Our objective was to evaluate the mRNA expression of TLR-2 and 4, expression and production of IL-12, IFN-γ, TNF-α, IL-17, IL-10 and TGF-β and NO production during infection with Leishmania chagasi and correlate TLR2 and 4 expressions with cytokines production and NO. Infection resulted in increased TLR2, TLR4, IL-17, TNF-α and TGF-β expression at the beginning of infection, with a decrease at the final phase in according the parasitic load; IFN- γ and IL-12 decreased at the peak of parasitemia and increased at the final phase; IL-10 increased during the whole period under analysis. With respect to cytokines and NO production, TGF-β, TNF-α and IL-17 showed high rates at the initial phase, and IFN-γ and IL-12 showed high rates at the final phase; IL-10 and NO showed increasing production during the infection period evaluated. There was a positive correlation of TLR2 and 4 with TNF-α, IL-17, NO, IL-10 and TGF-β at the beginning of infection, and with TNF-α, IL-17 and TGF-β at the end. Our data suggest that L. chagasi was in contact with host’s cells via TLR2 and 4, which resulted in cytokine modulation. This interaction could be considered as pathogenic mechanism in visceral leishmaniasis

Previous issue date: 2012 / Resumo: A endodontia assim como qualquer outra área da Odontologia está sujeita a insucessos. Atualmente estamos verificando um aumento significativo de processos judiciais na Odontologia e também em relação à especialidade de Endodontia. Frente a esses fatos este trabalho teve como objetivos fazer um levantamento no SEAP (Serviço de Assessoria Pericial da FOP/UNICAMP) num período entre 2005 e 2010 de processos envolvendo a Endodontia. Dentre esses processos envolvendo a endodontia, foi levantado o que se segue: a incidência de processos judiciais junto ao SEAP; a existência de prontuários e seus documentos constitutivos enviados para a perícia; o motivo principal que uma lide judicial e também discussão dos aspectos éticos e legais inerentes ao tema. Obtiveram-se como resultados: a incidência de processos judiciais em endodontia no período proposto (2005 a 2010) foi da ordem de 6,97% (n=6) do total de processos arquivados (n=86); os documentos (prontuários) apresentados pelos Cirurgiões-Dentistas nas perícias se encontravam incompletos, sem anuência do paciente em relação ao tratamento proposto e sem contrato de prestação de serviço; o principal motivo que gerou a ação judicial foi a quebra da relação paciente/profissional, além dos motivos de ordem técnica; os Cirurgiões- Dentistas deveriam realizar diariamente um prontuário completo, pois a falta de documentos deixa o C.D mais propenso a uma demanda judicial / Abstract: The Endodontics as well as any other area of Odontology is subjected to failures. We are currently observing a significant increase in Odontology litigation and also in the specialty of Endodontics. Given these facts, this study aimed to survey the SEAP for a period from 2005 to 2010 of processes involving Endodontics. Among these processes involving Endodontics were raised: the incidence of cases at the SEAP; the existence of records and its constituent documents sent to the medical examination; the main technical reason involving Endodontics that motivated the lawsuit and also the discussion about legal end ethical issues inherent in the subject. Were obtained as a result: the incidence of litigations in endodontics in the period proposed between 2005 and 2010 was 6,97% (n=6) of a total number of cases filed ( n=86); documents ( medical records) submitted by Surgeon Dentists to medical examination were incomplete, without consent of the patient regarding the treatment proposed, and no service contract; the main reason which generated the lawsuit was the breach of relationship patient/professional, besides of technical reasons, the Surgeon Dentists should daily fill up complete medical record, as the lack of documentation makes the Surgeon Dentists more vulnerable to a lawsuit / Mestrado / Odontologia Legal e Deontologia / Mestre em Biologia Buco-Dental

M.A. / With the advances achieved in medical technology in recent years, society is increasingly faced with the issue of determining how fit individuals are for reintegration in society following the event of traumatic brain injury. Together with Neurology and it's diagnostic capabilities, many other disciplines such as Neuropsychology, Physiotherapy and Occupational Therapy, to name but a few, have placed strong emphasis on the rehabilitation of individuals with brain damage, with a view to reintegrate them into society. Besides the physical rehabilitation which aims at normalizing the individual's physical health, issues such as psychological ,cognitive and social functioning have been the focus of a great body of research in recent years. Whereas in the past the prognosis for many such patients in respect of returning to a normal life style were discouraging, health workers are now more optimistic in most cases regarding the individuals' probability to achieve a higher level of functioning after recovery. In this context, the individuals' ability to function in society, their ability to be integrated into the family and working environment and competence to be active and valued members of society have thus become the new aims which the therapeutic team together with the patient strive to achieve. With this in mind, rehabilitation teams now place greater emphasis and effort on optimizing recovery in the areas of cognitive and emotional competence. Amongst these issues, the individuals' competence to make moral judgements and socially adequate decisions in everyday living is of the utmost importance. This impacts on their ability to abide by social rules and norms which will determine their competence and desirability as active members in the family, social and working environments. This is the main focus of the present study. The issue of moral judgement and behaviour is central to this discussion, and a delineation of what defines morality and moral judgement is mandatory. Contributions to this analysis were found in the writings of the various authors and schools of thought presented here, and each of them has dealt with the theme of morality, moral behaviour and judgement from a different theoretical perspective. Theorists such as Lawrence Kohlberg made a major contribution to the understanding of the development of moral judgement through his stage theory, which forms one of the cornerstones of the present study. The site and the severity of the brain injury is paramount to the prognosis for rehabilitation. The frontal lobes have been described as the structures responsible for the widest range of symptoms and seen as the seat of intellect, responsible for deviations in moral and emotional functioning. An overview of the frontal lobes and associated cognitive functions as well as behaviors observed as a result of injury to these structures is presented. The existential-phenomenological system of inquiry has been described by its proponents as the most adequate to investigate phenomena relating to human behaviour, when an understanding of the subjective experience of the individuals involved is desired. For this reason this methodology is used in the present study to facilitate a qualitative analysis of the experience of brain injury particularly in respect of possible alterations in moral judgement and behaviour following the accident. The present investigation is based on interviews with three subjects and their primary caretakers, who answered questions regarding moral dilemmas. These dilemmas aim at tapping the individual's underlying reasoning processes when faced with the decision of what is right and wrong. The analysis and integration of the data obtained through these interviews reveals a complex cluster of alterations in thought and behaviour, much in line with those expected from patients suffering traumatic injury of the frontal lobes. Although different behaviours were revealed by each participant in this study, they were generally in line with the alterations described in the literature resulting from injury to the described brain areas. The value of this study lies in projecting a clearer picture of the experience of brain injury from the point of view of the victims as well as the close family members. Furthermore, analysis of data obtained in this investigation highlights certain typical behaviors and responses observed in individuals with such brain injuries, as described in the literature presented here.

On micro to mesoscale homogenization of electrical properties for damaged laminated composites (and their potential applications in electrical tomography)Selvakumaran, Lakshmi 12 1900 (has links)
Efficient and optimal use of composites in structures requires tools to monitor and capture the complex degradation that can occur within the laminates over time.
Structural health monitoring (SHM) techniques uses sensors/actuators on the structure to progressively monitor the health of the structure with minimal manual intervention. Electrical tomography (ET) is a SHM technique that uses voltage measurements from the surface of the laminate to reconstruct a conductivity map of the structure. Since damage has been shown to modify the conductivity of the laminate, the conductivity map can provide an indirect measure of the damage within the material. Studies have shown the capability of ET to identify macroscale damage due to impact. But, little has been done to quantitatively assess damage using ET.
In this work, we present a theoretical framework to link degradation mechanisms occuring at the microscale to the conductivity at the mesoscale through damage indicators.
The mesoscale damage indicators are then shown to be intrinsic to the ply. Next, we use the knowledge obtained through mesoscale homogenization to study the detectability of transverse cracks. Last, we show how the mesoscale homogenization participates in regularization of the inverse problem and in the quantitative assessment of the reconstructed conductivity map. This is as such the first step towards turning ET into a viable quantitative health monitoring technique.

Legal institute of damage compensation is of cardinal and irreplaceable importance in the area of Civil law. The thesis examines this institute from its material aspect, namely in terms of the mode and amount of compensation. First part of the work deals with the introduction of the major changes brought by the new regulation of Czech Civil law into the area of damage compensation and their potential impacts on this sphere. Subsequently, there are further definitions of compensation's content, mode and amount, their mutual relations and historical development. Firstly, I tried to explain every institute separately and in the end of my work I dealt with their interrelation. In the first Chapter I outline a picture of civil liability in the level of juristic theory and I bring a view on the origin and content of civil liability. In this Chapter I point out that the understanding of the term of civil liability is non-uniform, which leads to serious opinion fragmentation. One of the main tasks of the legal order is to ensure the protection of the society and individual from the conception of damage with the aid of legal rules together with coercive state authority. It means that the protection of their health, property, as well as nature and the environment should be provided. Structural analysis of induced mutagenesis A’ protein from mycobacterium tuberculosis and of a thermophillic GH9 cellulaseAnye, Valentine January 2014 (has links)
Masters of Science / The three-dimensional structures of proteins are important in understanding their function and interaction with ligands and other proteins. In this work, the structures of two proteins, ImuA’ from mycobacterium tuberculosis and GH9 C1 cellulase from a metagenomic library, were analysed using structural biological and modelling techniques. The gene encoding ImuA’ was amplified by two-step PCR, cloned, and expressed in E. coli. The recombinant ImuA’ produced was found to be largely insoluble. The insoluble protein was successfully solubilized in 8M urea but refolding the protein to its native structure was unsuccessful. By homology modelling, a 3D model of ImuA’ was obtained from a partly homologous protein RecA. In comparison to RecA, ImuA’ appears to lack some loop amino acids critical for DNA binding. Hence ImuA’ is postulated to not bind DNA. The second protein, GH9 C1 cellulase, was produced in E. coli. The protein was purified by chromatographic techniques and crystallized in a precipitant to protein ratio of 1:2 by hanging and sitting drop crystallization methods. The reservoir solution was made up of 15-30% (w/v) PEG 3350, 200 mM salt and 100 mM Tris-HCL pH 7.5-8.5. The protein crystals only diffracted x-rays to 4 å resolution which could not be used to obtain a crystal structure of the protein. The diffraction data, however, showed the crystal to be monoclinic with space group P2. Homology modelling revealed GH9 C1 cellulase to be a two domain protein with a smaller N-terminal Ig-like domain and a larger catalytic domain.The catalytic domain retains two ca2+ binding sites, which potentially stabilize the active site conformation and increase thermostability of the protein. Overall GH9 C1 cellulase is structurally similar to other GH9 cellulases, suggesting that its catalytic mechanism may be conserved.
