Forensic sample analysis using supercritical fluid extraction coupled with solid phase microextraction

A third of adults in the UK admit to using controlled substances at least once hi their lives and the most commonly abused drug was found to be cannabis, followed by the opiates and ecognine derivatives. Forensic laboratories are continually trying to reduce the turnaround time for samples submitted for analysis, to less than the current time of four days. One of the main aims of this work was to reduce the time taken to prepare, extract and analyse hair for target analytes. A new method combining supercritical fluid extraction and solid phase micro-extraction (SFE/SPME) is introduced. This coupled procedure utilises the advantages of using supercritical carbon dioxide as an extraction solvent and collecting the extract on a selected SPME fibre. Together the combined techniques provide a method that is solvent-free and highly selective, whilst reducing sample preparation to a minimum. Digested hair samples are examined by SEM to find the best method. Alkali hydrolysis and enzyme digestion using proteinase K were found to be the most effective pre-extraction methods. Solid hair samples with the addition of a derivatization reagent (BSTFA) gave the best results using the coupled technique. After optimisation and validation, the extraction from both real and spiked samples resulted in recoveries between 60% to 80% for the target analytes A9 -tetrahydrocannabinol (A9- THC) , cannabinol (CBN) and cannabidiol (CBD). Other forensic applications such as the extraction of polycyclic aromatic hydrocarbons and pesticides from environmental and food samples and the examination of fire debris for the presence of accelerants are also carried out. PAH could easily be extracted from complex matrices to give recoveries of up to 97% for some analytes at the optimized conditions of 90°C at 4500 psi over a 10 minute dynamic extraction period. Pesticide extraction gave recoveries of 60 - 80% while the extraction of accelerants from fire debris clearly showed that accelerants such as diesel can be identified over the coincidental PAH and hydrocarbon peaks found in burnt debris. The coupling of SFE to SPME provides an exciting technique that has a wide range of applications and creates potential for a fully automated on-line system.

I have chosen the title for this thesis, "Reactions in the Solid State", for two reasons: Firstly, it is broad enough to cover all of my areas of research, which have been: • Effects of irradiation on solids (PhD topic) • Silver refining (while at the Chamber of Mines) • Kinetics of decomposition of solids (with Dr A.K. Galwey and various others) • Techniques of thermal analysis • Pyrotechnic delay systems (with support from AECI Explosives) • Thermal and photostability of drugs (with Prof B.D. Glass) and, secondly, it was the title of the very successful book co-authored by Drs Andrew Galwey, David Dollimore and me. A large part of my research has been involved in the writing and editing of books, so these are covered in a separate commentary, while commentary on the more than 100 papers to which I have contributed forms the main part of this compilation. It is hoped that the electronic format will enable ready access of to all aspects of my research, including electronic versions of the original papers. The reader will need a copy of Adobe Acrobat Reader to access these.

Tais avaliações impactam diretamente nos referenciamentos de crianças e adolescentes para a rede de atendimento, ocasionando decisões equivocadas e fazendo com que as notificações sejam realizadas a partir de critérios fatuais, caracterizando a falta de um trabalho em rede e de ações intersetoriais. Neste ponto, ferramentas de corte quantitativo para a tomada de decisão dos sujeitos que compõem a rede de atendimento é uma lacuna que pode ser explorada. Sendo assim, o objetivo da pesquisa é elaborar um protocolo que auxilie as instituições integrantes do Sistema de Garantia de Direitos dos municípios de Uberaba/MG e Bauru/SP a gerenciar as notificações de violência contra crianças e adolescentes. A pesquisa utiliza abordagem quantitativa, com a utilização da técnica de modelagem. Nesse sentido, o Analytic Network Process (ANP) torna-se um método de apoio à decisão apropriado, relacionado os direitos fundamentais das crianças e adolescentes com os tipos de violência cometidos e selecionando o setor prioritário que os sujeitos devem ser atendidos. A pesquisa apontou que, de um ponto de vista geral, o setor de Saúde deve ser o encaminhamento prioritário para as vítimas. Nos casos específicos de Negligência e Violência Física, a Assistência Social e o Conselho Tutelar são os setores prioritários, respectivamente. O setor de Saúde obteve a prioridade para os casos de Violência Psicológica e Violência Sexual. O protocolo de atendimento foi desenvolvido com base nas análises especificas de cada tipologia, apresentando os fluxos de encaminhamentos dentro das redes de proteção das cidades. A pesquisa quantitativa, com a utilização do ANP, proveu uma maior cientificidade ao assunto e pode ser considerada um avanço para futuras políticas públicas de proteção. / Violence against children and adolescents in Brazil presents a historically complex scenario, in which problems are often subjected to individual evaluations, counteracting violence, which presents many meanings. These evaluations directly impact on children and adolescents referrals to the service network, causing mistaken decisions and making notifications based on factual criteria, characterizing the lack of networking and intersetorial actions. At this point, quantitative cutting tools for the decision making of the subjects that make up the service network is a gap that can be explored. Therefore, the aim of the research is to elaborate a protocol that will help the institutions that are part of the System of Guarantee of Rights of the municipalities of Uberaba / MG and Bauru / SP to manage the notifications of violence against children and adolescents. The research uses a quantitative approach, using the modeling technique. In this sense, the Analytic Network Process (ANP) becomes a method to support the appropriate decision, related the fundamental rights of children and adolescents with the types of violence committed and selecting the priority sector that the subjects should be attended. The research pointed out that, from a general point of view, the Health sector should be the priority route for the victims. In the specific cases of Neglect and Physical Violence, Social Assistance and the Guardianship Council are the priority sectors, respectively. The Health sector has given priority to cases of Psychological Violence and Sexual Violence. The service protocol was developed based on the specific analysis of each typology, presenting the routing flows within the protection networks of the cities. Quantitative research, using ANP, has moved a greater scientific relevance to the subject and can be considered as an advance for future public protection policies.
