Resistência térmica de esporos bacterianos em néctar e suco de manga / Thermal resistance of bacterial spores in mango nectar and mango juice

Os valores D 85°C para esses esporos em polpa de manga, com pH 3,96, foram de 19 e 32 minutos, respectivamente; em néctar de manga, com pH 3,98, foram 31 e 28 minutos, respectivamente. Ao se adicionar em 12,5 mg de nisina/kg de produto observou-se que, em polpa de manga, os esporos de B. licheniformis e B. coagulans, apresentaram os valores D 85°C 18 e 31 minutos; e em néctar de manga, foram 20 e 31 minutos, respectivamente. A nisina adicionada na mesma concentração ao ágar nutriente, impediu a recuperação dos esporos da polpa e néctar de manga. Sugere-se que em tratamentos térmicos brandos de polpa e néctar de manga, a nisina possa ser usada para prevenção de defeitos decorrentes de contaminação por esporos bacterianos. / Thermal resistence of spores of Bacillus licheniformis and Bacillus coagulans, was determined in mango pulp and nectar at differents temperatures. D 85°C values in mango pulp, pH 3,96, were 19 and 32 minutes, respectively; in mango néctar, pH 3,98, D 85°C for those spores were 31 and 28 minutes, respectively. The addition of 12,5 mg nisin per kg pulp rendered D 85°C of 18 and 31 minutes for B. licheniformis and B. coagulans, respectively. In mango néctar added of the same concentration of nisin, D 85°C for those spores were 20 and 31 minutes, respectively. Nisin added to Nutrient Agar at the same concentration prevented spore recovery from heat treated mango pulp and nectar. It is suggested that nisin be further studied as a means of preventing spoilage due to spore forming bacteria in pasteurized mango pulp and nectar.

Clostridioses em ruminantes na região sul do Rio Grande do Sul / Clostridial diseases in ruminants in southern Rio Grande do Sul

Out of 5.446 bovine specimens sent by practitioners or from necropsies performed 91 (1,67%) were diagnosed as clostridiosis. Of those, 30 (0.55%) were diagnosed as by black leg, 15 (0.27%) as bacillary hemoglobinuria, 9 (0.16%) as botulism, 9 (0.16%) as malignant edema, and 28 (0.51%) as tetanus. Out of 860 sheep specimens 49 (5.69%) were bacterial diseases and 14 (28,57%) of those were diagnosed as clostridiosis, being 10 (1.16%) cases of tetanus, 3 (0.34%) of malignant edema, and 1 (0.11%) %) of enterotoxemia. It was concluded that some clostridiosis cause important losses in livestock in Rio Grande do Sul, mainly black leg. The vaccination of young stock against black leg is recommended in the whole state. Vaccination against botulism is recommended in areas of native pastures where phosphorus deficieny occurs, and vaccination against bacillary hemoglobinuria is recommended in low lands where fasciolosis is enzootic. In cattle, tetanus may occur as outbreaks with significant economic losses, recommending also the vaccination of animals. In sheep clostridioses are less important being tetanus the more frequent. Good hygienic practices, mainly during castration or tail are recommended to prevent the disease. Enterotoxemia is a rare disease in the region, because most sheep are raised in native pastures with limited grass production; vaccination is only recommended in sheep grazing in cultivate pastures or supplemented with concentrates. / Foi realizado um estudo retrospectivo dos diagnósticos de clostridioses ocorridos na região sul do Rio Grande do Sul correspondente a área de influência do Laboratório Regional de Diagnóstico da Faculdade de Veterinária da Universidade Federal de Pelotas entre janeiro 1978 e janeiro de 2008 e acompanhados os casos de clostridioses diagnosticados nos anos 2009 e 2010. De um total de 5.446 materiais de bovinos provenientes de necropsias ou órgãos enviados 91 (1,67%) correspondiam a enfermidades causadas por bactérias do gênero Clostridium sendo 30 (0,55%) de carbúnculo sintomático, 15 (0,27%) de hemoglobinúria bacilar, 9 (0,16%) de botulismo, 9 (0,16%) de edema maligno e 28 (0,51%) de tétano. De 860 materiais de ovinos 49 (5,69%) diagnósticos corresponderam a doenças bacterianas e desses 14 (28,57%) foram de surtos de clostridioses. Dos 14 surtos de clostridioses observados em ovinos 10 (1,16%) foram de tétano, três (0,34%) de edema maligno e um (0,11%) de enterotoxemia. Concluiu-se que algumas clostridioses causam prejuízos econômicos à bovinocultura da região, principalmente o carbúnculo sintomático, recomendando-se a vacinação sistemática dos bovinos jovens contra esta clostridiose. Recomenda-se, também, a vacinação contra botulismo, em áreas de campo nativo onde ocorre carência de fósforo e contra hemoglobinúria bacilar em áreas de campos baixos onde ocorre infecção por Fasciola hepatica. Em bovinos o tétano pode ocorrer como surtos com prejuízos econômicos importantes, recomendando-se, também, a vacinação dos animais. Em ovinos as clostridioses têm pouca importância, sendo o tétano a mais frequente. Para a profilaxia desta doença em ovinos recomenda-se utilizar práticas de manejo, como descola e castração, em condições adequadas de higiene. Enterotoxemia é uma enfermidade rara na região por que a maioria dos ovinos permanece em campos nativos com limitada produção de foragem; recomenda-se a vacinação somente em animais em pastagens cultivadas ou suplementados com concentrados.

<p>ABSTRACT</p><p>Anaerobic digestion is one way of handling biowaste and generating energy in the form of methane, biogas.</p><p>This study shows that spore forming bacterias survive the process of pasteurisation and anaerobic digestion in biogas plants. It has also been established that both the nonpasteurised-and digestion- waste contains pathogen spore forming bacterias. Two Swedish full-scale</p><p>commercial biogas plants were sampled before pasteurisation, after pasteurisation and after digestion on 10 occasions with one week intervals. The samples were analysed quantitatively</p><p>and qualitatively, with biochemical methods, for Clostridium spp and Bacillus spp.</p><p>Polymerase Chain Reaction, a biomolecular method, was used for</p><p>C. chauvei analysis, with C. chauvei specific primers. For this analyse the biogas plants were sampled at 11 occasions.</p><p>Survival of pathogenic spore forming bacteria in digestion residue may be a health risk for both humans and animals. The digested residue may be used as fertiliser on arable land and the risk of contamination by pathogenic Clostridium spp and Bacillus spp is hard to assess, but can not be neglected.</p>

Characterization of Clostridium spp. from “blown-pack”, chill-stored, vacuum packaged beef

The objectives of this study were to determine the microbial ecology of “blown-pack” fresh beef obtained from a federally inspected facility and to use biopreservation to prevent spoilage of vacuum packaged chilled beef. Organic acids and alcohols in the purge obtained from the “blown” beef packages were detected using HPLC. PCR analysis indicated that Clostridium spp. were present in the purge of the commercial sample. Biochemical tests, RFLP and 16S rDNA sequencing were used to identify organisms isolated from the meat. Out of 66 isolated strains, 26 isolates were strict anaerobes and RFLP indicated that all were clonal isolates. Based on sequence analysis, the isolate was identified as Clostridium putrefaciens. The isolate caused “blown-pack” spoilage and produced butyric and propionic acids when inoculated onto fresh meat and Carnobacterium maltaromaticum UAL307 prevented the production gas and obvious signs of spoilage. / Food Science and Technology

Orientador: Márcio Garcia Ribeiro / Resumo: Os patógenos entéricos são um grupo complexo de micro-organismos que causam doenças em animais e humanos. Os gatos domésticos podem ser considerados carreadores de patógenos entéricos com potencial zoonótico devido ao hábito de auto-limpeza, estabelecer amplo território, pelo instinto de caça e eliminar subclinicamente patógenos. O presente estudo investigou a ocorrência de Escherichia coli, Salmonella sp., Clostridium spp. e Rhodococcus equi nas fezes de 200 gatos domésticos sem sinais entéricos, dos quais 100 pertenciam ao ambiente urbano e 100 à áreas rurais. Marcadores de virulência de E. coli, Clostridium spp. e R. equi também foram investigados. Do total de 200 amostras fecais avaliadas foram identificadas 175 (88%) linhagens de E. coli das quais 93 (93%) de gatos de ambiente urbano e 82 (82%) de gatos de área rural. Houve diferença significante (p=0,03) entre os grupos para frequência mais alta de isolamento de E. coli em gatos de ambiente urbano. Dentre os marcadores de virulência de E. coli foram identificados os genes eae (13%), escN (13%), stx1 (1%), stx2 (0,6%), aatA (0,6%) e ipaH (0,6%). Foram isoladas 86 (43%) linhagens de C. perfringens A, das quais em 21 (24%) foi detectado o gene cpb2, que codifica a toxina beta 2 e em uma (1%) linhagem foi identificado o gene cpe, responsável pela codificação da enterotoxina. Domestic cats may be considered carriers of enteric pathogens with zoonotic potential, due to habit of self-cleaning, to establish wide territory of life, by hunting behavior, and subclinical elimination of pathogens through feces. The aim of present study was investigate the occurrence of Escherichia coli, Salmonella sp., Clostridium spp., and Rhodococcus equi in 200 fecal samples of cats without enteric signs, of which 100 cats belonging to urban area and 100 to farm environment. Virulence markers from E. coli, Clostridium spp. and R. equi were also tested. Among 200 fecal samples were identified 175 (88%) E. coli strains, from these, 93 (93%) were from feces of cats breeding in urban areas, whereas 82 (82%) were from animals from farms. Statistical difference was observed (p=0.03) to higher identification of E. coli from cats breeding in urban area.
