Analýza nelineárních jevů v ultrazvukové diagnostice / Analysis of Nonlinear Effects in Ultrasound Diagnostics

The model study of nonlinear effects in the ultrasound diagnostics is focused on developing and experimental verification of new computer algorithms for numerical modeling of the ultrasound field including nonlinear effects accompanying the ultrasound propagation in biological tissues. A theoretical analysis of nonlinearities in ultrasound propagation and their contribution to thermal and mechanical effects of diagnostic ultrasound applications was conducted. A numeric model of ultrasound propagating through a tissue model formed of a set of homogenous layers with defined physical parameters was developed in Matlab environment. The computation of ultrasound field spatial distribution is based on the angular spectrum method. For simulating of nonlinearities Burgers nonlinear wave equation is used. The resulting spatial distribution of the ultrasound field is described by the amplitudes of selected number of harmonic components of the acoustic pressure, by the mechanical index, by the harmonic distortion of the signal and by the spatial peak temporal average intensity of all considered components. The numerical model was experimentally verified using an unique ultrasound measurement system – the ultrasound measurement basin. The verified numeric model was used for assessing the veracity of mechanical index and tissue heating prediction during ultrasound applications in cases when the nonlinear effects are neglected in computations. The reliability of the mechanical index indicated by the diagnostic ultrasound instruments is also evaluated using the designed numeric model. The aim was to compare the indicated value usually obtained by derating the measurements in a water medium to the actual mechanical index present in the insonated tissue.

This diploma thesis deals with the constructional design, followed by the manufacturing of the tester for non-invasive diagnostics of cars shock absorbers. It is a resonant adhesive tester with 3 mm lift of measuring platform, in contrast with standardly manufactured testers with 6 mm lift. By reducing lift of measuring platform is eliminated failure of EUSAMA methodology for problematic vehicles. In the thesis are also included verifications of tester function and comparative measurements with standardly manufactured tester.

Title: The exercise diagnostics of women ice-hockey players - Wingate test Objectives: The study is focused on anthropometric characteristics and anaerobic efficiency comparing with using 30s Wingate test parameters. For the test groups of women ice-hockey players and boys ice-hockey players ages 15, 16 and 17 have been selected. The purpose of the work is forming a picture about efficiency level of women players from the national team and comparing with the results of these three selected groups of boys. The study includes a data evaluation of questionnaire about hockey experiences of women players. The exercise anaerobic 30s Wingate test has been implemented on selected group of women ice-hockey players from 14 to 26 years (x = 17.8 ± 3.7). The results of this group have been compared with results of three groups of hockey players ages 15, 16 and 17 (x = 14.98 ± 0.81; x = 16.03 ± 0.41; x = 17 ± 0.35). Working hypothesis supposed no differences between anaerobic training facility indices of women and boys ages 15, 16 and 17. Methods : Basic statistical items (x-arithmetic mean, SD-standard deviation) for all 4 groups (women, boys age 15, 16 and 17) have been evaluated. The ball-pen probe is a unique probe recently developed at the Institite of Plasma Physics in Prague. It has been designed for direct measurement of plasma potential at the CASTOR tokamak. Nowadays, it is used routinely at several tokamaks in Europe, and the first tests in low-temperature plasma have also already been performed. The aims of the thesis are primarily experimental. A ball-pen probe has been constructed from available materials, which is suitable for systematic measurements of radial profiles in the low-temperature plasma of a cylindrical magnetron. By means of comparison to other diagnostics, it was proved that ball- pen probe is able to directly measure plasma potential in a certain range of plasma parameters even though its current-voltage characteristic is not symmetric, which is in contradiction with the simplified theory for ball-pen probe. Cancer is one of the leading cause of death in modern world, so there is an emerging demand for better diagnostic tools and more specific less toxique therapeutics. Nanoparticles offers characteristics that could fullfill such perspectives. They can easily target tumor by ehanced permeation and retention effect (EPR). Nanoparticles can combine more than one imaging properties, so we can say that they are multimodal, some of them could combine diagnostic and therapeutic molecules in one nanoparticle, which is now highly popular topic of nanoparticles for theranostics . The aim of this thesis was to characterize new multimodal glycogen-based nanoparticle. Glycogen is an ideal structure for nanoparticle design. Glycogen is part of natural dendrimers group which are easily to modify. Glycogen's size is suitable for EPR effect. We have evaluated biological characteristics of five different types of modified glycogen. The in vitro experiments were carried on HepG2 cells. We have set time curve of cellular uptake of this glycogen probes, evaluated cytoplasmatic localization and for the first time we have carried MTT assay. The data will be collected in the form of structured questionnaires filled by a selected group of respondents. The thesis is, in the outcome, an educational material that can be used for comprehensive understanding of this issue in the adaptation process. Title: Influence of different way of strength training on body composition, postural stability and muscle and explosive strength in Men's Physique and Street Workout Athletes Objectives: The aim of the thesis was to find out the level and the difference between the parameters of body constitution, postural stability and muscular and explosive force of upper and lower limbs with groups of Men's Physique and Street Workout athletes. Methods: The research sample was represented by two groups (Men's Physique competitors and Street Workout athletes). We assessed chosen parameters of body constitution by Tanita MC-980MA, postural stability by RS Footscan, explosive force (Kistler) and muscular force (Cybex Humac Norm and Takei A5401). Assessed parameters of body constitution were percentage of body fat and fatless matter. In the tests of postural stability, we assessed total travel way of the center of pressure (TTW) in chosen stands (open/closed eyes and one- leg left/right). When testing explosive force, overall produced maximal force and height of the leap were assessed. Liver flukes of the family Fasciolidae are parasites of mammals including human. Fascioloides magna and Fasciola hepatica are considered as a veterinary and medically important species occurring also in the Czech Republic. Fascioloides magna and F. hepatica infect wide spectrum of wild and domestic ruminants and in case of F. hepatica human can be also infected. Both flukes are responsible for damage of liver tissue and/or bile-ducts of their definitive hosts causing weight lose, anemia, reduced productivity and in specific cases the death of the host. Effective diagnosis plays the key role in control of F. hepatica and F. magna infections. Current diagnostics is predominantly based on serodiagnostic methods using specific antigens e.g. from excretory-secretory products (ESPs). Due to heterogenity of ESPs, such diagnostic markers can lack the specificity and also the reproducibility of the method is poor. Particular proteins of ESPs are often used in diagnostics of fasciolid flukes. Such approach requires biological material and laboratory procedures associated with identification, purification and antigenicity testing of selected proteins.
