"Fools for Christ": An Examination of the Ministerial Call in Three Novels by William Golding

This thesis examines the ministerial call in three novels by William Golding, specifically The Spire, Darkness Visible, and Rites of Passage. The central character of each novel, a Christian minister, has a vision, or series of visions, which dominates his life. The call and vision(s) of Golding's ministers are examined in light of Jacques Ellul's The Humiliation of the Word, a work examining the differences between the word and the image. The ministerial call, in this thesis, is linked to Ellul's ideas about the word; the vision, in this thesis, is linked to Ellul's ideas of the image. As a result of following their vision(s) rather than their call, the ministers fail, and their lives end in despair and ruin.

Analyse qualitative du contenu des signalements retenus à la protection de la jeunesse et leur évaluation pour les enfants de minorités visibles

Cette recherche propose une analyse qualitative du contenu de signalements retenus à la protection de la jeunesse et leur évaluation pour des enfants de minorités visibles. Les premiers objectifs de cette recherche sont de décrire les deux premières étapes de l’intervention en protection de la jeunesse, soit la réception du signalement et l’évaluation du besoin de protection. Par la suite, elle vise l’élaboration d’une typologie de familles de minorités visibles signalées à la protection de la jeunesse. Une analyse qualitative de contenu est conduite en utilisant une grille d’analyse des dossiers de 48 situations d’enfants signalés à la protection de la jeunesse. Il s’agit d’une analyse secondaire de données d’une étude plus large portant sur l’intervention en protection de la jeunesse auprès de familles de minorités visibles. Les résultats de la présente étude montrent que les informations consignées aux dossiers par les intervenants respectent les dimensions légales du jugement sur la compromission du développement ou de la sécurité d’un enfant. Toutefois, l’absence d’éléments ethnoculturels dans la majorité des dossiers, ou leur traitement superficiel, sont surprenants. La typologie découlant de cette analyse décrit quatre profils présentant des dynamiques familiales distinctes : les familles traversant une crise, les familles qui abdiquent leur rôle parental, les familles isolées et les familles éclatées. Ces résultats rendent compte de la diversité des dynamiques familiales et par conséquent, des besoins de ces familles quant aux interventions des services de protection. Enfin, étant donné l’importance de l’évaluation et de ses conclusions dans la trajectoire de ces familles à l’égard de la DPJ, l’intégration des éléments ethnoculturels devrait être systématique afin de leur offrir une réponse adaptée et culturellement sensible. / The first objects of this research are to describe the first two youth protection intervention stages, namely report notification and assessment of the need for protection. Afterwards, the research deals with the development of a typology of visible minority families reported to youth protection services. A qualitative content analysis is conducted using an analysis grid of records dealing with 48 situations of children reported to youth protection services. It is a secondary analysis of data from a broader study dealing with child protection intervention with visible minority families. The results of the present study show that the information recorded by child-care workers complies with the legal aspects of the judgment on the potential harm to a child’s development or safety. However, the absence of ethnocultural elements in most records or a superficial treatment thereof, are surprising. The typology flowing from this analysis describes four profiles presenting distinct family dynamics: families going through a crisis, families abdicating their parenting responsibilities, isolated families and broken families. These findings account for the diversity of family dynamics and consequently, those families’ needs in terms of child protection intervention services. Finally, given the importance of the assessment and of these conclusions in the path of those families towards child protection intervention services, the integration of ethnocultural elements should be systematic in order to provide them with an adapted, culturally sensitive response.

The skin has a system that can detect light in a fashion similar to the retina. Although its presence was initially reported almost 20 years ago, only in 2011 functional studies started to be reported. The biological clock of the skin has also been reported in the beginning of the century, but its function and relevance still remain unexplored. Thus, this Ph.D. project was designed to explore the functionality of both systems in melanocytes, and whether the disruption of these systems is associated with the development of melanoma cancer. Using in vitro, in vivo, and bioinformatics approaches, we have shown that: 1) the biological clock of malignant melanocytes is more responsive to visible light, UVA radiation, estradiol, and temperature compared to normal cells; 2) UVA radiation is detected by melanopsin (OPN4) and rhodopsin (OPN2), which triggers a cGMP related cascade that leads to immediate pigment darkening (IPD) in normal and malignant melanocytes; 3) in addition to detecting UVA radiation, OPN4 also senses thermal energy, which activates the biological clock of both normal and malignant melanocytes; 4) regarding the biological clock, we have provided several layers of evidence that proves that in melanoma a chronodisruption scenario is established compared to healthy skin and/or normal pigment cells; 5) in vivo tumor samples display a low amplitude circadian rhythm of clock gene expression and an ultradian oscillatory profile in melanin content; 6) a non-metastatic melanoma leads to a systemic chronodisruption, which we suggest that could favor the metastatic process; 7) in human melanoma, we demonstrated the role of BMAL1 as a prognostic marker and a putative marker of immune therapy success. Taken altogether, these results significantly contributed to the literature as it brought to light new and interesting targets and processes, which will be explored in future projects / A pele possui um sistema que pode detectar luz de forma análoga à retina. Embora a presença deste sistema tenha sido inicialmente descrita quase há 20 anos, apenas no ano de 2011 estudos funcionais começaram a ser relatados. Sabe-se que o relógio biológico da pele também foi identificado no início do século, mas sua função e relevância ainda continuam pouco exploradas. Diante deste cenário, este projeto de doutorado foi desenhado para investigar a funcionalidade de ambos os sistemas em melanócitos e se perturbação dos mesmos estaria associada com o desenvolvimento de melanoma. Através do uso de abordagens in vitro, in vivo e de bioinformática, nós demonstramos que: 1) o relógio biológico de melanócitos malignos é mais responsivo à luz visível, radiação UVA, estradiol e temperatura comparado ao de células normais; 2) a radiação UVA é detectada por melanopsina (OPN4) e rodopsina (OPN2), que ativam uma via de sinalização dependente de GMPc, levando ao processo de pigmentação imediata (IPD) em melanócitos normais e malignos; 3) além de detecção de radiação UVA, a OPN4 também detecta energia térmica que, por sua vez, ativa o relógio biológico de melanócitos normais e malignos; 4) relativo ao relógio biológico, provamos por diferentes abordagens que, no melanoma, um cenário de cronoruputura está estabelecido em comparação a pele saudável e/ou melanócitos; 5) tumores in vivo apresentam um ritmo circadiano de baixa amplitude na expressão dos genes de relógio e um ritmo ultradiano oscilatório no conteúdo de melanina; 6) um melanoma não metastático leva a um quadro sistêmico de cronoruptura, o qual sugerimos favorecer o processo de metástase; 7) em melanoma humano, demonstramos o papel do gene BMAL11 como marcador de prognóstico e um possível indicador de sucesso de imunoterapias. Portanto, este projeto contribuiu de forma significante para a literatura científica uma vez que trouxe à luz novos e interessantes alvos terapêuticos e processos, os quais serão explorados em projetos futuros

O uso crescente de dispositivos móveis e o advento da Internet das Coisas têm demandado esforços para prover alternativas de comunicação sem fio. A utilização de sistemas óticos para a transmissão de dados apresenta-se como uma tecnologia promissora e complementar à comunicação por radiofrequência, especialmente devido aos desenvolvimentos consideráveis na tecnologia de iluminação por estado sólido e sua adoção em vários domínios. Os diodos de emissão de luz podem ser utilizados na promoção de comunicação por luz visível em ambientes internos e externos. Nos ambientes externos há inúmeras oportunidades a serem exploradas no que concerne aos Sistemas Inteligentes de Transporte, visto que os diodos de emissão de luz têm sido utilizados em luminárias públicas, semáforos e veículos. Esta pesquisa propôs o desenvolvimento e a avaliação de um sistema de comunicação por luz visível entre elementos da infraestrutura urbana e veículos, mais especificamente, luminárias públicas baseadas em diodos de emissão de luz e o modal de transporte público representado pelos ônibus que circulam em faixas exclusivas. A metodologia foi composta por simulação computacional e pela construção de um protótipo para auxiliar na validação das soluções propostas. O sistema complementar de comunicação sem fio por meio da luz proposto fornece pode fornecer subsídios à determinação do posicionamento preciso dos ônibus, suprindo as deficiências do sistema GPS em locais menos favoráveis à recepção de seus sinais em uma cidade. Essas informações auxiliam os usuários e os gestores do transporte público, atendendo alguns domínios de serviço em Sistemas Inteligentes de Transporte, de acordo com a norma ISO 14813. Os resultados obtidos nos testes com o protótipo, bem como, aqueles observados na simulação, comprovaram a viabilidade do sistema proposto, com o estabelecimento da comunicação entre luminária e ônibus, inclusive quando trafegando a 60 km/h. / The increasing use of mobile devices and the advent of the Internet of Things have called for efforts to provide alternatives for wireless communication. The use of optical systems for data transmission is a promising and complementary technology to radiofrequency communication, especially due to the considerable developments in solidstate lighting technology and its adoption in various domains. Light-emitting diodes can be used to promote visible light communication in indoor and outdoor environments. In outdoor environments there are numerous opportunities to be explored with regard to Intelligent Transport Systems, since light-emitting diodes have been used in public luminaires, traffic lights and vehicles. This research proposed the development and evaluation of a visible light communication system between elements of the urban infrastructure and vehicles, more specifically, public luminaires based on light-emitting diodes and the mode of public transport represented by buses that circulate in exclusive lanes. The methodology was composed by computational simulation and the construction of a prototype, to assist in the validation of the proposed solutions. The proposed complementary wireless communication system through light provides subsidies to determine the precise positioning of buses, supplying deficiencies of the GPS system in places less favorable to the reception of their signals in a city. This information helps the users and the managers of the public transport, attending some service domains in Intelligent Transport Systems, according to the ISO 14813 standard. The results obtained in the tests with the prototype, as well as those observed in the simulation, proved the feasibility of the proposed system, with the establishment of communication between luminaire and bus, even when traveling at 60 km/h.

A sua quantificação, é fundamental para o estudo da evolução química dessa substância. O objetivo deste trabalho foi caracterizar as amostras de ácidos húmicos (AHs) e quantificar o grau de humificação do solo e das frações físicas (por tamanho de partículas). As análises de Espectroscopias de Fluorescência de Raios X por Dispersão de Comprimento de Onda, Fluorescência na região do Visível, Espectroscopia Vibracional na região de Infravermelho e de Absorção na região do Ultravioleta e Vísivel, Espectrocospia Raman, acompanhadas das técnicas de Análise Térmica, Microscopia Eletrônica de Varredura com Canhão Eletrônico por Efeito de Campo e Espalhamento Dinâmico da Luz foram utilizadas. A quantificação do grau de humificação mostrou que AHs extraídos da profundidade (10- 20 cm) possuem estruturas aromáticas mais condensadas. A fração de AH que apresentou maior grau de humificação foi a extraída do solo com a adição de calagem, o que demonstra uma influência do calcário na aceleração do processo de humificação. O estudo das frações granulométricas do solo, determinou, com espectroscopia de infravermelho, os atributos orgânicos que estão associados as frações do solo e, que a maioria dos minerais presentes nas frações granulométricas, foram identificados como sendo caulinita, haloisita, quartzo e gibbsita. Já para o estudo da humificação dos AHs extraídos das frações granulométricas, a quantificação do grau de humificação, do maior para o menor, é procedente da fração silte, seguidos da fração argila e, por fim, a fração areia. / The humification process, conversion of organic waste into humus, is an important factor to understand the organic matter dynamics. Its quantification is vital for the study of this substance chemical evolution. This study aimed at characterizing humic acid (HA) samples and quantify the degree of soil humification and physical fractions (particle size). The analysis of Spectroscopies of wavelenght dispersive X-ray fluorescence, visible fluorescence, vibrational infrared region, ultraviolet and visible light absorption and Raman along with the techniques of Thermal Analysis, scanning electron microscopy with field effect gun and dynamic light scattering were employed. The humification degree quantification revealed that the HA extracted from the 10-20cm depth presented more polycondensed structures. The HA fraction that presented the highest humification degree was extracted from the soil with the addition of liming, which demonstrates the limestone influence in the acceleration of the humification process. The study of the soil granulometric fractions determined, with infrared spectroscopy, the organic properties that are associated to soil fractions and that most of the minerals present in the granulometric fractions, were identified as kaolinite, halloysite, quartz and gibbsite.The study of the humification in the HA extracted from the granulometric fractions quantified the humification degree and revealed the highest values in the silt fraction followed by the clay fraction and, finally, the lowest values were found in the sand fraction.

Mechanistic approaches towards understanding particle formation in biopharmaceutical formations : the role of sufactant type and level on protein conformational stability, as assessed by calorimetry, and on protein size stability as assessed by dynamic light scattering, micro flow imaging and HIAC

Control and analysis of protein aggregation is an increasing challenge to biopharmaceutical research and development. Therefore it is important to understand the interactions, causes and analysis of particles in order to control protein aggregation to enable successful biopharmaceutical formulations. This work investigates the role of different non-ionic surfactants on protein conformational stability, as assessed by HSDSC, and on protein size stability as assessed by Dynamic Light Scattering (DLS), HIAC and MFI. BSA and IgG2 were used as model proteins. Thermal unfolding experiments indicated a very weak surfactant-immunoglobulin IgG2 interaction, compared to much stronger interactions for the BSA surfactant systems. The DLS results showed that BSA and IgG2 with different surfactants and concentration produced different levels of particle size growth. The heat treatment and aging of samples in the presence of Tween 20, Tween 80, Brij 35 and Pluronic F-68 surfactants led to an increase in the populations of larger particles for BSA samples, whereas IgG2 systems did not notably aggregate under storage conditions MFI was shown to be more sensitive than HIAC technique for measuring sub-visible particles in protein surfactant systems. Heat treatment and storage stress showed a significant effect on BSA and IgG2 protein sub-visible particle size stability. This work has demonstrated that both proteins with different Tween 20, Tween 80, Brij 35 and Pluronic F-68 concentrations, have different level of conformational and size stability. Also aging samples and heating stress bears the potential to generate particles, but this depends on surfactant type. Poor predictive correlations between the analytical methods were determined.

近年來,全球氣候暖化問題嚴重,造成地球環境的變遷,加上能源安全威脅與經濟、環境永續發展的考量,世界各國對於節能減碳的議題日漸重視。車輛電動化即在減少溫室氣體排放、改善生活環境、加強能源安全及掌握產業發展轉型契機等因素驅動下,成為全球車輛發展的重要趨勢,全球電動車產業結構與產業網絡正在逐漸成形中。 本研究係針對全球主要國家政府在面對電動車這個新興產業發展時,所採取的政策進行分析與探討,並且以日本、美國、中國、德國政府政策為探討目標,從文獻回顧及專業機構報告中整理分析各國政府的產業政策,試圖探討政府的角色與定位,進一步描繪出台灣政府應該扮演的角色,並且提出建議,作為政府於制定產業政策時之參考,期能對電動車產業做出貢獻,讓台灣電動車產業在此波革命浪潮中,創造出更高的價值及競爭力。 本研究發現,電動車新興產業在發展過程中充滿許多經濟面、市場面、環境面以及技術面的發展限制,本研究在羅列出的十六項發展限制中,有高達十一項限制與政府的政策息息相關。汽車百年產業革命成功與否,業界對於政府是否能創造具有競爭優勢的發展環境要求殷切,於是在電池研發技術未臻成熟、產業各項標準尚未建立之際,各國政府紛紛伸出<看得見的那隻手>,從產業研究發展獎勵、消費市場購車、用車以及充電基礎建設普及的促成與補助,配合業者在關鍵技術發展的努力、商業模式的發展、產業群聚的形成、充電基礎建設的普及、維修體系的建置、社會大眾對於環境品質的需求、用車習慣的改變、電動車節能形象的認同……等種種助力,形成產業能量面的推升力量以及市場需求的拉力,形塑出電動車未來發展的良性循環,試圖在此領域內創造出比較優勢,促使這一波新的汽車產業革命形成。   值此關鍵時期,台灣政府如何提供本國產業一臂之力,期於未來世界汽車產業佔有一席之地?這是許多業者關心的議題。目前,各國政府紛紛制訂獎勵政策協助業者以及消費者對於電動車發展與使用的認同;尤其德國政府作法積極,從保持人民生活品質、解決石油倚賴問題、強化經濟傳統命脈的汽車工業,從環境結構、基礎設施到社會大眾的消費習慣宣導著手,具體規劃國家新經濟遠景,將德國塑造成世界電動車大國,這是非常值得台灣政府參考的典範。 本研究建議,電動車產業處發展初階段,眾家車廠以及各國政府皆在摸索前行中,台灣政府應參考各國政府政策後,了解台灣產業的優勢與機會,從基礎設施到新能源政策、基本軟硬體投資環境的建置著手,協助業者發展創造出友善的投資環境並且積極發展國內電動車運行,累積出屬於台灣自己的<台灣EV經驗>,協助業者強化電動車相關技術,除藉由電動車的普及來降低國內對於石油的依賴外,倘若能成功推行電動車成為國際典範,讓世界看到台灣先進的科技與環保觀念,提升國際形象,在時機成熟時,提供關鍵零件技術,利用中國的潛在市場,參與國際標準制定,創造出台灣電動車產業的發展空間。 / The worsening problem of global warming in recent years has led to the environmental changes on Earth. This fact, along with the emerging threats to energy security and the call for sustainable economic and environmental development, has drawn increasing worldwide attention on the issues of energy saving and carbon reduction. As a result, the EV (Electric Vehicle) industry is gaining increasing importance in the global automobile market, driven by various motives including reducing greenhouse gas emissions, improving living environment, safeguarding energy security and seizing the opportunities of industrial transformation. The industrial structure and network of the global EV market is gradually taking shape. The purpose of this research is to explore and analyze the policies adopted by the governments of the world’s leading countries on the emerging EV industry, focusing on Japan, the USA, China and Germany. By reviewing historical documents and professional reports, the author has probed the industrial policies of these countries, endeavoring to identify the roles of the governments in this process and, furthermore, to depict the roles and positions of the government of Taiwan. The author has also provided recommendations to the government for developing industrial policies, with the hope that the research results will contribute to the growth of the EV industry in Taiwan and the creation of higher value and competitive advantages in this revolutionary process. It is discovered in this research that a great deal of development restrictions are involved in the growing process of the emerging EV industry, respectively in the economic, market, environmental and technological fields. Among the sixteen development restrictions outlined in this research, there are as many as eleven items which are closely bound up with government policies. The success of this unprecedented industrial revolution, therefore, relies on whether the government can create a development environment with sufficient competitive advantages, which is anxiously longed for by the EV industry. Given the facts that the R&D technologies for the battery industry are not fully mature and various industrial benchmarks are yet to be established, the “visible hands” are offered by governments, one after another, to support the industry. These government initiatives include: providing incentives for industrial R&D, stimulating the consumer market, as well as facilitating and subsidizing the EV car use and recharging infrastructure. Other supportive policies include assisting the EV industry in developing key technologies, building up business models, shaping industrial clusters, expanding recharging infrastructure, establishing the maintenance/repair systems, accommodating to the public demand on environmental quality and the changes in car-using habits, promoting the identification with the energy-saving image of EV’s, etc. All these measures have turned into a pushing force that gives momentum to the industry, as well as a pulling force that booms the market. They contribute to forming a virtuous development cycle for the EV industry in future; help to create comparative advantages in this specific field; and make impossible this new wave of industrial revolution for automobiles. At this critical stage, how would the Taiwan government lend a helping hand so that the local EV industry could gain a stake in the international automobile market in future? This is an issue widely concerned by the industry. Now, many countries are offering incentive policies to help with the development of the industry and build up consumer identification with the electric cars. The German government, particularly, has taken aggressive actions. Apart from the efforts in maintaining/improving the life quality of its people, the government also endeavors to reduce the dependency on imported oil and strengthen the competitiveness of its automobile industry, which is vital to the economic growth of the country. To realize the new economic vision of the country, the German government has developed specific plans, covering various initiatives including environmental structure, infrastructure construction and consumer education, etc. The purpose is to make Germany a leading country for the EV industry. These we believe are the “best practices” to be considered by the Taiwan government. The EV industry is yet at its preliminary stage of development. Governments, as well as EV manufacturers around the world, are all endeavoring to find the right path ahead. It is recommended, therefore, that the government of Taiwan should firstly study the policies of other countries and understand the strengths and weakness of local industries. The initial efforts should be focused on constructing infrastructures, setting up new energy policies and installing the fundamental software/hardware for a friendly investment environment, so as to support the growth of the industry. The government should, in the meantime, actively promote the electric vehicles in the domestic market, build up the unique “EV Experience of Taiwan” to help improving the relevant EV technology for the local industry. Apart from reducing the country’s dependency on imported oil, the successful popularization of EV in Taiwan will also become one of the best practices in the world and contribute to the elevation of Taiwan’s international image, having showcased its advanced technologies and strong environmental awareness. With increasing sophistication, Taiwan’s EV industry also has the capabilities to provide critical spare parts technologies and to play a role, through the potential China market, in the international standard-setting process, thereby creating further expanding possibilities for the industry.
