Etude de la relation structure - toxicité des protéines amyloïdes en interaction avec des membranes modèles

Ce mémoire rapporte les études de protéines amyloïdes en interaction avec des membranes modèle afin d’établir une relation structure toxicité. Nous avons choisi différents modèles membranaires (monocouches, bicouches) de composition lipidique et charges différentes et utilisé différentes méthodes physico-chimiques afin de caractériser les interactions des protéines amyloïdes avec les membranes.Nous avons montré l’importance de la contribution électrostatique dans les interactions de la protéine amyloïde HET-s (218-289) et ses mutants avec les membranes modèles.L’ellipsométrie a démontré que les mutants toxiques de HET-s (218-289) (M8, WT.Y1Y2V2) perturbentfortement les monocouches lipidiques à l’interface air-eau. La structure riche en feuillets β antiparallèles des protéines àl’interface air-eau et dans l’interaction avec les monocouches de lipides a été démontrée par la spectroscopie PMIRRAS (Polarization Modulation – Infrared Reflection Absorption Spectroscopy). Nous avons établie que l’interface air-eau peut modifier l’agrégation des protéines amyloïdes. A l’aide de la spectroscopie de fluorescence, la spectroscopie PWR (Plasmon-Waveguided Resonance) et la spectroscopie ATR-FTIR (Attenuated Total Reflection – Fourier Transform Infrared), nous avons mis en évidence que la protéine toxique M8 adopte une structure riche en feuillets β antiparallèles en altérant fortement l’intégrité des bicouches lipidiques. Au contraire, la protéine non toxique WT se structure en feuillets β parallèles dans ces interactions et elle ne perturbe pas l’homogénéité des membranes. La toxicité de la protéine M8 semble liée à son organisation différente et à sa capacité à réorganiser les membranes.Nos résultats confortent également l’hypothèse de la toxicité des oligomères amyloïdes.Une étude sur la fabrication d’une cellule microfluidique pour la séparation de différents types d’autoassemblage afin de les détecter et de les étudier en interaction avec des liposomes par spectroscopie infrarouge est présentée. Une cellule microfluidique de CaF2 de 8 μm d’épaisseur de canaux est obtenue et est utilisée pour la détection d’une protéine de test. / This manuscript reports the studies of amyloid proteins in interaction with membrane models in order to establish their structure-toxicity relationship.Membrane models (monolayer, bilayer) of different charge and lipid composition were used. We used various physico chemical methods to characterize the interaction of these amyloid proteins with membranes.We showed the importance of the electrostatic contribution in the interactions of the amyloid protein HET-s(218-289) and its mutants with model membranes.Ellipsometry showed that the toxic mutants of HET-s (218-289) (M8, WT.Y1Y2V2) strongly disturbed thelipid monolayers at the air-water interface. The structure rich in antiparallel β sheets of auto-assembled proteins at theair-water interface and in interaction with lipid monolayers at the air-water interface has been demonstrated by the PMIRRAS spectroscopy (Polarization Modulation - Infrared Reflection Absorption Spectroscopy). We established that theair-water interface can change the aggregation properties of amyloid proteins.By using fluorescence spectroscopy, PWR spectroscopy (Plasmon Resonance-Waveguided spectroscopy) and ATR-FTIR spectroscopy (Attenuated Total Reflection - Fourier Transform Infrared spectroscopy), we found that thetoxic protein (M8) adopted a structure rich in antiparallel β sheets greatly altered the integrity of lipid bilayers. Incontrast, the protein non-toxic (WT) organized in a structure rich in parallel β sheets in these interactions and it did notdisturb the homogeneity of the membranes. The toxicity of the protein M8 appears to be related to its differentorganization and its ability to rearrange membranes.Our results also support the hypothesis of the toxicity of amyloid oligomers.A study on the fabrication of a microfluidic cell for the separation of different aggregation states of amyloidproteins in order to detect these assemblies and to study their interaction with liposomes by infrared spectroscopy is presented. A CaF2 microfluidic cell with channels of 8 μm of thickness was obtained and was used for the detection of atested protein.

EXPERIMENTAL STUDY OF A LOW-FREQUENCY THERMOACOUSTIC DEVICE

An experimental study of a low-frequency transcritical thermoacoustic device has been conducted at Purdue University's Maurice J. Zucrow Laboratories. The purpose of this study was to characterize the thermoacoustic response of transcritical R-218 and asses it's feasibility for energy extraction and waste heat removal. This rig operated as a standing-wave configuration and achieved pressure amplitudes as high as 690 KPa (100 psi) at a temperature difference of 150 K and a bulk pressure of 1.3 P/P<sub>cr </sub>(3.43 MPa). To the author's knowledge, this is the highest ever thermoacoustic pressure amplitude achieved in a non-reacting flow. The thermoacoustic response was characterized by varying temperature difference and bulk pressure parametrically. The effect of resonator length was characterized in a set of tests where resonator length and bulk pressure was varied parametrically at a single temperature difference. Finally, the feasibility for energy extraction was assessed in a set of tests which characterized the ability of the working fluid to pump itself through a recirculation line with check valves. This set of tests showed that the working fluid was able to create self-sustained circulation by inducing a pressure differential across the check valves with the thermoacoustic response. This circulation was induced while still maintaining a significant pressure amplitude, demonstrating promising results as a feasible method for energy extraction and waste heat removal.

Objective: The aim of this study was to evaluate the expression of hsa-miR-205ab and hsa-miR-218a microRNAs and BRCA1 protein in breast cancer samples and their associations with the pathological and prognostic clinical aspects of tumors. Method: A group of 92 breast cancer cases was evaluated for clinical-pathological aspects, five-year survival, expression of BRCA1, hsa-miR-205ab and hsa-miR-218a. BRCA1 expression was assessed by immunohistochemistry and the quantification of the miRNAs by quantitative PCR in real time. The clinical-pathological aspects were compared in relation to the expression of BRCA1, hsa-miR- 205ab and hsa-miR-218a using the Mann-Whitney test. Survival curves were generated by the Kaplan-Meyer method and compared by the long-rank method. Results: The group of tumors comprised 56 cases of non-triple-negative breast carcinomas and 36 tumors with triple-negative phenotype. Hypoexpression of hsa-miR-205ab and hsa-miR-218a was associated with larger tumors (> 2 cm), presence of lymph node metastasis and distant metastasis, the highest histological grade, triple negative phenotype, absence of expression of BRCA1 and lower survival. (P = 0.044), more advanced stages (p = 0.005), lymph node involvement (p = 0.038), presence of distant metastasis (p = 0, 0008) and absence of BRCA1 expression (p = 0.039). Conclusion: The hypoexpression of hsa-miR-205ab and hsa-miR-218a was associated with factors of poor prognosis, absence of BRCA1 expression and the lowest survival rate in breast cancer patients evaluated in this study. / Introdução: Os microRNAs são pequenas moléculas de RNAs não-codificantes de cadeia simples, que regulam a expressão gênica na fase pós-transcricional e vem sendo avaliados como marcadores prognósticos e preditivos no câncer de mama. Objetivo: O objetivo deste estudo foi avaliar a expressão dos microRNAs hsa-miR-205ab e hsa-miR-218a e da proteína BRCA1 em amostras de câncer de mama e suas associações com os aspectos clínico patológicos e prognósticos dos tumores. Método: Um grupo de 92 casos de câncer de mama foi avaliado quanto aos aspectos clínico-patológicos, a sobrevida em cinco anos, a expressão de BRCA1, hsa-miR-205ab e hsa-miR-218a. A expressão de BRCA1 foi avaliada por imuno- histoquímica e a quantificação dos miRNAs por PCR quantitativa em tempo real. Os aspectos clínicos-patológicos foram comparados em relação à expressão de BRCA1, hsa-miR-205ab e hsa-miR-218a, utilizando o teste de Mann-Whitney. As curvas de sobrevida foram geradas pelo método de Kaplan-Meyer e comparadas pelo método de long-rank. Resultados: O grupo de tumores compreendeu 56 casos de carcinomas de mama não triplo-negativos e 36 tumores com fenótipo triplo-negativo. A hipoexpressão de hsa-miR-205ab e hsa-miR-218a foi associada aos tumores maiores (> 2 cm), à presença de metástase linfonodal e de metástase a distância, ao grau histológico mais elevado, fenótipo triplo negativo, ausência de expressão de BRCA1 e menor sobrevida. A sobrevida das pacientes em função das características clínico patológicas foi influenciada pelo fenótipo triplo-negativo (p=0,044), estádios mais avançados (p=0,005), acometimento linfonodal (p=0,038), presença de metástase à distância (p=0,0008) e ausência da expressão de BRCA1 (p=0,039). Conclusão: A hipoexpressão de hsa-miR-205ab e hsa-miR-218a foi associada aos fatores de pior prognóstico, ausência de expressão de BRCA1 e à menor sobrevida nas pacientes com câncer de mama avaliadas neste estudo.

Packages that are left unattended in public spaces are a security concern and timely detection of these packages is important for prevention of potential threats. Operators should be always alert to detect abandoned items in crowded environments. However, it is very difficult for operators to stay concentrated for extended periods. Therefore, it is important to aid operators with automatic detection of abandoned items. Most of the methods in the literature define abandoned items as items newly added to the scene and stayed stationary for a predefined time. Hence other stationary objects, such as people sitting on a bench are also detected as suspicious objects resulting in a high number of false alarms. These false alarms could be prevented by discriminating suspicious items as living/nonliving objects. In this thesis, visible band and thermal band cameras are used together to analyze the interactions between humans and other objects. Thermal images help classification of objects using their heat signatures. This way, people and the objects they carry or left behind can be detected separately. Especially, it is aimed to detect abandoned items and discriminate living or nonliving objects
