Constituíram-se dois grupos (n=12). O primeiro grupo recebeu 30 μl de cetorolaco de tometamina 0,5%, sem conservante (OT), a cada seis horas, em uma das córneas ulceradas e 30 μl de solução salina fisiológica 0.9% (OC) na córnea adelfa, totalizando 4 aplicações diárias, por um período de 24 horas. O limiar de sensibilidade corneal foi avaliado por estesiometria antes e posteriormente a lesão ser realizada, a intervalos regulares de quatro horas. Avaliou-se ainda o edema corneal, a hiperemia conjuntival, o blefarospasmo e o diâmetro das úlceras, empregando-se, biomicroscópio com lâmpada em fenda, teste do tingimento pela fluoresceína, imagem digitalizada para posterior avaliação em programa Image J, até a reepitelização completa. Decorridas 24 horas da abrasão nos animais do primeiro grupo (n=6) e 55 horas nos animais do segundo grupo (n=6), estes foram submetidos à eutanásia. Ambas as córneas (OT e OC) foram avaliadas histologicamente e à microscopia eletrônica de varredura. Utilizou-se Análise de Variância para Medidas Repetidas, considerando significativos os valores de p ≤ 0,05. Conclui-se que mesmo sem a adição de conservantes e a despeito do bom efeito analgésico, o cetorolaco de trometamina a 0,5%, sem conservante, retarda a reparação cicatricial do epitélio corneal em coelhos submetidos a abrasões com hidróxido de sódio 1 mol/L. / Studied the effects of ketorolac tromethamine 0.5% without preservatives on ocular inflammation, evaluating the sensitivity and corneal epithelial repair in rabbits subjected to chemical corneal ulceration with sodium hydroxide (NaOH) 1 mol / L. Constituted two groups (n = 12). The first group received 30 μl of tometamine ketorolac 0.5%, without preservative (OT) every six hours in one of ulcerated corneas and 30 μl of saline solution 0.9% (OC) in the other cornea a total of four applications daily for a period of 24 hours. The threshold of corneal sensitivity was assessed by esthesiometry before and after the injury to be held at regular intervals of four hours. It was also evaluated corneal edema, conjunctival hyperemia, blepharospasm and the diameter of the ulcers, using, biomicroscopy with slit lamp, the fluorescein dye test, the scanned image for further evaluation in program Image J, until complete reepithelialization. After 24 hours of abrasion in the animals of first group (n = 6) and 55 hours in animals of the second group (n = 6), they were euthanized. Both corneas (OT and OC) were evaluated histologically and scanning electron microscopy. We used ANOVA for repeated measures, considering significant p values ≤ 0.05. We conclude that even without the added preservatives and despite the good analgesic effect of ketorolac tromethamine 0.5%, without preservative, slowing the repair of corneal epithelium wound healing in rabbits subjected to abrasion with sodium hydroxide 1 mol/L.

The effects of Nd:YAG laser cyclophotocoagulation on corneal sensitivity, intraocular pressure, aqueous tear production and corneal nerve morphology in the canine eye

Corneal ulceration with prolonged healing following Nd:YAG laser cyclophotocoagulation in dogs is a frequent complication. It is hypothesized that these corneal ulcerations may be a form of neurotrophic keratitis due to laser-induced damage to corneal innervation. Fifteen clinically normal dogs had the neodymium:yttrium aluminum garnet(Nd:YAG) laser cyclophotocoagulation performed on the left eye. Each treated eye received 100 Joules of laser energy. Corneal touch threshold (CTT) and Schirmer I tear tests (STT) were performed before the surgery and on days 1,3,5,7,9,11, and 13 post-laser treatment. Applanation tonometry was performed before surgery and twice daily for 14 days post-laser treatment. Eyes were enucleated after 14 days and corneal nerves were stained using a gold chloride technique. Major nerve bundles entering the cornea were quantitated by quadrant, using camera lucida reproductions. Nerve bundle diameters were measured using NIH image computer software on computer-scanned images. Statistical methods included repeated values for analysis of variance for CTT, STT and IOP, and a paired t-test for nerve diameters and bundles. All laser treated eyes had significantly higher CTTs (P<0.05) compared to control eyes for all measurements. Six out of fifteen dogs had evidence of ulcerative keratitis. Intraocular pressure was significantly lower in laser treated eyes compared to control eyes in the a.m. on days 2-9,and 14, and in the p.m. on days 2-11 using a Bonferroni-corrected alpha level (P<0.0039). A significant decrease of one nerve bundle per corneal quadrant was found between the laser treated and control eyes. There was no significant difference in STT or nerve bundle diameters between laser treated and control eyes. Nd:YAG laser cyclophotocoagulation effectively reduces IOP while increasing CTT. The procedure also causes a significant decrease in the number of major nerve bundles entering the cornea, but has no effect on the diameter of those bundles. These findings support the hypothesis that nerve damage and corneal hypoesthesia are etiologic factors in ulcerative keratitis following Nd:YAG laser cyclophotocoagulation. / Master of Science

Testes diagnósticos oftalmológicos em sagui de tufo preto (Callithrix penicillata)

Orientador: Cláudia Valéria Seullner Brandão / Resumo: O estudo objetivou descrever valores de referência para importantes testes diagnósticos oftalmológicos, como sensibilidade corneal, produção lacrimal, pressão intraocular, espessura corneal e microbiota conjuntival em saguis de tufo preto (Callithrix penicillata) saudáveis e sem alterações oculares. Foram utilizados 13 saguis de tufo preto, totalizando 26 olhos, provenientes do CEMPAS da Faculdade de Medicina Veterinária e Zootecnia - UNESP, Botucatu, São Paulo, Brasil. Os saguis foram submetidos a contenção química com anestesia dissociativa à base de cetamina e midazolam, para realização dos exames. Os resultados obtidos para sensibilidade corneal com estesiômetro Cochet-Bonnet foi de 5,8 ± 0,6cm; produção lacrimal com ponta de papel absorvente endodôntica estéril de 13 ± 3mm/min; pressão intraocular com tonômetro de rebote Tonovet Plus Icare® de 22,2 ± 2,9mmHg; espessura corneal 340,7 ± 25,5µm; e predomínio de bactérias Gram-positivas nas amostras de microbiota conjuntival. Não houve diferença significativa considerando lateralidade e sexo dos animais (P>0,05). Os resultados obtidos nos testes diagnósticos oftalmológicos poderão auxiliar médicos veterinários oftalmologistas a diagnosticar e tratar doenças oculares em saguis de tufo preto, além de contribuir para pesquisas na área de visão. / Abstract: The study aimed to describe reference values for important ophthalmic diagnostic tests, such as corneal sensitivity, tear production, intraocular pressure, corneal thickness and conjunctival microbiota in black tufted marmosets (Callithrix penicillata). Thirteen black tuft marmosets were used, totaling 26 eyes, followed by CEMPAS by the Faculty of Veterinary Medicine and Zootechnics - UNESP, Botucatu, São Paulo, Brazil. The procedures were used for chemical containment with dissociative anesthesia based on ketamine and midazolam, to perform the exams. The results obtained for corneal sensitivity with a Cochet-Bonnet esthesiometer were 5.8 ± 0.6 cm; tear production with sterile endodontic absorbent paper tip of 13 ± 3mm / min; intraocular pressure with Tonovet Plus Icare® rebound tonometer of 22.2 ± 2.9mmHg; corneal thickness 340.7 ± 25.5 µm; and predominance of Gram-positive bacteria in conjunctival microbiota. There was no significant difference considering the laterality and sex of the animals (P> 0.05). The results obtained in ophthalmological diagnostic tests can help veterinary ophthalmologists to diagnose and treat eye diseases in the black tufted marmosets, in addition to contributing to research in the field of vision. / Mestre

Régression non linéaire entre les motifs des fibres nerveuses et la sensibilité cornéenne en utilisant l'apprentissage automatique

Notre projet vise à élucider la relation complexe entre la morphologie des nerfs cornéens et la sensibilité cornéenne, afin d'améliorer la compréhension et le diagnostic des pathologies oculaires. En utilisant deux types d'esthésiomètres : l'esthésiomètre sans contact (NCCA) et le Cochet-Bonnet (CBA) pour mesurer la sensibilité, et en analysant les images de microscopie confocale (IVCM) via le logiciel CCMetrics, nous avons étudié 23 individus, y compris ceux souffrant de diabète et de kératite neurotrophique. Des corrélations négatives significatives entre certains attributs neuronaux et la sensibilité cornéenne ont été identifiées. L'utilisation d'algorithmes d'apprentissage automatique, tels que K-Plus Proches Voisins (KNN), les Réseaux de Neurones (MLP), la Régression à Vecteurs de Support (SVR) et les arbres de décision, a révélé des relations non linéaires complexes. Notre étude encourage l'utilisation de l’apprentissage automatique pour détecter ces relations complexes dans le domaine médical en général et en ophtalmologie en particulier. / Our project aims to clarify the complex relationship between the morphology of corneal nerves and corneal sensitivity, to improve understanding and diagnosis of eye pathologies. We used two types of esthesiometers: a non-contact esthesiometer (NCCA) and Cochet-Bonnet (CBA) for sensitivity measurement and analyzed confocal microscopy (IVCM) images using the software CCMetrics. We studied 23 individuals, including those with diabetes and neurotrophic keratitis. Significant negative correlations between certain neuronal attributes and corneal sensitivity were identified. The use of machine learning algorithms, such as K-Nearest Neighbors (KNN), Neural Networks (MLP), Support Vector Regression (SVR), and decision trees, revealed complex non-linear relationships. Our study advocates using machine learning to detect these complex relationships in the medical field, especially in ophthalmology.
