Skogsfinnarna i arkiven : En undersökning om spåren av skogsfinnarna i de digitaliserade arkiven

Since the year of 1580 until about the year of 1680 Forest Finns have immigrated to a large part of middle Sweden working mainly with burn-beating but also with cattle raising, hunting and fishing. There have been studies made for quite some time about the Forest Finns family names, last names and location names and also about their food culture, craftsmanship and building constructions. Still there hasn´t seen to be any research about how you can use the digitized archives for further research about Forest Finns and this is where I’ve found an interesting research gap to explore further. This essay has been delimited to the areas of Dalby, Norra Ny and Gräsmark based on Carl Axel Gottlund´s notes on the forest Finnish family names that he recorded in the household records in 1821 within the given area. After this study, it has become clear that the vast majority of sites with forest Finnish settlements contain both Finnish family names and Finnish place names. The vast majority of the sites also had croft remains and stone wall cairns. It has also become clear during this study that the remains of farm buildings can be found at several of these sites. Remains of mills have also been found at several sites while only a few of the sites investigated had a sauna or ria. Barns are also less common on the sites, as are carvings and natural formations. To summarise, the house interrogation lists in the church books that Carl Axel Gottlund has recorded can provide information about the Forest Finnish family names the families have had, and which Forest Finnish place names exist in the area. But the information can also be used to calculate how much of the population in these areas had Forest Finnish roots and how they chose to settle in the areas. the traces of the Forest Finns in the material remains or other cultural heritage remains can give us more information about the building skills of the Forest Finns but also more information about their cultural heritage and living conditions.

Effects of Isoproterenol on IhERG during K+ changes in HEK293 cells

Yes / Introduction:The human ether-a-go-go related gene (hERG) encodes the pore forming protein which mediates the rapid delayed rectifier K+ current in the heart (IKr). Together with other ion channels hERG determines the cardiac action potential and regulates the heart beating. Dysfuction of the hERG ion channel will lead to acquired long QT syndrome (LQTS). Therefore, new drug candidates must pass the test for a potential inhibitory effect on the hERG current as a first step in a nonclinical testing strategy. Arrhythmias in patients with LQTS are typically triggered during physical or emotional stress, suggesting a link between sympathetic stimulation and arrhythmias. It is well known that potassium level can affect the QT interval through affecting IhERG both in vivo and in vitro.In this study, we try to find out whether the trigger effect still exist when K+ changes violently in a short time period. In other words, whether the risk of TdP aggravate when patients suffer from acute water electrolyte balance disorder, which is a common symptom in hot weather. Methods: HEK293 Cell line stably expressing hERG channel were cultured in DMEM supplemented with 10% of fetal bovine serum.Whole-cell patch-clamp method was applied for ionic current recordings. The compositions of pipette was (in mM) 125 KCl, 5 MgCl2, 5 EGTA-K, 10 HEPES-K and 5 Na-ATP adjusted to pH 7.2 with KOH. The bath solutions for recording the IhERG currents was 136 NaCl, 4 KCl, 1 MgCl2, 10 HEPES-Na, 1.8 CaCl2 and 10 glucose, pH 7.4 with NaOH. The low extracellular K+ solution was 115 KCl, 5 MgCl2, 5 EGTA-K, 10 HEPES-K and 10 Na-ATP adjusted to pH 7.2 with NaOH. Patch-clamp experiments were performed at room temperature (22 ± 1°C). The recording of low K+ current was carried out immediately after the original normal K+ solution has been totally replaced. Isoproterenol (ISO) 100nM was added into both kinds of K+ solution to apply the effect of β1-AR stimulation. Results: We found that low K+ solution increased IhERG from 907.39±18.68to 1620.08±249.44pA(n=30,P<0.05); Low K+also shifted the I-V curve to the left. IC50 in control is 10.31±5.52 mV, low K+ is -6.15±1.58 mV. When adding ISO 100nM to extracellular solution, same effects were shown for both groups.ISO decreased Imax for both group. In control group, Imax reduced from 907.39±18.68to493.16±54.41pA (n=30, P<0.01), while in low K+ group, I max decreased Imax from 1620.08±29.44to 488.48±81.87pA(n=30,P<0.05). At the same time, ISO shifts the I-V curve to the right for the control group and shift the curve to the left for low K+ group. IC50 in control when added ISO is 22.25±3.80 mV, while IC50 in low K+ group after adding 100nM ISO is -31.00±5.73 mV. Conclusion: The results from this study is contradict to those in our previous study where low K+ combined with ISO can lead to temporarily increase of QT interval in vivo.It is reported that an increase in net outward repolarizing current, due to a relatively large increase of IKs, is responsible for the changes of QT interval in response to beta-adrenergic stimulation in vivo(2). Venöse Revaskularisation bei Vorliegen einer Mikro- und Makroangiopathie

In der vorliegenden Arbeit wurde die venöse Revaskularisation bei Vorliegen einer Mikro- und Makroangiopathie untersucht. Die Zielgruppe dieser Methodik sind Patienten, die aus kardiologischer und herzchirurgischer Sicht austherapiert sind, für die weder ein Stent noch eine konventionelle Bypassoperation in Frage kommt. Das betrifft Patienten mit disseziierten Gefäßen, mit schwerer diffuser koronarer Herzerkrankung oder small vessel disease. Für diese Patienten ist derzeit keine optimale Therapie vorhanden. Es handelt sich somit um eine ultima ratio-Therapie. Zur Prüfung der Effektivität der Methode bei beschriebenem Krankheitsbild wurde in Minipigs eine Mikroangiopathie durch Injektion von Mikrosphären (Durchmesser 100µm) in den linken Hauptstamm erzeugt. Nach 7 Wochen Krankheitsentwicklung der Mirkoangiopahtie wurde eine hochgradige Stenose des Ramus interventricularis anterior (RIVA) hervorgerufen, welches die Makroangiopathie simulieren sollte. Anschließend wurden die Tiere der Therapiegruppe mit einem Bypass von der Arteria mammaria auf die Begleitvene des RIVA versorgt. Dabei wurde die Begleitvene proximal der Anastomose ligiert. Die Kontrollgruppe blieb ohne Therapie. Die Stenose und der Bypass wurden angiographisch dargestellt. Nach 17 Wochen wurde bei allen Tieren eine Herzkatheteruntersuchung durchgeführt, um die Stenose und den Bypass zu beurteilen. Als Maß für die Herzleistung wurde zu allen 3 Versuchsteilen die Ejektionsfraktion bestimmt. Anhand der Ejektionsfraktion konnte die Überlegenheit der venösen Revaskularisation als Therapie gezeigt werden. 7 Wochen nach Injektion der Mikrosphären fielen die Werte der Ejektionsfraktion beider Gruppen ab. Nach 17 Wochen nahm die Ejektionfraktion der Therapiegruppe, die in der Zwischenzeit mit einem Bypass versorgt wurden, wieder deutlich zu und die der Kontrollgrupppe sank weiter ab. Zusätzlich wurden die entnommenen Herzen histologisch untersucht. Dabei zeigte sich ein Umbau des Gefäßsystems im Bereich der angeschlossenen Vene.:1. Einleitung 7 1.1. Danksagung 92 / Objective: Many patients with significant arterioclerosis of the heart cannot benefit from a coronary artery bypass and other methods because they have macroangiopathy combined with microangiopathy. We evaluated the efficiency of venous revascularization in minipigs with macroangiopathy combined with microangiopathy in a chronic model of 3 months. Histological analysis of arterial and venous vessels of the heart was conducted. Methods: In left anterior descending artery (LAD) microspheres (diameter 100µm) were injected in 24 minipigs (12 control group, 12 therapy group). 7 weeks later a stenosis of the LAD was performed in both groups with an average of 84,6 ± 4,3%. In therapy group left internal thoracic artery was anastomosed to the concomitant vein of the LAD in beating heart surgery. The flow of bypass was proved in angiography and the flow rate was measured by ultrasound. 10 weeks later bypass of therapy group and stenosis of both groups were verified in angiography. At the beginning of every part of the experiment the ejection fraction in both groups was evaluated with echocardiography. Hemodynamic monitoring was performed throughout the experiment. In histological analysis arterial and venous vessels in left atrium, right atrium, septum and area of anastomosis were evaluated in the thickness of wall and area of lumen. Results: In ejection fraction a significant difference between control and therapy group was seen. After performing the bypass, the ejection fraction in therapy group increased, while it decreased in control group in the same period of time of 10 weeks. In the histological analysis a non-classifiable type of vessel was found only in the area of the anastomosis. The vessel had a a large area of lumen and a thick wall due to media hyperplasia. Conclusion: Venous revascularization of the concomitant vein of the LAD via bypass of the left thoracic artery in a chronic model improves cardiac funcion and is therefore an effective method when having microangiopathy and macroangiopathy combined. The non-classifiable type of vessel are most likely arterialized veins which underwent a structural change of the wall due to the arterial blood pressure.:1. Einleitung 7 1.1. Quasi-phase-matching of high-harmonic generation

This thesis describes the use of counterpropagating pulse trains to quasi-phase-match high-harmonic generation (HHG). Two novel techniques for generating trains of ultrafast pulses are described and demonstrated. The first method makes use of a birefringent crystal array to split a single pulse into a sequence of pulses. The second method makes use of the time-varying polarisation of a chirped pulse passed through a multiple-order wave plate to generate a train of pulses by the addition of a polariser. It is demonstrated that this second technique can be used to make pulse trains with non-uniform pulse separation by using an acousto-optic programmable dispersive filter to manipulate the higher-order dispersion encountered by the chirped pulse. The crystal array method is used to demonstrate quasi-phase-matching of HHG in a gas-filled capillary, using one and two counterpropagating pulses. Enhancements of up to 60% of the intensity of the 27th harmonic of the 800,nm driving laser light are observed. Information on the spatial and dynamic properties of the HHG process is obtained from measurements of the coherence length in the capillary. Simulations of HHG in a capillary waveguide have been performed. These agree well with the results of the quasi-phase-matching experiments. The effect of mode-beating on the generation process in a capillary and its use as a quasi-phase-matching mechanism are investigated.

Composição e estrutura da fauna araneológica (Arachnida, Araneae) associada ao estrato arbóreo-arbustivo de matas ciliares e seus microambientes no Rio Grande do Sul, Brasil / Composition and structure of the spider fauna of the tree-shrub strata of riparian forests and its microhabitats in Rio Grande do Sul, Brasil

A intenção deste estudo foi conhecer a composição e a diversidade da araneofauna relacionada a matas ciliares no Rio Grande do Sul, em diferentes localidades: rio Piratini no município de Arroio Grande; rio Camaquã em Cristal; rio dos Sinos em Parobé e rio Maquiné em Maquiné. Foi comparada a riqueza de espécies, abundância e similaridade da fauna de aranhas e a sazonalidade, avaliando se fatores ambientais bióticos e abióticos influenciam na assembleia de aranhas das matas ciliares. Também foram avaliadas as guildas de aranhas, tanto aspectos espaciais, quanto temporais, e a influência de fatores abióticos e de complexidade ambiental sobre as mesmas. As aranhas foram separadas e classificadas em quatro guildas: construtoras de teias orbiculares, construtores de teias irregulares, caçadoras cursoriais e caçadoras emboscadoras/espreitadoras. Para a amostragem da araneofauna foram efetuadas duas coletas por estação durante dois anos em cada mata ciliar. As coletas foram realizadas em transecções, por um período de 45 minutos, utilizando como método o guarda-chuva entomológico (GCE) para amostrar a araneofauna do estrato arbóreo-arbustivo. Em cada mata ciliar foram delineadas seis transecções, duas em cada microambiente: borda com o campo, interior da mata e borda com o rio. O esforço amostral foi de 288 horas de coletas. No total foram coletadas 42057 aranhas: 8851 adultas (21%) e 33206 jovens (79%). A família predominante foi Theridiidae, com mais de 50% das aranhas amostradas em cada mata, a única exceção foi a mata ciliar do rio Maquiné que dominou Linyphiidae. A abundância de aranhas adultas foi diferente entre as matas ciliares, com maior abundância no rio Maquiné (34,58% das aranhas) e menor no rio dos Sinos (18,43%). Dos microambientes, as maiores abundâncias foram no interior da mata, seguido pelas bordas, muito próximas entre si. Foram determinadas 440 espécies/morfoespécies de aranhas, entre 168 gêneros. No total 232 espécies foram nominais (53%) com 14 novos registros para o Rio Grande do Sul e três para o Brasil. No rio Maquiné foi registrada a maior riqueza de espécies (S=218) e menor no rio dos Sinos (S=179). Entre os microambientes, as maiores riquezas de espécies de aranhas foram observadas nas bordas (campo: S=298 e rio: S=279) e após o interior da mata (S=246). A riqueza de espécies foi diferente entre os rios, já as riquezas entre os microambientes não diferiram. As espécies raras foram representadas por 130 singletons (32,5% das espécies do inventário). Para a sazonalidade, as aranhas adultas foram mais abundantes no verão e menos no outono; as aranhas jovens predominaram no outono e menos na primavera. A riqueza não diferiu entre os dois anos de amostragem. Já entre as estações a diferença foi significativa, maior na primavera e menor no outono. Não foi encontrada uma relação entre os fatores abióticos (temperatura e precipitação) com a araneofauna, tanto para abundância, quanto para a riqueza. As variáveis bióticas da vegetação não foram suficientes para explicar as variações na comunidade de aranhas, não influenciando nas mesmas. A análise multivariada de similaridade (ANOSIM), pelo índice quantitativo de Morisita e qualitativo de Simpson, mostra que as composições de espécies das araneofaunas diferem entre as matas ciliares. Para os diferentes microambientes não foram encontradas diferenças na composição. Prevaleceu a guilda das construtoras de teias irregulares e uma menor quantidade das caçadoras cursoriais. A MANOVA indicou diferença significativa na proporção das guildas entre todas as matas ciliares e entre os microambientes. A partir da análise de correspondência canônica (CCA), as guildas não estão correlacionadas com as variáveis de complexidade ambiental avaliadas. As curvas de rarefação e os estimadores analíticos de riqueza de espécies indicam que a fauna de aranhas ainda não foi suficientemente amostrada para as matas ciliares, revelando a urgência de estudos futuros, principalmente, abrangendo outros métodos, para diferentes estratos das florestas. / The aim of this study was to characterise the spider fauna associated to riparian forests in Rio Grande do Sul. Four different sites were sampled: Piratini River in Arroio Grande; Camaquã River in Cristal; Sinos River in Parobé and Maquiné River in Maquiné. Diversity components such as species richness, abundance and faunal similarity were compared among rivers, among microhabitats (forest edges and forest interior) and yearly seasons, and also against biotic and abiotic environmental factors. Spider foraging guilds were also evaluated, again for spatial and temporal scales, and again against biotic and abiotic environmental factors. Spiders were separated in four guilds: orbicular web builders, sheet web builders, running hunters and ambush hunters. Two samples per season were taken during two years for all forest sites. Sampling consisted of using beating tray on the tree-shrub strata across linear transections (50 X 2 m) for 45 min. On each site six transects were used, two per microhabitat: grassland edge, forest interior and river edge. Sampling effort totalled 288 h of sampling. Overall we obtained 42,057 spiders: 8,851 adults (21%) and 33,206 juveniles (79%). The predominant family was Theridiidae, with more than 50% of all spiders for all sites except Maquiné River, in which Linyphiidae dominated. Adult spider abundance differed among sites, with more spiders in Maquiné River (34.58% of the spiders) and less in Sinos River (18.43%). Among microhabitats, largest abundances occurred in the forest interior, with the edges having less spiders. A total of 440 spider species/morphospecies were recognised within 168 genera. Only 232 of those species were nominal (53%) with 14 new records for Rio Grande do Sul state and three for Brazil. In Maquiné River we found the largest richness (S=218) and in dos Sinos River the lowest (S=179). Comparing microhabitats, the largest species richness were observed for the edges (grassland edge: S=298; river edge: S=279) with the forest interior below (S=246), but not significantly. Rare species were represented by 130 singletons (32.5% of inventoried species). Adult spiders were more abundant during Summer and less during Autumn; while young spiders occurred more during Autumn and less during Spring. Species richness also differed among seasons with more species during Spring and less during Autumn. A multivariate analysis of similarity (ANOSIM), either using Morisita quantitative index or Simpson qualitative index, shows spider species composition to differ among sites, but not among microhabitats within a site. There were no significant relationships between spider diversity components and abiotic (temperature and precipitation) factors, either for abundance or species richness. Biotic factors related to vegetation complexity (vegetation density, vegetation height, vegetation structure, canopy cover) were insufficient to explain variation in spider assemblages. Sheet web builders was the prevailing guild and running hunters were relatively rare. A MANOVA indicated significant differences in guild proportions among sites and among microhabitats. A canonical correspondence analysis (CCA) showed the guilds not to be correlated with the vegetation complexity variables evaluated. Analytical species richness estimators indicate that the spider fauna is still not sufficiently sampled, revealing the urgency of continuing studies, especially involving other sampling methods so as to evaluate other forest strata.

A intenção deste estudo foi conhecer a composição e a diversidade da araneofauna relacionada a matas ciliares no Rio Grande do Sul, em diferentes localidades: rio Piratini no município de Arroio Grande; rio Camaquã em Cristal; rio dos Sinos em Parobé e rio Maquiné em Maquiné. Foi comparada a riqueza de espécies, abundância e similaridade da fauna de aranhas e a sazonalidade, avaliando se fatores ambientais bióticos e abióticos influenciam na assembleia de aranhas das matas ciliares. Também foram avaliadas as guildas de aranhas, tanto aspectos espaciais, quanto temporais, e a influência de fatores abióticos e de complexidade ambiental sobre as mesmas. As aranhas foram separadas e classificadas em quatro guildas: construtoras de teias orbiculares, construtores de teias irregulares, caçadoras cursoriais e caçadoras emboscadoras/espreitadoras. Para a amostragem da araneofauna foram efetuadas duas coletas por estação durante dois anos em cada mata ciliar. As coletas foram realizadas em transecções, por um período de 45 minutos, utilizando como método o guarda-chuva entomológico (GCE) para amostrar a araneofauna do estrato arbóreo-arbustivo. Em cada mata ciliar foram delineadas seis transecções, duas em cada microambiente: borda com o campo, interior da mata e borda com o rio. O esforço amostral foi de 288 horas de coletas. No total foram coletadas 42057 aranhas: 8851 adultas (21%) e 33206 jovens (79%). A família predominante foi Theridiidae, com mais de 50% das aranhas amostradas em cada mata, a única exceção foi a mata ciliar do rio Maquiné que dominou Linyphiidae. A abundância de aranhas adultas foi diferente entre as matas ciliares, com maior abundância no rio Maquiné (34,58% das aranhas) e menor no rio dos Sinos (18,43%). Dos microambientes, as maiores abundâncias foram no interior da mata, seguido pelas bordas, muito próximas entre si. Foram determinadas 440 espécies/morfoespécies de aranhas, entre 168 gêneros. No total 232 espécies foram nominais (53%) com 14 novos registros para o Rio Grande do Sul e três para o Brasil. No rio Maquiné foi registrada a maior riqueza de espécies (S=218) e menor no rio dos Sinos (S=179). Entre os microambientes, as maiores riquezas de espécies de aranhas foram observadas nas bordas (campo: S=298 e rio: S=279) e após o interior da mata (S=246). A riqueza de espécies foi diferente entre os rios, já as riquezas entre os microambientes não diferiram. As espécies raras foram representadas por 130 singletons (32,5% das espécies do inventário). Para a sazonalidade, as aranhas adultas foram mais abundantes no verão e menos no outono; as aranhas jovens predominaram no outono e menos na primavera. A riqueza não diferiu entre os dois anos de amostragem. Já entre as estações a diferença foi significativa, maior na primavera e menor no outono. Não foi encontrada uma relação entre os fatores abióticos (temperatura e precipitação) com a araneofauna, tanto para abundância, quanto para a riqueza. As variáveis bióticas da vegetação não foram suficientes para explicar as variações na comunidade de aranhas, não influenciando nas mesmas. A análise multivariada de similaridade (ANOSIM), pelo índice quantitativo de Morisita e qualitativo de Simpson, mostra que as composições de espécies das araneofaunas diferem entre as matas ciliares. Para os diferentes microambientes não foram encontradas diferenças na composição. Prevaleceu a guilda das construtoras de teias irregulares e uma menor quantidade das caçadoras cursoriais. A MANOVA indicou diferença significativa na proporção das guildas entre todas as matas ciliares e entre os microambientes. A partir da análise de correspondência canônica (CCA), as guildas não estão correlacionadas com as variáveis de complexidade ambiental avaliadas. As curvas de rarefação e os estimadores analíticos de riqueza de espécies indicam que a fauna de aranhas ainda não foi suficientemente amostrada para as matas ciliares, revelando a urgência de estudos futuros, principalmente, abrangendo outros métodos, para diferentes estratos das florestas. / The aim of this study was to characterise the spider fauna associated to riparian forests in Rio Grande do Sul. Four different sites were sampled: Piratini River in Arroio Grande; Camaquã River in Cristal; Sinos River in Parobé and Maquiné River in Maquiné. Diversity components such as species richness, abundance and faunal similarity were compared among rivers, among microhabitats (forest edges and forest interior) and yearly seasons, and also against biotic and abiotic environmental factors. Spider foraging guilds were also evaluated, again for spatial and temporal scales, and again against biotic and abiotic environmental factors. Spiders were separated in four guilds: orbicular web builders, sheet web builders, running hunters and ambush hunters. Two samples per season were taken during two years for all forest sites. Sampling consisted of using beating tray on the tree-shrub strata across linear transections (50 X 2 m) for 45 min. Remote Ischemic preconditioning (PC) has been shown to protect the liver from ischemia-reperfusion injury. However little is known about the usefulness of ischemic precondition as a strategy to improve tolerance of non-heart-beating liver grafts to warm ischemia. Therefore we designed an experimental study aiming the effects of ischemic preconditioning on liver transplantation from non-heart-beating donors. Methods: Protocol was approved by Federal University of Goias Ethics Committee. Twentyfour Landrace pigs were assigned into 4 groups: I: after cardiac arrest livers were procured and transplanted; II: after PC and cardiac arrest livers were procured and transplanted; III: after PC and cardiac arrest, a 15 minutes period of warm ischemia was observed before livers were procured and transplanted; IV: after PC and cardiac arrest, a 30 minutes period of warm ischemia was observed before livers were procured and transplanted. Donors: cardiac arrest was obtained by ligation of coronary arteries and interruption of ventilation. PC (10` x 10`) and warm ischemia waiting time were observed accordingly. Livers were flushed with 4°C HTK solution cooled and procured. Recipients: standard technique was used. After one hour of reperfusion, blood and liver samples were collected and euthanasia was carried out. Non-parametrical statistical tests were applied. Results were expressed as medians. A value of p<0.05 was considered statistically significant. Results: AST, ALT, lactic acid and factor V levels were similar for the different studied groups by the end of one hour of reperfusion. A trend towards a deteriorated liver function was observed in group IV. When histological variables were addressed, vacuolization of hepatocytes and cell dropout scores were similar in all groups. Congestion score, vacuolization of hepatocytes and cell dropout scores tended to be higher in animals from group IV, but with no statistical significance. Conclusions: Remote ischemic preconditioning showed no benefits to liver grafts from nonheart- beating donors. Even though it did not result in benefits, no prejudice could be noticed in this research, as well. / Introdução: A melhora na função hepática em doadores de coração parado parece estar relacionada a curtos períodos de isquemia normotérmica, logo a lesão de isquemia e reperfusão é uma questão crucial não resolvida. O pré-condicionamento isquémico remoto (PC), promove proteção ao fígado da lesão de isquemia e reperfusão. No entanto, pouco se sabe sobre a utilidade do PC como uma estratégia para melhorar a tolerância de enxertos de doadores em parada cardíaca frente à isquemia normotérmica. Portanto, foi desenvolvido um estudo experimental visando os efeitos do pré-condicionamento isquêmico no transplante de fígado de doadores em parada cardíaca.Métodos: O protocolo do estudo foi aprovado na Universidade Federal de Goiás pelo Comitê de Ética em Pesquisa. Vinte e quatro suínos da raça Landrace foram divididos em 4 grupos: I: após parada cardíaca, o fígado foi captado e transplantado; II: após o PC, promoveu-se a parada cardíaca, seguida de captação e transplante hepático; III: após o PC, promoveu-se a parada cardíaca, seguida de um período de 15 minutos de isquemia normotérmica, com captação e transplante hepático em seguida; IV: após o PC, promoveu-se a parada cardíaca, seguida de um período de 30 minutos de isquemia normotérmica, com captação e transplante hepático em seguida. Nos doadores, a parada cardíaca foi obtida pela ligadura das artérias coronárias e interrupção do suporte ventilatório. O PC (10 'x 10') e o tempo de isquemia normotérmica foram observados em conformidade com a padronização. O fígado foi perfundido com solução HTK resfriada a 4 ° C e posteriormente captado. Para o receptor foi utilizada a técnica padrão. Após uma hora de reperfusão, amostras de sangue e de tecido hepático foram coletadas, seguida da realização da eutanásia. Foram aplicados testes estatísticos não-paramétricos. Os resultados foram expressos em medianas. O valor de p <0,05 foi considerado estatisticamente significativo. Resultados: Os valores encontrados de AST, ALT, lactato e fator V foram semelhantes para os diferentes grupos estudados até o fim do experimento. No entanto, foi observado uma tendência para piora da função hepática no grupo IV. Em relação às variáveis histológicas, a vacuolização dos hepatócitos e a desestruturação celular foram semelhantes em todos os grupos; houve uma tendência de aumento da congestão no grupo IV, sem significância estatística. Conclusões: O pré-condicionamento isquêmico remoto não mostrou benefício para enxertos de fígado de parada cardíaca. Mesmo que o PC não tenha promovido benefícios nesta pesquisa, também não foram observados danos.

ObjectifsL’objectif de ce travail a été d’évaluer la faisabilité d’utiliser le gel LeGoo pour occlure temporairement les artères coronaires afin de réaliser des pontages coronariens à cœur battant initialement chez le porc puis chez l’humain. Les objectifs secondaires ont été d’étudier l’impact de l’injection du gel sur l’endothélium et le myocarde.MéthodesLa première étude a eu pour but d’évaluer la possibilité de réaliser chez le cochon des pontages coronariens sur l’artère interventriculaire antérieure ou l’artère coronaire droite avec une artère thoracique interne, et d’évaluer la fonction endothéliale des artères coronariennes au niveau de la zone d’injection du gel.La deuxième étude a analysé chez le porc la fonction endothéliale des artères thoraciques internes au niveau de la zone d’occlusion par du gel LeGoo chez le porc.Après l’obtention du marquage CE, la troisième étude a permis de prouver la faisabilité de l’utilisation de LeGoo dans la réalisation de pontages coronariens à cœur battant chez l’humain et d’évaluer l’impact myocardique de cette utilisation.La dernière étude était une étude randomisée, multicentrique, internationale, comparant l’utilisation du gel pour la réalisation des anastomoses, versus les occlusions conventionnelles par élastiques péri-coronaires. RésultatsPour la première étude, 14 cochons ont été pontés à cœur battant et sacrifiés à la 3ème h (n=8) ou au 3ème jour (n=6). L’étude de la réactivité endothéliale n’a pas mis en évidence de différence entre les artères coronaires occluses par le gel et l’artère coronaire circonflexe qui servait de témoin. L’endothélium ne présentait pas de lésions histologiques en microscopie. Les mêmes résultats ont été obtenus dans la deuxième étude, sur les 9 artères thoraciques internes qui avaient été occluses durant 15 minutes avec le gel LeGoo.Les premières utilisations humaines ont permis de réaliser 99 anastomoses chez 50 patients. Le taux de satisfaction sur la qualité de l’hémostase obtenue a été de 91%. Un patient a présenté une arythmie, qui a nécessité de dissoudre le gel et de mettre en place un shunt intra-coronaire. Un autre patient a présenté un infarctus du myocarde à J1 lié à une sténose sur l’artère thoracique interne en amont de l’anastomose, nécessitant une réfection du pontage. Conclusions L’utilisation du gel LeGoo (Poloxamer 407) permet de réaliser des anastomoses en diminuant les temps d’occlusion et de manière sécuritaire sans altérer les fonctions de l’endothélium coronarien. / ObjectifsThe objective of this work is to assess the feasibility of using the LeGoo gel to occlude the coronary arteries temporarily, to facilitate the performance of coronary bypass (off-pump technique), initially in pigs then in human. A Secondary objective was to evaluate the impact of the injection on the endothelium and myocardium.MethodesThe first study evaluated in pigs the feasibility of using LeGoo, while performing coronary bypass on the left anterior descending or on the right coronary artery and to evaluate the endothelial function of coronary arteries occluded by the gel.The second study analyzed endothelial function of internal thoracic arteries, in the area occluded by the gel.After obtaining the CE mark, a third study demonstrated the feasibility of the utilisation of LeGoo for temporary coronary artery occlusion during the performance of beating heart coronary bypass surgery in human.The last study was a randomized, multicenter and international clinical study that compared the gel with conventional vessel loop during the performance of off-pump coronary artery bypass anastomoses.ResultsIn the first study 14 pigs received coronary bypass and were sacrificed after 3 hours (n=8) or 3 days (n=6). The study of endothelial reactivity did not reveal any difference between the coronary arteries occluded by the gel and the circumflex coronary artery, which served as a control. The endothelium showed no histological damage. Identical observations were made on the 9 internal thoracic arteries, used in the second study and occluded during 15 minutes by a LeGoo plug.In the first human use, 99 anatomoses were performed in 50 patients. The level of satisfaction on the quality of haemostasis was 91%. One patient presented arrhythmia which required the introduction of an intracoronary shunt after rapid dissolution of the gel. Another patient had a myocardial infarct on day 1 associated with stenosis on the internal thoracic artery upstream of the anastomosis, requiring a refection of this bypass (new off pump procedure). Other complications were identical to what is reported in the literature.In the randomised study, anastomoses times were shorter in LeGoo group than with vessel loops (12,8 ± 4,7 min vs 15,4 ± 6 min, p<0,001). The differences were 2 minutes for anterior bypass and 4 minutes for the lateral and posterior territories.The satisfaction rate of haemostasis was 88% with the gel against it was 60.7% with the vessel loop (p<0,0001). Other peri-operative measurements were statistically identical between the 2 groups.ConclusionsLeGoo (a poloxamer based formulation) can be used safely for the performance of coronary anastomoses. Anastomotic times are shorter than with traditional vessel loops, without any alteration of the endothelial function.

Caractérisation des mécanismes du battement ciliaire dans le cadre du transport mucociliaire normal et pathologique / Characterization of ciliary beat mechanisms under normal and pathological mucociliary clearance

L'épuration mucociliaire est la première ligne de défense de l'appareil respiratoire. L'altération de l'épuration mucociliaire se traduit par une stagnation et/ou une accumulation de mucus, conduisant potentiellement à des obstructions plus ou moins partielles et à des infections chroniques des voies aériennes. On compte deux grands types d'altération de l'épuration mucociliaire. Le premier est lié aux caractéristiques du mucus, comme dans le cas de la mucoviscidose. Le deuxième est lié à des anomalies de l'ultrastructure du cil et /ou de son battement regroupées sous le terme de "ciliopathies"qui peuvent être soit innées soit acquises.Ce travail, qui s'inscrit dans le cadre clinique de la problématique des ciliopathies respiratoires et de leur prise en charge, a pour objectif de mieux caractériser, à partir des outils de la Biomécanique, les mécanismes du battement ciliaire (incluant une mesure de l'efficacité globale du battement des cils) et à transférer ces connaissances au profit de la clinique.Pour cela une méthodologie de caractérisation systématique de la mécanique ciliaire à partir de prélèvements biologiques issus de patients observés par vidéo-microscopie à haute vitesse a été développée parallèlement au développement d'un modèle numérique intégrant le couplage entre battement ciliaire et transport de fluide. Cette association entre expériences et modèle numérique a permis de proposer un nouvel index utilisable par les cliniciens pour caractériser l'efficacité du battement ciliaire. Alteration of this clearance leads to a stagnation and/or accumulation of mucus, potentially resulting in more or less partially obstructions and in chronic infections of the airways. There are two main types of mucociliary clearance alterations. The first one is linked to the mucus characteristics, as in cystic fibrosis. The second one is connected to cilium ultrastructure abnormalities and/or ciliary beating defects encompassed by the term of "ciliopathies" which may be primary or acquired.This work, which takes place in the clinical issue of respiratory ciliopathies and their care, aims to better characterize, through biomechanics tools, the mechanisms of ciliary beating (including a measurement of the global efficiency of cilia) and to transfer these knowledges in favor of the clinical management.A methodology of systematic characterization of the ciliary mechanics, from biological samples derived from patients observed through high-speed video-microscopy analysis, has been developed as the same time as the development of a numerical model incorporating the coupling between ciliary beating and fluid motion. This association between experiments and numerical model allowed to propose a new index usable by the clinicians to characterize the ciliary beating efficiency. This index has the advantage of not requiring a modification of the clinical practice of biological data collection
