Efeito de tratamentos químicos, revestimentos comestíveis e irradiação na conservação de mamões minimamente processados / Effect of chemical treatments, edible coatings, and irradiation on fresh-cut papaya conservation

Avaliou-se o efeito de tratamentos químicos, revestimentos comestíveis e irradiação na conservação de mamões processados minimamente. Após seleção, lavagem e sanitização, os mamões foram descascados e cortados em meias rodelas, as quais foram submetidas a diferentes tratamentos, embaladas e armazenadas a 5±1°C e 90±2%UR. Os mamões PM foram avaliados após 1, 3, 6, 9, 12 e 15 dias. As análises microbiológicas foram fundamentadas na quantificação de coliformes totais e termotolerantes, bactérias psicrotróficas, bolores e leveduras, assim como na verificação da presença de Salmonella. As avaliações físico-químicas basearam-se na determinação da concentração de CO2 no interior das embalagens, perda de massa, cor, firmeza, sólidos solúv eis, acidez titulável, ratio e pH. As características sensoriais aparência, aroma, sabor e textura foram avaliad as por meio de testes com escala hedônica. No primeiro experimento os tratamentos foram: controle, aldeído cinâmico a 0,1%, cloreto de cálcio a 0,75% e combinação de aldeído cinâmico a 0,1% com cloreto de cálcio a 0,75%. O uso de tratamentos químicos em mamões PM resultou em: maior controle de coliformes totais para os mamões PM tratados com aldeído cinâmico e com a combinação de aldeído cinâmico e cloreto de cálcio; menor concentração de CO2 e maior manutenção da firmeza para mamões PM tratados com a combinação de aldeído cinâmico e cloreto de cálcio; e maior concentração de CO2 para os mamões PM tratados apenas com aldeído cinâmico. A imersão nos tratamentos químicos resultou em maior descoloração da polpa dos mamões PM e redução do teor de sólidos solúveis ao longo do armazenamento. No segundo experimento foram utilizados os tr atamentos: controle, amido de arroz a 3%, alginato de sódio a 0,5% e carboximetilcelulose a 0,25%. O uso desses três tipos de revestimento resultou em maior controle de coliformes totais do que o observado no controle. Mamões PM revestidos com amido de arroz e carboximetilcelulose apresentaram redução e aumento da concentração de CO2, respectivamente. Os mamões PM revestidos apresentaram menores teores de sólidos solúveis e seus valores de pH se tornaram menores após 9 dias de armazenamento refrigerado. O r evestimento com carboximetilcelulose proporcionou maior firmeza da polpa no 15° dia. No terceiro experimento foram utilizados os tratamentos: controle, radiação nas doses de 2kGy e 4kGy. O uso de radiação gama em mamões PM resultou em: maior controle de coliformes totais; menor concentração de CO2 nos mamões PM tratados com 2kGy; maior concentração de CO2 e maior descoloração da polpa nos mamões PM tratados com 4kGy; redução da firmeza nos mamões PM tratados com 2kGy e 4kGy; ligeira redução do teor de sólidos solúveis e pequenas variações da acidez titulável em todos os tratamentos. As características sensoriais dos mamões PM tratados com radiação gama não diferiram significativamente do controle durante os 15 dias de armazenamento / The effect of chemical treatments, edible coatings , and irradiation on fresh-cut papaya conservation was evaluated. After selection, washing, and sanitation the papayas were peeled and cut into half slices, which were submitted to different treatments, packed, and stored at 5±1°C and 90±2%RU. The fresh-cut papayas were evaluated after 1, 3, 6, 9, 12, and 15 days. The microbiological analyses were based on the count of total coliform, thermotolerant and psychrotrophic bacteria, molds and yeasts, as well as on the presence of Salmonella. The physicochemical evaluations were based on the determination of CO2 concentration inside the package, weight loss, color, firmness, soluble solids, titratable acidity, ratio, and pH. The sensory characteristics appearance, aroma, flavor, and texture were evaluated using a hedonic scale. In the first experiment, the treatments tested we re: control, cinnamic aldehyde 0.1%, calcium chloride 0.75%, and the combination of cinnamic aldehyde 0.1% and calcium chloride 0.75%. Using chemical treatments to preserve fresh cut papaya resulted in: higher control of total coliforms in fresh-cut papayas treated with cinnamic aldehyde and with the combination of cinnamic aldehyde and calcium chloride; lower CO2 concentration and increased maintenance of firmness in freshcut papayas treated with the combination of cinnamic aldehyde and calcium chloride; and increased in the CO2 concentration in fresh-cut papayas treated only with cinnamic aldehyde. Immersion in chemical treatments caused higher pulp discoloration and reduction in solu ble solids during storage. In the second experiment, the treatments tested were: control, rice starch 3%, sodium alginate 0.5%, and carboxymethylcellulose 0.25%. The use of these three coatings resulted in higher control of total coliforms compared to the control treatment. The fresh-cut papayas coated with rice starch and carboxymethylcellulose presented reduction and increase in the CO2 concentration, respectively. Coated fresh-cut papayas presented lower soluble solids and pH values were lower after 9 days of cold storage. Carboxymethylcellulose coating increased firmness maintenance at day 15. In the third experiment, the following treatments were used: control, radiation at the doses of 2kGy and 4kGy. The use of Gamma radiation in fresh-cut papaya resulted in: higher control of total coliforms; lower CO2 concentration in fresh-cut treated with 2kGy; increased CO2 concentration and increased pulp discoloration in fresh-cut papayas treated with 4kGy; reduction in firmness in fresh-cut papayas treated with 2kGy and 4kGy; slight reduction in soluble solids and small changes in titratable acidity in all treatments. The sensory characteristics of fresh-cut papayas treated with gamma radiation did not significantly differ from the control during the 15 days of storage

Indução de aneurisma em aorta abdominal de porcos: um modelo endovascular / Endovascular model of abdominal aortic aneurysm induction in swine

A ruptura do aneurisma da aorta abdominal está entre as principais causas de óbito. A alta morbi-mortalidade associada à ruptura e tratamento dos aneurismas representa um grande desafio aos médicos e um alto risco aos pacientes. Apesar dos modelos experimentais serem úteis para compreendermos, treinarmos, testar novos métodos diagnósticos e terapêuticos para esta doença, os modelos existentes até o momento ainda não são os ideais. Nos modelos existentes, os animais são muito pequenos e não representam a doença nos humanos, ou o procedimento envolve laparotomia, ou o comportamento do aneurisma criado não é semelhante ao de um aneurisma verdadeiro. Desenvolvemos, a partir de uma abordagem minimamente invasiva, um método eficiente de induzirmos a formação de um aneurisma verdadeiro na aorta abdominal infrarrenal de porcos Large White. Os animais foram submetidos a indução química a partir de uma aplicação por via endovascular de cloreto de cálcio a 25% ou elastase pancreática suína. Os animais controles foram submetidos a tratamento com soro fisiológico (NaCl 0,9%). Todos os animais foram submetidos à mesma técnica operatória, sob anestesia geral. Os animais foram acompanhados com exames ultrassonográficos com Doppler semanalmente, e as aortas colhidas para testes biomecânicos e análise histológica após 4 semanas. Apesar das aortas tratadas com elastase apresentarem apenas dilatação, estudos de imagens, histológicos e biomecânicos mostraram que as aortas tratadas com cloreto de cálcio evoluíram para aneurismas verdadeiros, com comportamento biomecânico semelhante ao dos aneurismas de humanos. Estes resultados/achados indicam que a abordagem endovascular para a indução de aneurisma é factível e não ocasiona uma fibrose retroperitoneal / Abdominal aortic aneurysms rupture are among the main causes of death. The high morbidity and mortality associated with aneurysm rupture and repair represents a challenge for surgeons and high risk for patients. Although experimental models are useful to understand, train, and develop new treatment and diagnostic methods for this disease, animal models developed to date are far from ideal. Animals are either too small and do not represent the pathology of humans, or the procedures employ laparotomy, or the aortic behavior does not resemble that of a true aneurysm. We developed a novel, less invasive and effective method to induce true aortic aneurysms in Large White pigs. Animals were submitted to an endovascular chemical induction using either calcium chloride (25%) or swine pancreatic elastase. Controls were exposed to saline solution. All animals were operated on using the same surgical technique under general anesthesia. They were followed weekly with ultrasound examinations and at 4 weeks the aorta was harvested. Although elastase induced only arterial dilation, imaging, histological, and biomechanical studies of the aorta revealed the formation of true aneurysms in animals exposed to calcium chloride. Aneurysms in the latter group had biomechanical failure properties similar to those of human aneurysms. These findings indicate that the endovascular approach is viable and does not cause retroperitoneal fibrosis

O estudo do comportamento dos biofilmes é fundamental para melhorar as formas de controle, especialmente durante infecções, tais como as doenças periodontais. No primeiro capítulo, foram avaliados os efeitos da aplicação tópica e frequente do digluconato de clorexidina (CLX) em diferentes concentrações na periodontite induzida por ligadura nos primeiros molares de ratos. As ligaduras receberam 10 µl de soluções de CLX à 0,2%, 2%, 10%, 20% ou diluente, de quatro em quatro dias, em um total de quatro aplicações. Após eutanásia, a quantidade de células bacterianas no biofilme formado sobre a ligadura foi estimada por cultura e por PCR quantitativo. A reabsorção óssea foi mensurada em altura e área por fotografia digital e em volume por microtomógrafo. Depois de quatro dias a partir da última aplicação da CLX, as reduções bacterianas mantidas pelos tratamentos com CLX atingiram até 10-6. O grupo que recebeu CLX a 20% teve, em média, logs bacterianos 3,3× menor (p<0.01, Kruskal-Wallis). Não houve diferença estatística entre os grupos em relação à reabsorção óssea por ambos os métodos testados (p>0.05, Kruskal-Wallis), embora 55% dos sítios apresentaram menor reabsorção óssea. No segundo capítulo, foi avaliada a influência de diversas substâncias na formação de biofilme por Prevotella intermedia. Os biofilmes foram formados em placas de 48 poços contendo tratamento de superfície prévio com DNA purificado, hemina, CaCO3, Ca(OH)2, CaCl2, soro, albumina, dextrana, metionina, glicose, glutamina, KCl, complexo vitamínico, cistina ou mucina. O biofilme formado foi corado e quantificado por espectrofotometria. A arquitetura do biofilme foi visualizada por microscopia confocal de fluorescência por varredura laser. O tratamento da superfície com CaCl2 a 1 mg/cm2 permitiu a formação do biofilme em quantidade de 0,3 OD490nm (p<0.01, ANOVA Dunnet), sendo esse valor 10× superior quando a superfície foi tratada com 2,5 mg/cm2 (p<0.01, ANOVA Dunnet). As demais substâncias tiveram pouco ou nenhum impacto sobre a formação do biofilme. A visualização por microscopia confocal revelou uma comunidade estruturada e com vitalidade em toda sua espessura. Conclusões: os dados indicam que a CLX concentrada diminui a carga bacteriana na região da periodontite induzida, que reflete em menor reabsorção óssea apenas em parte das amostras. O pré-revestimento da superfície de crescimento com cálcio promove a formação de biofilme por P. intermedia / Abstract: Biofilms are biological communities with a high degree of organization, in which micro-organisms form structured, coordinated and functional communities. These biological communities are embedded in a self-created extracellular matrix. Biofilm is also associated with a high level of antimicrobial tolerance of the associated organisms. Understanding biofilm behavior is crucial to develop ways for its control during infections, such as periodontal disease. In the first chapter, topical and frequent application of various concentrations of chlorhexidine digluconate (CLX) where evaluated. Periodontitis were induced by ligature on first molars. Then, ligatures were treated with 10 µl of chlorhexidine solutions at 0.2%, 2%, 10%, 20% or diluent, every four days in a total of four applications periods. After euthanasia, bacterial loads on ligatures were estimated by both culture and qPCR. Bone resorption height and area were measured by digital photography and its volume by microtomography. Treated sites had bacterial reductions up to 10-6 cells. Treatment with 20% CLX showed mean of 3.3× lower bacterial levels (p<0.01, Kruskal-Wallis). There was no statistical difference between groups regarding bone resorption (p>0.05, Kruskal-Wallis), although 55% of the treated sites had some lower bone resorption. In the second chapter, substances that may enhance biofilm formation by Prevotella intermedia were investigated. Wells of 48-well plates were coated with DNA, hemin, CaCO3, Ca(OH)2, CaCl2, serum, albumin, dextran, methionine, glucose, glutamine, KCl, vitamin complex, cystine or mucin. Biofilms were grown for 24 h, washed, stained and quantified by spectrophotometry. Biofilm architecture and its viability were visualized by Confocal Laser Scanning Microscopy. Surfaces treated with 1 mg/cm2 of CaCl2 enhanced biofilm amount by 0.3 OD490nm (p<0.01, ANOVA Dunnet), while 2.5 mg/cm2 yielded 10-fold more biofilm mass (p<0.01, ANOVA Dunnet). Other substances had modest or no impact in biofilm mass. Confocal microscopy images showed structured and alive biofilms with no dead areas. Conclusions: concentrated CLX reduces bacterial load, which reflects in lower bone resorption in few sites. Micro-organismos bioprotetores em salames com baixo teor de sal e seus efeitos sobre a qualidade global e sensorial / Bacteriocin-producingmicroorganismsin low-sodium salami and their effects on global and sensory quality

As mudanças dos hábitos de consumo do consumidor, força o desenvolvimento de produtos industrializados mais saudáveis. Assim, pesquisadores em ciência e tecnologia de produtos cárneos, têm focado parte de suas pesquisas no desenvolvimento de produtos com níveis reduzidos de sódio. O presente estudo teve como objetivo buscar por micro-organismos bioprotetores e proteolíticos para o uso em salames com baixo teor de cloreto de sódio (NaCl). O primeiro capitulo consiste em uma revisão de literatura, sendo o segundo o desenvolver salames italianos com 1,0% NaCl com a adição ou não de cloreto de potássio (KCl) e cloreto de cálcio (CaCl2) com características sensoriais semelhantes aos descritos no salames comerciais com 2,5% de NaCl adicionado. Os resultados mostraram diferenças de pH e Aw, devido à redução de NaCl. As misturas substitutas de sal (KCl/CaCl2) nos salames não afetaram tecnologicamente a elaboração de salames. Para os consumidores, esta substituição não acarretou diferenças significativas (p> 0,05) na aparência (cor), porém observou-se diferenças nos demais atributos. Com relação ao teor de sal, os consumidores consideraram os tratamentos com baixo teor de NaCl adicionados dos substitutos KCl e CaCl2 como ideais. No terceiro capitulo, o objetivo foi analisar os aldeídos produzidos pela lipólise e proteólise assim como a aceitação sensorial de salames tipo Italiano com reduzido conteúdo de sódio. Das seis amostras elaboradas, todas foram monitoradas quanto ao pH e atividade de água e após a maturação do salame, foi realizada analise do perfil de voláteis e comparado com a sensorial. Os resultados mostraram a existência de uma relação entre a concentração de aldeídos e aceitação sensorial. Com a finalidade de elucidar a proteólise dos micro-organismos escolhidos, foi realizado no quarto capitulo um estudo com modelos Beaker Sausage (BS) para simular a produção de salame com baixo conteúdo de sódio para avaliar a produção de aminoácidos e peptídeos de culturas starters bacteriocigenicas. O BS contendo Enterococcus mundtii CRL35 apresentou a maior variedade de peptídeos pequenos, seguido pela BS contendo L. curvatus CRL1862. Embora o modelo de carne inoculadas com Lactobacillus plantarum CRL681 produziu o maior número de peptídeos derivados de proteínas sarcoplasmáticas. A pequena quantidade considerável de péptido produzido no BS inoculado permitiu inferir que os conteúdos mais baixos de sódio não afetam negativamente a ação das peptidases microbianas. O objetivo do último capitulo, foi avaliar o efeito das alegações de saúde sobre as aceitação dos salames tipo Italiano com reduzido conteúdo de sódio. Neste estudo, foi avaliada a expectativa gerada pelas alegações de saúde e os resultados mostraram que as alegações tiveram pouco efeito na aceitação dos consumidores decorrentes em valores de aceitação baixo para o protótipo de laboratório. Assim, pode-se concluir que a produção de salames tipo Italiano com teores reduzidos de NaCl, adicionados de CaCl2+ KCl é viável com a pouca adição destes substitutos e o uso de culturas starters bacteriocigênicas, que também são altamente proteolíticas de ser melhor estudado a fim de adequar a proteólise ideal para a formação dos compostos desejáveis no salame com baixo teor de sódio. / Changes in consumers\' eating habits and lifestyle have required the development of healthier processed products. Thus, researchers in science and meat products technology has focused their studies on the development of products with reduced sodium contents. This study investigated the use of bioprotectants and proteolytic microorganisms for low-sodium NaCl in salami. The chapter 1 consists in a literature review. In the second chapter, the objective was to develop Italian salami with 1.0% NaCl with or without the addition of potassium chloride (KCl) and calcium chloride (CaCl2) with sensory characteristics similar to those described in the commercial salami with 2.5% NaCl added. The results showed differences in pH and water activity (Aw) due to the reduction of NaCl. Salt substitutes chloride (KCl / CaCl2) in salamis did not technologically affectsalami preparation. For consumers, this replacement did not cause significant differences (p> 0.05) in appearance (color), but there were differences in other attributes. Regarding the salt content, consumers considered treatment with low NaCl content and added with KCl and CaCl2 as ideal substitutes. In the third chapter, the goal was to analyze the aldehydes produced by lipolysis and proteolysis as well as sensory acceptance of the Italian salami type with reduced sodium content. Allsix samples preparedwere monitored for pH and Aw and after salami maturation, thevolatile profile analysis was performed and compared with the sensory analysis. The results show the existence of a relationship between the concentration of aldehydes and sensory acceptance. In order to elucidate proteolysis of selected microorganisms, in the fourth chapter a study of models Beaker Sausage (BS) was conducted to simulate production of low-sodium salami to evaluate the production of amino acids and peptides of bacteriocigenicstarter cultures. The BS containing Enterococcus mundtii CRL35 showed the greatest variety of small peptides, followed by BS containing Lactobacilluscurvatus CRL1862. However, the meat model inoculated with Lactobacillus plantarum CRL681 produced the largest number of sarcoplasmic proteins derived from peptides. A considerable number of peptides produced in the BS inoculated allowed to infer that lower sodium content did not affect the action of microbial peptidases. The purpose of the last chapter was to evaluate the effect of health claims on the acceptance of Italian-type salami with reduced sodium content. In this study, the expectation generated by health claims wereanalyzed and the results showed that health claims had little effect on the increase of consumer acceptance for the prototype developed in the laboratory. Thus, it can be concluded that the production of Italian-type salami with reduced levels of NaCl, KCl + CaCl2 added is feasible but low addition of these substitutesmust be considered. Moreover, the use of bacteriocigenicstarter cultures, highly proteolytic to achieve optimal proteolysis in the formation of the desired compounds, should adjusted to low-sodiumsalami.

This thesis deals with optimization of MRI measuring of phantoms containing the chicken embryo. The theoretical part of thesis is dedicated to the MRI fundamentals and description of pulse sequences. In the next part the options of keeping and monitoring the temperature are being discussed. The thesis also deals with design and realization of phantoms which are used for measuring. A browser of images captured by MRI was programmed in Matlab environment. The program allows users to look through the images with different settings of pulses sequences or kind of phantom. The final part of thesis describes the anatomical structures of chicken embryo and discussion of collected results.

Nucleation, milk and membranes as modifications to enhance biological phosphorus removal in activated sludge

Enhanced biological phosphorus removal (EBPR) was researched from the performance of a modified University of Cape Town (UCT), anaerobic-anoxic/nitrifying-aerobic process. The work focussed on high P influent where milk was compared to carbohydrates as exogenous added carbon and typical settled sewage. The results confirmed that at equal COD load in the influent (minimum COD:P (250:5) ratio for EBPR), milk always provided sufficient soluble substrate than the carbohydrate mix, but also improved the EBPR performance. The laboratory scale treated 10L/day where 2 parallel treatment trains for milk and an equivalent carbohydrate mix as supplement to compare and study the P sequestration from hypothesised P ligands in milk and easily assimilable carbon (AOM) after fermentation for biological P uptake. The aerobic bioreactors used submerged flat sheet membranes (AeMBR) to improve the effluent quality and reduce the suspended solid residues. The results suggested extra benefits from adding calcium chloride (CaCl2) (200 ml at 250 mM/day or 200 mg/L treated) to form P complexes both in the anaerobic and aerobic zones (100 ml CaCl2 250mM/zone/day). To complete P removal a calcium phosphate (CaPO4) further treatment stage (post membrane final effluent (F.E.)) was added for nucleation. The combination of, A2O-N, exogenous carbon and calcium addition improved the performance of the EBPR, and enabled the laboratory units to achieve less than the 1 mg/L P required by the EU Directive. The process was tested at higher than normal P loads (maximum 100 mg/L) (domestic wastewater influent 15 mg/L). Experiments with influent P load ≤ 50mg/L, with 1% milk as AOM were compared to the carbohydrate mix and could remove soluble P to less than 1mg/L above 97% and less than 2 mg/L more than 99% of the in the time respectively. With an influent P load of 60mg/L (maximum 100 mg/L), the soluble P in the F.E. with milk was below 5 mg/L and below 8 mg/L with carbohydrates mix. The results showed that most of the phosphorus was retained by the sludge during the anoxic-aerobic phases. The remaining phosphate in the F.E. was able to pass through AeMBR pore size (0.4 μm) and needed to be chelated by the nucleation process. The results indicated this A2O-N modifications achieved stable nutrient removal and also offered the potential for more sustainable phosphorus recovery. The EBPR without AOM was 25% less efficient compared to milk and never achieved the E.U standard of 1mg/L in final effluent. The flat sheet membrane always achieved a NTU final effluent below 1 and the TOC always greater than 90% removal or less than the EU 125 standard regardless of the feeding COD/P ratio.

O agregado de trióxido mineral (MTA) tem-se demonstrado aplicável em diversas situações que exigem reparação dos tecidos dentais e periapicais. Neste sentido, um novo cimento de aluminato de cálcio e aditivos (CAC+) foi desenvolvido para superar algumas características negativas do MTA, mantendo, no entanto, suas propriedades satisfatórias. O objetivo deste estudo foi avaliar a progressão de culturas de células osteogênicas e de células indiferenciadas da polpa dental (linhagem OD-21) expostas ao CAC+, utilizando MTA como controle, ou a preparações alternativas do CAC+, com maior conteúdo de cloreto de cálcio (CaCl2) e/ou com substituição do óxido de zinco por óxido de bismuto como radiopacificador. Para isso, células osteogênicas derivadas de calvárias de ratos ou células da linhagem OD-21 foram crescidas sobre Thermanox® por 24 h e expostas, por períodos de até 14 dias, a amostras dos diferentes materiais, posicionadas sobre Transwell®. Em células osteogênicas, apesar de a proximidade com as amostras de CAC+ e MTA inibir o crescimento celular, nas áreas periféricas das lamínulas, foram observados proliferação, viabilidade celular e expressão de marcadores-chave da diferenciação osteoblástica, que precederam a mineralização da matriz extracelular. Culturas expostas ao CAC+, comparativamente ao MTA, exibiram aumentos na viabilidade celular, atividade de fosfatase alcalina e na expressão de marcadores de diferenciação, o que não se repetiu para células OD-21. Além disso, demonstrou-se que os efeitos destes cimentos sobre a osteogênese in vitro variaram de acordo com o período de exposição das culturas, sendo mais favoráveis durante sua fase proliferativa. Entre as preparações de CAC+, verificou-se que o aumento no teor de CaCl2 promoveu maior disponibilização de Ca2+ no meio de cultura, o que correspondeu a maior diferenciação celular e formação de matriz mineralizada em culturas de células osteogênicas e OD-21, e à redução de efeitos negativos da adição de óxido de bismuto sobre osteoblastos. Conclui-se que o CAC+ favorece o desenvolvimento do fenótipo osteogênico, atuando principalmente em células em estágios iniciais de diferenciação e que a adição de CaCl2, independentemente do agente radiopacificador, potencializa os efeitos benéficos sobre células osteogênicas e favorece o desenvolvimento e diferenciação de células indiferenciadas derivadas do tecido pulpar, podendo ser considerado como material alternativo ao MTA. / Mineral trioxide aggregate (MTA) has been successfully applied in endodontic procedures in which dental and periapical tissue repair are required. However, some drawbacks of MTA as high cost, physicochemical properties and difficulty in handling have limited its use. In this context, a novel calcium aluminate cement plus additives (CAC+) has been developed to overcome some negative features of MTA. The aim of this study was to evaluate the progression of either osteogenic cell cultures or undifferentiated dental pulp-derived ones (OD-21 cell line) exposed to CAC+ or to alternative formulations of CAC+ with a higher content of calcium chloride (CaCl2) and/or replacement of zinc oxide by bismuth oxide as radiopacifier agent. Rat calvaria-derived cells or OD-21 cells were grown on Thermanox® coverslips for 24 h and exposed to samples of CAC+ or MTA (control) placed on Transwell® for periods of up to 14 days. In osteogenic cell cultures, the proximity to MTA or CAC+ samples inhibited cell growth, whereas at distance it was observed cell proliferation, cell viability and expression of differentition markers prior to mineralization of the extracellular matrix. Comparatively to MTA, osteogenic cell cultures exposed to CAC+ exhibited higher cell viability, alkaline phosphatase activity and expression of key osteoblast markers, contrary to what was observed for OD-21 cells. Furthermore, it was demonstrated that the effects of these cements on in vitro osteogenesis varied according to the timing of exposure, with a more favorable impact during the proliferative phase of cultures. Among the diverse formulations of CAC+, it was found that the increase in the CaCl2 content promoted greater availability of Ca2+ in the culture medium, which corresponded to higher cell differentiation and mineralized matrix formation in osteoblastic cell cultures and OD-21 cells, while reducing the negative effects of bismuth oxide on osteoblasts. In conclusion, CAC+ supported the acquisition of the osteogenic cell phenotype, mostly for cells in early stages of differentiation. Additionaly, the increase in the CaCl2 content, regardless of the radiopacifier agent, potentiates the beneficial effects on osteogenic cells and promotes the growth and differentiation of OD-21 cells, rendering CAC+ a potential alternative material to replace MTA in endodontic procedures.

The aim is to develop intensive methods for producing powdered iron or titanium, by mixing melts containing chlorides with molten salt reducing Ca-CaCl2 and Na-NaCl. As a result of this work was the possibility of obtaining metallic iron powder and products containing nedovosstanovleniya titanium compound. Conducted microscopic, X-ray, mikrorentgenoskopicheskie and particle size analysis of powders obtained. To discover the causes of titanium nedovosstanovleniya that need to be addressed in the future. The developed method can improve the performance as compared with the Kroll method because the reduction process within the melt, and not only at the interface between the salt and metal phases. / Целью работы являлось разработка интенсивных способов получения порошкообразных железа или титана, при смешении расплавов содержащих их хлориды с расплавленными солевыми восстановителями Ca–CaCl2 и Na–NaCl. В результате проведенной работы была показана возможность получения порошкообразного металлического железа и продукта, содержащего недовосстановленные соединения титана. Проведены микроскопические, рентгенофазовые, микрорентгеноскопические и гранулометрические анализы полученных порошков. Вскрыты причины недовосстановления титана, которые необходимо устранить в будущем. Разрабатываемый способ позволяет увеличить производительность по сравнению с методом Кролла благодаря тому, что процесс восстановления протекает в объеме расплава, а не только на границе раздела солевой и металлической фаз.

Evaluation of Dust Suppressants for Gravel Roads : Methods Development and Efficiency Studies

Approximately 75 percent (300 000 km) of the total Swedish road network and 20 percent(20 000 km) of the national road network consists of gravel roads. One of the most significantproblems associated with gravel roads is traffic-generated dust emission, which contributes tothe deterioration of the road surface and acts as a major source of particulate matter releasedinto the atmosphere, thereby involving public economics, road safety, human health, andenvironmental quality. In order to bind the fine granular material, which is prone to rise into theair, dust suppressants are applied on roads on a yearly basis. Methods for evaluating the efficiency of dust suppressants will facilitate in the selection of themost appropriate product and its optimal application rate. For example, methods forsupervision of residual dust suppressant concentration are valuable tools for estimatinglongevity and optimal application rates, and, consequently, effectiveness of different products. Application of the proper dust suppressant to a gravel road ensures road safety and ridingcomfort as well as creating a cleaner and healthier environment for residents in buildingsadjacent to the road. It also reduces the need and cost for vehicle repair, road maintenanceactivities, and aggregate supplementation. Both field-based and laboratory research were performed to evaluate the efficiency of varioussuppressants and the influence such factors as product concentration, leaching, and fine materialcontent have on the efficiency of different products. Within the field-based research, a newlydeveloped mobile methodology was used to measure dust emission on numerous test sectionstreated with various dust suppressants. In general, all dust suppressants tested, except apolysaccharide (sugar) and products, which form a brittle surface crust, i.e. lignosulphonate andbitumen emulsion, showed acceptable dust reduction. Test sections treated with a magnesium- or calcium chloride solution were the most effectivelydust suppressed. The application of solutions instead of a solid salts achieves a more uniformproduct distribution and, therefore, probably a more efficient performance. By applying acalcium- or magnesium chloride solution instead of traditionally used solids, the cost for annualdust control, as well as the environmental impact from the release of these chemicals in theenvironment, can be reduced by 50 percent. A significant problem when using dust suppressants is their tendency to leach during rainfalldue to their soluble properties. Residual chloride could be detected in the gravel wearing courseover a longer period of time than lignosulphonate and, therefore, showed more effective longtermperformance. Optimal percentages of fine material for minimal lignosulphonate andchloride leaching were found to be 15 percent by weight and 10-16 percent by weight,respectively. Ions of calcium chloride seemed to initiate flocculation of clay particles, therebypreventing them from leaching. Still, the fine material in gravel wearing courses has to be replenished regularly as indicated by studies of the longevity of fine material. Loss up to80 percent was found after two years. Toxicity tests show that dust suppressant application for dust control purposes, at traditionallyused application rates, does not constitute a threat to sensitive aquatic life. Tests on subsoilwater samples indicated elevated chloride levels, which possibly could cause corrosion to pipes,but not high enough to flavour drinking water. / QC20100616
