A Radical Approach to Syntheses and Mechanisms

The critically important nature of radical and radical ion mechanisms in biology and chemistry continues to be recognized as our understanding of these unique transient species grows. The work presented herein demonstrates the versatility of kinetic studies for understanding the elementary chemical reactions of radicals and radical ions. Chapter 2 discusses the use of direct ultrafast kinetics techniques for investigation of crucially important enzymatic systems; while Chapter 3 demonstrates the value of indirect competition kinetics techniques for development of synthetic methodologies for commercially valuable classes of compounds. The mechanism of decay for aminyl radical cations has received considerable attention because of their suspected role as intermediates in the oxidation of tertiary amines by monoamine oxygenases and the cytochrome P450 family of enzymes. Radical cations are believed to undergo deprotonation as a key step in catalysis. KIE studies performed by previous researchers indicate N,N-dimethylaniline radical cations deprotonate in the presence of the bases acetate and pyridine. By studying the electrochemical kinetics of the reaction of para substituted N,N-dimethylaniline radical cations with acetate anion, we have produced compelling evidence to the contrary. Rather than deprotonation, acetate reacts with N,N-dimethylaniline radical cation by electron transfer, generating the neutral amine and acetoxyl radical. Transport properties of reactants and solvent polarity changes were investigated and confirmed not to influence the electrochemical behavior forming the basis for our mechanistic hypothesis. To reconcile our conclusion with earlier results, KIEs were reinvestigated electrochemically and by nanosecond laser flash photolysis. Rather than a primary isotope effect (associated with C-H bond cleavage), we believe the observed KIEs are secondary, and can be rationalized on the basis of a quantum effect due to hyperconjugative stabilization in aromatic radical cations during an electron transfer reaction. Product studies performed by constant potential coulometry indicate N,N-dimethylaniline radical cations are catalytic in carboxylate oxidations. Collectively, our results suggest that aminyl radical cation deprotonations may not be as facile as was previously thought, and that in some cases, may not occur at all. Interest in design and synthesis of selenium containing heterocycles stems from their ability to function as antioxidants, anti-virals, anti-inflammatories, and immunomodulators. To establish synthetic feasibility of intramolecular homolytic substitution at selenium for preparation of selenocycles, we set out to determine what factors influence cyclization kinetics. A series of photochemically labile Barton and Kim esters have been syntheisized and employed as radical precursors. The effect of leaving radical stability on kinetics has been investigated through determination of rate constants and activation parameters for intramolecular homolytic substitution of the corresponding radicals via competition experiments. Notable leaving group effects on measured kinetic parameters show more facile reactions for radical precursors with more stable leaving radicals. Moreover, cyclizations to form six-membered (as opposed to five- membered) ring systems exhibited order of magnitude decreases in rate constants for a given leaving radical. Our results are congruent with expectations for radical cyclizations trends for the varied experimental parameters and suggest homolytic substitution affords a convenient means for synthesis of selenocycles. / Ph. Studies of particle and atom manipulation using free space light beams and photonic crystal fibres

Light can exert optical forces on matter. In the macroscopic world these forces are minuscule, but on the microscopic or atomic scale, these forces are large enough to trap and manipulate particles. They may even be used to cool atoms to a fraction of a degree above absolute zero. This thesis details a number of experiments concerned with the optical manipulation of atoms and micron-size particles using free space light beams and photonic crystal fibres. Two atom guiding experiments are described. In the first experiment, a spatial light modulator is used to generate higher blue-detuned azimuthal Laguerre-Gaussian LG) beams, which are annular beams with a hollow core. These LG beams are then used to guide laser cooled rubidium-85 atoms within the dark core over a distance of 30 mm. The second atom guiding experiment involves attempting to guide laser cooled and thermal rubidium atoms through a hollow-core photonic crystal fibre using red-detuned light. Hollow-core photonic crystal fibres are fibres that are able to guide light with low attenuation within a hollow core. For this experiment a hot wire detection system was designed, along with a number of complex vacuum systems. The first dual-beam fibre trap for micron-size particles constructed using endlessly single-mode photonic crystal fibre (ESM-PCF) is described. The characteristics of dual-beam fibre traps are governed by the fibres used. As ESM-PCF has considerably different properties in comparison to conventional single- or multimode fibres, this dual beam ESM-PCF trap exhibits some novel characteristics. I show that the dual beam ESM-PCF trap can form trapping, repulsive and line potentials; an interference-free ‘white light’ trap; and a dual-wavelength optical conveyor belt.

Structural Investigations of the Italian Trap Allochthon, Redington Pass, Pima County, Arizona

Italian Trap Allochthon is a rare upper-plate exposure of Paleozoic metasedimentary and Precambrian to Tertiary crystalline tectonites in the Santa Catalina-Rincon metamorphic core complex. Elsewhere in the complex, metasedimentary tectonite is usually restricted to an autochthononous position. The internal structures of the allochthon consist of numerous low-angle faults, tear faults, and overturned asymmetric and upright folds. Close association of the low-angle faults and asymmetric folds, and vergence of the folds, indicates that these folds were formed during westward transport along the low-angle faults. The structures of the allochthon are truncated and rotated to the northeast by a listric (?) normal fault. The probable shape of the fault surface, together with the northeastward rotation of the internal structures, suggests translation of the allochthon from the northeast to the southwest. The fact that metasedimentary tectonites are found in upper- plate position indicates that the listric (?) normal faulting post-dates the metamorphism of the Paleozoic and Mesozoic strata. Metamorphism in turn was part of the development of the Santa Catalina-Rincon metamorphic core complex. It is inferred that the Italian Trap Allochthon was emplaced in the final stages of profound regional extension which prevailed during the mid-Tertiary in southern Arizona.

Essais sur des questions internationales en économie des ressources naturelles

Cette thèse s'articule autour de trois essais portant sur des questions internationales en économie des ressources naturelles. Le premier essai examine la production et l'échange des ressources non-renouvelables dans un modèle spatial et souligne le rôle de la superficie des pays et du coût des transports dans la détermination du sens de l'échange. Le deuxième essai considère le tarif d'extraction de la rente de rareté liée aux ressources naturelles non-renouvelables avec le modèle spatial développé dans premier essai. Le cadre spatial (plus général) permet de représenter des pays qui sont à la fois importateurs et producteurs de la ressource, ce qui n'est pas possible dans les modèles traditionnels de commerce international où les pays sont traités comme des points (sans dimension). Le troisième essai aborde la question des droits de propriétés sur les zones maritimes et examine l'allocation d'une population de pêcheurs entre les activités productives et non-productives dans une communauté côtière. Le premier chapitre propose un modèle spatial de commerce international des ressources non-renouvelables. Le cadre spatial considère explicitement la différence de taille géographique (superficie) entre les pays et permet ainsi de tenir compte du fait que les gisements naturels et leurs utilisateurs soient dispersés dans l'espace, même à l'intérieur d'un pays. En utilisant un modèle spatial à la Hotelling, nous examinons l'évolution dans le temps du sens de l'échange entre deux pays (ou régions) qui diffèrent du point de vue de leur technologie de production, de leur superficie et de leur dotation en gisement d'une ressource naturelle non-renouvelable. Le chapitre met en évidence le rôle de la taille géographique dans la détermination du sens de l'échange, à côté des explications traditionnelles que sont l'avantage comparatif et les dotations des facteurs. Notre analyse est fondamentalement différente des autres contributions dans la littérature sur le commerce international des ressources naturelles parce qu'elle souligne l'importance de la taille géographique et du coût de transport par rapport à d'autres facteurs dans la détermination des flux de ressource à l'équilibre. Le coût unitaire de transport joue un rôle capital pour déterminer si la différence de superficie entre les pays influence le sens de l'échange à l'équilibre plus que les autres facteurs. Le chapitre discute aussi du caractère régional des échanges qui a été observé pour certaines ressources telles que le minerai de fer et la bauxite. Le chapitre deux aborde la question de la répartition de la rente de rareté liée aux ressources naturelles non-renouvelables entre les pays producteurs et les pays consommateurs. Cette question a été abordée dans la littérature sous une hypothèse quelque peu restrictive. En effet, dans la plupart des travaux portant sur ce sujet le pays importateur est automatiquement considéré comme dépourvu de gisement et donc non producteur de la ressource. Pourtant la réalité est qu'il existe des ressources pour lesquelles un pays est à la fois producteur et importateur. Le cadre d'analyse de ce second essai est le modèle spatial développé dans le premier essai, qui permet justement qu'un pays puisse être à la fois importateur et producteur de la ressource. Le pays importateur détermine alors simultanément le tarif optimal et le taux d'extraction de son propre stock. Nous montrons que le tarif optimal croît au taux d'intérêt et de ce fait, ne crée aucune distorsion sur le sentier d'extraction de la ressource. Le tarif optimal permet de récupérer toute la rente lorsque le pays exportateur ne consomme pas la ressource. Néanmoins, la possibilité pour le pays exportateur de consommer une partie de son stock limite la capacité du pays importateur à récupérer la rente chez le pays exportateur. La présence de gisements de la ressource dans le pays importateur réduit la rente du pays exportateur et de ce fait renforce la capacité du pays importateur à récupérer la rente chez le pays exportateur. Le tarif initial est une fonction décroissante du stock de ressource dans le pays importateur. Cet essai aborde également la question de la cohérence dynamique du tarif obtenu avec la stratégie en boucle ouverte. Le troisième chapitre examine un problème d'allocation de l'effort entre les activités productives (par exemple la pêche) et les activités non productives (par exemple la piraterie maritime) dans une population de pêcheurs. La répartition de la population entre les activités de pêche et la piraterie est déterminée de façon endogène comme une conséquence du choix d'occupation. Nous établissons l'existence d'une multiplicité d'équilibres et mettons en évidence la possibilité d'une trappe de piraterie, c'est-à-dire un équilibre stable où une partie de la population est engagée dans les actes de piraterie. Le modèle permet d'expliquer l'augmentation significative des attaques de piraterie dans le Golfe d'Aden au cours des dernières années. Le chapitre discute aussi des différents mécanismes pour combattre la piraterie et souligne le rôle crucial des droits de propriété. / This thesis consists of three essays on international issues in natural resource economics. The first essay proposes a spatial model of trade in exhaustible resources and emphasizes the role of geographical size and transport costs in the determination of trade patterns. The second essay considers the rent-extracting tariff in a spatial (more general) framework in which the importing country can be simultaneously a producer and an importer of the resource, a feature which is not possible in the traditional trade model, where countries are assumed dimensionless. The third essay tackles the issue of property rights in maritime zones and examines the allocation of a population of fishermen between productive and unproductive activities in a coastal community. The first chapter proposes a model of trade in exhaustible resources that explicitly accounts for the fact that countries have different geographical sizes while resource sites and their users are spatially distributed, even within a country. Using a spatial model à la Hotelling, we examine the evolution over time of the pattern of trade between two countries (or regions) which differ in terms of their technology, their geographical size, and their endowment of some nonrenewable natural resource. The model emphasizes the importance of geographical size in determining trade patterns besides the traditional explanations of comparative advantage and factor endowments. Indeed, three forces influence the direction of international trade in the presence of transport costs. The analysis fundamentally differs from other contributions in the natural resource literature because it emphasizes the importance of geographical size and of transport cost relative to other factors in the determination of the equilibrium resource flows. The unit cost of transport is shown to play a decisive role in determining whether the international asymmetry in terms of geographical sizes of countries has a greater influence than other factors on the equilibrium pattern of trade. The chapter also discusses the regional character of trade which has been observed for some resources such as iron ore and bauxite. Most findings in the literature on tariff and exhaustible resources have been derived under a serious abstraction. Indeed, virtually all contributions on that issue have assumed that no stocks of the resource are available within the importing country's borders and therefore the importing country is not itself a producer. Reality is in fact quite different: there are many instances of countries that are simultaneously importers and producers of a natural resource. The second chapter makes use of the spatial trade model of chapter one to depart from the usual assumption and allow the importing country to have access to a stock of the resource of its own and to determine simultaneously the optimal tariff and the rate of depletion of its own stock. The optimal tariff is shown to increase at the rate of interest and is therefore nondistortionary. Moreover, the optimal tariff captures all the rent if the exporting country gets no utility from consuming the resource. Allowing the exporting country to consume the resource restricts the ability of the importer to capture all of the foreign rent. The presence of resource deposits in the importing country reduces the available rent to foreign producers and, in essence, reinforces the ability of the importer to capture the foreign rent. In effect, the initial tariff is shown to be a decreasing function of the initial resource stock in the importing country. The essay also discusses the time consistency of the open-loop tariff. The third chapter examines how agents in a coastal community allocate effort between productive (fishing) and unproductive (piracy) activities. The allocation of population between fishing activity and piracy attacks is determined endogenously as a consequence of the occupation choice. We prove the existence of multiple equilibria and emphasize the possibility of a piracy trap, that is a steady state equilibrium where part of the population is engaged in piracy acts. The chapter offers an explanation for the significant increase in piracy attacks in the Gulf of Aden in the recent years. The chapter also discusses different schemes in combating piracy and highlights the crucial role of property rights.

Single molecule studies of F1-ATPase and the application of external torque

F<sub>1</sub>-ATPase, the sector of ATP synthase where the synthesis of cellular ATP occurs, is a rotary molecular motor in its own right. Driven by ATP hydrolysis, direct observation of the rotation of the central axis within single molecules of F<sub>1</sub> is possible. Operating at close to 100% efficiency, F<sub>1</sub> from thermophilic Bacillus has been shown to produce ~40pN&dot;nm of torque during rotation. This thesis details the groundwork required for the direct measurement of the torque produced by F<sub>1</sub> using a rotary angle clamp, an optical trapping system specifically designed for application to rotary molecular motors. Proof-of-concept experiments will be presented thereby demonstrating the ability to directly manipulate single F<sub>1</sub> molecules from Escherichia coli and yeast mitochondria (Saccharomyces cerevisiae), along with activation of F<sub>1</sub> out of its inhibited state by the application of external torque. Despite in-depth knowledge of the rotary mechanism of F<sub>1</sub> from thermophilic Bacillus, the rotation of F<sub>1</sub> from Escherichia coli is relatively poorly understood. A detailed mechanical characterization of E.coli F<sub>1</sub> will be presented here, with particular attention to the ground states within the catalytic cycle, notably the ATP-binding state, the catalytic state and the inhibited state. The fundamental mechanism of E.coli F<sub>1</sub> appears to depart little from that of F<sub>1</sub> from thermophilic Bacillus, although, at room temperature, chemical processes occur faster within the E.coli enzyme, in line with considerations regarding the physiological conditions of the different species. Also presented here is the verification of the rotary nature of yeast mitochondrial F<sub>1</sub>. The torque produced by F<sub>1</sub> from thermophilic Bacillus, E.coli and yeast mitochondria is the same, within experimental error, despite their diverse evolutionary and environmental origins.

CHUDOBA A PŘEDLUŽENOST ČESKÝCH DOMÁCNOSTÍ / Poverty and overdebtedness of Czech households

The diploma work deals with the problem of poverty following from overdebtedness of Czech households. Poverty is defined as a strong social handicap leading up to a social exclusion. Overdebtedness means such a financial situation when households are not able to fulfill their financial obligations and thus it concerns especially households with low incomes. In the Czech Republic this fact has been distinguished since the year 2000. The topic of the diploma work points at the development of overdebtedness of Czech households and its evaluation, the size of this problem and the estimation of its further development. It analyses the threat to households leading to a debt trap. It deals with economic impact of state debtedness in connection with that of Czech households. Some attention is paid to the increasing unemployment as one of the strongest causes leading to a state of poverty and consequently to undesirable debtedness showing the evident link between unemployment and overdebtedness. At the same time, the emphasis is put onto individual responsibility and lifelong education as the prevention and solution to unemployment and doubtedness. The phenomenon of overdebtedness of Czech households is understood as a warning signal of the present.

Le développement de différentes techniques pour contrôler les degrés de liberté internes et externes des molécules et pour produire (ultra-) froide, piège des moléculaire ensembles ouvrir des voies différentes à la physique et la chimie dans le régime de basse température. Il s'agit notamment de nombreux territoires en physique comme, phases quantiques de la matière, traitement de l'information quantique, les froides collisions moléculaires, les chimies froides et aussi de divers tests de haute précision pour la physique fondamentale. Cette thèse décrit diverses expériences de guidage et de décélération des faisceaux supersoniques d'atomes paramagnétiques à l’aide de champs magnétique inhomogène dépendent du temps. Ces champs magnétiques inhomogènes ont été utilisés pour exercer une force sur les atomes ou les molécules paramagnétiques, qui résultent de l'effet Zeeman. Le principe du ralentisseur Zeeman nouvellement développé est de produire un déplacement tridimensionnel du piège magnétique, à la vitesse initiale du faisceau. Le contrôle de la dépendance temporelle du champ magnétique nous permet de contrôler la vitesse du piège magnétique co-mobile, procurant ainsi une décélération d'une classe de vitesse du faisceau supersonique. Le piège magnétique co- mobile est déduit à partir d'une onde magnétique mobile, offrant un minimum de distorsion du piège lors de sa propagation. Les propriétés transverses du piège sont réglables grâce à un champ magnétique transversal quadrupolaire, qui peut être ajusté indépendamment des propriétés de vitesses et l'accélération du piège. Une grande part du travail de thèse a été consacrée à la conception, la réalisation et la construction du montage expérimental, consistant en un jet supersonique et en un dispositif complexe de bobines pour réaliser l’onde magnétique progressive, formant un piège magnétique mobile. Le jet froid pulsé d'atomes métastables est produit par expansion supersonique à travers une valve refroidie à l'azote liquide, excités dans l'état métastable par une décharge électrique. Nous avons guidé le jet d'argon au travers d’un tube capillaire le guidage et la décélération ont été démontrés. Le piège magnétique mobile est formé par la combinaison d'un champ magnétique quadrupolaire et d'un champ magnétique axial modulé spécialement. Le champ quadrupolaire est continu et un gradient de champ est dirigé seulement dans la direction transverse du jet. Le circuit plan produit une onde magnétique sinusoïdale avec un gradient de champ dans la direction axiale. Avec l'électronique fabriquée au laboratoire, ou peut produire une onde magnétique progressive d'amplitude 0.69T (avec un courant AC de 300A) et de fréquence 40 kHz. On obtient ainsi une onde qui se déplace à une vitesse de 464m/s. Plusieurs expériences de principe ont été réalisées en utilisant le jet froid pulsé d'argon métastable. Nous avons étudié les propriétés de guidage du quadrupole pour divers courants et pour différents atomes (hélium et argon) et comparé les résultats aux prédictions théoriques de simulations numériques. Le jet d'argon métastable a été guidé en 3D à des vitesses variées (464m/s, 400m/s, 392m/s) avec un décélérateur de 28cm de long. La température observée du paquet guidé est de 100mK. L'expérience de décélération a été réalisée avec le jet d'argon métastable depuis la vitesse de 400m/s jusqu’à 370m/s et depuis la vitesse de 392m/s jusqu’à 365m/s. Les résultats expérimentaux sont comparés avec les simulations numériques. / The development of various techniques to control both the internal and external degrees of freedom of molecules and to produce (ultra-) cold, trapped molecular ensembles open various avenues to physics and chemistry in the low temperature regime. These include many territories in physics like, quantum phases of matter, quantum information processing, cold molecular scattering, cold chemistry and also various high precision tests for fundamental physics.This thesis describes various guiding and deceleration experiments of supersonic beams of paramagnetic atoms using inhomogeneous time-dependent magnetic fields. Inhomogeneous magnetic fields have been used to exert a force on paramagnetic atoms or molecules, which derives from the Zeeman effect. The principle of the newly developed Zeeman decelerator is to produce a moving tridimensional magnetic trap, which moves at the initial velocity of the beam. The control of the time dependence of the magnetic field allows us to control the velocity of the so-called co-moving magnetic trap, thereby affording for a deceleration of a velocity class of the supersonic beam. The co-moving magnetic trap is inferred from a moving magnetic wave, offering a minimal distortion of the trap during its propagation. The transverse properties of the trap are tunable through a transverse quadrupolar magnetic field, which can be adjusted independently of the velocity and acceleration properties of the trap.Much of this thesis was devoted to the design, development and construction of the experimental setup consisting of a supersonic beam and complex coils to achieve a traveling magnetic wave. Using home-made electronics operating 300A AC currents at frequencies up to 40 kHz, the coils can produce a magnetic wave of amplitude 0.7T, moving at a controllable velocity up to 464m/s. Several proof-of-principle experiments have been carried out using a pulsed, cold beam of metastable atoms, excited in metastable states by an electric discharge during the supersonic expansion. We have studied the guiding properties of the quadrupolar magnetic field alone on two atomic beams (metastable helium and argon) and compared with the theoretical prediction of tridimensional numerical simulations. A supersonic beam of metastable argon atoms has been trapped in a co-moving trap at a constant velocity (464m/s, 400m/s, and 392m/s) using a 28cm-long prototype decelerator. The temperature of the guided beam packet is observed to be 100mK. Finally, Zeeman deceleration experiments have been done on metastable argon beams with an initial velocity of 400m/s, decelerated to various final velocities (392m/s, 370m/, and 365m/s).
