Giglioti, Rodrigo. January 2010 (has links)
Nowadays, his control is complicated by the slowness that acaricides molecules are generated, synthesized through and by the fast development of the resistance of these ticks to the market available products. This work aims to study the effect of four extracts of Neem seed oil (Azadirachta indica) containing 2.000, 5.000, 9.000 and 10.000 ppm of azadirachtin (AZA) quantified by high performance liquid chromatography (HPLC) on engorged females and larvae of R. (B.) microplus, in vitro. For the tests with engorged females, the Adult Immersion Test was used and for the larvae was used the Larvae Package Test. The test results with the engorged females showed that the main toxic effect produced by the extracts is related to the inhibition of the parasite reproduction. Were observed reductions in oviposition and hatchability rates of the larvae derived from the female treated with several dilutions of extracts, compared to controls. In the Product Effectiveness (PE) for all the test solutions, the 10.000ppm (N10) solution showed the highest efficiency. The analysis results from the tests showed significance (p<0,01) of the two effects included in the model, indicating variations both due to AZA and to the test performed. In the lethal concentration to the engorged females was verified that the CL50 did not show great variation, while the CL90 showed great variation between the studied extracts. In the tests with larvae was not observed mortality of them, indicating zero effectiveness of all tested extracts. The results of the tests with R. (B.) microplus engorged females demonstrated that the extract of Neem seeds oils showed acaricide activity. Thus, it is suggested that further studies can be conducted, such as development of the methods and of excrection, and characterization of other secondary ... Also planned is the introduction of the Environmental and Human Health Risk Assessment for pesticides both based on study results that follow internationally recognized protocols, however, all these those study protocols, including bee toxicity, may not be adequate to represent the Brazilian reality, once they are applied mostly in temperate countries. In this sense, there is growing concern about bees and other pollinators that possess universal importance to the economy, agriculture and environmental balance on the planet. The disappearance of these insects of the hives, a phenomenon commonly known as CCD (Colony Collapse Disorder) has been discussed nowadays in Brazil and worldwide. This study aimed to identify and study the existing protocols for assessing the toxicity of pesticides to bees, as well as to discuss alternative studies to better adapt the protocols used abroad to the Brazilian scenario. In order to do that, studies were conducted on survival, walking behavior, proboscis extention response (P.E.R), nutrition and morphology of hypopharyngeal glands in africanized Apis mellifera exposed to the insecticide Azamax®. Results have demonstrated that bee survivial can be diminished when those insects are exposed to the insecticide at field doses and also that bees present morphological changes in the areas of acini hypopharyngeal glands, preventing its development when young worker bees were exposed to three different doses of insecticide. These findings indicated the importance of exploring pesticides sub lethal effects on insects, observing parameters such as walking behavior, PER (Proboscis Extension Response), nutrition and morphology of hypopharingeal glands, instead of only consider acute toxicity studies, like it happens nowadays, when reviewing the test protocols that already exists, considering not only hazard evaluation, but also the dose and exposition scenery.

Desempenho de leucochrysa sp. (Neuroptera Chrysopidae) em diferentes presas e sua seletividade a produtos fitossanitários / Performance of Leucochrysa sp. (Neuroptera: Chrysopidae) on different prey and theselectivity of phytosanitary products

Costa, Simone Silva da 19 December 2016 (has links)
Green lacewings (Neuroptera: Chysopidae) are predatory insects with potential to be used in biological control programs of agricultural pests. This work aimed to evaluate the biological aspects of Leucochrysa sp. on different prey species and to verify the selectivity of phytosanitary products. The bioassays were conducted in climatized rooms regulated to a temperature of 25 ± 2 °C, 70 ± 10% R.H. and 12h photophase. The insects were individualized in rectangular boxes of 220 mL until the adult emergence. The duration and viability of larval instars and pupal period were evaluated. The formed couples (n = 20) were kept in PVC cages internally coated with sulphite paper. A period of 120 days was established to evaluate the fecundity and fertility of females. The predator had its best performance when fed with eggs, nymphs and pupae of Aleurocanthuswoglumi Ashby, 1915 (Hemiptera: Aleyrodidae); nymhs and adults of Toxopteracitricida (Kirkaldy, 1907) (Hemiptera: Aphididae) and Brevicorynebrassicae (Linnaeus, 1758) (Hemiptera: Aphididae), respectively, whose larval-adult duration ranged from 34.00 to 37.77 days. When submitted to eggs, nymphs and pupae of Aleurodicusmagnificus Costa Lima, 1928 (Hemiptera: Aleyrodidae) as food source, the green lacewing did not complete its cycle.Regarding the polynomial regression analyzes performed to investigate the functional response patterns on the aphids B. brassicae and T. citricida, a type II response was revealed for the second and third instar of the predator, which is represented by an asymptotic curve, in which the predator is increasing the prey consumption until saturation occurs. The functional response pattern was not altered by the influence of different prey stages used in this study. In the IPM the use of agrochemicals should be performed when they present some type of selectivity, thus, Neem based products were evaluated upon eggs and larvae of second and third instars of Leucochrysa sp. The insects were kept in Petri dishes, sprayed and individually transferred to plastic boxes until adult emergence. The larvae were fed with eggs of Anagastakuehniella(Zeller, 1879) (Lepidoptera: Pyralidae). The formed couples (n = 15) from treated eggs, were fed with beer yeast and honey (1: 1).Azamax®; Nim-I-Go® and Emulzinim® were classified as harmless; Ethanolic extract of neem and NeenMax® as slightly harmful, when sprayed on eggs of the predator. Azamax®; Nim-I-Go®, Emulzinim® and Ethanolic extract of neem were classified as harmless when applied to second and third instar larvae. Deltamethrin was considered harmful to eggs and second instar larvae and moderately harmful to third instar larvae.

Nouveau procédé de fractionnement de la graine de Neem (Azadirachta Indica A. Jussi) sénégalais : production d'un bio-pesticide d'huile et de tourteau / New fractionation process of Neem (Azadirachta Andica A. Jussi) seed : production of bio-pesticide, oil and cake

Faye, Mamadou 29 November 2010 (has links)
Originally from India, neem (Azadirachta indica Juss.) is a tree found in the dry tropics and subtropics of Asia, America and Africa. In Senegal, it produces an average of 50 kg per year of a fruit as a drupe ellipsoid, which generally contains a seed, with an average weight of 270 ± 30 to 91% dry matter. The variability of seeds (45 to 48% of capsular cellulose 52-54%, 24-28% hemicellulose, lignin 7-9% 52-55% almonds: 45 to 58% lipids, proteins 20 to 23% j) and extracted oils (palmitic acid: 17 to 19%, stearic: 14.5 to 17.5%, oleic acid: 42 to 48.5%, linoleic: 16.5 to 20% total sterols: 3 5 g / kg) was analyzed for five regions of Senegal. Extraction of azadirachtin, the main active ingredient of the plant seed by different solvents (methanol, ethanol, water) shows a content of 2 g per kg of seed, mainly localized in the kernel (3.9 g / kg ). The expression conditions of neem seed oil were studied in single-screw press OMEGA 20 and twin-screw CLEXTRAL BC 21. Under optimum conditions, 65 and 68% of the oil is expressed, from 7 to 10% of azadirachtin are entrained with the oil and meal contains 14% fat, 14% protein and about 2 g / kg azadirachtin. Watery splitting under intense shear seeds, led to the extraction of 55% lipids, 50% protein and 80% of azadirachtin, separated as a stable emulsion, composition: water 70% oil 25%, 4% protein, 6 g azadirachtin / kg, and an aqueous phase. The study of the implementation of the method of extraction and separation, twin-screw extruder and then CLEXTRAL BC 21 BC 45, produces continuous stable emulsion 0.3 kg per kg of treated seed (20 kg / h) with a ratio water / seed of 2 and a temperature of 60 ° C. The cake contains only 0.6 g / kg of azadirachtin, and stable emulsion which contains 5 g / kg can be directly implemented in the formulations of insecticide biosourced completely.

Due to the social concern about environmental sustainability, new compounds have been used in an attempt to reduce the environmental impact of insecticides mainly regarding the risks to mammals, environmental persistence, pollution and development of insecticide resistance. Within this scenario new compounds and particularly bioinsecticides have been increasingly used. Several studies have been carried out to measure the efficacy of pest control agents and its effects on non-target organisms. However, beyond the investigation of the lethal effects of these compounds, the study of sublethal effects on natural enemies is also necessary and circumscribed to few species. Pirate bug predators (Heteroptera: Anthocoridae) play an important role as biological control of many insect pests, some which of great economic importance like the tomato leafminer Tuta absoluta. Blaptostethus pallescens Poppius (Anthocoridae) is a little-known species, but frequently found in tomato fields exposed to insecticides used for pest control, but little is known about those effects in this predator. This study assessed the lethal effects of three insecticides - chlorpyrifos, deltamethrin and azadirachtin - to B. pallescens using their field label rates. Azadirachtin was subsequently selected for assessment of its sublethal effects on behavior and reproduction due to tis lower acute mortality (< 25%) to predator adults after 48 hours of exposure. The insecticides chlorpyrifos and deltamethrin caused 100 % mortality within 4 hours, whereas the survival of insects exposed to azadirachtin was similar to the untreated control. Thus, further studies were conducted with azadirachtin to investigate its sublethal effects. Aiming to evaluate the effects of this insecticide in behavior, adult predators were placed in arenas treated with and without azadirachtin residues for the measurement of parameters such as number of stops, resting time, distance walked and walking velocity. There were no significant differences between treatments. Irritability and repellence to azadirachtin were assessed by the period and the proportion of individuals respectively remaining in the area containing insecticide residue on partially treated arenas. The results indicated that B. pallecens do not exhibit irritability nor repellence to azadirachtin. Subsequently predator couples were subjected to four treatments with azadirachtin untreated couple, treated female (and untreated male), treated male (and untreated female), and treated couple - where female longevity data, daily fecundity, fertility and F1 sex ratio were recorded. These data were used in a projection matrix that generated data of net reproductive rate, population growth rate and generation time for each treatment. The results indicated that there was difference between treatments. Thus, azadirachtin seems to be safer for the predator B. pallescens and therefore a good candidate for the use in pest management programs against T. absoluta.

Merr.] is an important crop in the world with high economic and social value. The carterpillar Anticarsia gemmatalis (Hübner) (Lepidoptera: Noctuidae) is the most important defoliator of soybean in North and South Americas and the search for useful insecticides to control this pest has been studied. The neem, Azadirachta indica A. Juss (Meliaceae) is the plant with high insecticide potential of the world. The azadirachtin, synthesized mainly in the fruits of this plant is the main component responsible for the lethal and sub-lethal action of the extracts obtained from neem. The objective of this study was evaluate the potential of products derived from A. indica on A. gemmatalis in the laboratory and greenhouse conditions. Third stage larvae of A. gemmatalis were fed for four days with artificial diet containing neem seed ethanol extract and oil trade, AzamaxTM. Mortality, food consumption, feces production, weight gain, weight and deformation of pupae at 24 hours old and the number of eggs per female emerged from larvae fed on sub-lethal doses of azadirachtin were evaluated in laboratory. The effects of neem seed extract and the oil trade on mortality and food consumption of larvae of A. gemmatalis in soybean plants in the greenhouse were also evaluated. The neem seed extract and trades Azamax, caused deleterious effects on the A. gemmatalis larvae, as larval and pupal mortality, reduction of food intake and weight gain, disruption of midgut cells and changes in the fat body. Lower dosages of neem seed extract reduced the egg productions of the A. gemmatalis moths. Soybean plants sprayed with both products from A. indica caused high mortality and reduced the damages by A. gemmatalis in greenhouse conditions. The neem products can be used in integrated program management of A. gemmatalis in the soybean crop. / A soja [Glicine max (L.) Merr.]
