MODULATION OF COCAINE-LIKE BEHAVIOURAL ACTIVITY BY SEROTONIN UPTAKE INHIBITION RELATIVE TO THE EFFECTS OF THE NOVEL AND SELECTIVE DOPAMINE TRANSPORTER INHIBITOR, D-84

Cocaine dependence is a major health concern worldwide, but despite this high rate of abuse there are currently no approved therapies for cocaine dependence. Replacement pharmacotherapies are one possible approach for treating cocaine dependence, and identification of such therapeutics for cocaine abuse is the long-term goal of this research. Cocaine binds to, and inhibits uptake at the dopamine (DAT), serotonergic (SERT) and noradrenaline (NET) uptake transporters, but studies have shown that cocaine produces its strong behavioural and positive reinforcing effects through inhibition of the DAT. To this end a great number of diverse, non-selective DAT-inhibiting compounds have been investigated as potential cocaine replacement therapies. It was the initial objective of this research to determine whether the behavioral profile of a novel, selective DAT inhibitor, D-84, fit with that thought for an ideal cocaine replacement therapy. Results indicated that D-84 stimulated locomotor activity, incompletely generalized to the cocaine cue in discrimination tests, attenuated cocaine-self-administration and was self-administered. These observations provide a profile consistent, although perhaps not ideal, with one possible treatment strategy for cocaine dependence. Although it is well established that cocaine predominantly produces its abuse-related effects through inhibition of the DAT, recent evidence suggests that inhibition at the SERT may have modulating effects on the pharmacology of cocaine-like compounds. The second part of this dissertation investigated what effects that increasing SERT inhibition had on the cocaine-like behavioural effects of DAT inhibitors, as a method of determining the fruitfulness of incorporating this feature into future drug candidates to improve them. RTI-55 (DAT Ki 2.7 nM SERT Ki 3 nM) and GBR-12909 (DAT Ki 4.3 nM SERT Ki 73 nM) were selected based on their high and intermediate SERT inhibitory effects, respectively. They were compared in behavioural studies with D-84, which is considered to be a selective DAT inhibitor. The results indicated that although increasing SERT inhibition attenuated locomotor activity effects, it had less effect on cocaine-like discriminative stimulus and reinforcing effects, at least with the doses tested

Recent data mining methods represent modern approaches capable of analyzing large amounts of data and extracting meaningful and potentially useful information from it. In this work, we discuss all the essential steps of the data mining process - including data preparation, storage, cleaning, data analysis as well as visualization of the obtained results. In particular, this work is focused on the data available publicly from the Insolvency Register of the Czech Republic, that comprises all insolvency proceedings commenced after 1. January 2008 in the Czech Republic. With regard to the considered type of data, several data mining methods have been discussed, implemented, tested and evaluated. Among others, the studied techniques include Market Basket Analysis, Bayesian networks and social network analysis. The obtained results reveal several social patterns common in the current Czech society.

The diploma thesis focuses on the youngest age cathegory of respondents in public opinion polls. The main goal is to examine character and quality of information about adolescent's attitudes and opinions obtained in public opinion polls that are held by The Public Opinion Research Centre. To achieve the main goal nonattitude is examined. The thesis will be divided into theoretical and practical part. Theoretical part stands on the basis of public opinion sociology and developmental psychology and the issue of attitude measurement is introduced along with adolescents developmental theory and characteristics. Practical part reflects the information summoned in theoretical part and test them on data collected by The Public Opinion Research Centre which were obtained in continuous research within project Our society. Analysis focuses on examination of nonresponse, don't know answers and neutral attitudes. Results are compared among all age groups.

DTM Improvement in Sandstone Cliffs Relief using precise Co-registration of Airborne Laser Scanning Data Abstract The topic of this thesis is to design innovation of methodology improvement of digital terrain models from airborne laser scanning data in sandstone rocks rugged relief. Theoretical part of this thesis presents the Project GeNeSiS and New Elevation Model Project of the Czech Republic Territory, in which the test datasets were acquired. Simultaneously this section describes in detail methods of co-registrations datasets from strip adjustment to 3D least squares matching method, which have been published until now. In practical part of the work the Pravčická brána model region is introduced firstly. The focus of this section lays on description of new methodology for co-registration and classification of point clouds using spatial query. Co-registration of point clouds, respective detection transform parameters in this thesis is solved by introducing ground control areas. For each such region displacement and rotations in each axis are detected which subsequently determines the global transformation key. Hodnotící tabulka jako nástroj pro měření makroekonomických nerovnováh / Scoreboard Indicators as a Measure of Macroeconomic Imbalances

This thesis examined an ability of the scoreboard indicators created by the European Commission to capture macroeconomic imbalances expressed as the changes of GDP. We conducted an empirical analysis for panel data of 27 EU countries in the 1997-2011 period. We adopted three different dynamic panel data models based on the three estimators: the Arrelano- Bond, the Arrelano-Bover and the corrected LSDV estimator. Our results suggest that despite some bad characteristics of our dataset we can conclude that some of the indicators such as 3- year average of current account balance or percentage change in export market shares seem to be inadequate for measuring the imbalances. Moreover, the indicators were proved not to be able to predict an occurrence of imbalances.

Sytnéza povrchových dat pro digitální modely terénu / Synthesis of digital landscape surface data

A procedural generation of landscapes often meets a need for real spatial data at finer resolution that data available at the moment. We introduce a method that refines the spatial data at the coarse resolution into the finer resolution utilizing other data sources which are already at the better resolution. We construct weighted local linear statistical models from both the coarse and utility data and use the by- models-learned dependencies between the data sources to predict the needed data at better resolution. To achieve higher computational speed and evade utility data imperfection, we utilize truncated singular value decomposition which reduce a dimensionality of the data space we work with. The~method is highly modifiable and its application shows plausible real-like results. Thanks to this, the method can be of practical use for simulation software development. The goal of this thesis is to design and implement a framework to support Legacy system access. Legacy systems are databases that use incompatible and obsolete technologies and can not be easily abandoned. The framework allows the abstraction of application logic from database platform and will enable full or incremental migration to a new, modern platform in the future. The framework also considers the option of encapsulation of an existing legacy application to be included in the new system as a black box. A system based on proposed framework has been succesfully deployed in a company. The system facilitated the migration of the company to a new information system with an entirely different database platform. The practice shows the viability of the framework design. 1

Ochrana soukromí v cloudu / Privacy protection in cloud

In the Master’s thesis were described privacy protection problems while using cloud technologies. Some of the problems can be solved with help of homomorphic encryption, data splitting or searchable encryption. These techniques were described and compared by provided security, privacy protection and efficiency. The data splitting technique was chosen and implemented in the C language. Afterwards a performance of the implemented solution was compared to AES encryption/decryption performance. An application for secured data storing in cloud was designed and implemented. This application is using the implemented data splitting technique and third-party application CloudCross. The designed application provides command line interface (CLI) and graphical user interface (GUI). GUI extends the capabilities of CLI with an ability to register cloud and with an autodetection of registered clouds. The process of uploading/downloading the data to/from cloud storage is transparent and it does not overload the user with technical details of used data splitting technique.
