Phytoremediation is an innovative technology employing plants for reclamation of soils that have been polluted by industrial contaminants. While it is frequently slower than traditional physicochemical remediation techniques, there are, however, many significant advantages including simplicity, low cost, improvement of soil structure and microbiological fertility. While various hyperaccumulators have been reported from Europe and the USA, there is a need to identify Australian species. This study reports a number of metal accumulators following a soil and vegetation survey of two metal contaminated sites in New South Wales, the spoil (filtercake) heaps of the BHP (Broken Hill Propriety ) steelworks at Port Kembla and the abandoned mine at Sunny Corner. BHP is the largest manufacturer of steel in Australia. The Port Kembla is the largest single steel manufacturing site. Solid by-products from this site totalled 3.1 million tons in 1988. The Sunny Corner mine site was mainly a silver mine which operated over approximately 100 hectares 1184 to 1922 and produced over 100 tonnes of silver during that period. The soil and water in and around the site is heavily contaminated with metals and is distinctly acidic. It was concluded that Ricinus communis, Sonchus oleraceus, baeckea utilis, Poa labillardieri and Pinus radiata are options for heavy metal phytoremediation of contaminated Australian soils. Arbuscular Mycorrhizal infection (particular with Glomus and Gigaspora strains) should be promoted for optimum growth. / Doctor of Philosophy (PhD)

O objetivo central da tese foi compreender como se configura a aprendizagem nos processos organizativos de desastres. Realizei uma pesquisa qualitativa à luz da teoria ator-rede como inspiração teórico-metodológica, a partir do crime-desastre da Samarco/Vale/BHP Billiton ocorrido em novembro de 2015. Na primeira discussão teórica da tese, busquei aproximar processualmente os atos de organizar e aprender, por meio do conceito de aprendizagem organizativa. Na segunda discussão teórica, analisei as questões contemporâneas sobre a temática dos desastres do Brasil, propondo que os desastres são efeitos disruptivos em uma rede de relações heterogêneas, desiguais, atemporais e multiterritoriais, sendo as vulnerabilidades e os perigos constituídos relacionalmente. A pesquisa foi constituída por uma fase preliminar/exploratória e três momentos de imersão no campo: na etapa preliminar realizei um aprofundamento empírico sobre os desastres, especialmente a partir de experiências brasileiras; os três momentos de imersão foram no caso do desastre das empresas, com enfoque para os desdobramentos do desastre no Estado do Espírito Santo. Ao longo do primeiro ano do desastre-crime da Samarco/Vale/BHP, coletei e analisei 1.297 documentos, entrevistas (formais e informais) e registros em diários de campo. Todos os dados foram inseridos no software de análise qualitativa Atlas.ti. A metodologia e a análise dos dados foram realizadas a partir do mapeamento das principais controvérsias que emergiram das relações entre os atores do estudo, uma das possibilidades da teoria ator-rede enquanto método e que se constitui uma das contribuições desta pesquisa A partir das discussões teóricas e empíricas, foi possível observar que os desastres geram uma latência organizativa, que se constitui num estado abstrato e transitório de disposição para aprender a organizar. Essa latência impulsiona os processos de aprendizagem organizativa, que são os modos de cooperar e se fazer coletivamente, em movimento e instáveis, cognoscíveis pelo aprender e pelos saberes-em-ato (knowing); eles são estabelecidos numa rede de relações heterogêneas em múltiplas formas de espacialidades, podendo se inscrever e formar uma tessitura de práticas, sendo que as condições de possibilidades para a sua realização e participação não são dadas, podendo também servir como um meio para superar, combater as desigualdades, e formar outros fazeres e práticas. I conducted a qualitative research in the light of actor-network theory as a theoretical-methodological inspiration, starting from the Samarco/Vale/BHP Billiton crime-disaster that occurred in November of 2015. In the first theoretical discussion of the thesis, I tried to procedurally approach the acts of organizing and learning, through the concept of organizational learning. In the second theoretical discussion, I analyzed contemporary issues on Brazil's disasters, proposing that disasters are disruptive effects in a network of heterogeneous, unequal, timeless and multi-territorial relations, with vulnerabilities and dangers being constituted relationally. The research consisted of a preliminary/exploratory phase and three moments of immersion in the field: in the preliminary stage I realized an empirical deepening on the disasters, especially from Brazilian experiences; the three moments of immersion were in the case of the disaster of the companies, with focus on the unfolding of the disaster in the State of Espírito Santo. Throughout the first year of the Samarco/Vale/BHP crime-disaster, I collected and analyzed 1,297 documents, interviews (formal and informal) and records in field journals. All data were entered into the qualitative analysis software Atlas.ti. The methodology and data analysis were performed from the mapping of the main controversies that emerged from the relations between the actors of the study, one of the possibilities of actor-network theory as a method and that is one of the contributions of this research From the theoretical and empirical discussions, it was possible to observe that disasters generate an organizational latency, which constitutes an abstract and transitory state of disposition to learn how to organize. Tres años han pasado desde la ruptura de la represa de Fundão en Mariana-MG, la mayor catástrofe de la historia de la minería en el país y la mayor catástrofe mundial en la que están involucradas represas. Hasta ahora, la inmensa mayoría de los daños causados a los antiguos habitantes de los distritos de Bento Rodrigues, Paracatu de Baixo y Gesteira, los cuales perdieron seres queridos, casas, propiedades y pertenencias, no han sido reparados. Según el pronóstico de los órganos públicos implicados, la reconstrucción de las comunidades devastadas por el mar de lodo de Samarco, ni siquiera iniciada, llevará años. Las familias afectadas, ubicadas en casas alquiladas por la empresa y con parca ayuda financiera, siguen sufriendo la prolongación continua del daño.

The effect of sulforaphane on oxidative stress and biotransformation in HepaRG cells / A. Crous.Crous, Ané January 2013 (has links)
Sulforaphane is an isothiocyanate found in high concentrations in cruciferous vegetables like broccoli. Sulforaphane has received much attention due to the evidence that it inhibits phase I carcinogen-bioactivating enzymes and/or induces phase II antioxidant enzymes as well as metallothioneins (MTs) (Perocco et al., 2006; Clarke et al., 2008; Yeh & Yen, 2009). Since MTs and antioxidant enzymes are involved in the scavenging of reactive oxygen species (ROS), the question was raised whether sulforaphane can provide protection against increased oxidative stress and if sulforaphane exposure of a human hepatocellular carcinoma cell line, like HepaRG cells, will have a negative impact on phase I and II biotransformation in these cells. Oxidative stress was exogenously induced in HepaRG cells with tert- Butyl hydroperoxide (t-BHP). Phase I and phase II biotransformation pathways were assessed with caffeine, paracetamol, aspirin, sodium benzoate, and paraaminobenzoic acid, respectively, as probe substances. Through the use of a liquid chromatography-electrospray ionization-mass spectrometry (LC-ESI-MS/MS) assay, the biotransformation of caffeine in phase I and the formation of paracetamol, aspirin, sodium benzoate and para-aminobenzoic acid conjugates in phase II were investigated. This involved elucidating the time it took for the whole probe to be completely biotransformed during phase I biotransformation and the unique conjugates formed during phase II biotransformation in HepaRG cells.
The optimal t-BHP concentration and exposure time in HepaRG cells were standardized with a 3-(4, 5-dimethylthiazol-2-yl)-2, 5-diphenyltetrazolium bromide (MTT) assay. LC-ESI-MS/MS assays to monitor phase I and phase II biotransformation were optimized and validated. The optimal sulforaphane concentration and exposure time in HepaRG cells were standardized with a MTT assay. To evaluate the possible protective effect of sulforaphane against oxidative stress, HepaRG cells were pre-incubated with sulforaphane followed by the induction of oxidative stress with t-BHP and the quantification of the amount of viable cells with a MTT assay. To investigate the effect of sulforaphane on phase I and phase II biotransformation pathways, HepaRG cells were first pre-incubated with sulforaphane followed by the addition of a specific probe substance and the assessment of the biotransformation of the probe with a LC-ESI-MS/MS assay.
The results partially supported the hypothesis of the study that sulforaphane will protect HepaRG cells against oxidative stress without negatively influencing phase I and phase II biotransformation. Le travail présenté vise une meilleure compréhension des phénomènes mis en jeu lors de l'exposition à haute température des bétons à haute performance. Nous nous sommes plus particulièrement intéressés au comportement mécanique. La première partie de ce document présente une synthèse des travaux réalisés sur l'évolution des propriétés physiques et mécaniques des bétons soumis à haute température. Les résultats présents dans la littérature sont variés, parfois contradictoires et confus notamment ceux concernant l'évolution des propriétés mécaniques au cours de l'échauffement. Les évolutions de la résistance en compression, résistance en traction et module d'élasticité sont présentés en fonction des différents paramètres influençant ces comportements. L'influence des conditions de réalisation des essais est notamment discutée. En outre, le comportement au feu est abordé sous l'angle du phénomène de l'écaillage. Nous présentons ainsi le rôle des fibres polypropylène, un moyen efficace de réduire les risques d'apparition du comportement explosif. La deuxième partie, consacrée à l'étude expérimentale. Dans cette partie nous exposons les matériaux testés, leurs compositions et les constituants utilisés, ainsi que la fabrication de ces matériaux et leur conservation. Ensuite nous présentons le banc d'essais développé, permettant de tester le comportement mécanique à chaud des bétons. Ce nouveau dispositif expérimental spécifique et relativement complexe, a été spécialement mis au point afin de réaliser l'étude de la déformation thermique libre, de la déformation thermique sous charge mécanique, et du comportement en compression et en traction à chaud. Les résultats expérimentaux obtenus, sont ensuite présentés et discutés. La majeure partie de cette étude a été consacrée à l'établissement des relations entre le comportement mécanique et la température des différents types de béton. La résistance en compression et le module d'élasticité varient non seulement avec la température d'exposition du matériau, mais aussi avec le scénario d'échauffement (vitesse de montée en température, essais à chaud/après refroidissement, etc.). Dans les observations que nous avons réalisé, l'influence du rapport E/C des bétons testés a été étudiée ainsi que l'influence de l'ajout des fibres polypropylène sur les propriétés mécanique des BHP testés à chaud. Les observations de l'évolution des propriétés mécaniques réalisées nous ont permis de constater que les résultats du comportement à haute température sont fortement influencés par la présence de l'eau dans le matériau, surtout dans la gamme de températures jusqu'à 300°C. En complément de l'étude du comportement en compression, nous avons étudié la possibilité de réalisation des essais de traction directe sur le matériau béton "à chaud". Diverses solutions ont été envisagées et testées, afin de retenir la technique plus adaptée à nos besoins. Les premiers résultats sont très prometteurs. Il faut souligner que le nombre d'études expérimentales du comportement en traction directe "à chaud" est extrêmement faible. Une grande partie de la recherche a été consacrée à l'étude de la déformation thermique transitoire (DTT). Ce comportement, encore mal connu et souvent controversé, appelé "fluage thermique transitoire" a été étudié sur trois bétons à haute performance (BHP) et un béton ordinaire (BO). Parmi les paramètres influençant la DTT nous pouvons citer: le taux de chargement, l'histoire du chargement thermo mécanique, la vitesse de montée an température ou la teneur en eau du béton. De plus, nous avons étudié de l'existence du phénomène de la DTT sous charge mécanique en traction.
