covariate analysis indicates vocabulary ability and personality as significantly different for students in the high science interest profile. Implications of these results and future research directions are discussed.
