As soluções avaliadas foram: G1: Hipoclorito de sódio (NaOCl) a 2,5%; G2: NaOCl a 2,5% + cetramida (CTR) a 0,2%; G3: Clorexidina (CLX) a 2%; G4: CLX a 2% + CTR a 0,2%; G5: CTR a 0,2%; G6: QMiX. Como controle negativo foi utilizada solução salina. O estudo foi dividido em dois capítulos. No capítulo 1, foi realizada a avaliação da atividade antibacteriana sobre os micro-organismos em fase planctônica e biofilme. Para o contato direto com E. faecalis em fase planctônica, 50 μl da suspensão bacteriana foram colocados em tubos contendo 1,45 mL de cada solução irrigadora ou associações e, após os períodos de 1 e 3 minutos foram utilizadas soluções neutralizantes da ação antimicrobiana dos irrigantes. Em seguida, foi realizada diluição decimal seriada e plaqueamento em meio Tryptic Soy agar (TSa). Para a indução dos biofilmes foram utilizados blocos de dentina bovina como substrato no período de 14 dias. Os biofilmes foram colocados em contato com as soluções avaliadas, e transferidos para tubos de ensaio contendo soluções neutralizantes. Após agitação por 60 segundos para a suspensão das células remanescentes, foi realizada diluição decimal seriada e plaqueamento em TSa em triplicata. A incubação foi realizada em microaerofilia a 37°C por 24 horas e a análise efetuada por meio da contagem de UFC mL-1. Os resultados foram submetidos aos testes ANOVA e Tukey, com nível de significância de 5%. Todas as soluções irrigadoras e associações avaliadas eliminaram completamente os micro-organismos em fase planctônica. Sobre biofilme, somente NaOCl e NaOCl+CTR apresentaram 100% de eliminação bacteriana... / The aim of this study was to evaluate the action of conventional endodontic solutions such as sodium hypochlorite and chlorhexidine associated with cetrimide and QMiX, a commercial product composed by chlorhexidine, EDTA and cetrimide, on biofilm and planktonic phase of Enterococcus faecalis (ATCC 29212). The evaluated solutions were: G1: 2.5% Sodium Hypochlorite (NaOCl); G2: 2.5% NaOCl + 0.2% cetrimide (CTR); G3: 2% Chlorhexidine (CLX) 2%; G4: 2% CLX + 0.2% CTR; G5: 0.2% CTR; G6: QMiX. Saline solution was used as negative control. The study was divided into two chapters. In Chapter 1, the antibacterial activity of the solutions on microorganisms in planktonic phase and biofilm was evaluated. For direct contact with E. faecalis in planktonic phase, 50 μL of bacterial suspension were placed in tubes containing 1.45 mL of each irrigation solution or associations and after 1 and 3 minutes, neutralizing solutions were used to stop of the antimicrobial action of endodontic solutions. Decimal serial dilutions were performed and the suspensions were plated in Tryptic Soy agar medium (TSa). Dentin bovine blocks were used as substrate for induction of biofilms for 14 days. The biofilms were placed in contact with the solutions evaluated and transferred to test tubes containing neutralizing solution. After agitation for 60 seconds to suspend the remaining cells, decimal serial dilutions were performed and the suspensions were cultivated in triplicate on TSA plates. The incubation was held in microaerophilic at 37 ° C for 24 hours and the analysis was performed by counting CFU mL-1. The results were submitted to ANOVA and Tukey test, with significance level of 5%. All irrigating solutions and associations evaluated completely eradicated the microorganisms in planktonic phase. Influência do preparo biomecânico na microdureza e selamento marginal do MTA e do polímero derivado da mamona em perfurações radiculares simuladas /

Orientador: Carlos Henrique Ribeiro Camargo / Banca: Márcia Carneiro Valera / Banca: Eloi Dezan Júnior / Resumo: Neste estudo foram utilizados 96 dentes incisivos laterais bovinos que foram padronizados por meio de perfurações circulares com 2mm de diâmetro na face vestibular. As raízes foram divididas em 4 grupos de acordo com o material utilizado: G1-mineral trióxido agregado (MTA) sem proteção, G2- MTA protegido com cianoacrilato (SB), G3- MTA protegido com cimento de ionômero de vidro, G4- cimento derivado do polímero da mamona (CPM) sem proteção. Em seguida os canais foram preparados pela técnica seriada. As raízes foram seccionadas no sentido longitudinal. Catorze hemisecções foram utilizadas para análise de microinfiltração e 10 para análise de microdureza, para cada grupo. Para a análise de microinfiltração, as hemisecções foram impermeabilizadas e colocadas em solução tamponada de Rodamina 2% por 24 horas. No teste de microdureza as hemisecções foram incluídas em resina acrílica e desgastadas na face vestibular. Os dados da microinfiltração e microdureza foram avaliados através da análise estatística em nível de significância de 5%. Pôde-se observar que apesar do CPM ter o menor índice de infiltração (0.652 mm), observando-se as médias correspondentes (1.143 mm -MTA, 1.302 mm -SB, 1.029 mm - CIV) estatisticamente não houve diferença significante entre os grupos. No teste de microdureza pôde-se observar que todos os grupos com MTA (94,69HV -MTA, 92,25 HV - SB, 97,68 HV -CIV) demonstraram maior dureza em relação ao grupo do CPM (8,477 HV) com diferenças estatisticamente significante. Pode-se concluir que não houve injúria dos materiais quando submetidos aos agentes químicos e mecânicos. A realização das proteções do cimento de MTA não proporcionou melhoria nos resultados de microdureza. Todos os materiais mantiveram médias baixas de infiltração demonstrando serem eficientes como materiais seladores. O CPM se mostrou um material com boas propriedades seladoras, porém apresentou baixa dureza. / Abstract: In this study 96 bovine lateral incisor teeth were used that were standardized through perforations with 2mm of diameter in the buccal surface. The roots were divided into 4 groups according to the used material: G1 - mineral trioxide aggregate (MTA) without sealing, G2 - MTA sealed with cianoacrilate (Super Bonder, Henkel, São Paulo, Brazil) - SB, G3 - MTA sealed with glass ionomer cement (Vidrion R, S.S.White, Rio de Janeiro, Brazil) - CIV, G4 - castor oil bean cement (CPM) without sealing. Afterwards the roots were prepared by stepback technique. The roots were split up in the longitudinal direction. Fourteen roots were used in microleakage analysis and ten in microhardness test of each group. For the microleakage the specimens were waterproof and put in buffer solution of 2% Rodhamina by 24 hours. In the microhardness test the specimens were included in acrylic resin and consumed in the buccal direction. The data of the microleakage and microhardness tests were appraised through the statistical analysis (level of significance 5%). It was observed that in spite of CPM having the smallest infiltration index (0.652 mm), was observed the corresponding averages (1.143 mm - MTA, 1.302 mm - SB, 1.029 mm - CIV) however there was no statistically significant difference among the groups. In the microhardness test the difference was analyzed among the materials. And all of the groups with MTA (94,69HV - MTA, 92,25 HV - SB, 97,68 HV - CIV) demonstrated higher hardness comparing to the CPM group (8,477 HV). It was concluded that there was no harmful of the materials when submitted to the chemical and mechanical agents. The performance of the sealing of the MTA cement did not provide improvement in the microhardness results. All the materials maintained low averages of infiltration. All materials demonstrated to be efficient and CPM was shown as a material with good sealing properties on the other hand showed low hardness. / Mestre

O selamento foi testado por meio de suspensão de Staphylococcus aureus e saliva artificial. Houve passagem do microrganismo ao longo dos canais radiculares, em todas as obturações, independente da técnica utilizada. Os resultados revelaram um percentual de 85,71%, 100% e 68,42% para a técnica condensação lateral, sistema Ultrafil e obturadores Thermafil, respectivamente. Abstract : This study has evaluated in vitro the sealing in the cervical-apical direction of filled root canals by the lateral condensation method of gutta-percha points, the Ultrafil system, and the Thermafil "obturators". With that end, 68 teeth had their crowns sectioned and were prepared by the step back technique. Next, were gathered 5 experimental groups, 2 of these with 4 elements each, (positive and negative control group) and the remaining 3 made up by 20 teeth each, filled by the lateral condensation method of gutta-percha points, the Ultrafil system, and the Thermafil "obturators". The root canals of the teeth belonging to the positive control group remained empty, whereas those that were part of the negative control one were filled by the lateral condensation technique. In all of the specimens, the sealer applied was the canal Fill. Upon conclusion of the filling stage, the cavities of access, except for those belonging to the teeth pertaining to the positive control group, were sealed with Lumicon and were exposed to 37° C, in 100 % humidity, for 48 hours. After this period, the sealing was removed, except in the teeth belong to the negative control group. The root surface of all the teeth, except for the final millimeter, was made impermeable. The sealing was tested through the Staphylococcus aureus suspension and artificial saliva, which, as they were placed in the internal part of the device set up for the accomplishment of the experiment, got in touch with the coronary portion of the filling. The entrance of the microorganism in the apical region was detected by the color alteration of the selective means, in which the apices were emerged and that in the presence of Staphylococcus aureus, changed from pink into yellow. All of the devices (test bodies), were incubated at 37° and observed daily during the 90 days with the purpose of detecting the presence of microorganism. Infiltration occurred in all of the elements of the positive control group. However, in the specimens that made up the negative control group, the central color remained unaltered during the entire experiment period. There was passage of microorganism along the root canal, in all of the fillings, regardless of the technique employed. The results revealed a percentage of infiltration of 85,71%, 100%, and 68,42% for the lateral condensation technique, the Ultrafil system, and the Thermafil "obturators", respectively.
