Epigenetic transcriptional repression of tumor suppresor genes and its reversion by drugs.

Genetic alterations and deregulation of the epigenetic mechanisms collaborate in the initiation and progression of cancer. In contrast to the genetic defects, the epigenetic abnormalities are potentially reversible. This fact has driven the search for drugs that induce selectively changes in the epigenetic patterns of the tumor cells and thus lead to differentiation, cell death and/or cell growth arrest. Inhibitors of DNA (cytosine-5)-methyltransferases (DNMTs) and inhibitors of Zn(II)-dependent histone deacetylases (HDACs) have been developed with this purpose. The DNMTs inhibitors allow the reactivation of genes, including tumor-suppressor genes, silenced through hypermethylation of the CpG island at their promoter. The HDACs inhibitors allow the reexpression of genes silenced by ypoacetylation of the histones associated at their promoters. Despite all these chemicals have promising effects on cultured cancer cells, many of them have side-effects that limit their use in anticancer chemotherapy. Because of that, we analyzed the properties of the anesthetic procaine (4-aminobenzoic acid 2-diethylaminoethyl ester) as a new inducer of DNA demethylation and we also compared the effects of seven HDACs inhibitors. In both cases, the breast cancer cell line MCF7 was the model system.1. Procaine reduces the proportion of 5-methylcytosine into global genomic DNA, achieving its maximum effect within 24-48 h of treatment. Low concentrations of procaine decrease the amount of 5- methylcytosine at RAR¦Â2 promoter, which is hypermethylated in MCF7 cells, and reactivate its expression with only small decrease in global DNA methylation. This fact could be an advantage, since global DNA hypomethylation leads to chromosomal instability. Finally, procaine reduces cell proliferation and arrests cell cycle in mitosis, but does not induce apoptosis in MCF7 cells after treatments 3 days long.2. The seven inhibitors of Zn(II)-dependent HDACs chosen for comparison were: two carboxylic acids (butanoic and valproic acid); one N-(2¡¯-aminophenyl) benzamide (MS-275); and four hydroxamic acids(trichostatin A, suberoylanilide hydroxamic acid, CX and CY). The results of in vitro HDAC activity assays performed on MCF7 nuclear extracts show the existence of a relationship between the chemical structure of these compounds and their activity: low micromolar concentrations of hydroxamic acids are sufficient for inhibiting almost completely the deacetylase activity, whereas millimolar concentrations of carboxylic acids are required for similar effects. Also the alterations that the drugs cause on cell growth and cell cycle arrest depend on its chemical structure. The IC50 for cell treatments 24 h long is in the range of millimolar concentrations for butanoic and valproic acids and low micromolar for the rest of the chemicals. At the IC50, MS-275 induces cell growth arrest in G1/G0, whereas the hydroxamic acids stop cell cycle mostly at G2/M and the carboxylic acids seems to arrest the cycle at both G1/G0 and G2/M. Despite all these inhibitors induce similar changes in the global acetylation of H4 and H3 when employed at their respective IC50, not all of them are able of reactivate the expression of the same genes. Moreover, it seems that the induced expression levels of CDKN1A and GADD45¦Â determine the alterations induced by the drugs on cell cycle. The changes on histone modifications at the promoters of six genes ( CDKN1A, GADD45¦Â, JunD, IGFBP3, MT1X and MT2A) upon CY treatment were studied. HDACs inhibition induces an increase in histone H4 tetraacetylation and in dimethylation of lysine 4 in H3, as well as a decrease in dimethylation of lysine 9 in H3. Additionally, HDAC2 is released from the promoters upon CY treatment. These changes take place also in the promoters of MT1X and MT2A, the genes whose expression remains unaltered after the treatment with CY. / Las alteraciones genéticas y la desregulación de los mecanismos epigenéticos colaboran en la iniciación y progresión del cáncer. Los defectos epigenéticos son potencialmente reversibles, lo que ha suscitado la búsqueda de fármacos que selectivamente causen cambios en los patrones epigenéticos de las células tumorales, con la consiguiente diferenciación, muerte y/o parada de crecimiento de las mismas. Se han estudiado especialmente inhibidores de metiltransferasas de DNA (DNMTs) e inhibidores de desacetilasas de histonas (HDACs) dependientes de Zn(II). Los inhibidores de DNMTs posibilitan la reactivación de genes silenciados mediante hipermetilación de la isla CpG de su promotor y los inhibidores de HDACs, de genes silenciados vía hipoacetilación de las histonas asociadas a su promotor. A pesar de sus prometedores efectos en cultivos celulares, muchas de estas sustancias presentan inconvenientes que limitan su aplicación en quimioterapia. Por ello, en esta tesis:1. Se ha estudiado por primera vez la capacidad del anestésico procaína para reducir la metilación de DNA genómico y la proliferación de la línea celular MCF7. Procaína reduce la cantidad de 5-metilcitosina en DNA, reactiva genes silenciados por hipermetilación ( RAR¦Â2) e induce parada del ciclo celular en mitosis.2. Se han comparado siete inhibidores de HDACs dependientes de Zn(II): ácido butanoico, ácido valproico, MS-275, tricostatina A (TSA), SAHA, CX y CY. Existe una relación estructura-actividad para los efectos de estas sustancias en ensayos in vitro y en el crecimiento de MCF7. Además, aunque estos inhibidores inducen cambios similares en acetilación global de histonas, no todos reactivan los mismos genes. Los niveles expresión de CDKN1A y GADD45¦Â parecen determinar los efectos de estos compuestos en el ciclo celular. En los promotores de los seis genes estudiados, la inhibición de HDACs aumenta la acetilación de H4 y la dimetilación de la lisina 4 de H3, mientras disminuye la dimetilación de lisina 9 de H3.

Estudio Teórico de Mecanismos de Oxidación de Interés en Química Atmosférica: Reacciones de Terpenos, Isopreno y Productos Derivados.

Durante el periodo de disfrute de la Beca Predoctoral de Formación de Personal Investigador de la Generalitat Valenciana, el becario D. Víctor Miguel Ramírez Ramírez ha trabajado sobre el proyecto de Tesis Doctoral correspondiente al estudio teórico de mecanismos de oxidación de interés en química atmosférica, concretamente las reacciones de oxidación de terpenos, isopreno, y compuestos relacionados.Los terpenos estudiados han sido el -pineno, el -pineno y el d-limoneno. Se ha dilucidado el mecanismo de degradación del -pineno iniciado por el radical OH, analizando las posibles vías de reacción, y poniendo especial énfasis en la reacción inicial de ataque del radical oxidante. Se han empleado los métodos de cálculo AM1 (para realizar la exploración preliminar de la hipersuperficie de energía potencial), y el funcional B3LYP. Además, se han realizado cálculos HF y UMP2 para el estudio de la reacción inicial de adición de OH o abstracción de H sobre este monoterpeno. Para el -pineno y el d-limoneno se ha realizado un estudio detallado de la reacción inicial de adición de OH a todos los átomos de carbono, a través de todas las caras posibles, de los dobles enlaces existentes en estas moléculas.Los metodología empleada ha sido el método UMP2 para realizar la optimización de geometrías, y cálculos puntuales QCISD(T) sobre las estructuras optimizadas UMP2, para el refinamiento de la energía potencial.Además de los terpenos mencionados, se ha realizado un estudio del mecanismo de oxidación del isopreno, iniciado por el radical OH, del mismo modo que para el -pineno. Tras una exploración preliminar de la hipersuperficie de energía potencial mediante el método semiempírico AM1, se han estudiado las reacciones de formación concernientes a algunos de los principales productos de degradación, como son la metilvinilcetona, el formaldehído y algunos de los compuestos hidroxicarbonílicos identificados experimentalmente.Sobre la metilvinilcetona, se ha estudiado la reacción de formación de los dos isómeros de la molécula, por medio de cálculos B3LYP y BHandHLYP, y con el método UMP2. También se muestra la reacción de interconversión de ambos isómeros, localizando el correspondiente estado de transición, no descrito hasta el momento.La formación de formaldehído se ha estudiado a partir de la reacción del radical CH2OH con O2. Nuestros resultados muestran la presencia de un estado de transición para la adición inicial de O2 no considerado hasta la fecha para esta reacción. Se han optimizado los puntos estacionarios de la hipersuperficie de energía potencial con los métodos UMP2 y QCISD. Además, se han realizado cálculos puntuales QCISD(T) sobre las geometrías optimizadas QCISD, para refinar los valores de energía potencial.Para el estado de transición TS1 se han hecho cálculos extra a nivel CCSD para la optimización, y cálculos puntuales CCSD(T) sobre la estructura optimizada QCISD. Asimismo, se han realizado cálculos Restricted Active Space SCF (RASSCF), con ayuda del programa MOLCAS.Finalmente, se ha estudiado la evolución de dos radicales dihidroxialquilo, originados como productos de la degradación intermedia del isopreno por el radical OH, resultando en la formación de dos hidroxicarbonilos identificados experimentalmente. La optimización de geometrías se ha realizado con el método UMP2, mientras que se ha utilizado el método QCISD(T) sobre las geometrías UMP2 para refinar los valores de la energía potencial.Además de estos trabajos, el becario D. Víctor Miguel Ramírez Ramírez ha colaborado activamente con otros miembros del grupo de investigación, en el estudio de la reacción entre el radical HCO y el O2, y la reacción entre el compuesto H2NO y el O3.

Characterization of the rpoN global regulatory gene of Pseudomonas syringae pv. syringae B728a and its impact on the plant-pathogen interaction

Gene regulation in bacteria is highly complex and requires the activity of sigma factors that function as transcriptional regulators. In Pseudomonas syringae pv. syringae B728a, 14 sigma factors have been identified. One of the more interesting is rpoN, encoding Sigma 54, which was initially described for its role in nitrogen utilization and later shown to be involved in regulating adhesion, motility, toxin production, and pathogenicity. The only commonality identified amongst these genes is that gene regulation by Sigma 54 is not essential for normal growth and development because mutational inactivation of rpoN is not lethal. Unlike Sigma 70, which recognizes promoter sites located at positions -10/-35 upstream of the transcription initiation site, Sigma 54 recognizes sites located at positions -12/-24. P.s. pv. syringae B728a encodes an RpoN that shares 80-98% identity with other Pseudomonas species. Promoter scans were conducted on the B728a genome to look for probable binding sites of RpoN. Analysis revealed that RpoN may be involved in regulating genes encoding ABC transporters, drug efflux pumps, flagella proteins, nitrate transporters, and several regulatory proteins. An insertional mutation in the rpoN gene was constructed in the B728a genome and a phenotypic analysis was initiated. Decreased swarming and adhesion ability of the rpoN mutant was observed as compared to B728a. The ability to utilize sole nitrogen sources was also affected. The rpoN mutant showed little or no growth on sole nitrogen sources such as alanine, histidine, lysine, and serine. Pathogenicity was shown to require a functional RpoN, as both HR and disease development was effected by an rpoN mutation. Pseudomonas syringae pv. syringae is most known for the production of two phytotoxins. Unlike RpoN in other species, in P.s. pv. syringae B728a it appears to indirectly down regulate toxin production of syringomycin and syringopeptin. The goal of this study was to characterize some of the important roles RpoN is known to possess and to understand its role in the plant pathogenic and epiphytic lifestyle of P. s. pv. syringae B728a.

Development of polymeric nanocomposites with enhanced distribution of catalytically active or bactericide nanoparticles

Esta tesis es un trabajo interdisciplinario dirigido a la obtención de nuevos nanocompuestos funcionales sintetizados a partir de materiales poliméricos intercambiadores de iones modificados por nanopartículas (NPs) de diferente composición. El desarrollo de nanomateriales es de gran interés en la actualidad dada sus especiales propiedades que difieren de las de la materia a escala macroscópica y que pueden resultar en ventajas tecnológicas. Sin embargo, existe cierta preocupación relativa a la toxicidad de estos. Por ello, en nuestro trabajo hemos desarrollado un nuevo método para la fabricación de nanomateriales con dos niveles de seguridad: la estabilización de las NPs en la matriz polimérica y la retención magnética. Por un lado, la metodología, desarrollada en este trabajo, de Síntesis Intermatricial (Intermatrix Synthesis, IMS) permite controlar el crecimiento y la estabilización de las NPs. Además, permite la obtención de NPs mono- o bicomponentes con estructura núcleo-envoltorio (core-shell). El core de esta estructura puede ser un material económico y/o con alguna propiedad adicional (p.e. magnetismo) y es recubierto con un material funcional específico para la aplicación de interés. El core magnético permite la recuperación de NPs que pudieran liberarse, por ejemplo, en un tratamiento de desinfección evitando así, la posible toxicidad por parte de las NPs. De la misma manera, se podrían recuperar nanocomposites con propiedades catalíticas con el objetivo de reutilizarlos en ciclos catalíticos sucesivos. Así, en este trabajo se ha logrado la optimización de la técnica IMS para la obtención de NPs monocomponentes (Ag, Co, Pd o Fe3O4) y con estructura core-shell (Ag@Co, Ag@Fe3O4 o Pd@Co) estabilizadas en matrices poliméricas de naturaleza diversas (membranas, fibras y resinas) y con diferentes grupos funcionales (sulfónicos, carboxílicos o de sales de amonio). Además, se han caracterizado los diferentes nanocomposites para evaluar sus propiedades. Las técnicas microscópicas (p.e. SEM y TEM) permiten conocer el tamaño y la localización de las NPs dentro del material. Otras técnicas (p.e. ICP-AES, XRD, XANES o SQUID) han permitido obtener información sobre la composición o las propiedades magnéticas de las NPs y nanocomposites. La distribución de las NPs concentrada en la superficie de la matriz es debido al efecto de exclusión de Donnan y a diversos parámetros optimizados. Así, las NPs son máximamente accesibles para las especies a tratar en la aplicación de interés. Por un lado, se ha demostrado y optimizado la actividad catalítica de nanocomposites basados en Pd y Pd@Co para la reacción de acoplamiento cruzado de Suzuki. También, se ha estudiado la aplicación de nanocomposites basados en Ag y con un núcleo magnético como agentes antibacterianos para el tratamiento de aguas obteniendo resultados claramente exitosos. Consideramos que este trabajo aporta y consolida conocimientos de interés relativos a la síntesis de materiales nanocomoposite polímero-metal y evaluación de sus aplicaciones. / This PhD thesis is an interdisciplinary work aimed at obtaining new nanocomposites synthesized from functional ion-exchange polymeric materials modified with nanoparticles (NPs) of different composition. The development of nanomaterials is of great current interest because of their special properties that differ from those of matter on a macroscopic scale and can result in technological advantages. However, there is some concern about the toxicity of those materials. Therefore, in this work we have developed a new method for the manufacture of nanomaterials with two levels of safety: the NPs stabilization in the polymer matrix and their magnetic retention. The Intermatrix Synthesis (IMS), method developed in this work, controls the growth and stabilization of the NPs. It also allows obtaining mono-or bicomponent NPs with core-shell structure. The core component of these structures can be an economic material and/or a material with some additional property of interest (e.g., magnetism). The core is coated with a specific functional material for the corresponding application of interest. Also, the magnetic core allows the recovery of the NPs which could be leached, for example, from a disinfection treatment and, thus, avoiding the possible toxicity from the NPs. Similarly, the recovery of NCs with catalytic properties in order to reuse them in successive catalytic cycles can also take place. Thus, in this work the IMS technique has been optimized for obtaining monocomponent NPs (Ag, Co, Pd or Fe3O4) and bicomponent core-shell NPs (Ag@Co, Ag@Fe3O4 or Pd @Co) stabilized in polymeric matrices of various physical types (films, fibres and resins) containing different functional groups (sulfonic, carboxylic or ammonium salts). Furthermore, we have characterized the different nanocomposites to evaluate their properties. Microscopic techniques (e.g., SEM and TEM) provide information about the size and location of the NPs within the material. Other techniques (e.g., ICP-AES, XRD, XANES or SQUID) have yielded information on the composition and magnetic properties of NPs and nanocomposites. The distribution of the NPs, concentrated on the surface of the matrix, is due to the Donnan-exclusion effect and some other various parameters optimized regarding the synthesis and polymer characteristics. Thus, the NPs are maximally accessible to the species involved in the application of interest. On one hand, the catalytic activity of Pd- and Pd@Co-based nanocomposites for the reaction of Suzuki cross-coupling has been shown and optimized. Also, the application of Ag and magnetic core-based nanocomposites as antibacterial agents for water treatment has been studied obtaining successful results. All in all, we believe this work provides and consolidates knowledge of interest related to polymer-metal nanocomposite materials and the evaluation of their applications.

Bifunctional nanocomposites: surface modification of reactive matrices with functional metal nanoparticles by intermatrix synthesis technique

En aquesta Tesi Doctoral s'ha desenvolupat la tècnica de Síntesi intermatricial (IMS), com a una metodologia factible per la modificació de matrius reactives amb nanopartícules (NPs) basada en les propietats d'intercanvi iònic de la superfície que es vol modificar; com ara: resines intercanviadores iòniques i la innovació en la modificació de nanofilms polimèrics funcionalitzats, nanotubs de carboni (CNTs) i nanodiamantes (NDs). L’efecte dels materials nanoestructutrats sobre el medi ambient és un dels temes més importants a la tecnologia en els darrers anys. A causa del seu alt grau de desenvolupament, producció, difusió i aplicació, les majors preocupacions associades als Nanomaterials (NMs) inclouen: a) l'elevada reactivitat i toxicitat de molts NMs en comparació dels seus anàlegs macroscòpics, b) l'absència de tècniques analítiques òptimes per a la seva determinació en el medi ambient i c) l'absència d'una legislació efectiva que regularitzi els nivells permesos de diversos NMs en sòl, aigua i aire. Per això és primordial la seguretat i estabilitat dels NMs a través de la seva incorporació en els Nanocomposites bifuncionals (BFNCs) La bifuncionalitat dels BFNCs és determinada per les propietats pròpies de la matriu en la qual les NPs metàl·liques (MNPs) són sintetitzades (per exemple polímers d'intercanvi iònic) i per les propietats respectives de les MNPs (magnetisme, activitat bactericida, nanocatlizadores). La modificació superficial de les matrius reactives amb MNPs es porta a terme a través de la IMS acoblada a l'Efecte d'exclusió de Donnan (DEE). D'aquesta manera, els BFNCs preparats amb valor afegit són caracteritzats, les seves propietats són avaluades i es comprova l'estabilitat i la distribució favorable de les MNPs principalment en la superfície dels BFNCs. La IMS inclou diferents possibilitats per a la preparació de MNPs i BFNCs. En una primera etapa es realitza un intercanvi iònic sobre la matriu per fixar els precursors de les NPs. Posteriorment la IMS pot incloure: a) Reducció dels ions a l'utilitzar un agent reductor com NaBH4 o b) precipitació de NMs com quantum dots (QDs) o òxids metàl·lics a l'afegir el contraió respectiu. Sigui quina sigui la ruta que s'empri, els grups funcionals d'intercanvi iònic propis de la matriu són regenerats; de manera que els cicles de IMS poden repetir-se per augmentar el gruix de les MNPs o bé per produir MNPs bimetàl·liques tipus core-shell. Una versió estesa de la IMS és presentada a l'utilitzar el desplaçament galvànic com a precursor per Au- MNPs i AgAu-MNPs a matrius catiòniques, utilitzant MNPs sintetitzades inicialment com nanoplantilles per a la cristal·lització de les noves MNPs mono- o bi- metàl·liques. Un avantatge molt important que aporta la tècnica IMS és la seva gran versatilitat, possibilitant la síntesi de BFNCs amb les propietats desitjades per a les diferents aplicacions: activitat bactericida, magnetisme, catàlisi heterogènia i electroquímica, entre d'altres. / En esta Tesis Doctoral se ha desarrollado la técnica de Síntesis Intermatricial (IMS), como una metodología factible para la modificación de matrices reactivas con Nanoparticulas(NPs) basada en las propiedades de intercambio iónico de la superficie que se desea modificar; tales como: como resinas intercambiadoras iónicas y la innovación en la modificación de nanofilms poliméricos funcionalizados, nanotubos de carbono (CNTs) y nanodiamantes (NDs). El efecto de los materiales nanoestructurados sobre el medio ambiente es uno de los temas más importantes de la tecnología en los últimos años. Dado su alto grado de desarrollo, producción, difusión y aplicación, las mayores preocupaciones asociadas a los Nanomateriales (NMs) incluyen: a) la elevada reactividad y toxicidad de muchos NMs en comparación con sus análogos macroscópicos, b) la ausencia de técnicas analíticas adecuadas para su determinación en el medio ambiente y c) la ausencia de una legislación efectiva que regularice los niveles permitidos de varios NMs en suelo, agua y aire. Por ello es primordial la seguridad y estabilidad de los NMs a través de su incorporación en los Nanocomposites Bifuncionales (BFNCs) La bifuncionalidad de los BFNCs es determinada por las propiedades propias de la matriz en la que las NPs metálicas (MNPs) son sintetizadas (por ejemplo polímeros de intercambio iónicos) y por las propiedades respectivas de las MNPs (magnetismo, actividad bactericida, nanocatlizadores). La modificación superficial de las matrices reactivas con las MNPs se lleva a cabo a través de la IMS acoplada al Efecto de Exclusión de Donnan (DEE). De esta manera, los BFNCs preparados con valor añadido son caracterizados, sus propiedades son evaluadas y se comprueba la estabilidad y la distribución favorable de las MNPs principalmente en la superficie de los BFNCs. La IMS incluye diferentes posibilidades para la preparación de MNPs en BFNCs. En una primera etapa se realiza un intercambio iónico sobre la matriz para fijar los precursores de las NPs. Posteriormente la IMS puede incluir: a) Reducción de los iones al utilizar un agente reductor como NaBH4 o b) precipitación de NMs como quantum dots (QDs) u óxidos metálicos al agregar el contraión respectivo. Sea cual sea la ruta que se siga, los grupos funcionales de intercambio iónico propios de la matriz son regenerados; por lo que los ciclos de IMS pueden repetirse para aumentar el grosor de las MNPs o bien para producir MNPs bimetálicas tipo core-shell. Una versión extendida de la IMS es presentada al utilizar el desplazamiento galvánico como precursor para Au- MNPs y AgAu-MNPs en matrices catiónicas, utilizando MNPs sintetizadas inicialmente como nanoplantillas para la cristalización de las nuevas MNPs mono o bimetálicas. Una ventaja muy importante que aporta la técnica IMS es su gran versatilidad, posibilitando la síntesis de BFNCs con las propiedades deseadas para las diferentes aplicaciones: actividad bactericida, magnetismo, catálisis heterogénea y electroquímica, entre otras. / In this Doctoral Thesis, the Intermatrix Synthesis (IMS) technique has been described as a feasible methodology for the modification of reactive matrices with Nanoparticles (NPs), which is based on the ion exchange properties of the NPs supporting surface, such as ion exchange resins and the innovation of the application of IMS on polymeric nanofilms, carbon nanotubes (CNTs) and nanodiamonds(NDs). The effects of nanostructured materials on the environment are one of the most important issues of technology in recent years. Given their high level of development, production, dissemination and application, the main concerns associated with nanomaterials (NMs) include: a) the high reactivity and toxicity of many NMs compared to their macroscopic analogues, b) the lack of appropriate analytical techniques for their determination in environment and c) the absence of effective legislation to regularize the convenient levels of various NMs in soil, water and air. Therefore, it is a must to ensure the security and stability of NMs through its incorporation into the Bifunctional Nanocomposites (BFNCs) The bifunctionality of BFNCs is determined by the specific properties of the matrix in which the metal NPs (MNPs) are synthesized (for example by ion exchange polymers) and by the respective properties of the MNPs (magnetism, bactericidal, nanocatlizadores). Surface modification of reactive matrices with MNPs is conducted through the IMS coupled to Donnan exclusion effect (DEE). Thus, the added value BFNCs prepared are characterized, their properties are evaluated and stability and the favourable distribution of the MNPs mainly on the surface of BFNCs is verified. The IMS includes different possibilities for the preparation of MNPs on BFNCs. In the first stage ion exchange is performed on the matrix to attach the NPs precursors. Subsequently, the IMS includes: a) Reduction of ions by using a reducing agent such as NaBH4 or b) precipitation of NMs as quantum dots (QDs) or metal oxides by adding the respective counterion. Whichever route is followed, the ion exchange functional groups of the matrix are regenerated; so IMS cycles can be repeated to increase the thickness of the MNPs or to produce bimetallic core-shell type MNPs. An extended version of the IMS is presented as using galvanic replacement for the preparation of Au- MNPs and AgAu-MNPs in cationic matrices using MNPs initially synthesized as nano-templates for crystallization of new mono or bimetallic MNPs. An important advantage provided by the IMS technique is its versatility, allowing the synthesis of BFNCs with the desired properties for different applications: bactericidal activity, magnetism, electrochemistry and heterogeneous catalysis, among others.

Membranes en la separació enantiomèrica del fàrmac propranolol. Desenvolupament i caracterització

Els sistemes de separació mitjançant membranes han guanyat importància com a tècniques de separació enantiomèrica. S'han investigat distintes configuracions de membranes.Per a la separació enantiomèrica de SR-propranolol, fàrmac usat per al tractament d'algunes anomalies cardiovasculars, s'han estudiat diferents transportadors selectius: N-alquil-hidroxiprolines i dialquil tartrats en membranes líquides, i el norbornadiè com a polímer quiral. Malgrat tots aquest sistemes mostraven resultats prometedors, cap d'ells ha estat investigat detalladament. La finalitat d'aquesta tesi ha consistit a desenvolupar i caracteritzar diferents sistemes de membrana per a la separació del fàrmac SR-propranolol. S'han investigat quatre sistemes distints de membranes: membranes líquides suportades (SLM), membranes compòsit activades (ACM), membranes activades quirals (CAM) i membranes basades en polisulfona quiral (CPS). En la majoria dels casos s'ha emprat com a transportador selectiu l'N-hexadecil-L-hidroxiprolina (HHP).L'estudi i la caracterització de cada sistema s'ha dut a terme mitjançant tècniques microscòpiques i experiments de transport usant quatre mòduls de membrana diferents: una cel·la de diàlisi, un mòdul micro espiral , un mòdul que permet el modelatge del transport i un mòdul de filtració frontal. L'estudi del sistema de SLM va permetre elucidar els dos mecanismes de transport implicats en aquest sistema de separació, així com discriminar els principals paràmetres que governen (en termes de flux i selectivitat) el comportament del sistema de membranes. En condicions òptimes de treball (fase de càrrega ajustada a pH 8 i concentració de transportador de C/A = 3) es va detectar un excés enantiomèric del 5.6%. Les membranes compòsit activades, formades per una capa porosa de polisulfona pel·liculada sobre un paper no teixit, i una capa densa superficial de poliamida, foren caracteritzades mitjançant distintes tècniques: microscòpia de força atòmica, transport de soluts, mesures d'angle de contacte, ressonància electrònica d'espí, i microscòpia electrònica d'escombratge. L'ús d'aquestes tècniques complementàries va permetre obtenir informació de la morfologia interna i superficial de les membranes, i alhora va confirmar la presència de transportador a l'interior de les membranes. Tot i així, al ser aplicades per a la separació enantiomèrica de propranolol no es va observar transport enantioselectiu. Les CAM, formades per un capa porosa de polisulfona pel·liculada sobre un paper no teixit, si que van permetre el transport enantioselectiu del fàrmac d'estudi. En aquest cas es va estudiar la influència sobre el sistema de la concentració de transportador selectiu en la membrana i del valor de pH de la solució receptora. L'estudi va revelar la relació entre el flux i la enantioselectivitat i va contribuir a proposar les preferències d'afinitat del transportador vers ambdós enantiòmers del fàrmac propranolol. Els bons resultats van esperonar el desenvolupament d'un nou sistema de membranes CAM emprant didodeciltartrat com a transportador selectiu. La comparació dels dos sistemes de CAM es va dur a terme mitjançant el modelatge del transport ambdós enantiòmers implicats. Els resultats mostraren que el segon sistema posseeix millors propietats per al transport enantioselectiu de propranolol.Per últim es van investigar les membranes basades en polisulfona quiral, que contenen el selector quiral enllaçat covalentment a la matriu polimèrica. La polisulfona fou derivatitzada seguint dues rutes sintètiques distintes, i van obtenir-se dos productes diferents: CPSA i CPSB. Ara bé, sols la CPSA va resultar adequada com a polímer. Les membranes obtingudes a partir de CPSA van ser caracteritzades. L'aplicació d'aquest tipus de membranes van donar lloc a resultats prometedors tan pel que fa al flux com a la enantioselectivitat, i van permetre confirmar la major afinitat del selector per a l'enantiòmer S-propranolol. En aquest cas, quan es va incorporant la quantitat més gran possible de transportador en la membrana es va obtenir un factor de separació de 1,1a les 96h d'experimentació. / Membrane systems have been gaining importance as enatioseparation techniques.Various membrane configurations, have been proposed to attain enantioseparation. Liquid membrane processes were firstly proposed, however they exhibit a rapid selectivity decrease with operating time and show low stability and short lifetime when tested under industrial separation conditions. Therefore, solid polymeric membranes have also been developed. For the case of SR-propranolol, a β-blocking drug used for treating certain cardiovascular anomalies different enantioselective carriers have been studied. N-n-alkyl-hydroxyprolines, and dialkyl tartrate were used in liquid membrane configurations, and norbornadiene was considered as chiral polymer. Although all these membrane systems showed promising results, none of them was investigated in detail. The purpose of the present work was the characterisation and development of membrane systems for the enantioselective separation of SR-propranolol. Four different membrane systems, based in supported liquid membranes (SLM), activated composite membranes (ACM), chiral activated membranes (CAM) and chiral polysulfone (CPS) membranes, have been investigated. In most of them, N-hexadecyl-L-hydroxyproline (HHP) was applied as chiral carrier.The study and characterization of each membrane type was carried out using microscopy techniques and transport experiments. The latest were conducted with four different membrane modules: a dialysis cell, a micro spiral module, a module capable of well-defining hydrodynamics conditions for transport modelling and a dead-end filtration module. The study of SLM, permitted the elucidation of the two transport mechanisms involved in these separation systems and allowed discriminating the principal parameters governing the performance of the membrane system, either in terms of flux as in terms of selectivity. Under operating conditions showing the best performance results (feed at pH 8, and carrier concentration up to C/A = 3) a 5.6% of enantiomeric excess was reached. Activated composite membranes (ACM) consisted of a porous polysulfone support casted on a none-woven fabric, with a top dense polyamide layer. ACM were characterized by different techniques, such as atomic force microscopy, solute transport, contact angle measurements, electron spin resonance, and scanning electron microscopy. The use of all these complementary techniques provided an extensive and concise description of ACM internal and surface morphology, together with the confirmation of the presence of the chiral carrier inside the membranes. However, when applied to the enantioseparation of propranolol enantioselectivity was not observed. CAM consisted in a porous polysulfone film, also casted on a non-woven fabric. This membrane type permitted the enantioselective transport of propranolol across the membranes. In that case, the influence of carrier concentration in the membrane as well as the striping phase pH were studied. These studies revealed the relationship between transport rate and enantioselectivity and contributed to propose the affinity preferences of the carrier by the two enantiomers. The good results obtained, motivated the development of another CAM system containing the chiral carrier didodecyltartrate. The comparison of the two equivalent systems was carried out by conveniently modelling the transport of both propranolol enantiomers. Results showed that the last system possesses better properties for the enantioseparation of propranolol.The last type of polymeric membranes assayed were based in chiral polysulfones CPS, containing the chiral carrier covalently bonded to the polysulfone polymeric structure. PS was chiraly derivatised following two different sysnthetic routes, and two corresponding chiral polysulfones were obtained, namely CPSA and CPSB. However when preparing the membranes, only CPSA was suitable as polymer. CPS membranes, obtained from CPSA were prepared and characterised. The application of these polymeric membranes showed promising results regarding both the transport rate and enantioselectivity, and confirmed the higher affinity of the carrier by the S-propranolol enantiomer. In that case, with membranes containing the highest CPSA amount, a separation factor of 1.1 was encountered at 96h of experiment.

Estudi computacional de mecanismes de reacció en sistemes organometàl·lics

La utilització de la química computacional com una eina addicional en l'estudi de mecanismes de reacció està esdevenint una pràctica cada cop més habitual. Aquesta tècnica es pot utilitzar per determinar el mecanisme d'una reacció per a la qual no es tenen prou dades experimentals, però també per donar suport o descartar un mecanisme concret. En aquesta tesi es presenten tres treballs que exemplifiquen cadascuna d'aquestes situacions. Per una banda s'ha estudiat el mecanisme d'isomerització d'al·lilamina a enamina catalitzat pel complex [(BINAP)Rh]+. Amb aquest estudi es descarta el mecanisme N-triggered proposat per Noyori i col·laboradors, i es proposa un nou mecanisme al·lílic que a diferència de l'anterior esta d'acord amb les dades experimentals. Per altra banda s'ha estudiat el mecanisme de la reacció del complex [(dppp)2Pt2H3]+ amb tiofè. Aquest estudi ha permès determinar perquè en aquesta reacció addicionalment al trencament del enllaç C-S que dóna al complex [(dppp)Pt(SC4H4-C,S)], es forma el complex [(dppp)2Pt2(μ-SC4H5-C,S)], producte de l'activació C-S i parcial hidrogenació del tiofè, observat per primera vegada amb aquest sistema. Finalment s'ha realitzat l'estudi teòric dels mecanismes cíclic i obert proposats pel procés de transmetal·lació en la reacció de Stille utilitzant [L2Pd(Ph)(X)] (L=PH3, AsH3) i Me2SnCH=CH2 com a reactius, i THF i clorobenzè com a solvent. Aquest estudi ha permès donar més detall als mecanismes proposats experimentalment i racionalitzar en quins casos la reacció anirà per un mecanisme cíclic i obert. / The application of Computational Chemistry as an additional tool in the study of reaction mechanisms is becoming a common practice. This technique can be used to determine reaction mechanisms when experimental evidence is not sufficient, or to support or rule out a specific mechanism. This thesis encloses three works that exemplify each of those situations. The isomerization of allylamines to enamines catalyzed by [(BINAP)Rh]+ has been studied. In this study the N-triggered mechanism proposed by Noyori and co-workers is ruled out by an allylic alternative more consistent with the experimental data. The C-S bond activation of thiophene by [(dppp)2Pt2H3]+ has been also studied. This reaction gives the mononuclear [(dppp)Pt(SC4H4-C,S)] metallacycle by activation of the thiophenic C-S bond exclusively, and the dinuclear [(dppp)2Pt2(μ SC4H5-C,S)]ClO4 complex by activation of the C-S bond and partial hydrogenation of thiophene. On the basis of our results we propose a plausible mechanism for the whole process. Finally, we also studied the cyclic and open mechanisms proposed for the transmetallation step in the Stille Cross-Coupling reaction, considering [L2Pd(Ph)(X)] (L=PH3, AsH3; X=Br,OTf) and Me2SnCH=CH2 as reactants, and THF and chlorobenzene as solvents. The mechanistic insight obtained in this study allows to rationalize in which experimental conditions one of those mechanisms (cyclic or open) will be preferred.

Estudi teòric de reaccions catalitzades per complexos d’or: cicloaddicions amb al·lens i oxidació d’alquins

En l’última dècada, el camp de la catàlisi homogènia amb complexos d’or ha experimentat un creixement exponencial degut principalment a la seva capacitat per catalitzar reaccions orgàniques via l’activació d’enllaços múltiples de carboni. Malgrat la rellevància d’aquestes reaccions, el seu funcionament, o més concretament el seu mecanisme és encara poc conegut. La causa principal resideix en que generalment són reaccions molt ràpides on és difícil caracteritzar intermedis de reacció, i per tant, les evidències experimentals que aporten informació de com evolucionen aquestes reaccions són escasses. En aquest context, aquesta tesi s’ha centrat en l’estudi dels mecanismes d’una sèrie de reaccions catalitzades per complexos d’or: cicloaddicions entre al·lens i diens, i oxidacions d’alquins terminals via la transferència d’un àtom d’oxigen d’un grup X-O. Aquests estudis s’han realitzat mitjançant l’ús de càlculs de la mecànica quàntica i en estreta col·laboració amb els grups experimentals que les han desenvolupat al laboratori. Per a les reaccions de cicloaddició intramoleculars d’al·lè-diens s’han investigat els mecanismes per a la formació dels productes de cicloaddició (4+3) i (4+2) emprant complexos d’or(I) i platí(II) com a catalitzadors. Els resultats obtinguts han permès identificar que ambdós mecanismes transcorren a través de la formació d’un intermedi heptacíclic producte d’una cicloaddició [4C+3C], i analitzar els efectes del catalitzador en la selectivitat d’un mecanisme en front de l’altre. També s’ha observat que els substituents en la posició terminal de l’al·lè provoquen unes diferències notables en el mecanisme. L’estudi per a les reaccions de cicloaddició intermoleculars entre al·lens i diens ha demostrat que el producte de cicloaddició (4+2) es pot obtenir via diferents mecanismes, cicloaddicions concertades ([4C+3C] o [4C+2C]) o per etapes, en funció de la disposició relativa entre els dos reactius, els substituents en el diè i la seva conformació (s-cis o s-trans). Amb el diè en la conformació s-trans s’han obtingut dos mecanisme de cicloaddició per etapes competitius per a la formació dels productes de cicloaddició (4+2) i (2+2). L’estudi destaca que modificant el lligand del catalitzador es pot afavorir un o altre producte de reacció. En el cas de les reaccions d’oxidació d’alquins terminals s’han investigat les reaccions on el grup X-O és un sulfòxid o un N-òxid. Per a la reacció d’oxiarilació entre un arilsulfòxid i un alquí terminal l’estudi ha permès determinar que per a les dues versions, intra- i intermolecular, el mecanisme transcorre a través d’un procés de reagrupament sigmatròpic [3,3’], en discrepància amb la proposta que existia a la bibliografia a través d’un intermedi del tipus carbè/carbocatió d’or. Aquest procés concertat està d’acord amb la regioselectivitat observada en el procés d’arilació, sempre en la posició orto del tiofenil. El mecanisme per a la reacció amb un grup N-òxid transcorre també via un procés concertat amb un cicle de sis membres en l’estat de transició. Per a aquesta reacció s’havia proposat un mecanismes per etapes via un intermedi carbè/carbocatió d’or, aquest mecanisme s’ha analitzat però tots els intents han estat infructuosos. A més, s’ha trobat un mecanisme alternatiu a través d’una espècie diradical, que tot i ser menys favorable que la via concertada, no es pot descartar ja que les diferències d’energia no són prou importants. Per tant, l’estudi permet concloure que les reaccions d’oxidació d’un alquí terminal amb un grup X-O transcorren via un procés concertat sense que intermedis carbè/carbocatió d’or formin part del camí de reacció. En conjunt, els resultats obtinguts en aquesta tesi demostren que els mètodes computacionals són una eina molt útil per estudiar mecanismes de reacció en l’àrea de la catàlisi homogènia amb metalls de transició. / In the last decade, the field of homogeneous catalysis with gold complexes has experienced exponential growth mainly due to their ability to catalyze organic reactions via carbon multiple bond activation. Despite the importance of these reactions, its operation, or more specifically its mechanism is still little known. The main cause lies in that these reactions are generally very fast and the characterization of reaction intermediates is difficult, hence experimental evidences, that provide information on how these reactions evolve, are rare. In this context, this thesis has focused on the study of the mechanisms for a series of reactions catalyzed by gold complexes: cycloadditions between allenes and dienes, and terminal alkynes oxidations via oxygen-transfer from a X-O group. These studies have been carried out using quantum mechanic calculations and in close collaboration with experimental groups that have developed them in the laboratory. The mechanism for the (4+3) and (4+2) cycloaddition products have been investigated for the intramolecular cycloadditions of allenedienes catalyzed by gold(I) and platinum(II) complexes. The results have made it possible to identify that both mechanisms are set through the formation of a heptacyclic intermediate, product of a [4C+3C] cycloaddition, and to analyze the effects of catalyst in the selectivity of each pathway. It has also been observed that the substituents in the terminal allene position induce notable differences on the mechanism. The study on the intermolecular cycloaddition reactions between allenes and dienes has shown that the (4+2) cycloaddition product can be obtained via different mechanisms, concerted ([4C+3C] or [4C+2C]) or stepwise cycloadditions, depending on the relative arrangement of both substrates, the diene substituents and its conformation (s-cis and s-trans). The reaction with s-trans diene can evolve through two competitive stepwise cycloaddition pathways obtaining (4+2) or (2+2) cycloaddition products. The study shows that, by modifying the catalyst's ligand, one or the other reaction mechanism can be favored. For the oxidative reactions of terminal alkynes, the particular cases where the X-O group is a sulfoxide or N-oxide have been investigated. The study of the oxyarylation reaction between a terminal alkyne and an arylsulfoxide has allowed to determine that for both versions intra- and intermolecular, the mechanism run through a [3,3’] sigmatropic rearrangement process. These results are in disagree with the previous mechanism on the bibliography, that proposed a type of gold carbene/carbocation intermediate. Moreover, the concerted mechanism is accorded with the regioselectivity observed in the arylation process, always in the tiofenil ortho-position. The reaction mechanism for the N-oxide group evolves also through a concerted process with a six member cycle. For this reaction was also previously proposed a stepwise mechanism involving a gold carbene/carbocation intermediate, this mechanism was investigated but all the attempts were unsuccessful. In addition, it has found an alternative mechanism through a diradical species; despite this alternative pathway is less favored than the concerted one, it cannot be ruled out due to the energy differences are not important enough. Therefore, the study allows concluding that oxidation reactions of terminal alkynes with a X-O group takes place via a concerted process without presence of gold carbene/carbocation intermediates on the reaction path. To sum up, the results obtained in this thesis demonstrate that computational methods are a very useful tool for studying reaction mechanisms in the field of homogeneous catalysis with transition metals.
