Blood constituents labeled with technetium-99m (99mTc) have also been employed as radiobiocomplexes and used also experimental model for evaluation of the biological effects of natural or synthetic drugs. The analysis of the morphology and the morphometrics parameters (perimeter/?rea ratio) can be used to evaluate the effects of drugs upon the structure of the membrane of red blood cells. Cinnamomum zeylanicum (cinnamon) is a spice used as herbal medicine to treat diseases. The aim of this study was to evaluate the effect of in vitro and in vivo treatment with an aqueous cinnamon extract on the labeling of blood constituents with 99mTc and on the morphology of red blood cells from Wistar rats. In the in vitro treatment, isolated blood sample from animals were incubated with cinnamon extract. In the in vivo treatment, blood samples were also withdrawn from animals treated with cinnamon extract. In both cases, the radiolabeling of blood constituents was done. The morphological analysis of red blood cells was also done. As control, blood or animals treated with NaCl 0.9%. The data obtained on the labeling of blood constituents with 99mTc experiments indicated that the in vitro treatment with cinnamon extract was capable to decrease signiicantly (p<0.05) the percentage of radioactivity in cellular compartments and on the fixation of cellular and plasma proteins. These effects were not observed on the in vivo treatment. The results obtained for the morphology of red blood cells suggest that the in vitro and in vivo treatments did not alter the morphology and the perimeter/area ratio. The in vitro treatment with aqueous cinnamon extract could affect the membrane structures related with the oxidation status of the stannous ion pertechnetate ion, altering the labeling of blood constituentes with 99mTc. This study was a multidisciplinary experimental research. It was developed with the contribution of the different Departments and Services of the Hospital Universit?rio Pedro Ernesto of the Universidade do Estado do Rio de Janeiro / Radiobiocomplexos s?o utilizados para obten??o de imagens em medicina nuclear e empregados na pesquisa b?sica. Among various species of plants containing essential oils in its composition and that may be used to fight various diseases highlight the essential oil extracted from the leaves of cinnamon (Cinnamomum zeylanicum Blume). The cinnamon oil can be used to prevention of schistosomiasis, an endemic disease that affects areas with poor basic sanitation. In this study, the essential oil was extracted from the leaves of cinnamon by hydrodistillation and its molluscicidal action has been assessed against the snail Biomphalaria glabrata (Say, 1818). The percentage of oil in the leaves was an average of 1.3 mL in the time interval ranging from 1 to 5 hours, after extraction the physico-chemical properties of oil were determined. The measurements of oil components were made by gas chromatography-mass spectrometry (GC-MS). The chromatographic analysis showed that oil has 82.67% of eugenol (major constituent), at retention time 22,63 min and 2.47% of humulene (minor component), at retention time 25.74 min. The cinnamon oil has been tested for toxicity and molluscicidal agent, calculating the lethal concentration (LC50). As for toxicity, the oil extracted from cinnamon leaf was considered moderately toxic LC50 = 162.1 mg.L-1 ± 2,80, with a confidence interval 95%. For the pilot test, the molluscicide activity of the oil presented a mortality of 100% in 72 h indicating the presence of toxic compounds in the oils against the snail indicating the presence of toxic compounds in the oils against the snail. The molluscicide activity of essential oil extracted from the leaves of cinnamon obtained results relative to the 50% lethal concentration LC50 of 18.62 mg L-1 ± 2,18, with a confidence interval 95%, lower the level recommended by the World Health Organization (WHO), showing that this oil is active against the snail Biomphalaria glabrata. / A utilização de plantas para fins medicinais é uma prática antiga. Dentre várias espécies de plantas que contém óleo essencial na sua composição e que podem vir a ser utilizadas no combate a várias doenças destaca-se o óleo essencial extraído das folhas da canela. O óleo da canela pode ser utilizado no combate da esquistossomose, doença endêmica que afeta aréas com saneamento básico precário. Nesta pesquisa, o óleo essencial foi extraído das folhas da canela (Cinnamomum zeylanicum Blume) por meio de hidrodestilação, sua ação moluscicida foi avaliada frente ao caramujo Biomphalaria glabrata (Say, 1818). O percentual de óleo nas folhas em média foi de 1,3 mL no intervalo de tempo variando de 1 a 5 horas, logo após o extraído, passou por caracterizações físico-químicas. As quantificações dos seus componentes foram feitas por cromatografia gasosa acoplada à espectrometria de massas (CG-EM). As análises cromatográficas mostraram que o óleo possui 82,67% de eugenol (constituinte majoritário), no tempo de retenção de 22,63 min; e 2,47% de Humuleno (componente minoritário), no tempo de retenção de 25,74 min. O óleo de canela foi testado quanto à toxicidade e como agente moluscicida, calculando-se a concentração letal (CL50). O óleo estudado, extraído da folha da canela quanto a toxicicidade foi considerado moderadamente tóxico apresentando CL50 de 162,1 mg.L-1 ± 2,80, com um intervalo de confiança a 95%. Para o teste piloto, a atividade moluscicida do óleo apresentou uma mortalidade de 100% em 72 h, indicando a presença de componentes tóxicos nos óleos frente ao caramujo. A atividade moluscicida do óleo essencial extraído das folhas da canela obteve resultado referente à concentração letal 50% (CL50) de 18,62 mg.L-1 ± 2,18, com um intervalo de confiança a 95%. , abaixo do preconizado pela Organização Mundial de Saúde (OMS); mostrando que este óleo é ativo frente ao caramujo Biomphalaria glabrata.

The new global trends of a concern for biodiversity and the ideas of sustainable development have brought new air to studies of Brazilian medicinal plants. New lines of research were established in Brazilian universities, some of them seeking solid basis for the validation of the use of medicinal plants, thus justifying this work, using aqueous extracts of plants place of occurrence, such as Cinnamomum spp., Mentha piperita and Passiflora foetida, inhibition of growth of some species of the genus Aspergillus tox?genas, previously studied in the literature and identified as antifungal and encouraging the use of plants to replace chemicals. Using the method of minimum inhibitory concentration in agar, with the technique of dilution plate (Pour-Plate), being held dilutions of different extracts, from a solution of 5% and resulting in the dilution of 2.5% and 1.25%. Batteries tests were incubated at the 36oC. It was held also test for sensitivity to antifungal commercial - Ketoconazole. It was observed that the aqueous extract of Cinnamomum spp. was able to inhibit the mycelial growth of A. flavus and A. carbonarius at concentrations of 2.5% and 5% in readings of 48 hours. The other aqueous extracts, used in this work, showed no inhibitory effect of mycelial growth of fungi tested. / Muitas ?reas est?o envolvidas na pesquisa de novas subst?ncias oriundas de plantas. Effect of Indole-3-butyric Acid on the Formation of Adventitious Roots in Cinnamomum kanehirae Cuttings

Synthetic auxin, indole-3-butyric acid (IBA), effectively promoted the rooting in Cinnamomum kanehirae cuttings. The easy-to-root genotype, H107, responded to IBA much earlier than the difficult-to-root genotype, L41. On day 5, the POD activity significantly decreased in the IBA-treated tissues as compared with the control. Similar phenomenon was observed in extract of L41 genotype showing that IBA inhibited POD activity. In addition, the variation in POD activity corresponds to an inverse variation in the concentration of free IAA. The levels of IAA in H107 genotype increased dramatically in IBA-treated tissues on day 5; while, in L41 genotype, the raise of IAA in IBA-treated tissues was observed on day 20. Moreover, the lignin content in IBA-treated H107 cuttings decreased is quite correlated with the decline of the POD activity; yet the lignin content in L41 genotype cuttings was almost unchanged. Hence, we suggest that the inhibition on POD may lead to the redifferentiation processes induced by IBA, then produce the new root primordia during the formation of adventitious roots.

Cloning and sequence analysis of the peroxidase genes in Cinnamomum kanehirae young roots

Auxin can induce adventitious rooting. Synthetic auxin, indole-3-butyric acid (IBA), effectively promoted the rooting in Cinnamomum kanehirae. The peroxidase (POX) activity significantly decreased in the IBA-treated tissues as compared with the control. Hence, I suggest that the inhibition on POX activity may lead to the redifferentiation processes induced by IBA, which produces the new root primordia during the formation of adventitious roots. On this investigation, I cloned POX cDNA from the young roots. Degenerate primers were designed from the conservative regions of other published POX to amplify the expectant DNA fragment. Full-length cDNA of the POX gene designated CKPX1 and CKPX3 were cloned by the method of 5'and 3' RACE. The deduced amino acid of CKPX1 and CKPX3 were compared with the previously reported POX and showed between 40% and 60% identity with those plant species. Further studies on the promoter elements of CKPX3 were found out that elements related to auxin response, lignification, pathogen invasion and stress response. The results suggest that CKPX3 may be involved in the regulate process of adventitious rooting and defense against pathogens and environment stress.

In Cinnamomum kanehirae, there are high (H) and low (L) rooting cultivar. The peroxidase (POX) activity significantly decreased in the IBA-treated tissues as compared with the control. The inhibition on POX activity may lead to the redifferentiation processes induced by IBA. In this investigation, we cloned POX cDNA from the young roots. Degenerate primers were designed from the conservative regions of other published POXs to amplify the expectant DNA fragment. We found that the H and L line have similar genes (>99%). The Full-length cDNA of the POX genes were cloned by the method of 5'and 3' Rapid Amplification of cDNA Ends (RACE). The deduced amino acid were compared with the previously reported POX and showed between 40% and 70% identity with other plant POXs. Further studies on the promoter elements of POX in High-rooting cultivar and Low-rooting line show that some elements are related to auxin response, lignification, pathogen invasion and stress response. The regulatory elements of the POX gene in High-rooting line contain sugar repression responsiveness (SRS) elements that might repress the expression of POX gene, causing the lower POX activity.

Its pathogenicity is related to virulence factors. The treatment of this disease is difficult because the drugs present high toxicity, resistance of the fungus and little therapeutic efficiency. Research with biologically active natural products has shown that essential oils (OEs) have important antifungal activity. Thus, the present study had as objective to evaluate the antifungal efficacy and action on the virulence factors in C. neoformans complexes of OEs of Cinamomum cassia (Cinnamon of China) and Cybopogom flexuosus (Lemon Grass). By means of the microdilution technique in broth, OEs had MIC ranging from 8 to 256 μg / ml and CFM from 16 to 512 μg / ml. C. cassia promoted a 47.8% reduction in capsule size and inhibited melanin synthesis in 30% of the isolates. The evaluation of this OE on the activity of exoenzymes phospholipase, urease and DNAse demonstrated inhibition in 15%, 30% and 25% of the evaluated isolates respectively. C. flexuosus reduced the size of the capsule by 48.7%, inhibited the production of melanin, urease, phospholipase and DNAse by 30%, 30%, 25% and 15% respectively. The data demonstrated that the isolates did not produce proteinase and the hemolytic activity was reduced by 15% for C. cassia and 45% C. flexuous. The results highlight the potential of these OEs in the treatment of cryptococcosis as well as the inhibition of virulence factors. These data are promising, and further research is needed to develop of a new drug. / O complexo Cryptococcus neoformans compreende duas espécies, responsáveis por causar criptococose, sendo a principal manifestação a meningoencefalite em imunocomprometidos. Sua patogenicidade está relacionada aos fatores de virulência. O tratamento desta enfermidade é dificultado devido aos fármacos apresentarem alta toxicidade, resistência do fungo e pouca eficiência terapêutica. Pesquisa com produtos naturais biologicamente ativos tem demonstrado que os óleos essenciais (OEs) apresentam importante atividade antifúngica. Assim, o presente estudo teve como objetivo avaliar a eficácia como antifúngico e ação sobre os fatores de virulência em espécies do complexo C. neoformans dos OEs de Cinamomum cassia (Canela da China) e Cybopogom flexuosus (Capim limão). Por meio da técnica de microdiluição em caldo, os OEs apresentaram CIM variando entre 8 a 256 μg/ml e CFM de 16 a 512 μg/ml. C. cassia promoveu uma redução de 47,8% no tamanho da cápsula e inibiu a síntese de melanina em 30% dos isolados. A avaliação deste OE sobre da atividade das exoenzimas fosfolipase, urease e DNAse demonstrou inibição em 15%, 30% e 25% dos isolados avaliados respectivamente. C. flexuosus reduziu o tamanho da cápsula em 48,7%, inibiu a produção de melanina, urease, fosfolipase e DNAse em 30%, 30%, 25% e 15% respectivamente. Os dados demonstraram que os isolados não produziram proteinase e a atividade hemolítica foi reduzida em 15% para C. cassia e 45% C. flexuosos. Os resultados enaltecem o potencial desses OEs no tratamento da criptococose assim como na inibição dos fatores de virulência. Esses dados são promissores, sendo necessário pesquisas mais aprofundadas para o desenvolvimento de um novo fármaco.

Camphor derivatives in asymmetric synthesis: a synthetic, mechanistic and theoretical study

A series of 3,3-ethylenedioxy-exo- and endo- bornyl esters have been prepared and subjected to α-benzylation using lithium diisopropylamide and benzyl bromide. In the exo-series of esters the diastereofacial selectivity of benzylation was found to improve (up to 34% d.e.) as the steric bulk of the O-alkyl group increased, whereas in the endo-series, a surprising decrease in stereoselectivity was observed as the steric bulk increased – an observation attributed to flexibility of the metal-coordinated endo-enolate system, compared to the relative rigidity of the exo analogues. The conformational options for each series was explored at the density functional theory level. Reductive cyclization of a range of specially prepared N-carbobenzyloxy-amino acid esters has been shown to afford the corresponding derivatives, contrary to previous reports that the cyclization is limited to the glycine derivative. The cyclization sequence has been explored in detail, and the yield has been shown to be critically dependent on the stereochemistry of the α-amino acid moiety. Moreover, it seems that reductive cyclization occurs more readily with the endo- rather than the exo-bornyl N-CBZ-amino acid esters. Molecular modelling of relevant transition states at the DFT levels indicates that L-amino acid-derived systems should cyclize preferably in the exo-series and D-amino acid-derived systems should cyclize preferably in the endo series. Studies of alkylation of an iminolactone system have reported an interesting anomaly - exo-methylation is observed while endo-alkylation predominates for larger alkyl groups. This has been studied in detail at the DFT level, and the anomaly is attributed to thermodynamic control in the methyl case, whereas kinetic control is the norm in this system. Preliminary computer modelling of the intramolecular rearrangement of a 3,3-xylylbornyl system at the HF/STO-3G level raised doubts concerning the structure assigned by Evans to one of the rearrangement products, prompting an X-ray crystallographic analysis and leading to the revision of its structure from a pinene to a camphene derivative. The previously elusive spiro[bornane-3,2’-indan]-2-exo-tosylate has been successfully isolated, and the kinetics of its ready decomposition to the two camphene products has been followed by 1H NMR spectroscopy. The endo-tosylate analogue, on the other hand, was found to be remarkably stable. Kinetic data obtained for rearrangement of this exo-bornyl tosylate have indicated the operation of tandem autocatalytic and pseudo-first-order transformations leading sequentially to the two isomeric camphene products. An extensive coset analysis of all possible rearrangement processes of the initially-formed cation formed from decomposition of the exo-tosylate has afforded a graph containing 336 classical cations (modelled at the AM1 and B3LYP levels) and 526 transition-state complexes (modelled at the AM1 level). This analysis afforded a viable 4-step classical mechanism connecting the first camphene product with the second. A more realistic study, involving non-classical carbocations, has afforded a graph of all possible (classical and non-classical) cations that could be formed by rearrangment of the initiallyformed cation. The resulting graph confirms that the only energetically feasible path corresponds to the classical mechanism, but simply involves two steps, including a novel, concerted Wagner-Meerwein – 6,2-hydride shift – Wagner-Meerwein rearrangement.

The effect of Cinnamomum verum 1X on glucose tolerance in non-diabetic males

M.Tech. / Impaired glucose tolerance, as determined by an oral glucose tolerance test, is the inability of the blood glucose regulatory mechanisms in the body to return the blood glucose levels to normal physiological levels after the ingestion of a meal (Guyton and Hall, 2006). Mollentze and Levitt (2006) report on various studies that were conducted in South Africa on the prevalence of impaired glucose tolerance and diabetes mellitus and they illustrate that there is an incidence of impaired glucose tolerance as high as 10.2% in certain communities and that impaired glucose tolerance may result in diabetes mellitus. A study has shown that cinnamon controls blood sugar levels by increasing insulin sensitivity resulting in better blood sugar level control (Jarvill-Taylor et al, 2001). The above research was done on cinnamon in the crude form but there is no research on Cinnamomum verum in a homoeopathic potency. The aim of the research was to evaluate the effect of Cinnamomum verum 1X on glucose tolerance in non-diabetic males after an oral glucose tolerance test. A double-blind pre-test post-test placebo controlled study on thirty healthy adult males between the ages of eighteen and forty years was conducted. The participants were recruited by means of an advertisement at the University of Johannesburg Health Clinic. Inclusion criteria comprised: no medication for one month prior to the study with a normal fasting blood sugar level of more than 3.0 but less than 6.0 mmol/l (Oussama, 2006); systolic blood pressure between 100 and 140 mmHg and diastolic blood pressure between 70 and 90 mmHg. At the first consultation the participant data was obtained and an oral glucose tolerance test was performed. The participants were issued with 100 ml of either Cinnamomum verum 1X or a placebo and a weekly checklist to complete. After two weeks the participant data was recorded again and the oral glucose tolerance test was repeated. The Mann-Whitney U-test was used to analyse between the group data statistically and the Wilcoxon Signed-Rank test was used to perform the within-group analysis. The mean fasting blood glucose level of the experiment group changed from 4.71 mmol/l to 4.49 mmol/l. This was a statistically significant reduction, p = 0.025 (< 0.05). The reduction in the blood glucose level after the oral glucose tolerance test of the experiment group was also found to be statistically significant, p = 0.001 (< 0.05) with the mean value changing from 5.86 mmol/l to 5.40 mmol/l. It was concluded that the homoeopathic remedy, Cinnamomum verum 1X taken orally twice daily, was more effective than the placebo in reducing the fasting blood glucose level and the blood glucose level after an oral glucose tolerance test in non-diabetic males between the ages of eighteen and forty years with normal initial fasting blood glucose levels within a time period of two weeks.
