Double-negative (CD27−IgD−) B cells are expanded in NSCLC and inversely correlate with affinity-matured B cell populations

Background: The presence of B cells in early stage non-small cell lung cancer (NSCLC) is associated with longer survival, however, the role these cells play in the generation and maintenance of anti-tumor immunity is unclear. B cells differentiate into a variety of subsets with differing characteristics and functions. To date, there is limited information on the specific B cell subsets found within NSCLC. To better understand the composition of the B cell populations found in NSCLC we have begun characterizing B cells in lung tumors and have detected a population of B cells that are CD79A(+)CD27(-)IgD(-). These CD27(-)IgD(-)(double-negative) B cells have previously been characterized as unconventional memory B cells and have been detected in some autoimmune diseases and in the elderly population but have not been detected previously in tumor tissue. Methods: A total of 15 fresh untreated NSCLC tumors and 15 matched adjacent lung control tissues were dissociated and analyzed by intracellular flow cytometry to detect the B cell-related markers CD79A, CD27 and IgD. All CD79A(+) B cells subsets were classified as either naive (CD27(-)IgD(+)), affinity-matured (CD27(+)IgD(-)), early memory/germinal center cells (CD27(+)IgD(+)) or double-negative B cells (CD27(-)IgD(-)). Association of double-negative B cells with clinical data including gender, age, smoking status, tumor diagnosis and pathologic differentiation status were also examined using the logistic regression analysis for age and student's t-test for all other variables. Associations with other B cell subpopulations were examined using Spearman's rank correlation. Results: We observed that double-negative B cells were frequently abundant in lung tumors compared to normal adjacent controls (13 out of 15 cases), and in some cases made up a substantial proportion of the total B cell compartment. The presence of double-negative cells was also found to be inversely related to the presence of affinity-matured B cells within the tumor, Spearman's coefficient of -0.76. Conclusions: This study is the first to observe the presence of CD27(-)IgD(-)double-negative B cells in human NSCLC and that this population is inversely correlated with traditional affinity-matured B cell populations.

Challenges of Experienced Nurses in a Full-Time Online RN-BS Nursing Program

Practicing nurses need to be qualified at the baccalaureate level at a rate of 80% by 2020 to meet the needs of the increasingly demanding healthcare industry. Designed to meet this need, the online RN-BS (registered nurses earning a Bachelor of Science) nursing program at a local university has a declining completion rate that needs to be addressed and increased. The conceptual framework for this project was a qualitative program evaluation with a case study design; the project was designed to determine if the structure of the program was limiting student success or if factors experienced by full-time working RNs in the program were impacting success. E-mail interviews were conducted with 29 students in the online RN-BS nursing program who were selected through purposeful sampling. Research questions for this study examined the barriers faced by experienced nurses when they studied in the online RN-BS nursing program while maintaining their work schedule as RNs. Predetermined, open-ended, semistructured questions produced data that were analyzed through coding and theme identification. Themes were validated through member checking and use of an external auditor. Data revealed that students preferred the online format of the program to a face-to-face program, and they felt the local university nursing faculty was responsive and supportive. Challenges the students faced were primarily time management, cost of the program, information technology issues including learning the learning management system, group projects, and study skills. Based on the findings, an orientation program was developed that will affect social change by helping the local university nursing students complete the baccalaureate preparation they need to comply with the increasing industry demands.

Cellular-connected UAV in Next-Generation Wireless Networks

Unmanned aerial vehicle (UAV) technology has gained a great interest in communication systems due to its ability to host a cellular base station (BS) and thus act as an aerial BS (UAV-BS). The inheritance of mobility in the airspace makes the deployment of UAV-BSs flexible and agile aiming to mainly complement the terrestrial network, extend its coverage, and serve as a capacity injector in high-throughput demand scenarios. Besides, a UAV can also act as an aerial user (UAV-UE) for various use cases, such as aerial data collection and cargo delivery. Such UAV-UE missions need reliable cellular communication links in order to safely operate in beyond visual line-of-sight (BVLoS). Since terrestrial networks were not primarily designed to serve aerial users, due to their down-tilted BS antennas, re-coursing solely to these networks for aerial users’ cellular connectivity might not be a viable approach as a long-term solution. Alternatively, deploying UAV-BSs in this context can substantially improve both aerial and terrestrial users coverage and capacity. One of the challenging issues is how to characterize the UAV-UE performance in integrated aerial/terrestrial networks, called vertical heterogeneous networks (VHetNets). First, we thoroughly study the aerial user’s performance in terms of coverage probability in a VHetNets setup. Under a more realistic system model, we revisit the coverage and throughput performances of an aerial user in VHetNets, considering LoS and non-LoS (NLoS) transmissions and under different spectrum sharing policies among separate aerial and terrestrial networks. Some insights have been concluded on the integration of aerial BSs and UAV-UEs with the existing terrestrial network. Specifically, optimal positioning of UAV-BSs for maximized aerial users coverage was investigated for various aerial users distribution and spectrum allocations. Moreover, visioning that UAV technology will revolutionize the cargo delivery industry, we proposed a new concept of 3D aerial highways, which designs coordinated routes for a massive number of UAVs used mainly for delivery purposes. In this context, multiple network technologies were proposed and discussed to guarantee the cellular connectivity of cargo-UAVs in 3D aerial highways. For the particular case of connectivity supported by terrestrial cellular networks, an optimal energy-efficient and low-handoff trajectory planning for a cargo-UAV mission was proposed, with respect to disconnectivity constraints. Consequently, associated design guidelines and recommendations have been drawn. Leveraging reinforcement learning (RL) tools, we proposed a novel algorithm for path planning and cell association for the cargo-UAV that maximizes its cellular service reliability and minimizes the handoff events. Finally, we introduced a new paradigm, intermittently tethered UAV (iTUAV), as a trade-off between mobility and energy availability for providing cellular connectivity in temporary events.

Considerando a importância de que a qualidade dos processos realizados em uma instituição seja mensurada e avaliada, tem por objetivo pesquisar sobre indicadores de desempenho para bibliotecas e propor sua aplicação. O objeto do trabalho está delimitado considerando-se, especificamente, os processos de desenvolvimento de coleções (mais relacionados à avaliação do uso do acervo) e de processamento técnico do Sistema de Bibliotecas da Universidade Federal do ABC (SisBi UFABC). Para tanto, revisa a literatura sobre os temas abordados (Qualidade, Gestão da Qualidade, Avaliação em Serviços de Informação e Indicadores de Desempenho), contextualiza o macro e o micro ambientes envolvidos (UFABC, SisBi UFABC, Divisão de Desenvolvimento de Coleções e Divisão de Processamento Técnico) e apresenta casos de aplicação de indicadores de desempenho em bibliotecas. Analisa a terceira edição da norma BS ISO 11620 Information and documentation - Library performance indicators, publicada em 2014, e a segunda edição das Diretrizes da IFLA intituladas Measuring quality: performance measurement in libraries, publicadas em 2007. Ambos documentos trazem orientações para a aplicação de indicadores de desempenho especificamente em bibliotecas. Após a análise dos dois documentos, apresenta a aplicação de seis indicadores de desempenho, sendo três para os processos de desenvolvimento de coleções (Disponibilidade de Títulos Solicitados; Taxa de Uso da Coleção e Porcentagem do Acervo não Usado) e três para o processamento técnico (Mediana de Tempo de Processamento dos Documentos; Produtividade da Equipe de Processamento e Custo por Título Catalogado). The fishes of this order have a compact DNA which favors molecular studies, as well as comparisons with more basal species. Model of genome evolution, there are still many gaps in knowledge about their chromosomal patterns and how evolutionary rearrangements influence the marked variation in DNA content of this order. In view of this, we present cytogenetic analyzes of the species Acanthostracion quadricornis (Ostraciidae), A. polygonius (Ostraciidae) Melichthys niger (Balistidae) Cantherhines macrocerus (Monacanthidae) and C. pullus (Monacanthidae), Lagocephalus laevigatus, Colomesus psittacus and Canthigaster figueiredoi (Tetraodontidae), to contribute with cytogenetic data for this group. The analysis was performed by C-banding, Ag-RONs, coloring with base-specific fluorochromes DAPI-CMA3, restriction enzymes AluI, EcoRI, TaqI, PstI and HinfI and in situ hybridization with probes for ribosomal DNA 18S and 5S. The heterochromatic ultrastructure of A. quadricornis and A. polygonius revealed a outstanding heterochromatin content, which may indicate that the accumulation or loss of extensive heterochromatin content could be responsible for large variations in genomic content displayed in different Tetraodontiformes families. The species Cantherhines macrocerus, C. pullus (Monacanthidae) and Melichthys niger (Balistidae) shows a huge karyotypic similarity both numerically and structural. L. laevigatus showed similar cytogenetic features (2n = 44 and single RONs) to the species of the genus Takifugu, which reinforces the idea of their phylogenetic relationships. C. psittacus presented the highest diploid number described for the family (2n = 56) and large amount of HC, features that related with its sister family Diodontidae. Cytogenetic analysis in C. figueiredoi revealed heterochromatic polymorphisms, RONs multiple and Bs chromosomes. These events are rare in marine fishes, and are possibly associated with the strong restructuring and genomic reduction that this family has been suffered. These features, plus the morphological and molecular data suggests that these species share the same ancestral branch, with a possible monophyletic origin. In this study, new contributions to the knowledge of evolutionary patterns facing by Tetraodontiformes are provided and discussed under cytotaxonomyc, genomic and evolutionary perspectives. / Frente ? grande diversidade de peixes, a Ordem Tetraodontiformes se destaca por exibir caracter?sticas gen?ticas e morfol?gicas bastantes singulares. Os peixes desta Ordem apresentam um DNA compacto o que favorece estudos moleculares, assim como compara??es com esp?cies mais basais. Modelo de evolu??o gen?mica, ainda existem v?rias lacunas de conhecimento sobre seus padr?es cromoss?micos e como os rearranjos evolutivos influenciaram na marcante varia??o no conte?do de DNA desta Ordem. As an?lises foram realizadas atrav?s do bandamento C, Ag-RONs, colora??o com fluorocromos base-espec?ficos DAPICMA3, enzimas de restri??o AluI, EcoRI, TaqI, PstI e HinfI e pela hibrida??o in situ com sondas de DNA ribossomal 18S e 5S. A ultra-estrutura heterocromat?nica de A. quadricornis e A. polygonius, revelaram um marcante conte?do heterocrom?tico, situa??o que pode indicar que o ac?mulo ou perda de extenso conte?do de heterocromatinas poderiam ser respons?veis pelas extensas varia??es no conte?do gen?mico exibidas nas diferentes fam?lias dos Tetraodontiformes. As esp?cies Cantherhines macrocerus, C. pullus (Monacanthidae) e Melichthys niger (Balistidae) apresentam uma grande similaridade cariot?pica, tanto num?rica, como estruturalmente. Lagocephaluslaevigatus mostrou caracter?sticas citogeneticas similares (2n=44 e RONs simples) as esp?cies do g?nero Takifugu, o que refor?a a id?ia de seu relacionamento filogen?tico, e Colomesus psittacus apresentou o maior n?mero dipl?ide descrito para a fam?lia (2n=56) e grande quantidade de HC, caracter?sticas que o relacionariam com a fam?lia irm? Diodontidae. An?lises citogen?ticas em C. figueiredoi revelaram polimorfismos heterocrom?ticos, RONs m?ltiplas e cromossomos Bs, sendo estes eventos raros para peixes marinhos, estando possivelmente associados ? marcante reestrutura??o e redu??o gen?mica que esta fam?lia sofreu. Estas caracter?sticas, somadas aos dados morfol?gicos e moleculares sugerem que estas esp?cies compartilham de um mesmo ramo ancestral, com poss?vel origem monofil?tica. Neste trabalho novas contribui??es ao conhecimento dos padr?es evolutivos enfrentados pelos Tetraodontiformes s?o fornecidas e discutidas sob perspectivas citotax?nomicas, gen?micas e evolutivas.

Mesure de la violation de CP dans les désintégrations Bs -> J/psi phi auprès du détecteur LHCb

La mesure de la phase phis violant la symétrie CP dans les désintégrations Bs -> J/psi phi est une des analyses phares de l'expérience LHCb. Le Modèle Standard prédit cette observable avec une faible incertitude théorique. La présence de Nouvelle Physique notamment dans les diagrammes en boucle de l'oscillation Bs - anti Bs peut significativement modifier la mesure de cette phase. Durant cette thèse, nous avons participé à différents aspects de l'analyse des désintégrations Bs -> J/psi phi. Nous avons proposé une sélection multi-variables, basée sur un algorithme d'arbres de décision boostés. Cette nouvelle sélection augmente le nombre de signal Bs -> J/psi phi de 19% mais nécessite une correction temporelle. Une étude portant sur l'origine des distorsions angulaires caractéristiques des désintégrations Bs -> J/psi phi a aussi été menée. Il s'avère que la principale source de distorsion est la géométrie du détecteur LHCb. Nous avons aussi contribué à un autre élément clef de cette analyse : l'étiquetage de la saveur. Afin de connaître la saveur initiale des mésons B, un algorithme d'étiquetage a été développé. Il exploite les traces chargées provenant du hadron B de côté opposé. Nous avons participé à son optimisation ainsi qu'à son étalonnage en étudiant les désintégrations Bd -> J/psi K*. En utilisant l'intégralité des données enregistrées en 2011, soit 1 fb-1, après optimisation et calibration de cet algorithme, la puissance d'étiquetage dans le canal Bs -> J/psi phi est : (2.29 +- 0.07 +- 0.26) %. La valeur de phis mesurée est alors : \phis = -0.001 +- 0.101 +- 0.027 rad dans les désintégrations Bs -> J/psi phi. L'analyse combinée de Bs -> J/psi phi avec les désintégrations Bs -> J/psi \pi+ \pi- donne la meilleure mesure mondiale de \phis = -0.002 +- 0.083 +- 0.027 rad. Cette mesure est compatible avec le Modèle Standard, mais ses incertitudes laissent possible la présence de Nouvelle Physique.

An examination of von Soden's I^bl group of manuscripts (Acts & Catholic Epistles only)

This collation of 8 of von Soden's manuscripts is a companion work to my earlier investigation into his manuscripts. A similar division of this thesis is employed, with the nucleus in sections III, IV & V. Section III, with the general and individual descriptions of the MSS involved, is an attempt to bring together in one place all the known details about the history, the format, the scribe, the script and any other relevant material. Section IV contains the Synoptic Collation of the Group in Acts and the Catholic Epistles. Section V is itself divided into six parts and is concerned with the singular and subsingular readings. The conclusion is that neither can logically be separated from one another as the interrelationship of individual MSS is far more complex than von Soden makes evident.
