Safety and Efficacy of Commercially Available Pre-Workout Supplements

Class of 2015 Abstract / Objectives: The purpose of this review was to determine the safety of various pre-workout supplements that utilize proprietary blends in comparison with some of the most common individual ingredients; caffeine, creatine, and B-alanine. We hypothesized that there will be a greater number of adverse events reported for proprietary products than for the individual active ingredients. Additionally, we also wanted to look at the efficacy of the same aforementioned products. We hypothesized that there would be no statistically significant differences in performance between the two arms. Methods: Four databases were searched for subjects that were 18-35 years of age that were already physically active. The number of participants included in each trial ranged from 6 to 98. Results: Caffeine was the only individual compound that affected health markers, increasing mean arterial pressure (MAP) (P<0.05), and HR in 2 of the 3 studies (P<0.05) significantly. Both caffeine and creatine showed a benefit in maximal exertion, but only caffeine improved endurance at doses of 3mg/kg (P<0.05). Proprietary blends did not show a benefit, but serious adverse events such as liver failure were reported. Conclusions: Individually caffeine, creatine, and B-alanine all look to be safe at the recommended doses in healthy and active individuals, with caffeine and creatine benefitting performance. Pre-workout blends should be safe in theory, but due to the unregulated nature of the supplement industry there are a number of serious adverse events that occur. Untested amphetamine-like compounds seem to be the most common addition, with contamination of other ingredients such as anti-depressants occurring as well.

The Prevalence and Operational Feasibility of Utilizing Pre-commercially Thinned Pine as a Woody Biomass Energy Source

The southern pine beetle (SPB) poses a significant threat to pine forests of the southeastern US. Pre-commercial thinning (PCT) is a commonly used silvicultural practice to mitigate and prevent SPB spread in young southern pine stands. Typically, PCT represents an added management cost to landowners and thinned material is not utilized for forest products. Increased demand for woody biomass energy may provide landowners and harvesting contractors an opportunity to utilize PCT residues as a woody biomass energy feedstock, which may wholly or partially offset PCT costs. However, little information is available regarding harvestable biomass quantities in PCT stands and few studies have assessed harvesting productivity and costs in very young pine stands. To develop estimates of biomass abundance in PCT candidate stands, loblolly pine (Pinus taeda) stands aging 5 to 12-years old, and enrolled in the Virginia Department of Forestry Pine Bark Beetle Prevention Program (VDOF PBBPP), were inventoried across the Piedmont and Coastal Plain regions of Virginia. To attain productivity and cost estimates of utilizing small-diameter stems for woody biomass energy, a biomass harvesting case study was then conducted on a 15-year old loblolly pine stand. Results of the inventory and case study indicate that stands at the upper age limit for the PCT program may contain harvestable quantities of biomass (39.63 green tons/acre), although high harvesting costs ($23.46/green ton) relative to regional delivered biomass prices may limit the economic feasibility of utilizing PCT biomass for energy. / Master of Science

Fate of vitamin C in commercial fruit juices

Vitamin C occurs in relatively high concentrations in fresh and processed fruits and vegetables but is found to a lesser extent in animal tissues and animal-derived products. Nearly 90 % of vitamin C in the human diet is obtained from fruits and vegetables but this can be indirect by way of commercially prepared fruit juices. These juices are often enriched with vitamin C which has been synthetically prepared. There is a wide range of such juices on the New Zealand market, and they are a significant source of dietary vitamin C for many in the population. The focus of this research is on the Keri range of juice products.The present study monitors the fate of vitamin C during storage of Keri juices up to the best-before date, and under a range of other storage and consumption situations. Two methods were adopted for determining ascorbic acid (AA, the chemical identity of vitamin C). These were the titrimetric method, which is based upon the reduction of the dye 2,6-dichlorophenolindophenol by AA in acidic solution, and liquid chromatography, which is used to separate AA from its immediate oxidation product dehydroascorbic acid. In the latter method these two analytes can be measured independently. The liquid chromatography was less successful than the simpler titrimetric method, so most of the work was done by titration. However, the concentration of dehydroascorbic acid, which has vitamin C activity in vivo, remained uncertain. Moreover, the titrimetric method could not be applied to juices with high purple anthocyanin concentrations, like blackcurrant, because the colour change at the titration end point could not be detected. pH adjustment to change colour was ineffective, and decolourisation with charcoal led to the rapid and complete destruction of AA. The concentration of AA in Keri juices at the time of manufacture were always much higher than claimed on the labels. Storage for up to nine months at room temperature resulted in a loss in AA of between 37 and 68 %, depending on the juice and exposure to fluorescent light. However, the time of storage was a much more dominant factor than light exposure. The kinetics of loss, straight lines, were most easily explained by an aerobic model of AA degradation from oxygen diffusing across the polyethylene tetraphthalate bottle wall. Overall, the label claims made were defensible in terms of the best-before date, because it took at least 100 days of storage before the AA concentration in the most susceptible juices fell below the claimed value. This is because these drinks are fast moving consumer goods and storage beyond 100 days is unlikely. (Nonetheless, the supplier (Keri Juice Company) has since adopted its new unitised method of formulating juice. This has resulted in an initially higher concentration of vitamin C as compared to the juices under investigation.) In the nine months storage experiment there was some evidence for the presence of dehydroascorbic acid in blackcurrant drinks, but not in another three juices. Pasteurisation during preparation of these drinks resulted in up to 7 % loss of AA, probably due to oxygen dissolved in water, and accelerated by heat of pasteurisation. Higher temperatures in later storage also accelerated losses. Progressive exposure of juice to air during simulated consumption of 3 L bottles over a week also accelerated losses. Finally, exposure to sunlight in a diurnal temperature environment accelerated losses five-fold higher than in total darkness. Filtration of ultraviolet light approximately halved the loss due to sunlight. Overall however, it can be concluded that AA in the Keri range of juices is very resistant to degradation of AA.

Short- and Long-Term Effects of Commercially Available Gold Nanoparticles in Rodents

Gold nanoparticles (GNPs) are currently being intensely investigated for their potential use in biomedical applications. Nanotoxicity studies are urgently needed to validate their safety in clinical practice. The objective of this research was to assess the acute, subacute, and chronic effects of a single intravenous exposure to commercially available GNPs in two in vivo models, mice and rats. Gold nanoparticles were purchased and independently characterized. Animals were exposed to either 1000 mg GNPs/kg body weight (GNP group) or an equivalent volume of phosphate buffered saline (PBS group) intravenously via the tail vein. Subsets of animals were euthanized 1, 7, 14, 21, 28 days (female BALB/c mice and female F344 rats) or 20 weeks (female and male C57BL/6 mice) post-exposure and samples were collected for biochemistry, histopathology, electron microscopy, and atomic absorption spectrometry analysis. Independent characterization demonstrated that the physicochemical properties of the purchased GNPs were in good agreement with the information provided by the supplier. Important differences in GNP-induced immune responses were identified when comparing mice and rats 1 to 28 days post-exposure. Gold nanoparticles stimulated the formation of liver microgranulomas in mice, along with transiently increased serum levels of the proinflammatory cytokine interleukin-18. No such alterations were found in rats. Species differences in GNP biodistribution and excretion were also detected, with higher relative accumulation of GNPs in spleen and longer fecal excretion in rats. In the long-term (20 weeks after dosing), exposure to GNPs incited chronic inflammation in mice, characterized by the persistence of microgranulomas in liver, spleen, and lymph nodes, as well as further increased serum levels of interleukin-18. Impairment of body weight gain was also observed in the GNP-exposed group. No sex differences were detected. In conclusion, GNPs are not innocuous and have the ability to incite a robust macrophage response in mice. However, considering the mildness of the toxic effects identified despite the high dose selected for the study, GNPs continue to have great potential for biomedical uses. Further studies are needed in order to determine specific mechanisms of toxicity and the role of chronic inflammation in the development of adverse effects after co- or post-exposures. / Ph. D.

Machining of powder metal titanium

Thesis (MScEng (Mechanical and Mechatronic Engineering))--University of Stellenbosch, 2011. / ENGLISH ABSTRACT: The purpose of this study is to investigate the machinability of commercially pure (CP) titanium, manufactured using the press-and-sinter PM process. To this end, CP titanium powder (-200 mesh) was compacted and sintered in vacuum (10-4 torr) for two hours at 1200°C. Small cylindrical samples were compacted at pressures varying from 350 to 600 MPa in order to determine the compressibility of the powder. Following these tests, four larger stepped-cylinder samples were compacted at pressures close to 400 MPa and sintered under similar conditions. These samples had sintered densities varying between 3.82 and 4.41 g/cm3. They were used to evaluate the machinability of the sintered titanium using face turning machining tests. The samples were machined dry, using uncoated carbide (WC-Co) cutting tool. Cutting speeds between 60-150 m/min were evaluated while keeping the feed rate and depth of cut constant at 0.15 mm/rev and 0.5 mm, respectively. The final machined surface finish and the tool wear experienced during the face turning machining tests were monitored in order to evaluate PM titanium’s machining performance. This study showed that it is possible to use the press-and-sinter PM process with CP titanium powder, with a particle size of less than 75 μm (-200 mesh), to manufacture sintered titanium. However, particle shape influences the compressibility of the powder and pressing parts of larger volume, such as the machining test sample shape, is challenging when using such small particle size powder. Processing conditions, such as compaction pressure, sintering temperature and sintering time, influence the sintered density. Results from the machinability tests show that tool wear increases with a decrease in the porosity of the sintered titanium. A more porous sintered material has both lower strength and thermal conductivity. As these factors have opposing effects on the machinability of materials, it is concluded that the strength of the sintered titanium has a stronger influence on its machinability than the thermal conductivity. The cutting tool wear was uniform but showed indications of crater wear. The machined surface of the denser parts had minimal defects compared to less dense parts. Chip shape is long for the dense parts, and spiral for the less dense parts. The chips formed were all segmented, which is typical for titanium. The machinability of the sintered CP titanium was compared to that of wrought titanium alloys. As expected, it was found that the machinability of the sintered titanium was poor in comparison. / AFRIKAANSE OPSOMMING: Die doel van hierdie studie is om die masjineerbaarheid van kommersieel suiwer (KS) titaan, wat deur die pers-en-sinter poeiermetallurgie (PM) metode vervaardig word, te ondersoek. Om hierdie doel te bereik, is KS titaan poeier (-200 ogiesdraad) gekompakteer en gesinter in ‘n vakuum (10-4 torr) teen 1200°C vir 2 ure. Klein silindriese monsters is tussen drukke van 350en 600 MPa gekompakteer om die samedrukbaarheid van die poeier te bepaal. Die vorm van die spaanders is lank vir digte parte, en spiraalvormig vir minder digte toetsmonsters. Die spaanders wat gevorm het, was almal gesegmenteerd, wat tipies is vir titaan. Die masjineerbaarheid van die gesinterde KS titaan is met dié van gesmede titaanallooie vergelyk. Soos verwag is, is gevind dat die masjineerbaarheid van die gesinterde titaan in vergelyking swak is.

Arthropods associated with commercial Proteaceae in the Western Cape Province, South Africa

Thesis (MSc)--University of Stellenbosch, 2011. / ENGLISH ABSTRACT: The commercial cultivation of Proteaceae is an important industry in the Western Cape, however, farmers are challenged with arthropod infestation which compels them to solely rely on chemical pesticides. Past studies in South Africa have shown that Proteaceae comprise a rich and diverse arthropod fauna. However, as most of these studies were conducted on wild Proteaceae, they may not be representative of cultivated proteas. Moreover, most of these species remained unidentified due to lack of identification expertise. These past studies, however, form a useful baseline for arthropod studies in proteas, e.g. the feeding guilds found in proteas. The aim of this research was to conduct an intensive and extensive survey of the arthropod-fauna associated with commercially-cultivated proteas across an entire year. Specifically, this survey was designed to document the composition of the arthropod fauna (creating a comprehensive reference collection for pest management purposes) and to assess whether the arthropod fauna differed between seasons and pesticide treatments. Infructescences, inflorescences and foliage of mainly commercial Proteaceae were sampled for arthropods seasonally for a period of twelve months by collection of plant material and direct searching. Seven commercial protea blocks, and a wild protea block (remnant patch of fynbos vegetation), were used as the sampling sites, and two sprayed blocks were used for assessing pesticide efficacy. Individual arthropods were identified as far as possible, with 37% identified to species level. A species accumulation curve showed that rare (minor) arthropod species made up of 70% of arthropods occurring in cultivated proteas. More than 8 700 individuals from more than 140 species and about 80 families were collected and identified, revealing that cultivated proteas have a rich and diverse insect fauna. These arthropods represent the full range of plant-feeding guilds: leaf miners, leaf chewers, flower bud borers, sap suckers and seed feeders. Flower visitors/free living guild was the most abundant (72%) and speciose (25%). In addition to phytophages, there was a large suite of insect predators and parasitoids. A large number of the arthropods were endemic to the Cape Floristic Region (CFR) and some (7.86%) have a pest status, in that they cause significant damage to the protea plants (for example, 60% of Safari sunset cultivar (Leucadendron salignum x L. laureolum) new flush stems and leaves were affected by Epichoristodes acerbella (Tortricidae). Capys alphaeus (Lycaenidae) and Phyllocnistis sp. (Phyllocnistidae) appear to be specialist pests, as they attack mainly Protea cynaroides and Susara cultivar (Protea magnifica x P. susannae) respectively. Arthropod abundance did not differ significantly between seasons, although significant seasonal effects were observed in species richness when the protea cultivars were examined separately. Pesticide application did not affect arthropod abundance, but did decrease species richness in sprayed blocks. Pesticides appeared to negatively affect minor (rare) species disproportionately, probably due to their lack of prior exposure to pesticides and hence sensitivity. Due to this inefficacy of pesticides in cultivated proteas, an increasing emphasis on the importance of non-chemical control measures, and our improved knowledge of the predatory and parasitic species in this system, integrated pest management strategies deserve greater research attention. Monitoring and use of threshold values for arthropod pests were suggested here, as well as the use of biological, cultural, physical and chemical (optimal use) control. For instance, in cultural control, polycropping and intercropping in proteas to increase plant diversity in the monocultures to promote a higher density of predators and parasitoids can be used. Certain flowering plants are known to provide greater temporal and spatial distribution of nectar and pollen sources, which can increase parasitoid reproductive potential and abundance of alternative hosts/prey when the pest species are scarce or at an inappropriate stage. / AFRIKAANSE OPSOMMING: Die kommersiële verbouing van Proteaceae (proteas) is 'n belangrike bedryf in die Wes-Kaap. Menige plantasie wemel egter van artropodes, wat boere noop om slegs van chemiese plaagdoders gebruik te maak. Vorige studies in Suid-Afrika toon dat proteas die gasheerplant vir 'n ryke en diverse artropodefauna is. Aangesien die meeste van hierdie studies egter op wilde proteas uitgevoer is, weerspieël dit moontlik nie die stand van sake met verboude proteas nie. Weens 'n gebrek aan kundigheid om die artropodes te eien word baie van die spesies boonop nooit uitgeken nie. Study of Void Growth in Commercially Pure Titanium

The ductile fracture process, which consists of the nucleation, growth and coalescence of microvoids, was extensively studied for materials deforming homogeneously. For materials with a non-homogeneous deformation behavior, such as those having hexagonal closed packed (HCP) crystal structure, experimental and numerical data is lacking. Therefore, the fracture properties of materials with such HCP structure, like titanium (used in aerospace and biomedical applications), zirconium (nuclear industry) and magnesium (manufacturing industry) are not well understood. The main research objective of this Ph.D. thesis is to better understand the mechanisms governing fracture in commercially pure (CP) titanium. In particular, the effect of grain orientation on void growth is investigated. The fracture process of CP titanium was visualized in model materials containing artificial holes. These model materials were fabricated using a femtosecond laser coupled with a diffusion bonding technique to obtain voids in the interior of titanium samples. Diffusion bonding was carried out either above or below the phase transformation temperature resulting in different microstructures. Changes in void dimensions during in-situ straining were recorded in three dimensions using x-ray computed tomography. Void growth obtained experimentally was compared with the Rice and Tracey model which predicted well the average void growth. However, a large scatter in void growth was observed experimentally and was explained in terms of differences in grain orientation which was confirmed by crystal plasticity simulations. It was also shown that grain orientation has a stronger effect on void growth than intervoid spacing and material strength. Intervoid spacing, however, appears to control whether the intervoid ligament failure is ductile or brittle. While this study showed a good agreement between experiments and simulations on average, there is no direct void growth comparison for particular grain orientations. In a follow-up study, an experimental approach was developed to directly relate the growth of a void to its underlying grain orientation. This is achieved by first annealing CP titanium samples below the α-β phase transformation temperature, then performing electron backscatter diffraction iii (EBSD) and finally diffusion bonding the samples together. Samples were then tested in x-ray tomography. This study showed the importance of the local state of strain on void growth. Crystal plasticity simulations that take into account the particular grain orientation and the local state of strain were found to predict well experimental void growth. Crystal plasticity simulations confirmed that the orientation of the voidcontaining grain is more important than the orientation of surrounding grains and more important than the volume fraction of voids, in order to determine void growth. This thesis on the growth and coalescence of voids is important to validate and improve the predictions of ductile fracture models and to design new materials with improved fracture properties.

Surrogatmodern - utnyttjad eller autonom? : En kvalitativ idéanalys om den svenska debattens syn på surrogatarrangemang

This essay is comparing the different perspectives of the debate on surrogacy in two Swedish Newspapers. The purpose of this study is to contribute with knowledge about the different perspectives of the debate about surrogacy in relation to women's autonomy. The theory is defined by decisional autonomy and practical autonomy, which is written by Jonathan Pugh. Through an idea analysis, the material of the essay is analyzed through an analysis tool created with Pugh’s theory. The delimitation of material is selected from articles from Dagens Nyheter and Svenska Dagbladet between 2010 and 2020. The result shows that 12 of the arguments in the articles are based on the decisional autonomy perspective, where six of them are based on both decisional and practical autonomy in their arguments. Practical autonomy is the main argument in four of them. The conclusion is that the side of the debate that is against surrogacy argues that the woman does not have enough autonomy to decide to be a surrogacy mother, without pressure or manipulation. The other side of the debate, that is for the altruistic model argues that she has enough autonomy to make her own decision. Another conclusion in this essay is that most of the arguments, even regarding practical autonomy, are based on decisional autonomy, since practical autonomy is mostly a tool for protecting women or helping them to implement their wishes. That's why decisional autonomy is most frequently used in the debate of surrogacy.
