Structural and functional properties of NMDA receptors in the mouse brain endothelial cell line bEND3Dart, Christopher F. 07 January 2011 (has links)
Previous work in our laboratory indicates that the diameter of brain arteries and arterioles can be increased by N-methyl-D-aspartate (NMDA) receptor activation. We looked for expression of NMDA receptors and endothelial cell responses to NMDA receptor agonists and antagonists in the mouse brain endothelial cell line bEnd.3.
Using RT-PCR and Western blotting we found evidence supporting the presence of NMDA receptor subunits NR1 and NR2C. Treatment of bEnd.3 cells with combinations of 100 μM glutamate and D-serine significantly increased intracellular calcium. However, we saw no direct evidence that NO was produced in response to NMDA receptor activation using the Griess method. We did observe an NMDA receptor-dependent increase in protein nitrosylation. This increase is unlikely related to enhanced NO levels since it was not correlated with NO production and was not inhibited by the endothelial NO synthase inhibitor L-NIO.

Title: Pedagogy of Yoga Asanas in Relationship to the Public Health Objectives: The aim of this thesis is to describe selected forward bend asanas according to the literature, compare the performance and instructions given during lessons with our research. We would like to find out whether a student's health condition is being asked of and considered by the tutor and whether there is a connection between the education of the tutor and their ability to provide appropriate directions. Evaluation of the possibilities for the use of yoga in physiotherapy. Methods: It is descriptive research of theoretical and empirical character, where research, observation, guided interview and comparison of acquired data were used. Results: Among the chosen asanas we found discrepancies in literature research. There were discrepancies among the theoretical background of asanas and their pedagogy in yoga classes. Two thirds of tutors do not ask about an individual's health limitations. A connection between tutors' education in yoga and ability to provide appropriate directions during classes has been not confirmed nor was a connection between medical education in the tutor and their ability to lead quality class. 20% of classes included less than three forward bend asanas and there was a class with none. Numeric Modelling of Water Hammer Effects in PenstocksBernard, Dominic January 2013 (has links)
Water hammer represents a complex hydraulic phenomenon with significant consequences on the proper functioning and safety of operation for pipe and conduit systems. The complexity and intricate physics of water hammer translated into significant difficulties associated firstly, with finding a proper solution for understanding the mechanism of its occurrence and, secondly, relating to proposing technically and economically viable design methods and devices that would help reduce and mitigate water hammer effects. In this context, the present thesis deals with the numerical modeling of the transient behaviour of water pipe segments. Following an extensive literature review of the state-of-the-art on the water hammer mechanisms and past work on experimental, analytical and numerical analysis of this phenomenon, a three dimensional numerical model of the water hammer in a pipe which considers the fluid-structure interaction (FSI) is developed using a Finite Element Method – Finite Volume Method (FEM-FVM) technique. Structural and fluid computational results based on rapid and slow gate closure scenarios are compared with existing closed-form solutions of the water hammer.
A parametric study is also performed on a simply supported pipe segment to determine the influence of various design parameter. A systematic sensitivity analysis was conducted and a ranking mechanism was established for the importance of each parameter on the fluid fields and structural response. A first comparative analysis is conducted on horizontally and vertically bent elevated pipe segments to quantify the influence of the bend angle on the results. A second comparative analysis is performed on a horizontally bent segment buried in soil to determine the influence of the pipe interaction with the soil on the response.
It is observed that the thickness, span, initial velocity and bend angle had a significant impact on the pressure and structural response. The presence of soil was observed to have a significant benefit in decreasing the von-Mises stresses.

The bend is made by the bending and the press tools. The press tools and the bending profile were suggested on the bases of the literature study of technology bending. In consideration of the calculated bending power and the dimension profile will be made profile on the hydraulic press CYS 320 ( the manufacturer ŠMERAL Brno a.s.). At the end of the the thesis there are performed calculations of the technical and economic evaluation for the machine press and the bending .

Analysis of the Influence of Negatively Buoyant Jets on Curved Open-Channel Flow by Means of Numerical and Experimental MethodsWang, Xueming 18 November 2022 (has links)
In urban areas, discharging wastewater into rivers is a common way to dispose of contaminants, and it is usually the most economical. Accurate information about how effluents are distributed in the receiving water body is desirable when designing industrial plants. Flow structures will be influenced by an effluent’s dilution processes during the mixing. Meanwhile, the cross-stream motions resulting from the streamline curvature can redistribute both the velocity and the shear stress, which favors the mixing behavior compared to a straight channel. However, the interactions between jet mixing behavior and the bend flow requires further investigation.
In the present study, jets with different densities were discharged horizontally into a laboratory flume with a 135-degree open channel bend, and both the main and secondary flow behaviors in the bend were observed after the introduction of effluents. The acquired three- dimensional velocity data were used to validate numerical models of the effluent-bend flow. Numerical turbulence models such as the standard k-ε eddy viscosity model, non-linear k-ε model (Shih quadratic k-ε), and the k-ω SST (shear stress transport) model were employed to evaluate their accuracy. OpenFOAM was selected in the analysis for proposing better numerical models since it gives high-quality results to individualized complex fluid flows, and as an open source CFD software it can be beneficial to further develop and maintain.
The first part of this study presents the implementation of the physical modelling of the proposed problems. Detailed descriptions of the experimental process were elaborated. Specifically, the three velocity components at four cross-sectional planes in the bend section were measured with and without saltwater jets by using the stereo Particle Image Velocimetry (PIV)

technique in the laboratory flume. The experimental results show that the more pronounced effects with the jets were found at the beginning and exit of the bend. Although the jets had little effect on the maximum streamwise velocity, it was found that the occurrence of the negatively buoyant jets would affect the patterns and properties of the secondary flow in the bend.
The second part of this study investigated the mechanisms underlying the two cells system, particularly when interacting with a discharged effluent jet. Detailed experimental data were used in interpreting the large center-region cell as well as small structures in the 135-degree open channel bend. A term-by-term analysis of the downstream vorticity equation was executed to investigate the various mechanisms underlying these cross-stream flow motions considering the influence of the negatively buoyant jets. The results indicated the generation and the dissipation of the streamwise vorticity with the effective terms of the vorticity equation.
The third part of this study evaluated the performance of three different turbulence models with the experimental measurements. It can be concluded that fully 3D numerical models are capable of simulating the primary flow pattern in a strongly curved channel with the presence of a negatively buoyant jet. The comparison also shows that, although the outer bank cell was not predicted, the k-omega SST model can satisfactorily predict some of the smaller flow features in bend flow, such as the inner bank circulation cell and the overall form of the vorticity distributions. The results enable more reliable predictions for the characteristics and development of jets in a bend.

The purpose of the study was to provide a complete historical account of the events which led to the rise and fall of the Studebaker Brothers Manufacturing Company of South Bend, Indiana. The study also evaluated the impact upon the ethnic development of South Bend from the years 1856 to 1966.The applicability of the Kuhnian paradigmatic process of culture change to the South Bend-Studebaker interaction sphere was considered as well.Ninety-seven employees of the company were selected and interviewed in regard to their knowledge of the company and its impact upon the city. Primary and secondary archival materials were utilized to supplement worker interviews.FINDINGS1. The Studebaker Brothers Manufacturing Company and the Studebaker Corporation acted as primary agents of ethnic development in the South Bend community.2.The interviewing process provided new data which supplemented and substantiated previous accounts.3. The Studebaker Company's success was founded upon intensive employer-employee involvement in the production process.4. The Studebaker Company's failure was brought about by the breakdown of the employer-employee relationship.CONCLUSIONS1. The study proved the significant impact of the Studebaker Company upon the American transportation industry.2. The Studebaker Company exerted a major influence upon the ethnic and cultural development of the city of South Bend.3. The "paradigmatic process of social change" model as postulated by Thomas Kuhn was appropriate to the Studebaker-South Bend situation.4. An ethnohistorical reconstruction technique proved successful in recounting the impact of the Studebaker Company.
