Data-driven outbreak forecasting with a simple nonlinear growth model

Recent events have thrown the spotlight on infectious disease outbreak response. We developed a data-driven method, EpiGro, which can be applied to cumulative case reports to estimate the order of magnitude of the duration, peak and ultimate size of an ongoing outbreak. It is based on a surprisingly simple mathematical property of many epidemiological data sets, does not require knowledge or estimation of disease transmission parameters, is robust to noise and to small data sets, and runs quickly due to its mathematical simplicity. Using data from historic and ongoing epidemics, we present the model. We also provide modeling considerations that justify this approach and discuss its limitations. In the absence of other information or in conjunction with other models, EpiGro may be useful to public health responders. (C) 2016 The Authors. Published by Elsevier B.V.

3,3'-diindolylmethane improves drought tolerance of Zea mays through enhancing antioxidant activity

This figure will have to increase in order to feed the ever-growing human population. One of the major environmental constraints that impact maize production is drought. Plants use antioxidant defences to cope with drought stress. Understanding and improving these defence mechanisms will be important to improve overall drought tolerance. A previous study done by Gokul and authors in 2016 showed that 3,3’-diindolylmethane (DIM) improves both seed germination and seedling shoot growth in Brassica napus. Plants belonging to the Brassicaceae family have the metabolic machinery to synthesize glucosinolates such as DIM, which play vital roles in physiological and stress responses. These responses have not been investigated in plants such as maize, which lack the machinery to produce DIM. Therefore, this study investigated the effects of exogenously applied DIM on the physiological and biochemical responses of maize under drought stress. Physiological parameters such as relative water content, chlorophyll content and lipid peroxidation, were determined in order to understand how drought and DIM , as separate or combined treatments, affected the plants. Additionally, proline accumulation was also assessed because free proline plays a role as an osmoprotectant during stress. The accumulation of ROS, namely hydrogen peroxide, was measured using spectrophotometric assays to determine how the above treatments affect ROS accumulation in maize. As a result of changes in the ROS content in due to the treatments, it would only be natural to investigate the changes in antioxidants as well. Given that hydrogen peroxide was the ROS to be measured, we therefore investigated the antioxidant enzymatic activities responsible for hydrogen peroxide scavenging. Therefore, changes in Ascorbate peroxidase (APX) and catalase (CAT) were assessed. An improved drought response was observed in maize plants treated with DIM as these plants had better ability to maintain their water status than when no DIM was applied. This is indicated by water-deprived plants treated with DIM having a higher RWC than water-deprived plant without DIM.

[pt] Os sistemas MIMO (Multiple In - Multiple Out) são o surgimento de novos sistemas de telecomunicações, tendo o recurso para gerar maior capacidade de canal com transmissores de baixa potência, criando uma utilização mais eficiente da energia. Como o objetivo é proporcionar uma maior taxa de dados, os métodos convencionais são muito limitados, assim os sistemas MIMO geram um aumento em capacidade e confiabilidade e prometem ser concorrência para sistemas 4G, aprimorando as técnicas de multiplexação e diversidade, ajudando a aumentar a taxa de transmissão, mantendo os mesmos níveis de potência de um sistema convencional. Desta forma, o estudo e aprimoramento das técnicas de medições do canal MIMO são de extrema importância para verificar a confiabilidade da transmissão de dados por múltiplas antenas, as técnicas de modulação assim como a capacidade do canal. Neste trabalho quatro antenas planares aplicadas ao sistema MIMO são dimensionadas, simuladas e realizadas, comparando resultados experimentais com simulados. Apresenta-se também o desenvolvimento de um transmissor e um receptor simulado pelo SystemVue (Agilent) e implementado em VHDL. / [en] MIMO systems (Multiple In - Multiple Out) are the rise of new tele- communication systems. They have the characteristic of generating greater channel capacity with low power transmitters, creating a more efficient use of energy. As the purpose is to provide higher data rate, conventional methods are too limited to improve this, and MIMO systems generate an increase in capacity and reliability, also they promise to be competition for 4G systems improving multiplexing and diversity techniques which helps to increase the transmission rate while maintaining the same power levels of a conventional system. Thus, the study and improvement of techniques for MIMO channel measurements are extremely important to check the reliability of data trans- mission from multiple antennas, modulation techniques as well as the channel capacity. In this work four planar antennas applied to the MIMO system are scaled, simulated and performed by comparing experimental results with simu- lations. Also present the development of a transmitter and a receiver simulated by SystemVue (Agilent) and implemented in VHDL.

Metabolic reprogramming of T cells to optimize adoptive T cell therapy

The clinical efficacy of adoptive T cell therapies including CAR T therapy are limited by poor in vivo persistence and moderate anti-tumor efficacy. According to the literature, metabolism plays a critical role in the phenotypic state and fate of T cells during antigen-driven expansion. During different stages of a T cell life cycle, the predominant pathway used for metabolism changes. Naïve T rely on oxidative phosphorylation, but as the T cells becomes activated, their metabolic profile switches to become more reliant on glycolysis. Most T cells become terminally differentiated and become senescent once they have performed their cytotoxic function. A minority of the activated T cells gradually start to rely on oxidative phosphorylation once again and become memory T cells. Memory T cells can become either effector memory or central memory T cells. These memory T cells, specifically central memory T cells, are the key to T cells persistence during both ex vivo and in vivo expansion and following disappearance of the antigenic stimulus. Since the metabolic profile of the T cells plays a critical role in its differentiation state, we tested the hypothesis that inhibitors of intermediary metabolism could promote a metabolic profile that is more desirable for the optimal phenotype consistent with the memory phenotype that would favor persistence in spite of strong activation signals. The four inhibitors screened were: a PFKFB3 inhibitor, an inhibitor of a key step in glycolysis; ibrutinib, an inhibitor of Bruton’s tyrosine kinase; idelalisib, an inhibitor of PI3K subunit; and duvelisib, an inhibitor of PI3K and PI3K gamma subunits. To test this hypothesis, T cells were cultured with or without each compound and then the analysis included: phenotypic analysis by flow cytometry, quantitative analysis by counting cells with ethidium bromide acridine orange, and metabolic profiling by the Seahorse assay. This study was conducted using T cells from a human healthy volunteer that were collected by apheresis. T cells were cultured in a G-Rex plate for 15 days with complete media supplemented with recombinant human IL-2 (30 U/mL). Cells were activated on day 1 and day 8 by the addition of anti-CD3/CD28 beads and test metabolic inhibitor compounds were added every 4 days. T cells cultured with idelalisib, duvelisib, and ibrutinib had increased expansion (approximately50-fold: idelalisib/ duvelisib and 21-fold ibrutinib) when compared to control (cells with beads alone) with only 6-fold expansion. Phenotypic analysis performed using flow cytometry showed an increased percentage of CD27+ CD28+ in the CD8+ and CD4+ T cell cell populations in the idelalisib treated group and decreased number of senescent T cells that are double negative for CD27 and CD28. Consistent with our hypothesis, metabolic analysis showed that cells treated with idelalisib and duvelisib were more reliant on oxidative phosphorylation, rather than glycolysis as compared to the control cultures. Cells treated with duvelisib also showed an increased spare respiratory capacity (SRC), which is associated with more efficacious memory T cells. The results of these studies show that metabolism plays a critical role in the long-term survival of T cells. We demonstrate that inhibiting intermediary metabolism, specifically inhibiting PI3K, favorably alters the metabolic state of the T cells leading to increased cell numbers and T cells with a phenotype consistent with enhanced ex vivo and in vivo proliferation and persistence.

Improving the implementation of the DBSA's environmental management capacity building at local government level

The increased importance of environmental management at local government level required a more consistent approach from the DBSA in order to improve delivery. This necessitated the identification of success criteria that the DBSA could implement to guide this improvement. In order to identify these criteria, the following main aspects were researched: The environmental mandate and capacity of local government in South Africa, the international perspective on capacity building, the DBSA’s mandate on capacity building and lastly the outcomes of four environmental capacity building projects implemented by the DBSA. The criteria identified focussed on two main areas, namely project management and environmental management. Furthermore, various steps were identified that the DBSA needs to take with relation to its own project cycle that might lead to improvements in this regard.

Identifying relevant factors in implementing a chemistry curriculum in Botswana

A general concern in science education is that change in the curriculum has had little impact on the classroom practice. Following the introduction of a new curriculum called the Botswana Government Certificate in Secondary Education (BGCSE) curriculum in Botswana senior secondary schools, this study set out to investigate issues relating to implementation of the proposed curriculum, that is, mapping teaching effectiveness intended to improve students’ learning of chemistry compared with teaching during the old curriculum. This thesis reports the extent to which some chemistry teachers in Botswana senior secondary schools are implementing classroom methodologies that focus on learner-centred and hands-on activities. This investigation involved working with eleven chemistry teachers in four secondary schools in Botswana. The case study research methodology included observations and semi-structured interviews. The sub-constructs from Rogan and Grayson’s (2003) theory were used to guide data collection as well as analysis and interpretation of data. From the categories, it was possible to identify level of operation for the schools as well as the Zone of Feasible Implementation (ZFI) which spelt possible routes for interventions in individual schools. There were common areas observed about the teachers and the schools, in that they · All possessed the right qualifications to teach chemistry at senior school level. · All were relatively young in the teaching field · According to Rogan and Grayson’s (2003) scale, the teachers are operating at a very low level in the science practical and contextualisation dimensions. · Classroom dimensions were characterized by patches of rich interactions for most teachers. These were evidenced by some teaching interactions at level 3 on Rogan and Grayson’s operational levels. Some teachers’ lesson activities were more interactive in that there were presentations that encouraged the use of various communication skills. Learners presented organised information to support their positions, showing there had been a search for information before presentations. The other teachers limited involvement by learners, as their focus was on articulating the content. · Otherwise teachers were generally operating at lower levels in Rogan and Grayson’s (2003) scale. This is a clear indication that to some extent the teachers are not doing much in implementing the teaching ideas intended by the new curriculum. Mind and hands-on activities are still limited. Though teachers possess the right qualifications, there seems to be little desire to try new things. According to the operational levels on classroom interaction, practical work and contextualisation, it was found that teachers generally operated moderately on Rogan and Grayson’ scale. Though interviews results showed that teachers saw the need to have practical work as a way to actively engage learners, classroom observations showed that it was not a favoured method. Classroom observations revealed that the lecture method was still the favoured approach. Some learner-engaging activities in the form of group discussions, group experiments and demonstrations were practiced. Contextualizing of science was still generally low in all schools. The study also looked at the construct of capacity to support innovation. The three sub-constructs which were investigated in this study were physical resources, teacher factors and ecology and management systems. These factors had a great impact on how the sub-construct on profile of implementation panned out. The three factors were found to be higher on the Rogan and Grayson’s (2003) scale than the profile of implementation. This was found to be linked with a great deal of improvement of infrastructure by the Botswana government soon after the curriculum implementation e.g. science related facilities (laboratories, apparatus, chemicals) and teacher qualifications. Though this was the case, it was found out that classroom implementation was not as inflated as the resources on ground seemed to suggest. Several influential factors were found to be linked to the unfolding of a curriculum inside the classroom such as departmental contexts. The contexts were found to relate closely with the nature of communities that existed in science departments, which were linked to how teachers were distributed within departments.

The contributions of and challenges faced by developmental Non-Governmental Organisations in Gauteng.

This research was aimed at establishing the contributions of developmental nongovernmental organisations (NGOs) to social welfare service delivery in Gauteng, South Africa, and to examine the challenges they have faced since the transition from Apartheid social welfare to a developmental welfare paradigm. This study revealed how developmental NGOs function to meet the needs of citizens, especially the marginalised and vulnerable class of the society. It also examined the contributions of developmental NGOs in terms of capacity building, physical infrastructure development and their sustainability. The research was qualitative and exploratory in nature, and a multiple case study design was applied. The research sample of 15 participants was drawn from three developmental NGOs registered with the Department of Social Development, Gauteng, South Africa. Five employees from each developmental NGO were interviewed using a semi-structured interview schedule. A face to face interview was the method of data collection. The data collected was analysed. The main findings showed that there is evidence of positive contributions to capacity building and infrastructural development at the grassroots level. However, funding remains the biggest challenge, which then affects the sustainability of development.

Desenvolvimento e aplicação de um novo ensaio para a determinação eletroquímica da capacidade antioxidante de compostos modelo e de matrizes complexas / Development and application of a new assay for the electrochemical determination of the antioxidant capacity of model compounds and of complex matrices

Este trabalho descreve o desenvolvimento e aplicações práticas de uma nova e simples metodologia eletroquímica para a determinação da capacidade antioxidante de moléculas modelo específicas e/ou algumas amostras complexas de alimentos normalmente consumidas no Brasil. Outros sistemas de interesse teórico ou tecnológico também foram investigados. O método se baseia no uso de uma quantidade conhecida de um íon inorgânico como oxidante e na determinação cronoamperométrica de sua concentração remanescente após reação com as espécies antioxidantes de interesse. Contudo, testes iniciais para diferentes marcas comerciais de sucos de laranjas usando Fe3+ como oxidante (ensaio FRAP modificado), só obtiveram êxito quando o antioxidante apresenta um comportamento eletroquímico totalmente irreversível como, por exemplo, o ácido ascórbico. Para superar esse problema, o ensaio foi então desenvolvido usando o Ce4+ como oxidante (ensaio CRAC) uma vez que sua redução após reação pode ser realizada em 0,8 V vs Ag/AgCl, uma região de potencial na qual não ocorre a redução das espécies formadas pela oxidação reversível ou quase reversível do antioxidante. Devido ao elevado potencial anódico requerido quando o Ce4+ é usado, foi necessário um filme de diamante dopado com boro como eletrodo de trabalho. Após uma rigorosa caracterização do sistema eletroquímico, foram realizadas determinações da capacidade antioxidante de oito compostos padrões (ácido ascórbico, ácido gálico, ácido tânico, BHA, catequina, quercetina, rutina e trolox), usando o ensaio CRAC. Os resultados mostraram uma correlação satisfatória com ensaios mais complexos reportados na literatura e foram aplicados em um conjunto de sucos de frutas industrializados, mostrando valores máximos com quase uma ordem de grandeza superior ao apresentado pelo composto de referência (trolox), com a seguinte seqüência de capacidade antioxidante: caju > goiaba > uva > manga > laranja > maracujá. Considerando a busca da indústria local de \"cachaça\" por madeiras alternativas em contrapartida aos tonéis de carvalho, o ensaio CRAC foi realizado usando quatro extratos etanólicos de madeiras brasileiras [Pequi (Caryocar brasiliense), Imbuia (Octea porosa), Cabreúva (Myrocarpus frondosus) e Cabreúva-vermelha (Myroxylon balsamum)] assim como um extrato de Carvalho (Quercus sp), para comparação. Os resultados indicaram um aumento na capacidade antioxidante na ordem apresentada acima e, apesar da melhor amostra (Cabreúva-vermelha) ter apenas 60% da capacidade antioxidante apresentada pelo carvalho, sua disponibilidade e preço despertam o interesse por pesquisas futuras. Uma avaliação comparativa dos resultados obtidos usando os ensaios CRAC e DPPH foi realizado para extratos metanólicos de cana-de-açúcar e polpa de maracujá. Essa comparação revelou uma diferença quantitativa entre os valores dos ensaios, porém, a hierarquia foi mantida para cada conjunto de resultado. Esse efeito foi atribuído às diferenças nos mecanismos dominantes para a desativação radicalar, bem como para as condições experimentais de cada ensaio. A correlação entre a estrutura e a atividade antioxidante de moléculas modelo de flavonóides sob investigação foi relatada devido à presença de certos grupos substituintes na estrutura de difenilpirano. A atividade hierárquica para tais grupos foi estabelecida como: OH(C2´C4´) > OH(C4´) ~ OH(C3´C4´) > C2=C3 + 4-oxo > OH(C3,C5) + 4-oxo > OH(C3) + 4-oxo > OH(C5) + 4-oxo > OH(C3,C5). A formação de complexos entre flavonóides e íons metálicos, tal como o Fe2+, tem um forte efeito sobre a capacidade antioxidante e o ensaio CRAC mostrou que para a morina, quercetina e fisetina, esse aumento foi de 15,3; 31,8 e 27,9%, respectivamente. Por outro lado, para a catequina e crisina, o aumento foi de apenas 1,8 e 7,8%, respectivamente. Esses aumentos foram relatados devido à presença de, pelo menos, um dos três tipos de sítios ativos na molécula polifenólica que interage com íons metálicos. Todos esses resultados confirmam que o ensaio CRAC é uma ferramenta simples e viável para a determinação da capacidade antioxidante de uma variedade de sistemas práticos e moléculas modelo. / This work describes the development and practical applications of a novel and simple electrochemical methodology for the determination of the antioxidant capacity of specific model molecules and/or some complex food samples currently consumed in Brazil. Other systems having either theoretical or technological interest were also investigated. The method is based on the use of a known amount of an inorganic ion as the oxidant and in the chronoamperometric determination of its remaining concentration after reaction with the chosen antioxidant species. However, initial tests for different commercial brands of orange juices using Fe3+ as the oxidant (modified FRAP assay) were only successful when the antioxidant has a totally irreversible electrochemical behavior as, for example, ascorbic acid. To overcome this problem, the assays were then performed using Ce4+ as the oxidant (the CRAC assay) since its reduction after reaction can be carried out at 0.8 V vs Ag/AgCl, a potential region where the reduction of species formed by the reversible or quasi-reversible oxidation of the antioxidant does not occur. Due to the high anodic potentials required when using Ce4+, it was necessary to have a boron-doped diamond film as the working electrode. After a rigorous characterization of the electrochemical systems, measurements of the antioxidant capacity of eight standard compounds (ascorbic acid, gallic acid, tannic acid, BHA, catechin, quercetin, rutin and trolox) were carried out using the CRAC assay. The results showed a satisfactory correlation with those reported in the literature using other more complex assays and these studies were then applied to a set of industrialized fruit juices showing maximum values almost one order of magnitude higher than that of the reference compound (trolox) and following the antioxidant capacity sequence: cashew>guava>grape>mango>orange>passion fruit. Considering that the local \"cachaça\" industry is looking for alternative woods to the use of oak barrels, CRAC assays were carried out using four ethanol extracts of Brazilian woods [Pequi (Caryocar brasiliense), Imbuia (Octea porosa), Cabreúva (Myrocarpus frondosus) e Cabreúva-vermelha (Myroxylon balsamum)] as well as an Oak (Quercus sp) extract, for comparison. The results indicate an increasing antioxidant capacity in the order presented above and, although the best sample (Cabreúva-vermelha) has only 60% of the capacity shown by oak, its local availability and price makes it interesting for further research. A comparative evaluation of the results obtained using the CRAC and the DPPH assays was carried out for methanol extracts of sugar cane juice and passion fruit pulp. That comparison revealed a quantitative difference between the assay values but the hierarchy was maintained for each set of results. Such effect was attributed to differences in the prevailing mechanism for radical deactivation, as well as, the experimental conditions used for each assay. The correlation between structure and antioxidant activity of model flavonoid molecules under investigation was related to the presence of certain groups in the diphenilpyrene structure. The activity hierarchy for them was established as: OH(C2´C4´) > OH(C4´) ~ OH(C3´C4´) > C2=C3 + 4-oxo > OH(C3,C5) + 4-oxo > OH(C3) + 4-oxo > OH(C5) + 4-oxo > OH(C3,C5). The complex formation between flavonoids and metal ions, such as Fe2+, has a strong effect on the antioxidant capacity and CRAC assay showed that for morin, quercetin and fisetin the increase was 15.3, 31.8 and 27.9%, respectively. On the other hand, for catechin and chrysin the increase was only 1.8 and 7.8%, respectively. These increases were related to the presence of, at least, one of three types of active sites in the polyphenolic molecule that can interact with metal ions. All these findings confirm that the CRAC assay is simple and convenient tool for the determination of the antioxidant capacity of a variety of practical systems and model molecules.

Leadership Practices that Affect Student Achievement: Actions of the School Principal When Building Capacity in a High Performing Urban Elementary School

Thesis advisor: Diana C. Pullin / It is widely accepted that school leadership has both a direct and indirect impact on student achievement. Hitt and Tucker’s (2016) unified leadership framework summarized a decade of work by numerous researchers identifying the five most effective leadership domains that influence student learning. Using that work as a conceptual framework, this qualitative case study analyzed one of the five interdependent leadership domains in an urban elementary school that succeeded in educating traditionally marginalized students and outperformed other schools with similar demographics in the district. This study identified and explored the second leadership domain which is described as building professional capacity, focusing specifically on the principal’s actions. Building professional capacity is defined in this study as developing the knowledge, skills, and dispositions of teachers and is important for expanding leadership responsibility for student learning and increasing student achievement. The study resulted in identifying all seven dimensions of the domain present in practice at the school and explored the actions taken to embed them into the culture. Data were gathered through the review of district and school level documents and open-ended interviews with district leaders and members of school administration, faculty, and staff. Findings indicated that the school principal performed several specific and tangible actions to build professional capacity in the school including implementing professional practice, messaging consistently high expectations with faculty and staff, modeling expected professional behaviors and habits, and coaching faculty and staff. Many staff believed the success of the school was a result of the culture of collaboration, dedication, and high expectations forged by the principal. Recommendations for building upon this work included selecting and hiring teachers who offer evidence of effective instruction despite different philosophical viewpoints, addressing levels of trust with those who express conflicting levels of collaboration and commitment, and expanding the effective leadership practices with other school leaders throughout the district. / Thesis (EdD) — Boston College, 2018. / Submitted to: Boston College. Este trabalho propõe um método de análise do trade-off entre o buffer de capacidade de produção e o buffer de estoque, necessários para garantir o bom desempenho de um sistema sujeito a variações de demanda. A análise envolve a definição de um sistema de produção e a construção de um modelo de simulação. A simulação é realizada num sistema formado por uma única estação de trabalho, com mix de 10 produtos, controlados por um sistema kanban, que é considerado por muitos autores inadequado para operar em ambientes cuja demanda não é estável. Os resultados mostram que o buffer de capacidade proporciona vantagens operacionais e de custo ainda que sujeito a maiores níveis de variação de demanda. / This research proposes a method to analyze the trade-off between the production capacity buffer and the inventory buffer, required to guarantee high performance levels in a system with demand variation. The analysis involves a production system delineation and the development of a simulation model. The system simulated is a single-stage production system, with mix of ten products, controlled by a kanban system, which is classified by several authors as not recommended in environments with unstable demand. The results indicate that the production capacity buffer enables operational and cost advantages, even if the production system is under higher demand variation.
