Využití a účinnost kolostra v kosmetických přípravcích / The use and effectiveness of colostrum in cosmetics

This thesis deals with efficiency of colostrum contained in cosmetics. The aim of study was to determine whether the colostrum contained in cosmetic preparations have any influence on the state of human skin. The theoretical part is a literary review and it deals with defining the basic concepts and phenomena, especially the colostrum, its composition and properties. There is also discussed the raw materials used for real samples preparation and efficiency evaluated on these samples. There are also described the methods used to evaluate cosmetic products. The practical part is focused on evaluating the influence of hydratation of prepared cosmetic products. The skin hydratation and transepidermal water loss was measured by MPA 5. Consumer tests are attached to this part. Chemical stability of these cosmetics is assessed in a special chapter.

Analysis of buffalo colostrum and blod serum from bubaline calves with enfasis on passive immunity transfer /

Orientador: José Jurandir Fagliari / Resumo: O objetivo deste estudo foi avaliar a transferência de imunidade passiva em bezerros bubalinos saudáveis e avaliar a influência das concentrações séricas de imunoglobulina G (IgG) às 24 horas no desempenho produtivo nos primeiros 90 dias de idade dos bezerros. Amostras de colostro de novilhas (sem parto prévio), primíparas e pluríparas; e amostras de sangue de seus respectivos bezerros foram coletadas no nascimento, antes da ingestão de colostro, e 24h, 48h e 72h após o nascimento para determinação das atividades séricas de gamamaglutamiltransferase (GGT) e fosfatase alcalina (ALP); e concentrações séricas de proteína total (PT), imunoglobulinas A (IgA) e G (IgG) e lactoferrina. As concentrações de imunoglobulinas de amostras de colostro de búfalas e amostras de sangue de bezerros foram determinadas pela técnica de eletroforese em gel de poliacrilamida contendo dodecil sulfato de sódio (SDS-PAGE). O peso foi medido a cada semana após o nascimento até os três meses de idade. Os resultados da transferência de imunidade passiva foram analisados como medidas repetidas no tempo e o peso e o ganho de peso diário (GPD) foram avaliados por regressão linear simples. As diferenças foram consideradas significativas quando P ≤ 0,05. Os búfalos nasceram hipogamaglobulinêmicos (4,23±0,33 mg/mL), e às 24 horas as concentrações séricas de IgG foram de 34,5±1,48 mg/mL, a transferência de imunidade passiva foi considerada bem-sucedida. Colostrum samples from heifers (without previous calving), primiparous and pluriparous dams; and blood samples from their respective calves, were taken at birth, before colostrum ingestion, and at 24h, 48h, and 72h after birth for determination of serum activities of gamma-glutamyltransferase (GGT) and alkaline phosphatase (ALP); and serum concentrations of total protein (TP), immunoglobulins A (IgA) and G (IgG) and lactoferrin. Immunoglobulin G Concentrations in Alpaca Colostrum during the First Four Days after Parturition

During the first days after parturition, mammalian milk (colostrum) is specifically formulated to nourish newborns. Immunoglobulins are a particularly important component for newborn New World camelids, as their immune system is almost totally dependent on the intestinal transfer of colostral immunoglobulins to acquire passive immunity. In this study, colostrum samples were collected from 20 alpaca mares in the first four days after parturition and analyzed for their immunoglobulin concentration. Sampling started on the day of parturition. The associations of immunoglobulins with other components were determined. The immunoglobulin G (IgG) concentrations decreased significantly within the first four days after parturition. The correlation coefficients between IgG content and the content of various minerals were significant but variable. The correlation between IgG content and fat and lactose content was negative but between IgG content and protein content was highly positive. This strong association could be used for a brief estimation of the IgG content of the colostrum based on the measured protein concentration. The results of the present study can be used for the development of colostrum replacers where motherless rearing is required. Abstract Colostrum provides the newborn with nutrients and immunoglobulins. Immunoglobulins and their intestinal transfer play a major role in the immune system of neonates since they are born agammaglobulinemic. In this study immunoglobulin G (IgG) content was determined in alpaca colostrum and the correlations of the IgG concentration by fat, protein, lactose and minerals were calculated. Colostrum samples were collected daily from 20 multiparous alpaca mares during the first four days after parturition. The IgG concentrations were determined by radial immunodiffusion using a Camelid IgG Test Kit. The IgG concentration decreased significantly from 26,319 mg/dL on day 1 to 3848.8 mg/dL on day 4. There were significant correlations between IgG concentration and the other components of the colostrum. While the correlations between IgG and fat (r = −0.69, p ≤ 0.001) and lactose (r = −0.64, p ≤ 0.001) were negative, the correlations with protein (r = 0.91, p ≤ 0.001), magnesium (r = 0.86, p ≤ 0.001) and cobalt (r = 0.87, p ≤ 0.001) were strongly positive. Due to the strong association, the colostrum protein concentration could be used for a brief estimation of the IgG content.

Concentrations of Fat, Protein, Lactose, Macro and Trace Minerals in Alpaca Colostrum and Milk at Different Lactation Stages

Although alpacas are not used for milk production a detailed knowledge on the composition of the colostrum and milk is needed for development of colostrum and milk replacers. The aim of the present study was to measure the concentration of fat, protein, lactose, and minerals in alpaca colostrum and milk. Colostrum samples were taken daily over four days after parturition from 20 multiparous alpaca mares. Milk samples were obtained monthly, during the first four months of lactation from 17 alpacas. Composition of colostrum and milk differed in numerous indicators. The concentrations of fat and lactose increased from day 1 (0.5%, 4.0%) to day 4 (5.3%, 5.0%), protein decreased from 20.4% on day 1 to 8.3% on day 4. In milk these three indicators did not change during the lactation. Minerals have been little studied in alpaca colostrum and milk in the past, many of which had the highest concentrations in colostrum immediately after birth. The results of the present study do not support that goat’s milk is the preferred substitute for feeding crias. This study contributes to the knowledge of the composition of alpaca colostrum and milk which can be of particular use in developing replacers.

Survey of colostrum quality and management practices on commercial dairy farms in the Eastern Cape Province of South Africa

Bovine maternal antibodies are not transferred across the placenta during pregnancy and newborn calves are unable to produce their own antibodies within the first weeks after birth. As neonates are born agammaglobulinemic they need to acquire immunoglobulins (Ig) from their dam’s colostrum to acquire passive immunity. Colostrum fed to dairy calves, which is not timeous, of inadequate quantity and of unverified quality, might result in decreased neonate health. The aim of this study was to conduct a survey of the management of colostrum on commercial dairy farms, to estimate the quality of the Ig titre of colostrum fed to neonates and finally to recommend methods and techniques critical to the successful management of colostrum. The methods used included a questionnaire which was conducted as a structured interview on a one-on-one basis among 50 randomly selected commercial dairy farmers in the Eastern Cape Coastal Region of South Africa. The estimation of the colostrum Ig titre of colostrum fed to neonates was made by the on-farm measurement of specific gravity (SG) by making use of a commercially available KRUUSE colostrometer (Fleenor and Stott, 1980). A pooled colostrum sample, from each of the four quarters, from 90 randomly selected post-partum cows was collected on a leader commercial dairy farm. This method was used to compare colostrum samples from cows run under similar management practices. These samples were collected for analysis within 6 hrs of calving and were done over 3 seasons (autumn, winter and spring). Survey - The colostrum mass and timing of the initial feed are the most important factors when aiming to achieve adequate passive immunity. The results of the survey indicated that most of the farmers of this region feed an inadequate mass of colostrum (volume and Ig concentration) and only 52% of farmers surveyed feed colostrum less than 6 hrs post- partum. The majority (78%) of surveyed farmers did not follow up their initial colostrum feeding.Colostrum sampling - At the trial site only 10% (9 from 90 colostrum samples measured), were found to be of adequate SG quality. Cow age (parity), season of calving and colostrum temperature had an influence on the estimated colostrum SG. However, season of calving was found to have the greatest influence on SG values. These results were consistent with findings from previous studies that SG values from the cooler months were higher than those of the hotter months. Tables 4.7 (P=0.330), 4.8 (P=0.012) and 4.9 (P=0.005) showed that regression analysis confirmed that LS means across seasons were inadequately below the required 50 mg/ml Ig required for sufficient passive immunity. Tables 4.1 (P=0.164), 4.2 (P=0.011) and 4.3 (P=0.021) shows that season of calving had a much greater effect on CR than did parity, Table 4.5 (P=0.177). Table 4.4 shows that colostrum temperature has an significant effect on SG value. Recommendations for methods and techniques critical to the successful management of colostrum were made. These recommendations were based on the analysis of the data obtained from the questionnaire and the on-farm colostrum sampling study. The most important and critical management practices surveyed includes the timing of the cow and calf separation where it was found that only 30 from the 50 (60%) of the farms surveyed separate calves and dams at day (0), 19 from 50 farms (38%) separate at day (3-5) and 1 from 50 farms (2%) separate only at day 7 or later. Thus 40% of surveyed farms allow cows to nurse their calves. With regards to early exposure to pathogens this is a high risk management practice. Further to that, only 2 from 50 surveyed farms (4%) measure the colostrum quality fed to their calves and 48 from 50 farms (96%) feed colostrum of unmeasured quality. The mass of colostrum fed to calves is an important parameter for successful transmission of Ig. In the survey it was found that 28 from 50 farms (56%) feed 2L – 4L of colostrum and 11 from 50 farms (22%) feed 2L of colostrum. Thus 78% of farms feed approximately 50% of the amount of colostrum required for successful transmission of Ig. Finally only 1 from 50 farms (2%) freeze excess colostrum and 1 from 50 farms (2%) pool excess colostrum. Small doses of colostrum are administered to the oral mucosa and upper respiratory tract of the infant, which may exert a protective effect on the mucosal membrane. In addition, immunoglobulin A, colostrum cytokines, antioxidant agents and all anti-infective agents may interact with lymphoid cells within the oropharynx, stimulating the infant's immune function. Recently, studies have reported the importance of colostrum administered by the oropharyngeal route, especially for extremely low birth weight preterm infants, with nutritional and immune defense effects already demonstrated. The more premature the baby, the more it will benefit from early exposure to colostrum. OBJECTIVE: To estimate the success rate after implantation of a colostrum administration protocol in low birth weight preterm infants at the Hospital Sa?de, in Caxias do Sul, RS. METHODOLOGY: The study took place between March and July 2017, after the ethical approval of the project. The participants were 41 mother / baby pairs whose mothers, after signing the free and informed consent, made attempts to exhaust the breast to provide colostrum to their children. Preterm infants between 24 and 32 weeks of age who had an indication of colostrum therapy prescribed by the physician were included, and attempts were made to exhaust the breast before six hours of the newborn's life. Two data collection instruments created by the researcher were used, one with information about mothers and newborns and the other for the Neonatal Intensive Care Unit nursing team to respond. After the observation of the attempt to obtain the colostrum, the questions of the collection instrument were considered, as if the puerpera managed to exhaust 0.2 mL of colostrum or not, or if the breast was exhausted before six hours. Through the charts of each baby, the data on administration were collected. It was observed if there was a decrease in oxygen saturation and / or an increase in respiratory and cardiac frequencies at the time of administration of colostrum. The success of colostrum therapy was considered to be the administration of at least one dose of colostrum within the first 72 hours of life, and the success rate was reported as a percentage of the cases that were successful among all included. RESULTS: Among the 41 premature infants included in the protocol, 19 (46.3%) received at least one dose of colostrum within the first 72 hours of life and 22 (53.7%) did not receive colostrum. There was an association between being able to deplete the breast earlier and the baby to be able to receive colostrum, regardless of when it was received. All 22 cases of colostrum failure were due to failure to obtain colostrum within 72 hours postpartum. There was no association between whether or not colostrum could be collected and maternal age or gestational age. There were no adverse events, such as alterations in the respiratory and cardiac frequencies or decrease in oxygen saturation, in the 19 infants who received oropharyngeal colostrum. Regarding the perception of the professional care team about colostrum therapy, more than half reported being partially knowledgeable about the practice and only one felt totally knowledgeable. However, most of the care team (83.4%) reported being satisfied, very satisfied or totally satisfied with the implementation of the colostrum protocol. CONCLUSIONS: In general, this study demonstrated difficulties in the implantation of the protocol of colostrum therapy to premature infants at Hospital Sa?de in Caxias do Sul. The obstacles to the success of a protocol of colostrum therapy were revealed, which mainly resided in the fact that the puerperae had difficulty in exhausting the breast in the first few days after preterm birth, and as a consequence, most newborns were unable to receive maternal colostrum within the first three days of life. In successful cases, the mother's satisfaction with the fact that the child received her colostrum was rewarding. There was a insufficient deepening in the training of the professional care team. These results brought important data that can be used in the execution of a new protocol, in the same unit as well as in other units with the same characteristics. Additional studies should be performed to reveal ways to achieve better success in the application of colostrum therapy protocol, supplanting the difficulties of early breast exhaustion, and can thus benefit from the administration of oropharyngeal colostrum to premature infants. / INTRODU??O: A colostroterapia, tamb?m chamada de terapia imunol?gica oral, ? a administra??o do colostro por via orofar?ngea para rec?m-nascidos prematuros e pode ser iniciada nas primeiras seis horas de vida. Foram inclu?dos prematuros entre 24 e 32 semanas que tinham indica??o de colostroterapia, prescrita pelo m?dico, sendo iniciadas as tentativas de esgota da mama antes das seis horas de vida do rec?m-nascido. Utilizaram-se dois instrumentos de coleta de dados criados pela pesquisadora, um com informa??es sobre as m?es e sobre os rec?m-nascidos e o outro para a equipe de enfermagem da Unidade de Terapia Intensiva Neonatal responder. Ap?s a observa??o da tentativa de se esgotar ou n?o o colostro, foram contempladas as quest?es do instrumento de coleta, como se a pu?rpera conseguiu esgotar 0,2 mL de leite ou n?o, ou se conseguiu esgotar a mama antes das seis horas. Atrav?s do prontu?rio de cada beb?, foram coletados os dados sobre a administra??o. Observou-se se houve queda de satura??o de oxig?nio e/ou aumento das frequ?ncias respirat?ria e card?aca na hora da administra??o da colostroterapia. Considerou-se como sucesso da colostroterapia a administra??o de pelo menos uma dose de colostro dentro das primeiras 72 horas de vida, sendo o ?ndice de sucesso referido como porcentagem dos casos que obtiveram sucesso entre todos os inclu?dos. RESULTADOS: Entre os 41 prematuros inclu?dos no protocolo, 19 (46,3%) receberam pelo menos uma dose de colostro dentro das primeiras 72 horas de vida e 22 (53,7%) n?o receberam a colostroterapia. Houve associa??o entre conseguir esgotar a mama mais precocemente e o beb? conseguir receber a colostroterapia, independentemente do momento em que a recebeu. Todos os 22 casos de insucesso da colostroterapia se deveram ? n?o obten??o de colostro dentro das 72 horas p?s-parto. N?o houve associa??o entre o fato de conseguir ou n?o coletar o colostro e a idade materna, ou a idade gestacional. N?o houve nenhum evento adverso, como altera??o nas frequ?ncias respirat?ria e card?aca ou diminui??o da satura??o de oxig?nio, nos 19 RN que receberam colostro por via orofar?ngea. Em rela??o ? percep??o da equipe assistencial quanto ? colostroterapia, mais da metade referiu ser conhecedor parcial da pr?tica e apenas um sentia-se totalmente conhecedor. Entretanto, a maior parte da equipe assistencial (83,4%) referiu estar satisfeita, muito satisfeita ou totalmente satisfeita com a implanta??o do protocolo da colostroterapia. CONCLUS?ES: Em geral, este estudo demonstrou dificuldades na implanta??o do protocolo de colostroterapia a prematuros no Hospital Sa?de de Caxias do Sul. Foram revelados os empecilhos para o sucesso de um protocolo de colostroterapia, os quais residiram principalmente no fato de que as pu?rperas tiveram dificuldade em esgotar a mama nos primeiros dias ap?s o parto prematuro e, como consequ?ncia, a maioria dos rec?m-nascidos n?o conseguiu receber o colostro materno dentro dos primeiros tr?s dias de vida. Nos casos de sucesso, a satisfa??o da m?e pelo fato do filho receber o seu colostro foi compensadora. Faltou maior aprofundamento no treinamento da equipe assistencial. Os resultados trouxeram dados importantes que podem ser aproveitados na execu??o de um novo protocolo, na mesma unidade, assim como em outras unidades com as mesmas caracter?sticas. Estudos adicionais devem ser realizados para revelar formas de obter melhor sucesso na aplica??o do protocolo de colostroterapia, suplantando as dificuldades da esgota precoce da mama, podendo assim auferir os benef?cios da administra??o de colostro por via orofar?ngea aos prematuros.

Thus 40% of surveyed farms allow cows to nurse their calves. With regards to early exposure to pathogens this is a high risk management practice. Further to that, only 2 from 50 surveyed farms (4%) measure the colostrum quality fed to their calves and 48 from 50 farms (96%) feed colostrum of unmeasured quality. The mass of colostrum fed to calves is an important parameter for successful transmission of Ig. In the survey it was found that 28 from 50 farms (56%) feed 2L – 4L of colostrum and 11 from 50 farms (22%) feed 2L of colostrum. Thus 78% of farms feed approximately 50% of the amount of colostrum required for successful transmission of Ig. Finally only 1 from 50 farms (2%) freeze excess colostrum and 1 from 50 farms (2%) pool excess colostrum. La naissance est l’une des étapes les plus délicates à laquelle les nouveau-nés mammifères doivent faire face. Le nouveau-né doit opérer des changements physiologiques et comportementaux pour s’adapter à l’environnement aérien, et l’un des premiers défis est d’ingérer du colostrum et du lait. Il est surprenant que la nature des stimuli et les mécanismes impliqués dans le déclenchement de la tétée soient encore mal connus, alors que la survie du nouveau-né est conditionnée par le succès de la première tétée. Par conséquent, cette étude a pour but de comprendre comment un nouveau-né immature et inexpérimenté réussit à s’orienter vers une tétine, à la saisir et à la téter de façon efficace? Cette question générale est posée chez la souris en focalisant sur : i) la nature des substrats chimiques utilisés par les souriceaux pour atteindre les tétines maternelles ; ii) la variation de la puissance attractive de ces substrats au cours du développement ; et iii) l’implication des effets de l’expérience dans l’établissement des réponses adaptatives précoces. Premièrement, nos résultats mettent en lumière que les odeurs mammaires de femelles allaitantes induisent plus d’approches et de saisies de la tétine chez les souriceaux que celles émanant de femelles non allaitantes. Deuxièmement, les odeurs de liquide amniotique et de lait déclenchent la première saisie orale de la tétine chez des souriceaux à la naissance, alors que les odeurs de salives maternelle et infantile n’induisent ce comportement qu’après une brève expérience de tétée. Troisièmement, les souriceaux âgés de 0, 2 et 6 jours postnatals (P), ayant eu une expérience de tétée, affichent une attraction sélective envers des odeurs de laits collectés en début de lactation plutôt qu’en fin de lactation, alors que les souriceaux plus âgés P15 ne montrent aucune réponse sélective envers ces odeurs. En résumé, certains substrats biologiques présents sur les tétines de femelles allaitantes sont immédiatement attractifs après la naissance, tandis que d’autres ont besoin d’être appris pour être réactogènes. Par conséquent, la réponse initiale de recherche de la tétine chez le souriceau est contrôlée par des processus d’apprentissage postnatal. A ce stade, l’implication de l’apprentissage prénatal et de processus prédisposés n’a pu être prouvée, bien qu’elle ne soit pas exclue. Newborns have then to adapt physiologically and behaviorally to the aerial environment, and one of their first challenges is to ingest colostrum and milk. It is paradoxical that the survival of pups is conditioned by the success of this first suckling, and that we have so little understanding of the stimuli that underlie and promote it. Thus, the present work aims to contribute to answer how immature and naïve newborns do manage to orient to a nipple, to grasp it, and to suckle efficiently? This general issue will be addressed in the mouse in focusing on: i) the nature of the chemical substrates that newborn mice use to reach nipples; ii) whether the attractive potency of these substrates changes as a function of development; and iii) whether exposure effects underly the establishment of early adaptive responses? The results highlight that mammary odors of lactating females are more behaviorally active for newly born pups than those of non-lactating females. Secondly, amniotic and milk odors provoke the first nipple grasping in newborns right at birth, while maternal and pup salivary odors induce this behavior after short sucking experience. Thirdly, younger pups on postnatal day (P) 0, 2 and 6 (with sucking experience), display a selective orientation toward the odor of milk collected during early-lactation rather than to the odor of late-lactation milk, whereas older pups P15 do not exhibit any selective attraction to these odors. To summarize, some of the biological substrates which are present on a nursing mouse nipples are attractive immediately after birth, while some others need to be postnatally learned to be active. Thus, the initial nipple search response of mouse pups is controlled by processes involving postnatal learning. At this stage, the involvement of prenatal learning and of predisposed processes that do not depend on previous exposure effect are not conclusive, although not excluded. O presente trabalho teve por objetivo verificar o efeito de diferentes níveis de IGF-I no colostro de vacas que receberam rbST durante o período pré-parto, no desenvolvimento do trato intestinal e na expressão do gene do IGF-I e de seu receptor no fígado e intestino de bezerros. Quarenta e duas vacas da raça Holandesa, gestantes e multíparas foram distribuídas ao acaso em dois grupos de 21 animais, o grupo rbST que recebeu hormônio de crescimento (500 mg rbST) e o grupo controle que recebeu injeção de vitamina E. As aplicações tiveram início aos 35 dias pré-parto, em intervalos de 14 dias até a data de parição. Os bezerros recém-nascidos foram distribuídos aleatoriamente, de acordo com as seguintes idades em que foram abatidos: após o nascimento e sem a ingestão de colostro, dois e sete dias de vida com ingestão de colostro das respectivas mães. Após o abate, amostras do fígado e segmentos do intestino delgado (jejuno e íleo) foram coletadas para quantificação de DNA, RNA, proteína total e o RNAm do IGF-I e do receptor tipo I. O delineamento experimental utilizado foi o inteiramente casualizado, numa estrutura fatorial 2x3, correspondendo aos grupos das mães (rbST ou controle) e as idades dos bezerros (após o nascimento, dois e sete dias de vida). No fígado, as concentrações de RNA e proteína (mg/g tecido) foram maiores no segundo dia de vida e a relação proteína/RNA aumentou no sétimo dia de vida (P<0,05). O jejuno apresentou interações entre os tratamentos nas concentrações de DNA, proteína e relações proteína/RNA e RNA/DNA (P<0,05). Os bezerros que consumiram colostro proveniente de vacas que receberam rbST apresentaram, no jejuno, maiores concentrações de DNA no segundo dia de vida e diminuição a níveis intermediários entre nascimento e dois dias, comparados aos sete dias de vida. Este efeito também foi observado na relação proteína/RNA. No grupo controle, também se verificou aumento de DNA aos dois dias, mas não houve diferenças aos sete dias, e a relação proteína/RNA foi semelhante entre as idades. A concentração de proteína no jejuno do grupo rbST aumentou no segundo dia de vida e diminuiu no sétimo, enquanto que no grupo controle, este aumento foi verificado apenas no sétimo dia de vida. A relação RNA/DNA diminuiu apenas no grupo controle (P<0,05). A expressão do gene do IGF-I foi maior ao nascimento e aos sete dias de idade no fígado dos bezerros do grupo rbST (P<0,05). As concentrações do gene do receptor tipo I diminuíram com a idade dos bezerros (P<0,05). Ao nascimento, o intestino dos bezerros apresentou condição de resposta celular à presença do IGF-I proveniente do colostro. Os resultados sugerem que as células do jejuno de bezerros do grupo rbST apresentaram um diferente estágio de maturação. / The objective of this study was to verify the effect of different levels of IGF-I in colostrum of cows treated with rbST during dry period, in the development of the intestinal tract and IGF-I and receptor gene expression in the liver and intestine of calves. Forty-two Holstein cows, in gestation and multiparous, were randomly assigned in two groups of 21 animals. Group rbST received injection of growth hormone (rbST), and group control received vitamin E injection. Both treatments started 35 days pre-partum, and were administered every 14 days until parturition. Newborn calves were randomly assigned to the following ages of slaughter: just after birth and without colostrum ingestion; two and seven days of life with colostrum ingestion from their respective mothers. After slaughter, samples from liver and small intestine (jejunum and ileum) were collected for quantification of DNA, RNA, total protein and mRNA of IGF-I and receptor type I. A completely randomized design was used with 2X3 factorial arrangements of treatments, which the factors were the mother’s group (control and rbST) and age (just after birth, two and seven days of life). In the liver, RNA and protein concentrations (mg/g tissue) were higher in the second day of age and protein/RNA ratio increased in the seventh day (P<0.05). The jejunum showed interaction among the treatments in the concentrations of DNA and protein, and protein/RNA RNA/DNA ratios (P<0.05). Calves fed colostrum from mothers that received rbST presented, in jejunum, higher DNA concentration in the second day of life, and decrease to intermediate levels between birth and two days, compared to seven days of age. This effect was also observed in protein/RNA ratio. In the control group, it was also verified DNA increase at two days, but there was not difference in the seventh day, and protein/RNA ratio was similar among ages. Protein concentration in jejunum of rbST group increased in the second day and decreased in the seventh, but in the control group, this increase was verified only in the seventh day of age. RNA/DNA ratio decreased in the control group (P<0.05). Expression of IGF-I was higher at birth and seven days old in the liver of the calves from rbST group (P<0.05). The concentrations of receptor type I mRNA decreased with calves’ age (P<0.05). At birth, the small intestine of calves showed a condition of cellular response to the presence of IGF-I in colostrum. These results suggest that cells in jejunum of calves from rbST group presented a different phase of maturation.

O presente projeto teve como proposta estudar a utilização do colostro bovino liofilizado como manejo alternativo para caprinos recém-nascidos. Estudos relacionados com a aquisição de proteção passiva e o desenvolvimento do epitélio intestinal, como aspectos citológicos e histológicos, atividade celular e expressão do receptor específico para o IGF-I (IGF-IR), foram realizados. Às 0, 7 e 14 horas de vida, 15 machos recém-nascidos receberam 5% do peso inicial de colostro bovino liofilizado (CBL) e 14 colostro caprino (CC), ambas as refeições com 55 mg/mL de IgG. Amostras de sangue foram coletadas às 0, 7, 14, 18, 24, 36, 48, 72 e 96 horas de vida para a quantificação da imunoglobulina G (IgG), proteína total (PT) e do fator de crescimento semelhante à insulina tipo I (IGF-I). Amostras do duodeno, jejuno e íleo foram coletadas às 18, 36 e 96 horas de vida para análises utilizando microscopia óptica e microscopia eletrônica de varredura e transmissão. A atividade de enzimas extracelulares, quantificação da proteína, DNA e RNA total e expressão do IGF-IR também foram determinadas. Amostras de um grupo adicional (0 h), que não ingeriu colostro, foram coletadas. No grupo CBL, as concentrações séricas de IgG às 14, 18, 24 e 48 horas foram maiores que às 0 e 7 horas, enquanto no grupo CC, apenas às 18 horas a concentração sérica foi maior que às 0 e 7 horas (P<0,05). A capacidade de absorção de IgG diminuiu entre 7 e 14 horas em CC (P<0,05), enquanto em CBL, manteve-se sem alteração nestes horários (P>0,05). As variáveis PT e IGF-I não foram influenciadas pela ingestão de colostro bovino liofilizado (P>0.05). Os indicativos de atividade celular, concentração de proteína, DNA e RNA total, proteína/DNA, proteína/RNA e RNA/DNA, e a atividade das enzimas aminopeptidase N, dipeptidil peptidase IV, aminopeptidase A, lactase, maltase e sacarase diferiram nos horários experimentais e em relação ao grupo adicional (P<0,05). Apenas as razões proteína/DNA, proteína/RNA no jejuno e a atividade da aminopeptidase A no íleo foram maiores no grupo CBL (P<0,05). A atividade da fosfatase ácida foi observada nas primeiras horas de vida, especialmente no duodeno. Com relação à morfologia do epitélio intestinal e densidade das vilosidades, o fornecimento de colostro bovino liofilizado não determinou alterações. As variações na ultraestrutura dos enterócitos revelaram a presença, no segmento jejuno, de material absorvido e do complexo endocítico apical às 0, 18 e 36 horas. No duodeno, a expressão gênica do IGF-IR ao nascimento foi maior que às 18 e 36 horas nos grupos CC e CBL (P<0,05). Enquanto, a expressão do receptor deste peptídeo bioativo no jejuno do grupo CBL foi maior às 18 e 36 horas em relação a 0 h (P<0,05). Os resultados obtidos do presente trabalho contribuem para que a fonte alternativa de imunoglobulinas, colostro bovino liofilizado, seja utilizada como um substituto para o colostro caprino sem alterações significativas na aquisição de proteção passiva e nas características histofisiológicas do tecido epitelial do intestino delgado de cabritos. / This project was proposed to study the use of lyophilized colostrum as an alternative management for newborn goat kids. Studies related to the acquisition of passive protection and development of the intestinal epithelium, as cytological and histological aspects, cellular activity and expression of the receptor specific for IGF-I (IGF-IR), were performed. At 0, 7 and 14 hours of life, 15 newborn males received 5% of initial weight of lyophilized bovine colostrum (LBC) and 14 goat colostrum (GC), both meals with 55 mg/mL of IgG. Blood samples were collected at 0, 7, 14, 18, 24, 36, 48, 72 and 96 hours of life to quantification of immunoglobulin G (IgG), total protein (TP) and insulin like growth factor type I (IGF-I). Samples of the duodenum, jejunum and ileum were collected at 18, 36 and 96 hours of life for analysis using optical microscopy and scanning and transmission electron microscopy. The activity of extracellular enzymes, quantification of total protein, DNA and RNA and expression of the IGF-IR were also determined. Samples from an additional group (0 h), not suckling, were collected. In LBC group, serum IgG concentrations at 14, 18, 24 and 48 hours were higher than at 0 and 7 hours, while in the GC group, only at 18 hours serum concentration was higher than at 0 and 7 hours (P<0.05). The capacity of IgG absorption decreased between 7 and 14 hours in GC (P<0.05), while in LBC, remained unchanged at these times (P>0.05). The variables TP and IGF-I were not influenced by ingestion of lyophilized bovine colostrum (P>0.05). The indicators of cellular activity, total protein, DNA and RNA concentration, protein/DNA, protein/RNA and RNA/DNA, and the enzyme activity of aminopeptidase N, dipeptidyl peptidase IV, aminopeptidase A, lactase, maltase and sucrase differed in sampling times and in relation to the additional group (P<0.05). Only the rations protein/DNA, protein/RNA in the jejunum and aminopeptidase A activity in the ileum were higher in the LBC group (P<0.05). The acid phosphatase activity was observed in the first hours of life, especially in the duodenum. Regarding the intestinal epithelium morphology and villi density, the supply of lyophilized bovine colostrum did not determine alterations. Changes in enterocytes ultrastructure revealed the presence in the jejunum segment of absorbed material and apical endocytic complex at 0, 18 and 36 hours. In the duodenum, IGF-IR gene expression at birth was higher than at 18 and 36 hours in the GC and LBC (P<0.05). While the expression of this bioactive peptide receptor in the jejunum of the LBC group was higher at 18 and 36 hours relative to 0 h (P<0.05). The results of this work indicate that the alternative source of immunoglobulins, lyophilized bovine colostrum, can be used as a substitute for goat colostrum without significant changes in the acquisition of passive protection and histophysiology characteristics of the small intestine epithelium of goat kids.
