Synthèse de nano-vecteurs dérivés des polydiacétylènes pour la co-délivrance d’un ARN interférent et d’un anticancéreux / Synthesis of polydiacetylenic nanovectors for intelligent co-delivery of siRNA and anticancer drug

Ripoll, Manon 11 December 2017 (has links)
In the nanomedecine field, a new approach consists in developing synthetic vectors able to co-deliver into a cancer cell, an antitumoral drug and siRNAs that target protein(s) involved in MDR. The work described in this manuscript was dedicated to the development of micellar nanovectors for the intracellular co-delivery of these two therapeutic agents. The first part details the synthesis and the formulation of nanometric photopolymerized diacetylenic micelles adapted for the delivery and intracellular release of the siRNA. Then, the encapsulation and delivery properties of these micelles, bearing histidine polar heads have been investigated in vitro and in vivo for the application of combination therapy. Finally, the last part presents the functionalization by electrostatic interaction of these cationic vectors with antibodies, priorly modified by anionic oligonucleotides. This original and versatile system allowed achieving an active targeting of tumoral cells.

Oncolytic Viral and Immunotherapy Models Combined with Strategies to Ameliorate Cancer Burden

January 2016 (has links)
abstract: Combination therapy has shown to improve success for cancer treatment. Oncolytic virotherapy is cancer treatment that uses engineered viruses to specifically infect and kill cancer cells, without harming healthy cells. Immunotherapy boosts the body's natural defenses towards cancer. The combination of oncolytic virotherapy and immunotherapy is explored through deterministic systems of nonlinear differential equations, constructed to match experimental data for murine melanoma. Mathematical analysis was done in order to gain insight on the relationship between cancer, viruses and immune response. One extension of the model focuses on clinical needs, with the underlying goal to seek optimal treatment regimens; for both frequency and dose quantity. The models in this work were first used to estimate parameters from preclinical experimental data, to identify biologically realistic parameter values. Insight gained from the mathematical analysis in the first model, allowed for numerical analysis to explore optimal treatment regimens of combination oncolytic virotherapy and dendritic vaccinations. Permutations accounting for treatment scheduled were done to find regimens that reduce tumor size. Observations from the produced data lead to in silico exploration of immune-viral interactions. Results suggest under optimal settings, combination treatment works better than monotherapy of either type. The most optimal result suggests treatment over a longer period of time, with fractioned doses, while reducing the total dendritic vaccination quantity, and maintaining the maximum virotherapy used in the experimental work. / Dissertation/Thesis / Doctoral Dissertation Applied Mathematics for the Life and Social Sciences 2016

Russellian Monism and Mental Causation

January 2018 (has links)
abstract: Russellian monism is a promising theory of consciousness that attempts to capture the strengths of both physicalism and dualism while avoiding their weaknesses. I begin by showing that the Russellian monist’s chief anti-physicalist rival, emergentism, is unable to give an adequate solution to the exclusion problem. Specifically, they fall prey to what I call “the opacity problem.” That is, because the emergentist is committed to there being both a sufficient physical cause and a sufficient mental cause for our actions, it is unclear what difference the mental cause is making in bringing about the effect. This is because, for the physical cause to truly be a sufficient cause, it must be sufficient to bring about the effect as it occurred. This distinguishes mental overdetermination from non-problematic kinds of overdetermination (like double rock throwing cases). I then show how the constitutive Russellian monist is able to avoid the exclusion problem, while the emergent Russellian monist faces similar opacity problems to emergentism. Finally, I give an account of how the constitutive Russellian monist can give a response to the strongest objection against—the subject-summing problem. I argue that we only have translucent access to our conscious states—that is, only part of the essential nature of the state is revealed to us through introspection. I then argue that we have reason to think that part of the essential nature of the conscious state not revealed to us is involved in subject-summing. / Dissertation/Thesis / Masters Thesis Philosophy 2018

Essência sobre a forma na interface jurídico-contábil: o ágio interno nas demonstrações individuais / Substance over form in an accounting and legal interface: the goodwill in business combination under common control in individual statements

Eduardo Guimarães Martinez 15 May 2015 (has links)
In the professional sphere as well as in the academy, business combinations require accountants, managers, economists, statisticians, lawyers and financiers due to the complexity and the high figures involved. However, relevant regulatory gap related to the recognition of eventual goodwill generated by transaction inside an economic group in the Individual Financial Statements provides thorny turmoil. In Brazil interpretations and even regulations have conflicted over the last few years. Further there is no international agreement regarding this issue. The question basically rests on the analogy of IFRS 3, CPC 15 in Brazil (addressed for business combination between third parties and Consolidated Statement). Both here and there, for different reasons, three possibilities for the Intragroup Goodwill are discussed: always recognize it, never recognize it, or the acclaimed \"it depends\". This theoretical and interdisciplinary study made use of theory and doctrine from Corporate Law and Accounting. It was also supported by economic theories, elements from General Theory of Civil Law and Obligation Law, as well as the Brazilian and international legislative standards. In the analysis of these 3 views, through this accounting and legal perspective, we sought to deepen the concepts of Goodwill and Economic Groups, guided by the structuring parameters of the IFRS. As a result, the normative research concluded in favour of \"it depends\". The application of the analogy, when legitimacy of intragroup goodwill is verified, is established not on the simple check of stock control, but through the parties\' independence. In most cases, the controlling stock holder ends up directing the transaction. Nevertheless, mismatch can occur between the stock control and the effective management control (transaction\'s direction) resulting in cases of independent parties inside the group and, on the other hand, dependent parties in external relations.

The Pardo River Basin provides a good representation of the situation of the region, where forest fragmentation is a result of the haphazard process of the land use and occupation. The aim of this study was to define the priority areas to facilitate connectivity between forest fragments in order to forest restoration. Multicriteria approach with Weighted Linear Combination was used for combination of factors (landscape features). To establish the factors, their weights compensation (expressing the relative importance of factors in decision making) and the restrictions (restrictive category of characteristics) it was used to Participatory Technical and Analytic Hierarchy Process. The selected factors were: proximity of fragments of larger nuclear area, proximity to forest area, proximity to water, distance to urban centers; soil erodibility, and slope. The result obtained was a map of priority areas, with levels of priority represented on a continuous way, from 0 to 255. To facilitate the visualization and the analysis of results, the map was reclassified, with only five levels of priority. It was concluded, from the results, that the Multicriteria Evaluation ,in a Geographical Information Systems (GIS), is appropriate to the mapping of priority areas for forest restoration in river basins, the method of Weighted Linear Combination is flexible, easy to be implement, and allows the interaction of knowledge (researchers, analysts) and landscape features, determining the robustness of the solution for decision making; multicriteria analysis, based on Analytical Hierarchy Process (AHP), is essential in evaluating the factors, it uses a pair wise comparison between factors to determine the relative importance of each one, reducing subjectivity and subsidizing the creation of better scenarios for the decision maker; and the map of adequacy, for forest recovery River Basin Pardo-SP... Reading practices in two urban multi-grade foundation phase classes

Sampson, Coleen Anthea January 2015 (has links)
A full dissertation submitted in fulfilment of the requirements for the degree of Masters in Education Presented to the Faculty of Education and Social Sciences 2015 / In Africa, throughout the history of schooling, many teachers have been confronted by the demanding situation of teaching two or more year groups in the same classroom although data on this multi-grade phenomenon is scarce. Although reading is a fundamental competency and the core of our curriculum, a gap exists in practice. In multi-grade classes the gap is wider as a result of the different grades and varying abilities within the grades. The present study was motivated by the researcher’s concern for the status of reading in all Foundation Phase classes. This study answers one main question: How do teachers in two urban multi-grade classrooms teach reading in the Foundation Phase? The two sub-questions are: What are the current reading practices in urban multi-grade classrooms in the Foundation Phase? What challenges do teachers of urban multi-grade classes face when teaching reading in the Foundation Phase? The conceptual framework that was central to answering the two sub questions includes four theorists namely: Lave and Wenger’s (1991) Vygotsky’s (1978); Bronfenbrenner’s (1990) and finally Piaget’s (1972) stages of cognitive development including pre-operational stage and concrete operational stage. The literature review highlights the physical setting of the multi-grade classrooms, debates the advantages and limitations of urban multi-grade teaching, compares the National Curriculum Statement (NCS) 2005 and the Curriculum Assessment Policy Statement (CAPS) reading curriculum, briefly deliberates the stages of reading development and finally discusses the variety of reading practices. A qualitative interpretive case study research design was formulated to explore the complex phenomenon of urban multi-grade reading practices in the Foundation Phase. The first research question explored the reading practices found in two urban multi-grade classes, and the findings include four themes which are: stories, vocabulary, comprehension and reading. The second sub-question, focussed on the challenges the urban multi-grade teachers experienced. In conclusion this study reveals that reading can be taught successfully in urban multi-grade classes. Teaching reading in multi-grade classes may foster the emotional, intellectual, social and academic well-being of learners. Secondly although the two urban multi-grade teachers faced many challenges, with the necessary support structures in place, these challenges could be minimized.

A strategy to assist rural multigrade schools to reduce the dropout rate experienced in high schools

van Niekerk, Susanna Elizabeth January 2014 (has links)
Thesis submitted in fulfilment of the requirements for the degree Master of Education in the Faculty of Education and Social Sciences at the Cape Peninsula University of Technology 2014 / Educationists and political leaders are concerned about the high dropout rates experienced in South African high schools. For the purposes of this research, “dropout” is defined as “one who has not graduated from high school”. Research indicates that the high school dropout rates, both in South Africa and globally, culminate in challenges for the school, the community and society. The multigrade primary school environment sees learners doing well academically, but when these learners move on to high school, the dropout rates increase for these learners. The purpose of this study is two-fold: to determine the reasons for dropout of learners who have ably and successfully completed their primary education within the multigrade system; and to propose a strategy to assist rural multigrade primary schools in reducing the foreseen dropout rates in high schools. A sequential explanatory mixed-method approach was designed in order to determine what strategy could assist rural multigrade schools in Circuit 2 of the West Coast Education District to prevent the dropout rate experienced in high schools. During the preliminary research a scrupulous literature study was done, to determine global trends and to determine which current intervention programmes exist. The quantitative phase of this study was conducted first and consisted of a content analysis of school documents to determine which learners did not complete high school. The qualitative phase followed and the data was collected through face-to-face interviews with principals of rural multigrade primary schools, and learners who had dropped out. This was done in order to determine the perceptions of the principals, and the former learners who had dropped out of the schooling system. This research elucidates the challenges – the inexorable odds – that these multigrade learners have had to overcome in order to complete Grade 12. In closing, it proposes pragmatic strategies which may decrease the high dropout rates that learners, from a multigrade primary setting, experience in future.
