Tato diplomová práce se zabývá návrhem IP bloku pro diskrétní kosinovou transformaci. V~teoretické části jsou shrnuty algoritmy pro výpočet diskrétní kosinové transformace a diskutována jejich použitelnost v~hardwaru. Zvolený algoritmus pro hardwarovou implementaci je modelován v jazyce C. Poté je popsán na RTL úrovni, verifikován a je provedena syntéza v~technologii TSMC 65 nm. Hardwarová implementace je poté zhodnocena s ohledem na datovou propustnost, plochu, rychlost and spotřebu.

Instruction-set-simulator-less Virtual Prototype Framework for Embedded Software Development

With continuous advancement in silicon technology and high feature demands on consumer electronics, the complexity of embedded software has led the software development effort to dominate System-On-Chip (SoC) design. Virtual Prototype (VP) addresses the problem by enabling early software development before hardware arrival. However, VP still poses challenges: 1) Instruction Set Simulator (ISS) degrades simulation time, 2) Development is restricted to embedded processor specific tools and 3) Applications and drivers are dependent on system software completion. In this work, we propose an abstraction framework which: 1) Removes ISS from VP, achieving native host software execution time, 2) Activates rich suites of desktop development tools in host by compiling embedded software in host binary and 3) Allows system software independent application and driver development. With this framework, we successfully demonstrated up to 2000% speed-up in VP run-time over conventional VP and improved software development productivity significantly.

Effects of the EPI-C Model upon Self-Actualization of Clients in Group Counseling

The problem with which this investigation is concerned is that of the evaluation of the effectiveness of the EPI-C model as a guide to group counseling. The purposes of the study are (1) to determine whether group counseling employing the EPI-C model results in positive gain in self-actualization, and (Z) whether group counseling using the EPI-C model is more effective than a topical discussion group or no treatment at all in producing greater positive change in subject self-actualization. This report concludes that the EPI-C model as a guide to group counseling is effective as a means of increasing reliance on inner support, increasing ascendance levels, emotional stability, and objectivity. The model also produced changes in assessed congruence of the emotional, physical and intellectual self. It demonstrated that the EPI-C model facilitated the growth of individuals on certain dimensions of self -actualization.

Novel mouse mammary cell lines for in vivo bioluminescence imaging (BLI) of bone metastasis

BACKGROUND:Tumor cell lines that can be tracked in vivo during tumorigenesis and metastasis provide vital tools for studying the specific cellular mechanisms that mediate these processes as well as investigating therapeutic targets to inhibit them. The goal of this study was to engineer imageable mouse mammary tumor cell lines with discrete propensities to metastasize to bone in vivo. Two novel luciferase expressing cell lines were developed and characterized for use in the study of breast cancer metastasis to bone in a syngeneic mouse model.RESULTS:The 4 T1.2 luc3 and 66c14 luc2 cell lines were shown to have high levels of bioluminescence intensity in vitro and in vivo after orthotopic injection into mouse mammary fat pads. The 4 T1.2 luc3 cell line was found to closely model the sites of metastases seen in human patients including lung, liver, and bone. Specifically, 4 T1.2 luc3 cells demonstrated a high incidence of metastasis to spine, with an ex-vivo BLI intensity three orders of magnitude above the commercially available 4 T1 luc2 cells. 66c14 luc2 cells also demonstrated metastasis to spine, which was lower than that of 4 T1.2 luc3 cells but higher than 4 T1 luc2 cells, in addition to previously unreported metastases in the liver. High osteolytic activity of the 4 T1.2 luc3 cells in vivo in the bone microenvironment was also detected.CONCLUSIONS:The engineered 4 T1.2 luc3 and 66c14 luc2 cell lines described in this study are valuable tools for studying the cellular events moderating the metastasis of breast tumor cells to bone.

Dynamiken hos organiskt kol i Mälarens avrinningsområde : flöden, drivande faktorer och modellering

I denna rapport undersöks hur mängden organiskt kol, TOC (Totalt organiskt kol), varierar i tid och rum i Mälarens avrinningsområde, samt vad det är som styr TOC-halten i Mälaren. Det är viktigt att förstå dynamiken hos TOC i Mälaren och i dess avrinningsområde eftersom ökat TOC i vattnet påverkar vattenkvaliteten och orsakar problem vid beredning av dricksvatten. TOC kan bland annat reagera med klor/UV-ljus och bilda cancerframkallande ämnen. Det kan också öka antal mikrober i vattnets distributionssystem. Arbetet omfattar analys av samband mellan elementen, transportberäkningar per ytenhet av elementen till Mälaren och en modelleringsansats för ett av avrinningsområdena. Rapporten innehåller även en jämförelse mellan de olika vattenföringsmodellerna samt uppmätt vattenföring för analys av eventuella systematiska skillnader mellan dessa som påverkar beräkningen av TOC och de andra elementens transport till Mälaren. Analysen av sambanden mellan variablerna TOC (mg/l), kaliumpermanganat förbrukning (KMnO4, mg/l), absorbans_F (F=filtrerad), järn (mg/l), mangan (mg/l) och SO4_IC (sulfat mätt med hjälp av jonkromatografi, mg/l), visade att vissa av dessa variabler är korrelerade med varandra. TOC mot KMnO4 och TOC mot absorbans_F hade de bästa anpassningarna med respektive R2- värden 0,65 och 0,59 och p-värden <0,001. Årsnederbörd är positivt korrelerad med TOC per ytenhet för Kolbäcksån med R2-värde 0,63 och p-värde <0,01, vilket innebär att sambandet är signifikant. Ökad årsnederbörd leder till ökad tillförsel av TOC till Mälaren. Det finns däremot inget signifikant samband mellan TOC-transport per ytenhet och årsmedeltemperatur. Arealflödesberäkningar tyder på att den största tillförseln av TOC- transport per ytenhet kommer från den nordöstra delen av Mälaren. Fyrisån står för den största tillförseln av TOC. Hydrologiska, kemiska och meteorologiska data inkluderades i modeller för att kunna skatta TOC-halten i Mälaren. Temperatur-, evapotranspirations- och nederbördsdata användes i en hydrologisk modell, HBV- modellen, för att simulera vattenföringen från avrinningsområdet. Sedan användes en processbaserad modell, INCA- C, som drivs av hydrologisk data och beräknade grundvattenbildning och markfuktighet för att simulera tidsmässiga mönster i TOC. Invariablerna till INCA-modellen, markfuktigheten och HER (grundvattenbildning), simulerades med hjälp av HBV- modellen. Dessa modeller tillämpades i Kolbäcksån (ett av Mälarens största avrinningsområden). Modelleringen av Kolbäcksåns TOC- halt resulterade i en modell som anpassade dynamiken mellan 1996 och 2009, men missar den mellan 2009 och juni 2010, med bäst anpassning mellan 2006 och 2008. R2- och NS värden som erhölls för modellen var 0,086 och -0,059. / In this report, it has been investigated how the amount of organic carbon, TOC, varies in time and space in the basin of Mälaren, and what controls the TOC content in the lake. It is important to understand the dynamics of the TOC in the lake and its catchment because increased TOC in the water affects water quality and causes problems in the preparation of drinking water. Particularly, it can react with chlorine / UV- light and form carcinogenic substances. It can also increase the number of microbes in water distribution systems. In addition the work includes analysis of the relation between water chemistry variables, annual fluxes calculations (g/m2/year) of element flows to the lake and a modeling approach to a watershed. Annual fluxes calculations (g/m2/year) indicate that the largest supply of TOC to the lake comes from the northeast of the lake. Fyrisån accounts for the largest input of TOC to the lake. The high TOC-flux is due to a small proportion of open water in the catchment. Hydrological, chemical and meteorological data have been included in models to estimate the TOC content in the Mälaren. Input data processing, especially precipitation data, has been an important part of the work as it affects the whole model. Temperature, evapotranspiration and precipitation data were used in a hydrological model, HBV model, to simulate the flow from the catchment area. Then a process-based model, INCA-C, operated by the hydrological data and soil moisture, has been used to simulate the temporal patterns in TOC. The input variables to INCA-C- model, soil moisture and HER (Hydrological effective rainfall), have been simulated using the HBV- model. Those models were applied in Kolbäcksån, one of the lake's largest catchments. The modeling of Kolbäcksån resulted in a model that captured the dynamics of a few periods of the whole time series. The modeling of Kolbäcksån TOC-concentration resulted in a model that captured the dynamics between 1996 and 2009, but misses it between 2009 and June 2010. R2 and NS values obtained for the model were 0.086 and -0.059, respectively.

Atualmente, o uso do solo e mudança de uso do solo emitem 1,3 ± 0,5 Pg C ano-1, equivalente a 8% das emissões globais. Técnicas como a agricultura de baixa emissão de C têm sido desenvolvidas para sequestrar C nos solos e reduzir a emissão de gases do efeito estufa. Porém, além dos desafios políticos e sociais envolvendo a doção destes sistemas, ainda há muita incerteza sobre o seu real potencial de mitigação. Assim, os objetivos desse estudo foram: i) Quantificar as fontes históricas e atuais de emissão de gases do efeito estufa na região dos Campos Gerais do Paraná, Brasil; ii) quantificar o potencial das melhores práticas de manejo agrícola baseadas nos três pilares da agricultura de conservação: Solo permanentemente coberto, plantio direto e rotação de culturas, em longo prazo (30 anos) para sequestrar carbono no solo, utilizando a fazenda Paiquerê (localizada na região dos Campos Gerais) como um modelo de sucesso; iii) estimar o impacto da adoção das melhores práticas de manejo nas áreas agrícolas da região e globalmente onde adequadas pelos próximos 100 anos utilizando os modelos Century e Roth-C. As fontes de gases do efeito estufa foram apresentadas como um inventário e mostraram que as emissões históricas (1930 – 2017) foram 412,18 Tg C, no qual as mudanças de uso do solo contribuíram com 91% (376,2±130 Tg C). As florestas sequestraram 51.7 ± 23.9 Tg C em 0.6 Mha em 47 anos (1.8 Tg C Mha-1 ano-1) e o plantio direto sequestrou 30.4 ± 23.9 Tg C em 1.9 Mha em 32 anos (0.5 Tg C Mha-1 ano-1). Ambos os modelos tiveram uma boa performance e o modelo Century foi mais eficiente em simular os estoques de carbono do solo, o resíduo médio da simulação foi 10 Mg C ha−1 (13%) para n = 91. O resíduo do modelo aumentou com a quantidade de óxidos no solo, sugerindo que a inclusão do controle mineralógico pode reduzir o viés de simulação. As predições do Century mostraram que o sistema tem potencial para mitigar 13 anos de emissões regionais (330 Tg C em 100 anos) ou 105 anos de emissões do setor agricultura, floresta e pecuária (40 Tg em 100 anos) na região. Da mesma forma, globalmente o sistema apresenta um potencial para sequestrar 2,5 ± 0.02 Pg C na profundidade 0–20 cm e 11,7 ± 3 Pg C na profundidade 0-100 cm em 86 milhões de ha distribuídos por todo o mundo. Este valor é equivalente à 11% das emissões globais dos setores agricultura, floresta e pecuária e mudanças de uso do solo. Assim, a nossa metodologia possa ser utilizada como um modelo para divulgar o potencial da agricultura conservacionista em sequestrar C nos solos e suportar políticas públicas que visem à mitigação das emissões de gases do efeito estufa. / Soils can be a source or sink of atmospheric CO2, according to land use and management. Currently the land use and land use change (LULUC) emits 1.3 ± 0.5 Pg carbon (C) year-1, equivalent to 8% of the global annual emission. Techniques such as low carbon agriculture, has been developed to sequester C in soils and reduce greenhouse gas (GHG) emissions. However, besides political and social challenges for the system adoption, there’s still great uncertainty related to its real mitigation potential. This study aimed: i) Quantify the historical and current main sources of GHG emissions for Campos Gerais region in Paraná state, Brazil; ii) quantify the potential of long term (30 years) agricultural best management practices, based on the three pillars of conservative agriculture: permanent soil cover, crop rotation and no-till, to sequester C in soils, using Paiquerê farm (located in Campos Gerais region) as a successful model; iii) estimate the impact of best management practices adoption in the region croplands and globally for the next 100 years where is suitable using Century and Roth-C models. The GHG emission sources were presented as an inventory and showed that historical (1930 – 2017) GHG emissions in the region was 412.18 Tg C, in which LULUC contributes 91% (376.2±130 Tg C). Forestry sequestered 51.7 ± 23.9 Tg C in 0.6 Mha in 47 years (1.8 Tg C Mha-1 year-1) and no-till sequestered 30.4 ± 23.9 Tg C in 1.9 Mha in 32 years (0.5 Tg C Mha-1 year-1). Both models performed well, and Century was more efficient for simulate the SOC stocks, the mean residue was 10 Mg C ha−1 (13%) for n = 91. The model residue increased along with the oxides content in the soil clay fraction, suggesting that mineralogical control inclusion can reduce the model simulation bias. Century predictions showed that the system currently practiced at Paiquerê farm have the potential to mitigate 13 years of regional total emissions (330 Tg C in 100 years) or 105 years of agriculture, forestry and other land use (AFOLU) sector emissions (40 Tg in 100 years) in the region. In the same way, it has the potential to sequester 2.5±0.02 Pg C at 0-20 cm and 11.7±3 Pg C at 0-100 cm soil depth in 86 million ha globally. This is equivalent to 11% of global annual emissions from LULUC sector. In this way, our methodology can be used as a model to access the potential of conservation agriculture to sequester C and support public policies aiming to mitigate GHG emissions.

Black community attitudes towards the disabled-educational implications

This study, BLACK COMMUNITY ATTITUDES TOWARDS THE DISABLED - EDUCATIONAL IMPLICATIONS, set out to investigate the attitude of the black community towards the disabled. Literature was consulted to provide a backdrop against which the empirical surveys could be measured. The study used systematic observation interviews for data collection. Appointments were made to conduct the interviews. The study obtained responses from 70 interviewees from four African cultures namely the Vend~ Pedi, Tsonga and Ndebele, who were divided into seven groups, ten per group, i.e. parents of the disabled, the disabled themselves, siblings of the disabled, parents of non-disabled, youth, old people and professional workers. Findings indicated that disabilities such as cerebral palsy, orthopaedic disabilities, blindness and deafness are undesirable and constitute problems in the life of a disabled, their families in general perspective and the community at large. Disabilities constituted different attitudes and were identified as varying from total rejection to acceptance which are brought about by the components of the TRIPARTITE MODEL I A B C MODEL being cognitive, affective and behavioural. Education of the whole black community concerning the issue of disabled people seems to be the only answer to bring about a change of attitude towards the disabled. Therefore a number of recommendations were included to serve as guidance and counselling so that myth and witchcraft ideologies and negative attitudes can be uprooted. The researcher hopes that the effort that went into this· study will be beneficial in the process of educating the black community about the disabled with the ultimate goal of changing existing negative attitudes towards the disabled into positive attitudes of acceptance and support. / Psychology of Education / D Ed.
