Caracterização do mecanismo de ação do composto natural plumieridina contra Cryptococcus gattii

As leveduras basidiomicéticas Cryptococcus neoformans e Cryptococcus gattii são os agentes etiológicos da criptococose. A infecção se desenvolve via inalação e disseminação até o sistema nervoso central, causando lesões pulmonares e meningoencefalite, a principal causa de morte da criptococose. Estes fungos patogênicos causam aproximadamente um milhão de casos por ano, principalmente entre pacientes imunocomprometidos, resultando em aproximadamente 625.000 mortes. O tratamento da criptococose emprega altas doses dos fármacos anfotericina B e 5-flucitosina, seguido de longa monoterapia com fluconazol. Este tratamento é caracterizado pela nefrotoxicidade e hepatotoxicidade. Além destes efeitos tóxicos, o surgimento de linhagens resistentes a estes compostos leva à necessidade de novas estratégias terapêuticas, dentre as quais podem ser citadas moléculas obtidas de fontes vegetais. O composto natural plumieridina, extraído de Allamanda polyantha (uma planta nativa brasileira), possui atividade antifúngica. Com o objetivo de elucidar as bases moleculares desta atividade, uma biblioteca de mutantes de C. gattii obtida por transformação mediada por Agrobacterium tumefaciens foi triada utilizando distintas concentrações deste composto, tendo como referencial o valor de MIC para a linhagem selvagem de C. gattii. Foram encontrados dez mutantes sensíveis e um mutante resistente a plumieridina e os fatores de virulência clássicos foram avaliados. A identificação dos genes inativados pelo T-DNA revelou o possível envolvimento de algumas proteínas no mecanismo molecular de plumieridina, tais como lisofosfolipídeo aciltransferase, transportador de colina, protease rhomboide, transportador de arsenito ATPase, proteína BAG e subunidade N6 do complexo regulatório 26S proteasoma. Estas proteínas estão relacionadas com diferentes processos biológicos ligados a via de sinalização Unfolded Protein Response (UPR). Esses resultados indicam um complexo processo celular desencadeado por estresse de retículo endoplasmático, afetando indiretamente várias vias, o que sugere um novo mecanismo de ação para um fármaco antifúngico em potencial. / The basidiomycete yeasts Cryptococcus neoformans and Cryptococcus gattii are the etiologic agents of cryptococcosis. The infection proceeds via inhalation and dissemination of yeast cells to the central nervous system, causing lung injury and meningoencephalitis, which is the major cause of death on cryptococcosis. These fungal pathogens cause one million cases per year among immunocompromised patients mainly, resulting in nearly 625,000 deaths. The treatment of cryptococcosis uses high doses of amphotericin B and 5-flucytoseine, followed by long fluconazole monotherapy. This regime is characterized by nephrotoxicity and hepatotoxicity. In addition to these toxic effects, the emergence of strains resistant to these compounds leads to the need of new therapeutic strategies, among which molecules obtained from plant sources. The natural compound plumieridine, extract from Allamanda polyantha (a Brazillian native plant), has antifungal activity. In order to elucidate the molecular basis of plumieridine antifungal activity, a C. gattii mutant library obtained by Agrobacterium tumefaciens-mediated transformation was screened using distinct plumieridine concentrations based on MIC value for C. gattii wild type. One resistant and ten susceptible mutants were found and the classical virulence factors were evaluated. The genome-wide screening revealed the possible involvement of some proteins in molecular mechanisms of plumieridine, such as lysophospholipid acyltransferase, choline transporter, rhomboid protease, arsenite transporting ATPase, BAG Protein and 26S proteasome regulatory subunit N6. These proteins are related with different biological process linked to Unfolded Protein Response (UPR) signaling pathway. This result suggests a complex cell process triggered by endoplasmic reticulum stress with many pathways indirectly affected by plumieridine, displaying a novel mechanism of action in a potential antifungal agent.

Análise de alterações morfológicas associadas a mutação de tropomiosina em Cryptococcus gattii

Processos celulares, incluindo crescimento celular, transporte de organelas, endocitose, exocitose e citocinese requerem um citoesqueleto intacto. A tropomiosina (Tpm1) é uma proteína dimérica de citoesqueleto altamente conservada, que forma polímeros que se associam aos filamentos de F-actina em células musculares e não musculares. Essa ligação estabiliza os filamentos de actina evitando a despolimerização por outros fatores. Assim, a tropomiosina desempenha um papel fundamental na integridade celular e também permite que processos como secreção, localização e função de muitas proteínas sejam exercidas, de acordo com a demanda celular exigida em diferentes situações. Em leveduras, Tpm1 participa da manutenção destes processos, assim como na integridade celular e morfologia da célula. Neste estudo, uma análise do gene que codifica para tropomiosina e a funcionalidade do produto codificado por ele foi realizada para a levedura Cryptococcus gattii. Foi realizada a construção de uma linhagem mutante de tropomiosina, que revelou que Tpm1 está envolvida na formação da cápsula polissacarídica, morfologia e virulência do fungo. Mutação no gene de Tpm1 leva a alterações no citoesqueleto celular, crescimento e morfologia, e, defeitos de citocinese. Estes resultados indicam um papel crucial para Tpm1 na formação de cápsula polissacarídica, morfologia e virulência de C. gattii. / Cellular processes including cell growth, organelle transport, endocytosis, exocytosis and cytokinesis require an intact cytoskeleton. Tropomyosin (Tpm1) is a dimeric protein of the highly conserved cytoskeleton forming polymers which are associated with the F-actin filaments in muscle and non-muscle cells. This binding stabilizes actin filaments preventing depolymerization by other factors. Thus, tropomyosin plays a key role in cellular integrity and also allows processes such as secretion, localization and function of many proteins to be performed according to the required cellular demand in different situations. In yeasts, Tpm1 participates in the integrity of these processes, cell integrity and cell morphology. Tropomyosin also plays an important role in the secretion, localization, and function of many proteins, in fungi. In this study we performed a construction of a Cryptococcus gattii tropomyosin mutant strain, which revealed that disruption of C. gattii tropomyosin gene leads to changes in cytoeskeleton, abnormal growth and morphology, defective cell separation, assembly of the polysaccharide capsule and virulence. Taken together, these results indicate a central role for Tpm1 in the assembly of the polysaccharide capsule, morphology and virulence of C. gattii.

Utilização de métodos de bioimagem para a análise ex vivo da infecção causada por cryptococcus gattii

O fungo patogênico Cryptococcus gattii é um dos agentes etiológicos da criptococcose, uma doença fatal que envolve a formação de criptococomas nos pulmões e cérebro em hospedeiros imunocompetentes. O monitoramento da patogênese de C. gattii e sua colonização nos diferentes órgãos em modelos animais geralmente são analisados pelo método laborioso e impreciso de contagem das unidades formadoras de colônias (UFC). Neste trabalho foi desenvolvido um modelo prático de monitoramento da infecção e da eficácia de tratamentos antifúngicos, através da construção de uma linhagem de C. gattii expressando constitutivamente o gene repórter mCherry e da avaliação por imagem baseada em fluorescência. Após a infecção intranasal de camundongos e do tratamento com anfotericina B (AMB) ou fluconazol (FLC) durante 4 a 7 dias, pulmões, rins, baço e o cérebro foram removidos e examinados em IVIS Lumina, e a infecção foi monitorada pelo método tradicional de contagem de UFC. As análises por imagem demonstraram a predileção de C. gattii pelo trato respiratório, com presença reduzida no sistema nervoso central. Estas observações foram consistentes com as obtidas pela determinação de UFC. As análises por imagem e a determinação por UFC corroboraram os resultados relativos aos efeitos do tratamento com FLC. Entretanto, os dados foram significativamente diferentes quando AMB foi utilizada. Os resultados obtidos sugerem esta técnica como um potencial método para a investigação da dinâmica da infecção e da sua resposta a tratamentos terapêuticos. / The pathogenic fungus Cryptococcus gattii is one of the etiological agents of human cryptococcosis, which may evolve to the formation of cryptococcomas in the lungs and brain of immunocompetent hosts. Monitoring the progression of pathogenesis and organ colonization in animal models is important in the study of infectious microorganisms. However, this monitoring usually involves laborious and imprecise methods, such as colony forming unit (CFU) determination. Here, we propose the use of a practical model to monitor C. gattii infections in mice and to evaluate antifungal treatment. Our approach involved the infection of mice with a C. gattii strain that constitutively expresses the mCherry reporter gene, followed by fluorescence-based imaging analysis of the colonized organs. Mice were infected intranasally with the fluorescent C. gattii strain and treated with amphotericin B (AMB) or fluconazole (FLC) for 4 or 7 days. The lungs, kidney, spleen, and brain were excised and examined in an IVIS Lumina instrument to determine fluorescence intensity and for CFU determination. Imaging analyses showed that C. gattii primarily colonizes the respiratory tract, with reduced dissemination to the central nervous system. This observation was consistent with the results of CFU determination. The evaluation of the effects of fluconazole treatment in infected mice revealed that both imaging and CFU methods produced similar results. However, the results of the two methods were considerably different when the effects of AMB were evaluated. Our results support the use of imaging-based techniques to monitor infections caused by C. gattii in mice, as well as the response of animal hosts to treatment with antifungals.

Análise de microRNAs-LIKE em cryptococcus gattii

O complexo Cryptococcus possui 37 espécies descritas, das quais se destacam C. neoformans e C. gattii devido a sua capacidade de causar doença tanto em humanos como animais. A fim de se prevenir de organismos patogênicos, o hospedeiro pode reduzir a disponibilidade de nutrientes em um processo conhecido como imunidade nutricional. Além disso, é bem descrito o papel central de metais essenciais em relação à expressão dos fatores de virulência nas espécies patogênicas do gênero Cryptococcus. MicroRNAs (miRNAs) são importantes mediadores da expressão gênica em resposta a estresses, incluindo privação de metais ou toxicidade causada pelos mesmos. Entretanto, pouco é conhecido sobre expressão de miRNAs ou miRNAs-like em C. gattii. Portanto, o objetivo desde trabalho foi determinar o perfil miRNAs ou miRNAs-like produzidos por C. gattii durante privação de ferro e privação de zinco. Células de C. gattii R265 foram cultivadas em três condições: privação de zinco, privação de ferro e condição controle. As sequências dos pequenos RNAs foram obtidas por RNA-Seq. A fim de identificar miRNAs em C. gattii, as sequências não redundantes foram alinhadas às sequências dos genomas das linhagens de C. gattii R265 e WM276. Foi realizada a predição de miRNAs ou microRNA-like com ferramentas de bioinformática que resultou em 31 possíveis miRNAs na linhagem R265 e 26 na linhagem WM276, respectivamente. Análises in silico da expressão diferencial dos miRNAs mostraram que a maioria está presente nas três condições, ao passo que apenas dois estão presentes apenas nas condições de privação de metais. Desta forma, estes miRNAs-like podem estar exercendo algum papel importante na homeostase de metais em resposta a condições que mimetizam o ambiente de infecção. Visto que a linhagem R265 de C. gattii não possui um gene codificador da proteína Argonauta, a qual desempenha um papel central na via de RNA de interferência (RNAi), esta via possivelmente não seria responsável pelo silenciamento de outros genes. Levanta-se a hipótese que estes miRNAs-like poderiam ser exportados da célula e, então, modular a expressão gênica no hospedeiro. Esta análise foi iniciada pela avaliação de possíveis alvos expressos pelos genomas de camundongo e humano. Alguns destes alvos mostraram relação com a resposta imune nos hospedeiros. Esta identificação deve esclarecer o papel desenvolvido por miRNAs na regulação da expressão gênica na interação patógeno-hospedeiro. / The Cryptococcus complex has 37 species described, from which C. neoformans and C. gattii stand out for their ability to cause disease in humans and animals. To prevent infection with pathogenic microorganisms, host may reduce the availability of nutrients in a process known as nutritional immunity. Furthermore, it is well described that essential metals have central roles related to the regulation of expression of virulence factors in pathogenic Cryptococcus species. MicroRNAs (miRNAs) are important mediators of gene expression in response to several stresses, including metal deprivation or toxicity. However, little is known about microRNAs or miRNAs-like expression in C. gattii. Therefore, the aim of this work was to profile miRNAs produced by C. gattii during iron and zinc deprivation. C. gattii R265 cells were incubated in three conditions: zinc deprivation, iron deprivation and control condition. The sequences of small RNAs were obtained by RNA-Seq. To identify miRNAs or miRNAs-like in C. gattii, non-redundant reads were aligned to sequences of C. gattii R265 and WM276 strains genomes. MiRNAs prediction was and resulted in 31 possible miRNAs in R265 strain and 26 in WM276 strain, respectively. In silico analysis of miRNAs differential expression showed that the majority of them are present in the three conditions, while two are present only in metal deprivation. Thereby, these miRNAs-like may be exerting some important role at metal homeostasis in response to infection environment mimicry conditions. As the C. gattii R265 strain does not have an Argonaut encoding gene, the protein that plays a central role in the RNA interference (RNAi) pathway, we assumed that probably this pathway would not be responsible for silencing of others genes. We hypothesized that these miRNAs-like could be exported and modulate the gene expression in host cells. This analysis was initialized by evaluating the potential targets expressed by human and mouse genome. Some of these targets were related to immune response in the host. This identification should clarify the role played by miRNAs in the regulation of gene expression in the host-pathogen interaction.

Influência da alteração dos níveis de ferro e cobre na fisiologia e patogenicidade de Cryptococcus gattii : uma abordagem proteômica

Os micronutrientes ferro e cobre possuem uma ampla distribuição na natureza e são utilizados por quase todos os organismos vivos. Em se tratando de patógenos, os mecanismos utilizados para a manutenção da homeostase desses metais possuem ligação com a expressão dos fatores de virulência e, por consequencia, auxiliam no estabelecimento do patógeno no hospedeiro e início da doença. Cryptococcus gattii é uma levedura encapsulada, que causa criptococose em indivíduos imunocompetentes e em animais. Essa espécie é responsável por um surto de criptococose nas ilhas Vancouver, Canadá, com altos índices de mortalidade e morbilidade registrados do ano de 1999 até o presente. Neste contexto, o presente trabalho teve por objetivo o esclarecimento dos papéis envolvendo a homeostase de ferro e cobre no patógeno C. gattii. Para isto, foi realizada uma análise proteômica global e comparativa em células de C. gattii cultivadas sob presença/limitação de ferro e/ou cobre. Essa análise foi feita utilizando a metolodogia de identificação multidimensional de proteínas (MudPIT), associada ou não com géis bidimensionais (2D-GE). Nossos resultados demonstraram que o fungo C. gattii sofre alterações metabólicas, principalmente envolvendo funções mitocondriais, na ausência dos micronutrientes ferro ou cobre. As análises também mostraram que o fungo responde a privação desses micronutrientes por um aumento na atividade de proteases, o que poderia vir a ser uma regulação pós-traducional utilizada por esse patógeno para manter a homeostase desses metais. Adicionalmente, funções específicas desempenhadas pelo fungo na ausência de ferro ou cobre também foram identificadas. / Iron and copper have a ubiquitous distribution in living organisms; the competition for such between the host and pathogen is related with disease establishment. Cryptococcus gattii is an encapsulated yeast that causes cryptococcosis in immunocompetent individuals and animals. This pathogen is responsible for an ongoing outbreak in Vancouver Island, Canada, which has substancial mortality and morbidity rates. In this context, the aim of this work was to clarify the mechanisms involved in the maintenance of iron and copper homeostasis in C. gattii. To accomplish that, a global and comparative proteomic analyses were performed in C. gattii cells grown in replete/depleted iron and/or copper conditions. For these analysis was used the multidimensional protein identification technology (MudPIT) associated, or not, with twodimensional gels (2D-GE). Our results demonstrated metabolic alterations in response to iron and copper deprivation in C. gattii, mainly associated with mitochondrial functions. The results also showed a high protease activity in cells of C. gattii grown in iron and copper deprivation in comparison with control. This might be a posttranslational regulation developed by C. gattii to maintain metal homeostasis. In addition, specific functions related to iron and to copper control were identified.

The emergence of <i>cryptococcus gattii</i> in British Columbia : veterinary aspects

A series of presumed or confirmed Cryptococcus gattii cases diagnosed between 1999 and 2003 was compiled through review of records from veterinary laboratories and human diagnostic services. There was a continual increase in the annual number of animal, but not human, cases diagnosed; no seasonality was observed. Animal cases exceeded human cases by almost 75% even though it was hypothesized that animal cases are more likely to go undiagnosed or unreported when compared to humans. Animal cryptococcosis cases were identified on Vancouver Island prior to 1999 suggesting the organism may have emerged in the region prior to its identification as a causative agent for human disease; therefore animals may serve as a good sentinel for human cryptococcosis infection. <p> There were 50% more feline than canine cases and disease appeared more commonly in middle aged cats and younger dogs. There was no sex predilection for either species. The primary system involved was most commonly respiratory, followed by central nervous system (CNS) in both cats and dogs. There was a higher proportion of CNS disease in dogs relative to cats, and cats were much more likely to have subcutaneous or dermal masses relative to dogs. Multivariate survival analysis identified only the presence of neurological symptoms as a statistically significant predictor of mortality; those animals exhibiting CNS symptoms were over four times more likely to die than those never showing neural signs. A case-control study identified host and environmental risk factors for clinical C. gattii infection in dogs and cats suggesting that where an infectious agent is not uniformly distributed, individual risk increases when the organism is re-distributed through large scale environmental disturbance, or when the animal has increased opportunities for exposure through travel or activity level. <p> Serum samples and material for fungal culture were collected from dogs, cats, horses and terrestrial mammal species residing within the region where clinical cases had been diagnosed. Nasal colonization was identified in squirrels (<i> Sciurus carolinensis</i>), horses, dogs and cats. Most of the animals sampled had no signs of systemic infection however asymptomatic infection, defined as the presence of cryptococcal antigen in the bloodstream in the absence of clinical symptoms, was identified in a small number of dogs and cats. Fourteen months of follow-up testing of asymptomatic animals revealed that animals can progress to clinical disease, remain sub-clinically infected, or clear the organism.

Identificação de proteínas envolvidas na resposta de cryptococcus gattii à temperatura de infecção

Cryptococcus gattii é uma levedura basidiomicética que possui uma cápsula polissacarídica robusta e é um dos agentes etiológicos da criptococose. A linhagem R265 de C. gattii foi responsável pelo surto de criptococose na Ilha de Vancouver e há vários estudos indicando sua dispersão e ocupação de novos nichos ecológicos. Fatores de virulência bem caracterizados deste patógeno incluem: a capacidade de sintetizar o pigmento antioxidante melanina, a produção de uma cápsula polissacarídica antifagocíticas e a capacidade de sobreviver e proliferar a 37°C. Neste sentido, o objetivo deste estudo foi identificar proteínas envolvidas na resposta da linhagem R265 de C. gattii à variação de temperatura experimentada quando o fungo deixa o ambiente para sobreviver no hospedeiro, usando análise in vitro e proteômica comparativa. Nossos dados demonstram que as proteínas com expressão aumentada a 25°C atuam principalmente em processos metabólicos de proteínas, carboidratos, lípidios, ácidos orgânicos e aminoácidos e que as proteínas com expressão aumentada a 37°C, em sua maior parte, participam de processos que incluem fosforilação, metilação, processos catabólicos de corpos cetônicos, GTP e ATP e processos metabólicos de nucleosidios. A diferença mais notável entre as condições testadas é a predominância, no desenvolvimento de C. gattii a 25°C, de proteínas envolvidas em processos de oxirredução e a presença de um número considerável de proteínas envolvidas na resposta ao stress e em processos catabólicos no desenvolvimento a 37°C. Em suma, o presente estudo é a primeira análise proteômica a fornecer informação sobre proteínas reguladas pela variação de temperatura e a primeira descrição da resposta da linhagem hipervirulenta R265 de C. gattii à esta condição. As proteínas identificadas podem ser alvos para novos estudos na identificação de proteínas-chave para o desenvolvimento do fungo a 37°C e para a pesquisa de biomarcadores que podem contribuir para o tratamento e prevenção da criptococose causada por este patógeno. / Cryptococcus gattii is a basidiomycetous yeast which has a robust polysaccharide capsule and is one of the etiological agents of cryptococcosis. The R265 strain of C. gattii was responsible for the Vancouver Island outbreak and there are various studies including their dispersal and occupation of new ecological niches. Well-characterized virulence factors of this pathogen include: the ability to synthesize the antioxidant pigment melanin; the production of an antiphagocytic polysaccharide capsule; and the ability to survive and proliferate at 37°C. In this sense, the objective of this study was identified proteins involved in the response of C. gattii strain R265 to the temperature variation experimented when the fungi leaves the environment to survive in the host, using in vitro analysis and comparative proteomics. Our data demonstrated that the proteins up-regulated at 25°C act mainly in protein, carbohydrate, lipid, organic acid and amino acid metabolic processes and proteins up-regulated at 37°C participate mostly in processes that include phosphorylation, methylation, ketone body, GTP and ATP catabolic process and nucleoside metabolic process. The most striking difference between the conditions tested is the prevalence of proteins involved in oxidation-reduction in C. gattii growth at 25°C and the presence of a relevant number of proteins involved in stress response and catabolic process in C. gattii growth at 37°C. In brief, this study is the first proteomic analysis to provided information about proteins regulated by the infection temperature and the first description of the response of C. gattii high virulent strain R265 to this condition. The proteins identified could be targets to further studies for the identification of key proteins for the development at 37°C and to the search for biomarkers that can contribute to the treatment and prevention of cryptococcosis caused by this pathogen.

Studies have shown that proton pump inhibitors (PPI´s) have an inhibitory effect on melanin production, in melanocytes,produced by a phenoloxidase, called tyrosinase. This effect on inhibition of melanogenesis was associated with inhibition of tyrosinase.Since melanocytes and Cryptococcus spp. produce the melanin with a similar oxidation action of phenolic compounds, the objective of this work was to test the effect of PPI´s on the inhibition of melanin production in Cryptococcusneoformans/gattii lato sensu. For this, a new methodology was initially developed for the quantification of melanin, through the gray scale. In this experiment, 12 strains of Cryptococcus spp., 1 strain of Candidaparapsilosis ATCC 22019 (negative control) and 1 strain of Hortaeawerneckii (positive control) were analyzed, resulting in an adequate production and quantification of melanin in testedstrains. Next, PPIs were tested (omeprazole, rabeprazole, lansoprazole, esomeprazole and pantoprazole), classical antifungal agents (fluconazole and amphotericin B) and glyphosate (inhibitor of melanogenesis in Cryptococcus spp.) on the susceptibility of 20 strains of Cryptococcus spp.and 1 strain of C. parapsilosis ATCC 22019, based on the protocol M27-A3 of the Clinical Laboratory Standart Institute (CLSI).The following MIC50 values were observed: for omeprazole 166.7 ± 91.29 μg/ml, esomeprazole 256 ± 115.1 μg/ml, lansoprazole 226 ± 200.1 μg/ml, rabeprazole 142.6 ± 74.6 μg/ml, pantoprazole 428.6 ± 297.3 μg/ml, glyphosate 952.4 ± 150.4 μg/ml, fluconazole 4,757 ± 1,6 μg/ml and amphotericin B 1,464 ± 2.6 μg/ml. Finally, subinhibitory concentrations of PPIs were tested on melanin production in Cryptococcus spp., resulting in an inhibition of melanin for protons pump inhibitors, the best effect being obtained with omeprazole, lansoprazole and pantoprazole. Therefore, PPI´s has an important effect on the inhibition of melanin in Cryptococcus spp., and further studies are needed to understand the mechanisms of inhibition and its potential in reducing virulence, dissemination and infection of the CNS. / Cryptococcus neoformans lato sensu e Cryptococcus gattii lato sensu são leveduras hialinas capazes de produzir melanina, estando associados auma doença chamada criptococose. Para sua instalação e disseminação, vários fatores de virulência têm sido associados, como a produção de melanina, que é produzida por uma fenoloxidase chamada lacase que oxida os compostos fenólicos. Estudos demostraram que os inibidores de bomba de prótons (IBP´s) têm efeitos inibitórios na produção de melanina, em melanócitos, produzidos por uma fenoloxidase chamada tirosinase. Este efeito inibitório da melanogênese foi associado ao bloqueio da tirosinase. Uma vez que os melanócitos e Cryptococcus spp. produzem melanina por uma via semelhante à oxidação de compostos fenólicos, o objetivo deste trabalho foi testar o efeito de IBP´s na inibição de produção de melanina em C.neoformans/C. gattiilatosensu. Para tanto, inicialmente foi desenvolvida uma nova metodologia para quantificação de melanina por meio da escala de cinza. Neste sentido, foram analisadas 12 cepas de Cryptococcus spp., uma cepa de Candidaparapsilosis ATCC 22019 (controle negativo) e uma cepa de Hortaeawerneckii (controle positivo), resultando em uma adequada produção e quantificação de melanina nas cepas testadas. A seguir, foram testados os IBP´s (omeprazol, rabeprazol, lansoprazol, esomeprazol e pantoprazol), os antifúngicos clássicos (fluconazol e anfotericina B) e glifosato (inibidor da melanogênese em Cryptococcus spp.) sobre a sensibilidade de 20 cepas de Cryptococcus spp. e uma cepa de C. parapsilosis ATCC 22019, baseado no documento M27-A3 do Clinical Laboratory Standart Institute (CLSI). Foram observados os seguintes valores de MIC50: para omeprazol 166,7 ± 91,29 µg/mL; esomeprazol 256 ± 115,1 µg/mL; lansoprazol 226 ± 200,1 µg/mL; rabeprazoL 142,6 ± 74,6 µg/mL; pantoprazol 428,6 ± 297,3 µg/mL; glifosato 952,4 ± 150,4 µg/mL; fluconazol 4,757 ± 1,6 µg/mLe; anfotericina B 1,464 ± 2,6 µg/mL. Finalmente, foram testadas concentrações subinibitórias dos IBP´s sobre a produção de melanina em Cryptococcus spp., obtendo como resultado uma inibição da melanina para os inibidores de bomba de prótons, sendo o melhor efeito obtido com omeprazol, lansoprazol e pantoprazol. Portanto, IBP´s têm um efeito importante na inibição de melanina em Cryptococcus spp., sendo necessário estudos mais aprofundados para compreender os mecanismos de inibição e seu possível potencial na diminuição de virulência, disseminação e infecção do SNC.

Avaliação da utilização de uma vacina de DNA contendo a região codificadora de 14-3-3 de Cryptococcus gattii como estratégia profilática para criptococose

Cryptococcus gattii é um patógeno emergente que infecta principalmente indivíduos imunocompetentes, causando criptococose. Dentre os tratamentos estão anfotericina B e fluconazol, porém são insuficientes na erradicação da doença. A estimulação da resposta imune por vacinas de DNA em diversos modelos fúngicos vem sendo relatados. Dentre as proteínas fúngicas candidatas no desenvolvimento de vacinas estão às proteínas 14-3-3, as quais foram identificadas como proteínas imudominantes em estudos de proteômica de Cryptococcus neoformans e C. gattii e em soro de pacientes com criptococose. O objetivo deste estudo foi realizar a construção do vetor recombinante contendo a região codificante do gene 14-3-3, seguido da análise preliminar da resposta imune em camundongos imunizados. A região codificante do 14-3-3 foi clonada em vetor de expressão eucariótico pcDNA3.1(+) e a funcionalidade do vetor recombinante foi confirmada a nível transcricional por RT-PCR a partir de células A549 transfectadas com a vacina de DNA e a nível traducional por imunobloting. A proteína 14-3-3 foi ainda localizada na parede celular da levedura, revelado por metodologias de microscopia por imunofluorescência. A porção codificante do gene 14-3-3 de C. gattii foi clonada em vetor de expressão pET23-d e a proteína recombinante foi expressa em células de E. coli BL21 e purificada por cromatografia de afinidade a níquel. A resposta imune humoral foi determinada por dosagem de imunoglobulinas, porém não ocorreu diferença estatística na indução de imunoglobulinas totais em camundongos inoculados com a vacina. Assim esse trabalho contribuiu para o estudo imunogenicidade da proteína 14-3-3 em C. gattii permitindo possíveis estudos em outros modelos animais usando a mesma estratégia profilática. / Cryptococcus gattii is an emerging pathogen that mainly infects immunocompetent individuals, causing cryptococcosis. Among the treatments are amphotericin B and fluconazole, but they have proven insufficient in eradicating this disease. The immune responses stimulation by DNA vaccines in several fungal models has been reported. Among fungal candidate proteins in vaccine development are the 14-3-3 proteins, which were identified as immunodominant sera proteins in Cryptococcus neoformans and Cryptococcus gattii proteomics and in serum of patients with cryptococcosis. The study aimed to construct a recombinant vector containing the 14-3-3 coding region, followed by the immune response preliminary analysis of immunized mice. The 14-3-3 gene was cloned into a eukaryotic expression vector pcDNA3.1(+) vector and RT-PCR confirmed the recombinant transcriptional level functionality from A549 transfected with DNA vaccine cells and translational level by immunoblotting. The 14-3-3 protein was localized associated with the yeast cell wall, as revealed by immunofluorescence microscopy. The coding portion of the 14-3-3 C. gattii gene was cloned into pET23-d expression vector and the recombinant protein was expressed in E. coli BL21 and purified by nickel affinity chromatography. The humoral immune response was determined by serum total immunoglobulin measurements, but no statistical differences were observed in total immunoglobulins from vaccine-inoculated mice. Thus, this research contributed to 14-3-3 C. gattii protein immunogenicity study, allowing possible researches in other animal models applying the same prophylactic strategy.
