Electron microscope observations and characterization of a reptilian neuromuscular junction

This thesis has investigated skeletal muscle fibers and neuromuscular junctions of the reptile Anolis carolinensis. The extensor digitorum communis and gastrocnemmius externus were used for observations and characterization. Cross sections were stained for oxidative and glycolytic activities and for contractile properties by the use of four histochemical stains. This revealed the extensor digitorum communis to be composed of predominately fast glycolytic fibers, with a central core of fast oxidative-glycolytic and slow oxidative fibers. The gastrocnemius externus exhibited a majority of fast oxidative-glycolytic fibers and random slow oxidative fibers.Ultra structural examination of the extensor digitorum communis revealed the nerve terminal of the neuromuscular junction to be smaller than that of the gastrocnemius externus, with junctional folds longer and straighter, and synaptic vesicles smaller and more closely packed. In both muscles, the nerve fibers terminated in a shallow groove of the muscle fiber. The characteristics and heterogeneity that exist are very similar to the homologous mammalian extensor digitorum longus and soleus skeletal muscle.Ball State UniversityMuncie, IN 47306

A comparative study of the effect of partial hepatectomy on the molecular form distribution of adenosine deaminase in various rat tissues

The molecular form distribution of adenosine deaminase has been characterized in nine rat tissues. This study has also investigated the effects of 70% partial hepatectomy in liver, spleen, and intestinal tissues. In all tissues studied, the molecular weight of the enzyme was found to be 35,000 daltons, as determined by gel filtration chromatography. Two prominent isozymes of this molecular weight were identified by means of thin-layer isoelectric focusing. Isozymes with pI's of 4.95 and 4.80 were present in tissues of liver, spleen, and intestine of normal and hepatectomised rats. An instrument measuring Perceived Vulnerability was administered to grades three through twelve across a rural school district. The background literature base for the concept and validity of the Perceived Vulnerability measurement is reviewed. A series of means is calculated for the above data and subjected to a simple analysis of variance. Resulting data support a relative unchanging level of Perceived Vulnerability with a possible rise in the fifth grade level only. It is concluded that an adequate control value for this rural school district is established which can be used to measure impact of teaching programs. Hemocytes of the fifth instar European borer, Ostrinia nubilalis Hubner (Lepidoptera: Pyralidae) were examined in stained and unstained preparations. Prohemocytes, plasmatocytes, granular hemocyter, and oenocytoids, and spherule cells were found in this stage, The prohemocytes are characterized by a scant intensely basophilic cytoplasm. The highly pleomorphic plasiratocytes are characterized by a punctate nucleus and production of cytoplasmic extensions in vitro. Granular hemocytes are characterized by a small nucleus, accumulation of lipid droplets, and the production of extremely fine cytoplasmic extensions in living preparations. The oenocytoid is characterized by a small eccentric nucleus in a large amount of homogeneous basophilic cytoplasm. The spherule cell is characterized by large spherules within the cytoplasm which may tend to mask the nucleus.Treatment of larvae with per os doses of Bacillus thuringiensis did not affect the differential hemocyte count. The subject of this thesis is the design of a FORTRAN compiler for the PDP-11/34 minicomputer owned by the Dept. of Mathematical Sciences of Ball State University, Muncie, Indiana. At the time that this project was undertaken, only the BASIC and ASSEMBLY languages were available on this machine.The problem consisted of writing a two-pass processer. The function of the first pass was to recognize FORTRAN statements, analyze them syntactically and semantically, build symbol tables, and construct an output file containing pointers to the symbol table, flags, and operators. Changes in muscle lipid metabolism with endurance training in man

Eight men were studied before and after an endurance training program to assess changes in the lipid metabolism of skeletal muscle. Training consisted of eight weeks of outdoor running with daily mileage increasing from one-half mile at the onset to four miles at the eighth veekof training. In vitro analyses of palmityl-CoA oxidation and the activities of carnitine palmityltransferase (CPT) and succinate dehydrogenase (SDH) were observed in muscle samples obtained by needle biopsy from the gastrocnemius before and after training.The subjects' maximal oxygen uptake increased from 3.47 1/min (± 0.26) to 3.82 1/min (± 0.21) as a result of the training. At the same time, muscle CPT and SDH increased (P <.01) 23% and 36%, respectively. The muscle's capacity to oxidize palmityl-CoA increased from 6.36 nmoles/min/g to 13.32 nmoles/min/g, a significant increase of 116% (P <.01).This investigation supports earlier findings with rats and emphasizes the influence of chronic endurance exercise on lipid metabolism in the skeletal muscle of man.Ball State UniversityMuncie, IN 47306

A comparison of selected enzyme activities in normal and tumorous mouse mammary tissue

The activities of phosphohexose isomerase (EC. 5.3.1.9), isocitrate dehydrogenase (EC. 1.1.1.42) and lactate dehydrogenase (EC. 1.1.1.27) were measured in 14 normal and 21 tumorous mouse mammary tissue samples. Methods of tissue extraction and activity determination in this study employed equipment found in most clinical laboratories.For each of the three enzymes there was a statistically significant (p <.05) elevation of the tumor sample group's activities. The activities of the normal tissues were: PHI x = 9.6 and SD = 4.6, ICD x = 13.2 and SD = 5.3, and IDH x = 10.9 and SD = 5.3. The activities of the tumorous tissues were: PHI x = 55.2 and SD = 29.9, ICD x = 40.5 and SD = 23.8, and IDH x = 55.8 and SD = 31.4.Using values of 20, 27, and 21 for the upper limit of normal activity (x + 2SD) for PHI, ICD, and LDH respectively, the tissue samples can be divided into normal and tumorous groups with l00% sensitivity and 85% specificity.Ball State UniversityMuncie, IN 47306

Model simulation of contaminant movement from a sanitary landfill

The potential for leachate movement out of a sanitary landfill was evaluated and simulated by a computer program model. Landfill operations and geohydrological conditions in the area were investigated in order to develop a conceptual model of the system and to obtain numerical data for the development of the simulation.The landfill is operated in a 20 to 27 foot thick clay till which overlies a 10 foot thick, continuous sand and gravel aquifer. Monitor wells located around the site indicate minimal changes in groundwater quality to this date. This is attributed to the slow movement of leachate through the confining layer which would take at least 5 to 10 years.Numerical parameters not readily determined were varied in 16 simulations. This procedure allowed analysis of the different plumes developed as well as observation of the program’s sensitivity to such changes. In simulations of five years from the time leachate would enter the aquifer, leachate plumes were developed which extended a mile and one half down the hydrologic gradient from the landfill. Physical and mineralogical variations in the Yorkville Till member, Grundy and adjacent counties, Illinois

This study has confirmed the existence of a distinct mineralogical component in the upper part of the Yorkville Till Member of the Wedron Formation of Wisconsinan age in northeastern Illinois. This upper till unit occurs within the area of, but is not bounded by, the older Marseilles Morainic System and the younger, overriding Minooka Moraine. The analyses of samples from 79 locations that were used to identify, compare, and differentiate the two tills were: matrix grain size, clay mineral composition, carbonate content, and color.The upper till is a grayish brown, silty clay till with 76% illite and a dolomite-calcite ratio of 2:1. The lower till is an olive gray, silty clay till with 81% illite and a dolomite-calcite ratio of4:1. Results of this study indicate that (1) the two tills are separable and mappable units with the aid of laboratory data; (2) the lower till composes the bulk of the Marseilles Morainic System; (3) the upper till is thin between the moraines and constitutes the till of the Minooka Moraine; and (4) the upper till apparently represents a minor readvance and a concurrent change in source material.Ball State UniversityMuncie, IN 47306
