DNA Haplotypes Determination for Members of Families with Phenylketonuria (PKU) / DNA Haplotypes Determination

Phenylalanine hydroxylase deficiency causes phenylketonuria (PKU) in humans. PKU is a recessive genetic disease that affects 1 in 10000 births among the Caucasian population. Its gene locus is highly polymorphic in its DNA sequence among different individuals and patients with PKU. DNA polymorphisms at the PAH gene locus are used to obtain haplotypes through restriction enzyme analysis. So far forty-six distinct RFLP haplotypes have been discovered in the human population. In theory, 384 distinct RFLP haplotypes can exist. This project is to develop a program to assist the geneticists by obtaining haplotypes for each member of the PKU family. It uses information obtained from digestion of the DNA samples from the family members with the restriction enzymes. The restriction enzymes employed for this purpose are Pvuii, Bglii, EcoRI, Mspi, Xmni, Hindiii, and EcoRV. The program "PKU" generates all possible haplotypes for each member of the PKU family. The generated haplotypes may include haplotypes from the forty-six defined haplotypes list or from the 338 other haplotypes that may fit the description from the restriction enzyme analysis. The program then carries out an elimination phase during which the "extra" haplotypes that had been generated for the family members but whose presence was not supported by the data from the other family members are eliminated from the individuals' haplotype lists. The remaining haplotypes are then used to determine a sibling's carrier status of the PKU disease, i.e., whether or not a sibling is a carrier of the PKU disease. / Thesis / Master of Science (MS)

Family Stress Factors Across Three Family Types

This study investigated the difference in stress levels of stress factors according to the structure or type of family. The relatedness of the ranking of stress factors within and across three family types and the relationship between level of stress and number of years in current family type were also examined. Important aspects of this study were using three family types, using families seeking counseling as the subjects, and investigating numerous stress factors across family types. These research techniques avoided the limitations of previous research which investigated only one family type, thus isolating special stress issues for a certain family type where those special issues actually might not differentiate among family types. Also, considering numerous stress factors at one time, rather than only a few factors, indicated relative levels of stress as well as absolute stresses that families are likely to experience. Targeting families who had sought counseling should give counselors a more realistic view of clients and their problems.

AN ATTITUDINAL STUDY OF SPOUSES OF PERSONS WITH APHASIA

The purpose of this investigation was to describe the attitudes of individuals who were spouses of patients with aphasia. Using modified Q-methodology, 15 spouses of fluent aphasic patients, 15 spouses of nonfluent aphasic patients and 30 matched controls completed a 70-item Q-sort constructed specifically for this study. The spouses of nonfluent aphasic patients were found to have a greater number of negative attitudes toward their spouses than were the spouses of fluent aphasic patients. The spouses of patients in both aphasia groups were found to have a greater number of negative attitudes toward their spouses than the matched controls. In addition, six factors (Compliance, Desirability, Egocentricity, Independence, Maturity, and Sociability) representative of the spouse-patient relationship were identified that incorporated the most commonly held attitudes by the spouses of patients in both aphasia groups.

Tiger; a stage play, and a reflective essay detailing the writing process

This thesis includes a full length play and a separate section describing the creation of this play. The play depicts family members struggling with the direct and indirect ramifications of alcoholism, depression, and suicide. The play is composed of two acts; act one contains eight scenes, and act two contains six scenes. It is set in the 1950s and 1960s and takes place in various areas of the family home, at a wedding reception, and at a funeral. The essay section includes a description of the process, a record of changes in the play's direction, notations of personal discoveries, and a self evaluation of the strengths and weaknesses of the play.
