Kan nätverk konstrueras? : En studie av relationerna och nätverksbyggandet kring Kista Science City AB

Ett väl fungerande nätverk kan hjälpa företag att förbättra sin produktivitet och skapa möjligheter till affärer. För att utnyttja fördelarna som nätverk kan skapa har Kista Science City AB sedan år 2000 aktivt arbetat med att skapa olika projekt och nätverk, med avsikt att stimulera tillväxt i regionen Kista Science City, framförallt hos de mindre företagen. Syftet med denna uppsats är att beskriva de relationer som företagen inom nätverken kopplade till Kista Science City AB har, med hjälp av ARA-modellen. För att besvara syftet så har en kvalitativ intervjubaserad fallstudie utförts. Uppsatsen visar att det finns störst potential till starka relationer gentemot externa aktörer och att det konstruerade nätverket är begränsat till någon del av de lager som nämns i ARA-modellen.

Remembering the River: The Retrieval of Rome's Forgotten Relationship with the Tiber

Ever since its founding, Rome has been vulnerable to the swelling waters of its Tiber River. This river was so important to the city that it was a defining character in Rome’s history for over two thousand years. However, this river-city relationship would be suddenly severed in the late 19th century as Rome was declared the Italian capital. And, with the creation of the capital, came the creation of the river walls. While this new infrastructure safeguarded the city from future flooding, it razed the relationship between the city and its river. It lost its use as a commercial trade route, transportation system and leisure landscape, and before long the Tiber was forgotten. This thesis proposes a design intervention at Rome’s historic river-city site, the Porto di Ripetta. Once the physical and symbolic gateway to the city, the Ripetta is presently the most disconnected site along the Tiber River. Not only does the proposed project aim to synergistically unite a series of complex archaeological layers from antiquity to present-day, it also aspires to reconnect the city to its historical relationship with its river, introduce another layer within Rome’s transportation network, and expand the city’s cultural agenda along its underutilized continuous corridor. The introduction of river-based programme is logical when created within a series of design solutions that both recognize and address the temporal nature of the riverscape. Through in-depth historical analysis, this thesis examines the complexities of the Tiber River’s existence and analyzes its sociological, physical and political importance to the Eternal City. This understanding of the Tiber River’s unique qualities reveals tangible opportunities for new public spaces connected the potential of the Tiber as part of an expanded network of new public transportation, leisure landscapes, and cultural institutions.

Syfte: Syftet med den här uppsatsen är att förstå lokala Resebyråers relationer och samverkan ur ett nätverksperspektiv, för att kunna analysera produktutvecklingen och marknadsföringen. Med koppling till de valda syfte har följande forskningsfråga valts: Hur kan resebyråer använda sina relationer med leverantörer och samarbetspartners för att förbättra marknadsföringen och produktutvecklingen? Metod: Uppsatsen utgår från en kvalitativ undersökningsmetod och har en abduktiv ansats. Uppsatsen behandlar relationer och nätverk, vilket behandlas i ett eget kapitel. Informationen till kapitlet är insamlat genom sekundärkällor. Där efter presenteras och analyseras resultatet, resultatet är material från semistrukturerade intervjuer och telefonintervjuer. Slutsatser: Under arbetsprocessen har vi slutligen kunnat se hur resebyråer använder sina relationer och nätverk i utvecklingen av marknadsföring och produktutveckling. Vi har identifierat både likheter och olikheter med resebyråernas relationer och nätverk, men även möjligheter för resebyråerna att utveckla marknadsföringen och produktutvecklingen.

O objetivo do presente estudo foi caracterizar a estrutura genética populacional de duas espécies de araras: Ara chloropterus e Ara macao. Foram analisadas amostras de sangue e penas de diferentes regiões no Brasil, de uma localidade na Bolívia e outra no Peru. Foram realizadas análises com DNA mitocondrial (região controladora e citocromo oxidase I) e nuclear (microssatélites) das duas espécies. Para A. chloropterus foram obtidos 2166 pares de base do DNA mitocondrial de 89 amostras e dados de seis locos de microssatélites de 95 amostras. A rede de haplótipos e a árvore de neighbor-joining construídas com dados mitocondriais e os índices de FST obtidos com os dois marcadores revelaram fraca estruturação genética. Isso pode ser devido a alto fluxo gênico apresentado ou retenção de polimorfismo ancestral. Portanto, a espécie parece se organizar em metapopulações (baixa estruturação genética e alto fluxo gênico). Nesse caso, seria interessante conservar indivíduos de diversas localidades e seus corredores. Para Muscular Dystrophy foram obtidos 2094 pares de base do DNA mitocondrial de 68 amostras e dados de sete locos de microssatélites de 64 amostras. A rede de haplótipos e a árvore de neighbor-joining construídas com dados mitocondriais e os índices de FST obtidos com os dois marcadores indicam ausência de diferenciação genética entre as localidades estudadas. A análise demográfica dessa espécie indica expansão populacional há pouco mais de 50.000 anos atrás e declínio populacional desde o último período máximo de glaciação. Estes resultados sugerem que essa espécie é constituída de uma única grande população que poderia ser considerada como uma única unidade de manejo caso outras diferenças (ex.: adaptações ecológicas locais) não sejam encontradas. Ambas as espécies estudadas apresentam alta diversidade genética, possivelmente devido a um intenso fluxo gênico dentro de cada uma. / The present study aimed to characterize the population genetic structure of two macaw species: Ara chloropterus and Ara macao. Caracterização de subpopulações de Leucemia Mielóide Aguda portadora do rearranjo MLL quanto à resposta diferencial ao tratamento em longo prazo com Citarabina / Characterization of subpopulations of Acute Myeloid Leukemia harboring MLL rearrangements according to differential response to the long-term treatment with Cytarabine

A natureza heterogênea da Leucemia Mielóide Aguda (LMA) tornou-se um desafio para o sucesso da quimioterapia convencional com o agente Citarabina (Ara-C), especialmente em leucemias com prognóstico desfavorável, como aquelas portadoras do rearranjo MLL. Visto que as células de LMA-MLL são consideradas sensíveis ao Ara-C quando comparadas às leucemias que não apresentam o rearranjo, mas a recaída à doença é frequente, a presente tese propôs estudar a relação entre características biológicas relacionadas às bases da resistêmcia ao Ara-C em LMA-MLL. A abordagem proposta foi a seleção de subpopulações de linhagens celulares portadoras do rearranjo MLL submetidas ao tratamento em longo prazo com Ara-C, comparando-as com as linhagens não expostas à droga. As células foram caracterizadas quanto: 1) ao potencial proliferativo na presença ou ausência de Ara-C; 2) a distribuição das células no ciclo celular; 3) a distribuição de marcadores clássicos de superfície de células-tronco hematopoiéticas, CD34 e CD38; e 4) o perfil de expressão global dos RNAs transcritos. O tratamento em longo prazo selecionou células mais resistentes ao Ara-C que as células parentais. Além disso, quanto ao ciclo celular, as células selecionadas com Ara-C apresentaram apoptose reduzida (fase sub-G1), acúmulo na fase de síntese (fase S) e aumento da capacidade proliferativa após reexposição à droga (fase G2-M). Quanto à análise de marcadores de células-tronco hematopoiéticas, observou-se que após o tratamento em longo com Ara-C, uma das linhagens celulares apresentou distribuição bimodal do marcador CD38. Quando separadas por sorting em citometria de fluxo, observou-se que as subpopulações com níveis distintos de expressão de CD38, denominadas MV-4-11 CD38High e MV-4-11 CD38Low apresentaram resposta distinta ao tratamento com Ara-C. Quando avaliadas quanto ao perfil global de expressão gênica, constatou-se que MV-4-11 CD38High eram mais semelhantes às células parentais, e que MV-4-11 CD38Low formavam um grupo isolado, distinto das outras duas populações celulares. A análise de ontologia gênica (GO) evidenciou que entre as categorias mais representativas de processos biológicos estavam atividades associadas à capacidade proliferativa, ao desenvolvimento e a resposta a estímulos. As análises de agrupamentos hierárquicos mostraram que: 1) o cluster de genes do desenvolvimento HOXA estava mais expresso nas células MV-4-11 CD38Low do que em MV-4-11 CD38High, que apresentaram expressão mais elevada do cluster HOXB; 2) o gene HOX mais diferencialmente expresso foi HOXA13, associado na literatura com prognóstico desfavorável em outros tipos de câncer; 3) dos genes associados a resposta a estímulos, o único relacionado à via de metabolização do Ara-C diferencialmente expresso entre as linhagens foi NME1; 4) aqueles que participam das vias de reparo de pareamento incorreto, reparo por excisão de bases e por excisão de nucleotídeos encontraram-se mais expressos nas células MV-4-11 CD38High que em MV-4-11 CD38Low. Além disso, diversas quinases dependentes de ciclinas (CDKs) também estiveram diferencialmente expressas entre MV-4-11 CD38High e MV-4-11 CD38Low. Sugere-se por fim, que o modelo in vitro proposto neste estudo para simular a situação de resistência ao Ara-C em subpopulações de LMA-MLL, demonstrou que os mecanismos de resposta à Citarabina nesta doença, vão além de alterações na detoxificação e metabolização da droga, e parecem mais associados a vantagens proliferativas e do desenvolvimento das células leucêmicas. Estas vias devem ser exploradas como alvos potenciais na terapia combinada ao Ara-C. / The heterogeneity of Acute Myeloid Leukemia (AML) became a challenge for the success of the conventional chemotherapy agent Cytarabine (Ara-C), especially in leukemias with poor prognosis, as those harboring MLL rearrangement. Since AML-MLL cells are considered sensitive to Ara-C when compared with leukemias that do not carry the rearrangement, but relapse is frequent, the present dissertation proposed to study the relationship between biological characteristics related to the basis of chemoresistance to Ara-C in AML-MLL. We proposed an approach based on the selection of subpopulations of cell lines bearing MLL rearrangement submitted to the long-term treatment with Ara-C, comparing them with the cell lines that were not previously exposed to the drug. The cells were characterized according to: 1) the proliferative potential in the presence and absence of Ara-C; 2) the distribution of the cells in the cell cycle; 3) distribution of hematopoietic stem cell classic surface markers, CD34 and CD38; and, 4) global expression profile of transcribed RNAs. The long-term treatment selected cells that are more resistant to Ara-C than the cells that were not previously treated (parental cells). Besides, according to cell cycle, the cells selected by Ara-C treatment present decreased apoptosis (sub-G1 phase), accumulation in the synthesis phase (S-phase) and increase in the proliferative capability after re-exposition to the drug (G2-M phase). Regarding the hematopoietic stem cell markers, we observed that after Ara-C long-term treatment, one of the cell lines exhibited a bimodal distribution of the CD38 marker. When sorted by flow cytometry, we observed that both subpopulations with distinct levels of CD38 expression, called MV-4-11 CD38High and MV-4-11 CD38Low also showed distinct response to Ara-C. When evaluated regarding to their global gene expression profiles, we verified that MV-4-11 CD38High were more closely related to the parental cells, and MV-4-11 CD38Low made up an isolated group, distinct of the other cell populations. Gene ontology (GO) analysis revealed that among the most representative categories of biological processes, activities associated with proliferative capability, development and response to stimuli were included. The hierarchical clustering analysis showed that: 1) the cluster HOXA of genes of development was more expressed in the MV-4-11 CD38Low than in the MV-4-11 CD38High cells, that presented increased expression of HOXB cluster; 2) the most differentially expressed HOX gene was HOXA13, which according to the literature is associated with poor prognosis in other types of cancer; 3) among the genes associated with response to stimuli, the only one related to Ara-C-metabolizing pathway that was differentially expressed between the cell lines was NME1; 4) those genes that take part in the mismatch repair, base excision repair and nucleotide excision repair pathways were more expressed in the MV-4-11 CD38High than in the MV-4-11 CD38Low cells. Additionally, several cyclin-dependent kinases (CDKs) were also differentially expressed between MV-4-11 CD38High and MV-4-11 CD38Low. Finally, we suggest that the in vitro model proposed in this study to mimic the situation of chemoresistance to Ara-C in subpopulations of AML-MLL, showed that the mechanisms of Ara-C response in this disease, go beyond changes in drug detoxification and metabolization, and seem more associated to proliferative and development advantages of the leukemic cells. These pathways should be explored as potential targets to Ara-C combination therapies.

Targeting the nucleotide metabolism of the mammalian pathogen Trypanosoma brucei

Trypanosoma brucei causes African sleeping sickness in humans and Nagana in cattle. There are no vaccines available against the disease and the current treatment is also not satisfactory because of inefficacy and numerous side effects of the used drugs. T. brucei lacks de novo synthesis of purine nucleosides; hence it depends on the host to make its purine nucleotides. T. brucei has a high affinity adenosine kinase (TbAK), which phosphorylates adenosine, deoxyadenosine (dAdo), inosine and their analogs. RNAi experiments confirmed that TbAK is responsible for the salvage of dAdo and the toxicity of its substrate analogs. Cell growth assays with the dAdo analogs, Ara-A and F-Ara-A, suggested that TbAK could be exploited for drug development against the disease. It has previously been shown that when T. brucei cells were cultivated in the presence of 1 mM deoxyadenosine (dAdo), they showed accumulation of dATP and depletion of ATP nucleotides. The altered nucleotide levels were toxic to the trypanosomes. However the salvage of dAdo in trypanosomes was dramatically reduced below 0.5 mM dAdo. Radiolabeled dAdo experiments showed that it (especially at low concentrations) is cleaved to adenine and converted to ATP. The recombinant methylthioadenosine phosphorylase (TbMTAP) cleaved methylthioadenosine, dAdo and adenosine into adenine and sugar-1-P in a phosphate-dependent manner. The trypanosomes became more sensitive to dAdo when TbMTAP was down-regulated in RNAi experiments. The RNAi experiments confirmed that trypanosomes avoid dATP accumulation by cleaving dAdo. The TbMTAP cleavage-resistant nucleoside analogs, FANA-A and Ara-A, successfully cured T. brucei-infected mice. The DNA building block dTTP can be synthesized either via thymidylate synthase in the de novo pathway or via thymidine kinase (TK) by salvage synthesis. We found that T. brucei and three other parasites contain a tandem TK where the gene sequence was repeated twice or four times in a single open reading frame. The recombinant T. brucei TK, which belongs to the TK1 family, showed broad substrate specificity. The enzyme phosphorylated the pyrimidine nucleosides thymidine and deoxyuridine, as well as the purine nucleosides deoxyinosine and deoxyguanosine. When the repeated sequences of the tandem TbTK were expressed individually as domains, only domain 2 was active. However, the protein could not dimerize and had a 5-fold reduced affinity to its pyrimidine substrates but a similar turnover number as the full-length enzyme. The expressed domain 1 was inactive and sequence analysis revealed that some active residues, which are needed for substrate binding and catalysis, are absent. Generally, the TK1 family enzymes form dimers or tetramers and the quaternary structure is linked to the affinity for the substrates. The covalently linked inactive domain-1 helps domain-2 to form a pseudodimer for the efficient binding of substrates. In addition, we discovered a repetition of an 89-bp sequence in both domain 1 and domain 2, which suggests a genetic exchange between the two domains. T. brucei is very dependent on de novo synthesis via ribonucleotide reductase (RNR) for the production of dNTPs. Even though T. brucei RNR belongs to the class Ia RNR family and contains an ATP-binding cone, it lacks inhibition by dATP. The mechanism behind the RNR activation by ATP and inactivation by dATP was a puzzle for a long time in the ~50 years of RNR research. We carried out oligomerization studies on mouse and E. coli RNRs, which belongs to the same family as T. brucei, to get an understanding of the molecular mechanism behind overall activity regulation.
