Thin Film Deposition on Powder Substrates using ALD and its Characterization using XPS, TEM, and SE

The major part of my dissertation consists of thin films deposited using atomic layer deposition on flat and powder substrates. It details the various optimization experiments for process parameters like dose time, purge time, temperature, and pressure on silicon shards and powder substrates. Spectroscopic ellipsometry (SE) was used to characterize these films over a wide wavelength range (191-1688 nm). An optical model with a BEMA (Bruggeman effective medium approximation) layer was used to fit the ellipsometric data to investigate the optical properties of the alumina surface. The optimized process parameters on the flat surfaces were used for coating powder substrates. I propose a set of experiments to optimize the conditions for coating of powders and high aspect ratio structures by atomic layer deposition (ALD). The coated powders were analyzed by surface analytical techniques like X-ray photoelectron spectroscopy, spectroscopic ellipsometry, transmission electron microscopy, energy X-ray dispersive spectroscopy (EDAX), and BET. The first chapter introduces the technique of atomic layer deposition, and details its advantages and limitations over conventional thin film deposition techniques like chemical vapor deposition and physical vapor deposition. The second chapter details the initial deposition experiments performed on flat surfaces and characterization of thin films using surface analytical tools. I conducted multi-sample analysis on eleven different thin films for calculation of optical constants of alumina. The third chapter introduces thin film deposition experiments performed on powder substrates, several challenges associated with achieving conformal thin films and characterization. The fourth chapter details the experiments to achieve unilateral ALD achieved on one side of the substrates. The fifth chapter details various unconventional materials including liquid water, Coca-Cola, a coffee bean, nitrogen gas, human tooth, and printed office paper, which were analyzed by near ambient pressure XPS (NAP-XPS). This dissertation contains appendices of other tutorial articles I wrote on obtaining optical constants liquid samples using spectroscopic ellipsometry, and good experimental techniques for maintenance of vacuum equipment.

T-bet-Mediated Tim-3 Expression Dampens Monocyte Function During Chronic Hepatitis C Virus Infection

Hepatitis C virus (HCV) induces a high rate of chronic infection via dysregulation of host immunity. We have previously shown that T-cell immunoglobulin and mucin domain protein-3 (Tim-3) is up-regulated on monocyte/macrophages (M/Mφ) during chronic HCV infection; little is known, however, about the transcription factor that controls its expression in these cells. In this study, we investigated the role of transcription factor, T-box expressed in T cells (T-bet), in Tim-3 expression in M/Mφ in the setting of HCV infection. We demonstrate that T-bet is constitutively expressed in resting CD14+ M/Mφ in the peripheral blood. M/Mφ from chronically HCV-infected individuals exhibit a significant increase in T-bet expression that positively correlates with an increased level of Tim-3 expression. Up-regulation of T-bet is also observed in CD14+ M/Mφ incubated with HCV+ Huh7.5 cells, as well as in primary M/Mφ or monocytic THP-1 cells exposed to HCV core protein in vitro, which is reversible by blocking HCV core/gC1qR interactions. Moreover, the HCV core-induced up-regulation of T-bet and Tim-3 expression in M/Mφ can be abrogated by incubating the cells with SP600125 – an inhibitor for the c-Jun N-terminal kinase (JNK) signalling pathway. Importantly, silencing T-bet gene expression decreases Tim-3 expression and enhances interleukin-12 secretion as well as signal transducer and activator of transcription 1 phosphorylation. These data suggest that T-bet, induced by the HCV core/gC1qR interaction, enhances Tim-3 expression via the JNK pathway, leading to dampened M/Mφ function during HCV infection. These findings reveal a novel mechanism for Tim-3 regulation via T-bet during HCV infection, providing new targets to combat this global epidemic viral disease.

Were they or weren't they? A study of possible sedentariness using faunal indicators at a coastal Thom's Creek site (38ch1693)

Site 38CH1693 is a coastal site located in Charleston County, South Carolina. Thom’s Creek ceramics place the site in the Late Archaic/Early Woodland Period and radiocarbon dating corroborates this, placing the features present between 3650 and 3950 BP. Faunal analysis was undertaken to assess seasonality in order to understand the occupations that occurred at the site. Faunal seasonality, botanical seasonality, and sedentariness indicators are used to determine the sedentariness of the site. The presence of certain marine fish species is one of the most important seasonality indicators used. It is determined from the available evidence that at least one short-duration, year-round occupation is represented by the materials recovered at 38CH1693. Site 38CH1693 does not fit into the current Late Archaic settlement pattern models for the coast, calling for a reevaluation of these settlement pattern models and the sedentary sites within them.

Biodiesel ses som ett av de främsta substituten för fossila bränslen, då den relativt enkelt kan appliceras i redan existerande dieselmotorer. Dagens produktion av biodiesel använder sig av homogena katalysatorer som inte återanvänds i processen, men för en mer cirkulär och i längden en billigare process ses heterogena katalysatorer som ett alternativ. Syftet med denna studie var att undersöka litium dopad kalciumoxid katalytiska egenskaper, de optimala förhållandena för reaktionen och även kinetiken för reaktionen. Den valda katalysatorn syntetiserades med kalcinering och sedan testades den katalytiska förmågan i reaktionen vid olika reaktionsförhållanden, för att finna de optimala förhållandena. Även katalysatorns fysiska egenskaper analyserades och kinetiken för reaktionen. De optimala förhållanden för transesterifieringen bestämdes till 3 h, 1:6 olja- metanolförhållande, 60°C och 5 vikts% katalysator, där 96% utbyte av biodiesel uppnåddes. Övriga utbyten från de andra försöken var betydligt lägre, vilket kan bero på icke optimala förhållanden, men även relativ dålig katalytisk förmåga. Ytarea och porstorlek konstateras vara små, vilket påverkar den katalytiska förmågan negativt. Anledningen till katalysatorns egenskaper beror huvudsakligen på tillverkningsprocessen, men även mängden litium i dopningen. Kinetiken visar en oväntad reaktionsutveckling med initialt hög koncentration FAME (Fatty acid Methylester), vilket troligen beror på felkällor.
